Welcome to Bid Nerds, the show where car enthusiasts John Polnik and Michael Deeb predict and reconcile the results of the latest car auctions. In this episode, we'll be discussing the BaT auction results of a 2000 Honda S2000. This iconic roadster is powered by a 2.0-liter VTEC inline-four engine that produces 240 horsepower and 153 lb-ft of torque, paired with a 6-speed manual transmission. The car is capable of reaching 60 mph in just 5.4 seconds and has a top speed of 150 mph. Abstract: Myocardial damage caused for example by cardiac ischemia leads to ventricular volume overload resulting in increased stretch of the remaining myocardium. In adult mammals, these changes trigger an adaptive cardiomyocyte hypertrophic response which, if the damage is extensive, will ultimately lead to pathological hypertrophy and heart failure. Conversely, in response to extensive myocardial damage, cardiomyocytes in the adult zebrafish heart and neonatal mice proliferate and completely regenerate the damaged myocardium. We therefore hypothesized that in adult zebrafish, changes in mechanical loading due to myocardial damage may act as a trigger to induce cardiac regeneration. Based, on this notion we sought to identify mechanosensors which could be involved in detecting changes in mechanical loading and triggering regeneration. Here we show using a combination of knockout animals, RNAseq and in vitro assays that the mechanosensitive ion channel Trpc6a is required by cardiomyocytes for successful cardiac regeneration in adult zebrafish. Furthermore, using a cyclic cell stretch assay, we have determined that Trpc6a induces the expression of components of the AP1 transcription complex in response to mechanical stretch. Our data highlights how changes in mechanical forces due to myocardial damage can be detected by mechanosensors which in turn can trigger cardiac regeneration. Copy rights belong to original authors. Abstract: The dynein motor complex mediates polarised trafficking of a wide variety of organelles, intracellular vesicles and macromolecules. These functions are dependent on the dynactin complex, which helps recruit cargoes to dynein's tail region and activates motor movement. How dynein and dynactin orchestrate trafficking of diverse cargoes is unclear. Here, we identify HEATR5B, an interactor of the AP1 clathrin adaptor complex, as a novel player in dynein-dynactin function. HEATR5B is one of several proteins recovered in a biochemical screen for proteins whose association with the human dynein tail complex is augmented by dynactin. We show that HEATR5B binds directly to the dynein tail and dynactin and stimulates motility of AP1-associated endosomal membranes in human cells. We also demonstrate that the HEATR5B homologue in Drosophila is an essential gene that promotes dynein-based transport of AP1-bound membranes to the Golgi apparatus. As HEATR5B lacks the coiled-coil architecture typical of dynein adaptors, our data point to a non-canonical process orchestrating motor function on a specific cargo. We additionally show that HEATR5B promotes association of AP1 with endosomal membranes in a dynein-independent manner. Thus, HEATR5B co ordinates multiple events in AP1-based trafficking. Abstract: FLT1/VEGFR1 negatively regulates VEGF-A signaling and is required for proper vessel morphogenesis during vascular development and vessel homeostasis. Although a soluble isoform, sFLT1, is often mis-regulated in disease and aging, how sFLT1 is trafficked and secreted from endothelial cells is not well understood. Here we define requirements for constitutive sFLT1 trafficking and secretion in endothelial cells from the Golgi to the plasma membrane, and we show that sFLT1 secretion requires clathrin at or near the Golgi. Perturbations that affect sFLT1 trafficking blunted endothelial cell secretion and promoted intracellular mis-localization in cells and zebrafish embryos. siRNA-mediated depletion of specific trafficking components revealed requirements for RAB27A, VAMP3, and STX3 for post-Golgi vesicle trafficking and sFLT1 secretion, while STX6, ARF1, and AP1 were required at the Golgi. Depletion of STX6 altered vessel sprouting in a 3D angiogenesis model, indicating that endothelial cell sFLT1 secretion is important for proper vessel sprouting. Thus, specific trafficking components provide a secretory path from the Golgi to the plasma membrane for sFLT1 in endothelial cells that utilizes a rare clathrin-dependent intermediate, suggesting novel therapeutic targets. “This is the company that really changed the access the rest of the United States had to get fresh, leafy greens.” Lori Taylor (5:28-5:35) Even though Tanimura & Antle wasn't “officially” launched until 1982, the families that make up this incredible fresh produce team have a legacy that stems from 100 years of growing experience. The Tanimura family and Antle family lived 40 miles apart from each other, with the Tanimura family emigrating from Japan and the Antle family migrating to California from Oklahoma during the Dust Bowl in the 1920's. During World War Two, the Antle family agreed to watch over the Tanimura's land while some of the Tanimura family was placed in internment camps and two of the brothers fought in the war. When the war was over, the two families had an unmatched partnership and bond and decided to join forces; the Tanimuras growing the lettuce and the Antles marketing and selling it under their brand. What started as a relationship between two families with a love for produce has now grown into a global company with 30,000 acres on the west coast, 7,000 employees annually, and sales across the Nation and internationally. Brian Antle, who got his start on the farm at the ripe age of 14 when he would get paid to move sprinkler pipes (a vital part of irrigation), jokes that “the plan was to get to 100,000 cases of produce a week.” The company now sells almost 140,000 boxes a day and about 40 million cases a year! Tanimura & Antle's core products are iceberg lettuce, romaine, red and green leaf, cauliflower, broccoli and celery. They also have organic produce, greenhouse grown produce, and an Artisan line of products that are uniquely branded for their different taste profiles, colors, qualities and crunchability. The Artisan family of specialty produce includes their Artisan Lettuce, Artisan Romaine, Artisan Sweet Gem, Artisan Baby Iceberg, Artisan Sweet Red Onions and Artisan Sweet Broccoli. “They were the first company that put iceberg lettuce on a rail car on ice and shipped it from California to New York City and it arrived fresh. From there, as they say, the rest is history.” Lori Taylor (5:45-5:56) The cool thing about Tanimura & Antle is the many ‘firsts' the company has ignited for the produce industry. Their family was the first to pack and wrap a head of lettuce in cellophane. They were also the first to use cardboard boxes and cool produce in vacuum cooling tubes prior to shipping, allowing the entire produce industry to move away from needing to ice everything and use rail cars. Speaking of innovation, Tanimura & Antle even breeds their own seed genetics! The company can tailor their lettuce, to have a certain color, size or taste. They can also breed plants to become more resistant to certain disease or mildew pressures. Innovations in seed genetics ultimately allows Tanimura & Antle to harvest more product for consumption and decrease food waste at the field level. This is all done through natural plant breeding, none of these seeds are genetically modified organisms (GMOs). Tanimura & Antle truly strives to find sustainability in every corner of their operations! Another innovative practice of the company is the use of automated technology. Tanimura & Antle has a group of diversified businesses, one of those being Stout, a company focused on “smart farm equipment”. With the cost and shortages of skilled agricultural labor, farmers are being challenged with doing more with less every day. Tanimura & Antle is investing in machines that can help supplement these labor shortages, such as machines that eliminate weeds in the field using artificial intelligence and computer vision systems. Machines like this will allow farmers to [AP1] utilize the employees in areas of agricultural production that cannot be automated, such as harvesting or operating these new machines. While this type of innovation can sometimes be viewed as eliminating jobs, they're not. Innovations like this allow farmers to supplement the shortages within their work force in order to continue to feed the world. “The ground is the most valuable asset we have next to our employees. Without either one, there is no sustainability.” Brian Antle (29:16-29:23) One key to Tanimura & Antle's success is how much they value the ground they grow on. They're constantly rotating their crops and focus on keeping the ground healthy. Without healthy soil, they know they cannot grow a healthy crop. In the third episode of Allocator's Corner, I had the opportunity to sit down with Helen Idenstedt who is portfolio manager idiosyncratic alternative investments at the first Swedish national pension fund - AP1. The role of the portfolio that Helen runs is to serve as a diversified source of alpha within the broader AP1 mandate – a mandate that currently has around 350 billion SEK in assets under management. During the session, we discussed how to manage so-called idiosyncratic risk using alternative investment strategies, the process behind her manager selection, what the portfolio looks like today and how ESG has received increased attention in the alternatives space as of late. We also had the chance to discuss a new mandate linked to private debt strategies that Helen has just started allocating assets to.

Once the car is in the field, it gets VERY expensive to fix, replace, or upgrade hardware. Software, thanks to OTA updates, can be fixed, and features can be added, but only to the limits of the hardware. Rich Fowler Hardware! Specifically, seats! Software can't fix a terrible seat. My analogy is that if you play cards, the hardware is the deck, and the software is the rule set. If the deck is missing half the cards, you can't play a lot of games you want to play with it, and you have to invent some other game with a different set of rules. That new game may or may not be more fun, but it's not the game you were looking for when you bought the deck in the first place. John Fruhwirth Since you are forcing us to choose between hardware and software as being more important in our EVs, I'm coming down on the side of hardware. Why? because hardware is the necessary basis/foundation for software. Without hardware to run on, software is useless. However, the reverse is not true. Hardware is definitely useful without software as we can compensate for the lack of software with our wet/brain ware. So, while software is nice, hardware is a must. However, the combination of hardware and software is where it's at and gives us the biggest bang for our transportation bucks. Ron KeRENski I think the right answer, for where we are in the EV market evolution, is actually hardware. Assuming a starting point with comparable software capabilities, the right hardware in a car rolling off of the line today means that software improvements over the next 5-10 years can reach backwards to owners. Nothing builds brand loyalty like not being left behind and improving the resale value of my trade-in due to a high demand, used market. Will Adams since you aren’t allowing the correct answer it has to be Software that is more important. Great software can make bad hardware work, but really bad software can make even the best hardware unusable Howard Smith Hardware is the most important part to get right on an EV. Hardware reliability provides value because the car will last longer. EV owners have to pay more upfront for their cars; they need to be durable over the long run. Software is easily updated; hardware is not. David Nye This is a very easy one to answer, no fence sitting necessary. Hardware is king, it must be right from day one. Software finessing and “polishing” can follow later with OTA updates. Tesla is of course the classic example- early release software in Teslas has at times had it’s issues, and of course critics, but it just keeps getting better and better. The Tesla you drive today will only be better in a year’s time. Imagine if you had to keep visiting the service centre for hardware fixes! Rob Crawford If your EV has FSD, then the hardware is more important. Simpley because it's the hardware that allows the car to drive in FSD mode. Without the hardware FSD isn't possible. And the hardware must be strong, rugged dependable to function perfectly, immatating what a driver can do only better. Now... I can answer the same question about the importance of the software. Software also plays an important part when it comes to FSD, without over the air updates, then the FSD will lag, and won't run correctly, as designed. So my answer is both Hardware and Software are important for the safest in functionality of an EV, especially pertaining to FSD. Software is important to the functionality of EVs in general. An ICE car couldn't even function as a self driving car. Tony Goodwin After a lot of thought, on balance hardware - in particular the fitment of sensors and processing power allowing for additional functionality - is much more important than software. Hardware changes post-delivery will likely be both difficult and expensive to fit and always require a dealer visit, whereas software can add capabilities to existing hardware as techniques are refined and "AI" knowledge iterates. It is also possible to do this OTA as well as being carried out at dealerships. Victor Wood Hardware is the most important to get correct. Software can always be updated relatively easily whereas hardware is not something that can be easily and cheaply swapped out. The cost of looking out say an autopilot hardware swap out must be sky high compared to the cost of pushing software over the air. For cars like mine where there is no OTA updates it’s still cheap to do a software update as in effect it’s “only” the time to put the software onto an SD card and to put that in the car that causes costs. The last 2 updates for my Ioniq I did myself after getting the software on an SD card from the dealer. It took 15 minutes. Andrew Beshera Hardware is more important over software because software is cheaper & easier to update. Andrew Jacobs Software is easily more important because you can have the best hardware in the world and bad software will render it useless. If you have great software and poor hardware, the software is going to improve the hardware limitations. Just look at the YouTube videos where people cover cameras to see how well autopilot works. Autopilot mainly uses the front forward stereo cameras. Even with the others blocked it can still work. Software is king. Bill Pollock HARDWARE is the most important thing to get right off the start. Hardware is expensive to upgrade, and often can not be done. I do however think that hardware should be designed to be as swapable as possible. For example, as battery tech improves, we should be able to upgrade with minimal intrusion. BTW-The Tesla Model 3 is not an easy battery to swap. The tech has to basically dismantle the interior of the car to get at fasteners. The S and X batteries were designed for easy replacement. I hope they Y has corrected this Model 3 shortfall. Michael McClure This is a no brainer. Hardware is static; software is not.I drive a 2015 Tesla Model S with AP1 that I picked up used last year. That’s old hardware, but I keep getting OTA updates all the time that make it feel ‘new’. Aaron Patel Hardware. You can't change the number of cells in your battery once it's sitting in your driveway. Autopilot software can be okay today but then gets incrementally better month after month until it's great. I just reached 1 year in my Model 3 and the day after I picked it up I got a software update that added automatic wipers. A new study reveals that the clathrin adaptor AP-1 and the kinesin motor KIF13A combine to sort melanosomal cargo and position endosomes near to developing melanosomes where they deliver the cargo via direct tubular contacts. Background: Total tau (T-tau) and beta-amyloid((1-42)) (A beta(1-42)) levels in cerebrospinal fluid (CSF) can differentiate Alzheimer's disease (AD) from normal aging or depressive pseudo-dementia. Differential diagnosis from dementia with Lewy bodies (DLB) in clinical settings is difficult. Methods: The analytical performance of the INNOTEST (R) PHOSPHO-TAU((181P)) assay was validated in terms of selectivity, sensitivity, specificity, precision, robustness, and stability. Clinical utility of the assay alone, or combined with T-tau and AP1-421 for discrimination of AD (n=94) from patients suffering from DLB (n=60) or from age-matched control subjects (CS) (n=60) was assessed in a multicenter study. Results: CSF concentrations of tau phosphorylated at threonine 181 (P-tau(181P)) in AD was significantly higher than in DLB and CS. Discriminant analysis, a classification tree, and logistic regression showed that P-tau(181P) was the most statistically significant single variable of the three biomarkers for discrimination between AD and DLB. Conclusions: P-tau(181P) quantification is a robust and reliable assay that may be useful in discriminating AD from DLB.

Pathogenetische Bedeutung der induzierbaren Stickstoffmonoxid-Synthase für den akuten hyperoxischen Lungenschaden Die längere Applikation von Sauerstoff in höheren Konzentrationen führt zu einer oxidativen Schädigung der Lunge. In den letzten Jahren gab es auch Hinweise darauf, dass Stickstoffmonoxid (NO), produziert durch die induzierbare NO-Synthase (iNOS), an der Pathogenese des hyperoxischen Lungenschadens beteiligt sein könnte. Ziel der Studie war es, die Beteiligung von NO an der Entstehung des akuten Lungenschadens unter Hyperoxie in vivo zu untersuchen. Dazu wurden unter standardisierten, experimentellen Bedingungen iNOS-defiziente und Wildtyp-Mäuse erhöhten Sauerstoffkonzentrationen (60 und 100 %)über 24 bzw. 72 h ausgesetzt. Die Ergebnisse zeigen, dass nach 72 h unter 100 % Sauer-stoff sowohl in der Wildtyp- als auch in der iNOS-defizienten Gruppe ein deutlich vermehrter Zellschaden im Sinne einer Abnahme der Zellvitalität und einer Zunahme der LDH-Aktivität auftritt. Als Zeichen einer verstärkten inflammatorischen Reaktion kam es zu einer Erhöhung der Gesamtzellzahl und -proteinkonzentration in der Bronchiallavageflüssigkeit sowie zu einer vermehrten TNF-alpha-Expression, welche durch die Transkriptionsfaktoren NF-Kappa B und Aktivatorprotein 1 (AP1) vermittelt wurde. Diese Auswirkungen waren auch in den histologischen Schnitten zu beobachten. Als Zeichen vermehrten oxidativen Stresses war eine erhöhte Lipidperoxidation zu verzeichnen. Die genannten Beobachtungen waren bei den Wildtyp-Mäusen wesentlich ausgeprägter als bei den iNOS-defizienten Tieren, was auf die erhöhte iNOS-Expression zurückgeführt werden könnte. Aus der Gesamtheit der Daten lässt sich schliessen, dass unter akuter Hyperoxie die proinflam-matorische Komponente der iNOS zu überwiegen scheint, dadurch wird das Ausmaß der inflammatorischen Reaktion und die resultierende Lungenschädigung potenziert.

EBV is a γ-herpes virus which is able to infect human resting B-cells and to transform them into permanently growing lymphoblastoid cell lines (LCLs). EBNA2 (Epstein-Barr virus nuclear antigen 2) is one of the first viral proteins expressed after in vitro infection and interacts with different cellular proteins like RBP-Jκ and PU.1. The EBNA2 protein acts as a transcriptional activator of the viral Latent Membrane Proteins 1 and 2 (LMP1 and LMP2) and the viral nuclear genes EBNA1, EBNA3A, -3B, -3C, EBNA-LP. Additionally EBNA2 is also able to transactivate cellular genes like CD21, CD23 or c-myc. To study the different EBNA2 target genes and the function of EBNA2 a LCL was established (ER/EB2-5 cells, Kempkes et al., 1995) harboring an estrogen-inducible EBNA2. In the presence of estrogen the ER/EBNA2 fusion protein (estrogen receptor binding domain) is located in the nucleus were EBNA2 can transactivate its target genes, whereas in the absence of estrogen the ER/EBNA2 fusion protein is kept in the cytoplasm and therefore inactive. The cells proliferate in the presence of estrogen and they arrest in the absence resulting in a phenotype similar to resting B-lymphocytes. By using the ER/EB2-5 cell line I could clearly show that the cell surface molecule CD83, belonging to the immunoglobuline superfamily (Zhou et al., 1992), is upregulated after the activation of EBNA2. By using a derivative ER/EB2-5 cell line that constitutively expressed LMP1 I could show that CD83 is still expressed even in the absence of functional EBNA2 suggesting that LMP1, the viral target gene of EBNA2, is responsible for the induction of CD83. Therefore I analysed the activation of the CD83 promoter by LMP1. LMP1 is a transmembrane protein with a short intracellular N-terminus, 6 hydrophobic transmembrane domains and a long intracellular C-terminus, containing C-terminal activator regions CTAR1, 2 and 3. The different CTAR regions are responsible for activating genes via NF-κB, ATF, AP1 and STAT signaling pathways. For the activation of its target genes LMP1 uses the same signaling molecules (TRAF, TRADD) as family members of the TNF-R family (CD40, TNF-R1, TNF-R2). The CD83 promoter was activated by LMP1 as shown by promoter luciferase reporter assays in 293-T cells. The induction was not observed in the absence of a NF-κB binding site in a CD83 promoter mutant. Furthermore LMP1 mutants which are mutated in the binding regions for TRAF2 (CTAR1) or TRADD (CTAR2) are not able to transactivate the CD83 promoter. By co-transfection of LMP1 and dominant/negative IκB the CD83 promoter could not be activated because of inactivation of NF-κB. These experiments clearly demonstrate that the CD83 promoter is transactivated by LMP1 via NF-κB. Additionally to the regulation of CD83 I was also interested in the functional role of CD83. Until now only little is known about the function of CD83. CD83 seems to have a specific role in the decision to single positive CD4+ T-cells in the thymus (Fujimoto et al., 2002). I have tested a possible co-stimulatory function of CD83 to CD4+ T-cells by retroviral expression of CD83 in non-professional antigen presenting cells (RCC). Indeed CD83 expression increased the CD4+ response in comparison to CD80 or GFP retroviral infected RCC cells. In mixed lymphocyte reactions this co-stimulatory effect could not be clearly demonstrated although a soluble CD83-Ig showed a small inhibitory influence. The identification of a CD83 ligand molecule could give new insights into the function of CD83. Therefore a CD83-Ig fusion protein as well as a CD83-tetramer construct were generated and used to screen for a potential ligand of CD83. First results showed that the CD83-Ig fusion protein and the CD83-tetramer construct bound to CD4+ and to CD8+ T-cells of isolated PBMCs as well as to activated T-cells in a culture of mixed T-cell populations.
