On this episode of This Thing Called Life, host Andi Johnson is talking with Sandra Wright. She has a special testimony to share. The goal of this podcast is to help listeners understand what organ donation is and isn't and how it truly impacts others. February is Black History Month and American Heart Month. Sandra is a transplant survivor and the founder of The Greater Cincinnati African American Heart Association. Tune in now for her special story! Episode Highlights:  Sandra is a heart transplant recipient. In 2014, she contracted a virus that was never named but lingered and left her with a side effect of shortness of breath. Later that year, she was on vacation when it all kind of climaxed. She shares her story of going to the hospital and the dire state she was in. She was told she had Heart Failure but she went into immediate denial. She cautions the listeners against denial because then you don't do what you need to do. There will be a book coming out to tell Sandra's full story in the future to help many. Christ Hospital in Cincinnati set her on the path of her life being saved.  Prior to Sandra's visit at Christ Hospital, every two months she would go for maintenance care to control the edema but other than that she wouldn't take the medicine or change her lifestyle until 2017 when her heart would no longer serve her. Sandra shares how her faith got her through the scary time of being in need of a heart. The Greater Cincinnati African American Heart Association was created by Sandra to help provide others support, to create a community where they feel respected, heard, and understood. She shares about the services and her vision for down the road. For those who hear they have CFH, Congestive Heart Failure, that is all they are going to think about and can come unexpectedly and at vulnerable times.  Andi shares how the healthcare system is not accessible for all and how detrimental it is for many, especially people of color.  Sandra shares a sad story of talking with her granddaughter who said she ‘hated being black' and how that ideology fuels her heart and mission to create an environment of support for the African American Community. Andi asks Sandra to share any encouraging thing she is experiencing or new strength she sees in the community. Andi reminds listeners that today, 106,494 people are waiting for life saving organ transplants and more than 3,400 need heart transplants. Can you think about your decision to be a donor and save a life?   3 Key Points: Sandra shares a shocking story of her journey with heart failure and the danger of being in denial of the diagnosis.  The African American community is at the highest risk and has the greatest fatality which led Sandra to create The Greater Cincinnati African American Heart Association as a community of support. Sandra shares that living 65 years in this world, we are beginning to understand the necessity of working together and supporting each other and that is encouraging and exciting. The goal of this podcast is to help listeners understand what organ donation is and isn't and how it truly impacts others. February is Black History Month and American Heart Month. Sandra is a transplant survivor and the founder of The Greater Cincinnati African American Heart Association. Tune in now for her special story!   Episode Highlights:  Sandra is a heart transplant recipient. In 2014, she contracted a virus that was never named but lingered and left her with a side effect of shortness of breath. Later that year, she was on vacation when it all kind of climaxed. She shares her story of going to the hospital and the dire state she was in. She was told she had Heart Failure but she went into immediate denial. She cautions the listeners against denial because then you don't do what you need to do. There will be a book coming out to tell Sandra's full story in the future to help many. Christ Hospital in Cincinnati set her on the path of her life being saved.  Prior to Sandra's visit at Christ Hospital, every two months she would go for maintenance care to control the edema but other than that she wouldn't take the medicine or change her lifestyle until 2017 when her heart would no longer serve her. Sandra shares how her faith got her through the scary time of being in need of a heart. The Greater Cincinnati African American Heart Association was created by Sandra to help provide others support, to create a community where they feel respected, heard, and understood. She shares about the services and her vision for down the road. For those who hear they have CFH, Congestive Heart Failure, that is all they are going to think about and can come unexpectedly and at vulnerable times.  Andi shares how the healthcare system is not accessible for all and how detrimental it is for many, especially people of color.  Sandra shares a sad story of talking with her granddaughter who said she ‘hated being black' and how that ideology fuels her heart and mission to create an environment of support for the African American Community. Andi asks Sandra to share any encouraging thing she is experiencing or new strength she sees in the community. Andi reminds listeners that today, 106,494 people are waiting for life saving organ transplants and more than 3,400 need heart transplants. Aging-US published a Special Collection on Eye Disease which included "Activation of C-reactive protein proinflammatory phenotype in the blood retinal barrier in vitro: implications for age-related macular degeneration" which reported that the retinal pigment epithelium (RPE) is considered one of the main targets of age-related macular degeneration (AMD), the leading cause of irreversible vision loss among the ageing population worldwide. Increased levels of circulating pentameric C-reactive protein (pCRP) are associated with higher risk of AMD. Monomeric form of pCRP has been detected in drusen, the hallmark deposits associated with AMD, and we have found that mCRP induces oBRB disruption Dr. Blanca Molins from The IDIBAPS, Hospital Clínic de Barcelona said, "Age-related macular degeneration (AMD) is the primary cause of irreversible vision loss among the ageing population worldwide." AMD presents RPE cell abnormalities, disruption of the outer blood-retinal-barrier (oBRB), and degeneration of photoreceptors. Altered immune responses are thought to contribute to the dry AMD phenotype. Loss of parainflammation control contributes to AMD by invoking a chronic, heightened immune response that causes tissue destruction. mCRP has been identified in ocular drusen and other subepithelial deposits, as well as in the choroid, and contributes to oBRB disruption in vitro. The "non-risk" Factor H (FH) variant can effectively bind to mCRP to dampen its proinflammatory activity. MCRP levels are elevated in individuals with the high-risk CFH genotype [29, 30] - this is because there is no CRP transcription in retinal tissue. Changing the Faces of Homelessness was founded by Katrina Williams, a Mental Health Case Manager who worked with the Regional Behavioral Health Authority under Value Options and Magellan Health Services for over 15 years in Phoenix, AZ. Katrina Williams recently relocated to Los Angeles CA and began working for Volunteers of America as a Case Manager for the Hope for Homes Shelter Program in Pomona CA. During her employment with VOA one of her clients ended their life via sucuide with her client's sudden death, she realized the severity of the Los Angeles homeless crisis and developed the CFH supportive housing Program where she provides realistic resources (employment, counseling etc.) to meet her residents' realistic goals with housing. Aging-US published a Special Collection on Eye Disease which included "The expression of C1 inhibitor (C1INH) in macrophages is upregulated by retinal pigment epithelial cells – implication in subretinal immune privilege in the aging eye" which reported that this study aimed to understand how complement expression in macrophages is regulated by retinal pigment epithelium (RPE) RPE can modulate macrophage complement expression at the retina-choroidal interface even under aging or oxidative conditions. During inflammation, they may promote the alternative pathway of complement activation through down-regulating CFH and CD59a and upregulating C3, CFB and C1INH. When BMDMs were treated with apoptotic RPE, the expression of C1qb, CFH, and CD 59a was reduced but increased in BMDM. TNF-α pre-treated RPE enhanced C1inH and CFB expression. Dr. Heping Xu from The Queen's University Belfast as well as The Central South University said, "The neuronal retina is segregated from the systemic immune system by the blood retina barriers (BRB) and is considered as an immune privileged tissue." The immune suppressive microenvironment of the eye is critical for retinal immune privilege. Despite the lack of systemic immune surveillance, the retina is well-protected by its own innate immune defence system, including innate immune cells and the complement system. During aging, the expression of complement proteins or fragments is increased in the retina, particularly at the retina-choroid interface. Subretinal macrophages in the healthy adult eyes (6 -12 months old) often have a small soma and long- fine-dendrites (Fig. 1A), whereas the cells in the aging eye (20 – 27 months) have a large cell body that often contains pigmented debris. This suggests that they are active phagocytizing debris released by stressed RPE cells. The Xu Research Team concluded in their Aging-US Research Output, "we show that RPE cells can modulate macrophage complement expression. Under normal aging conditions, RPE cells may convert macrophages into a phenotype that can suppress complement activation with enhanced phagocytosis. This immune regulatory function of RPE cells on macrophages may be lost under inflammatory conditions. Instead, inflammatory or apoptotic RPE cells promote macrophages to produce complement components necessary for the AP activation. RPE cells together with subretinal macrophages critically control complement activation at the retina-choroid interface in the ageing eye." At a time, when metal was on the decline, these guys brought in your face riffs packed with heavy Texas groove! The Dallas boys, Diamond Darrell, or rather Dimebag Darrell who he affectionately became to be known, was a master shredder and his brother Vinnie Paul on pounding drums; along with Rex on a thumping bass and Phil Anselmo on some screaming (ass) vocals made up this band. They had many albums. some in the glam rock vein, full of KISS and Van Halen love. But it wasn't until the release of Cowboys from Hell in 1990, that they broke down the metal walls. They were able to cement their legacy with a few more great albums, before they went on hiatus and broke up in 2003, way too soon. Phil and his drugs, were pinned as the bad guy in the breakup. But he went on to front many other bands. On a side note: Phil along with his biographer Cory Mitchell, hosted the Housecore Horror Film Festival in Austin, Texas in 2014 and 2015, in which I was lucky to have been a part of. It was a fantastic blend of horror movies and metal! However, the event died along with Cory Mitchell when he had a heart attack loading out and died in the parking lot the morning after Danzig closed down the festival. But today we are honoring Dimebag as today is the 15 anniversary of his passing. Tragically, in 2004 in front of crowd in Columbus Ohio, Dimebag was shot point blank just after taking the stage in with his new band, Damageplan, that he had formed with his brother on drums. The gunman, the coward, was said to be distraught over the breakup of Pantera when got on stage and killed 3 people, before he too was gunned down by a fast acting police officer. His brother Vinnie Paul, carried on with the band, but he too passed away too soon, just last year. To start us off, is Shedding Skin off of Far Beyond Driven, which celebrated it's 25th Anniversary this year: That was Cemetery Gates off of CfH and before that was Hollow, the song that closed 1992's Vulgar Display of Power album. I was at the GNR reunion tour in Dallas a few years ago. The next day, friends and I make the pilgrimage to Dimebag's grave. Upon arriving at the cemetery gates and not knowing where to go next, we asked a landscaper, he immediately knew who we were looking for and replied "Dimebag aqui, Dimebag aqui", pointing to his gravestone. Gathered around the grave were many concert goers from the night before and many joints layed as an offering on his headstone. 6 feet deep lies the man, in a gifted KISS Kasket and with Eddie Van Halen's personal yellow-and-black-striped Charvel electric guitar from Van Halen II, Bumblebee. fitting, since Dimebag's last words on Earth were, "Van Halen!" moments before he was gun down. Somebody near us said that they ran into Vinnie Paul at the concert last night and shows us proof, a photo together on their cell phone. Pantera and the legend of these 2 men captivated us all. Age-related macular degeneration (AMD) is a prevalent cause of blindness in Western societies. Variants in the genes encoding complement factor H (CFH), complement component 3 (C3) and age-related maculopathy susceptibility 2 (ARMS2) have repeatedly been shown to confer significant risks for AMD; however, their role in disease progression and thus their potential relevance for interventional therapeutic approaches remains unknown. Here, we analyzed association between variants in CFH, C3 and ARMS2 and disease progression of geographic atrophy (GA) due to AMD. A quantitative phenotype of disease progression was computed based on longitudinal observations by fundus autofluorescence imaging. In a subset of 99 cases with pure bilateral GA, variants in CFH (Y402H), C3 (R102G), and ARMS2 (A69S) are associated with disease (P = 1.6x10(-9), 3.2x10(-3), and P = 2.6x10(-12), respectively) when compared to 612 unrelated healthy control individuals. In cases, median progression rate of GA over a mean follow-up period of 3.0 years was 1.61 mm(2)/year with high concordance between fellow eyes. No association between the progression rate and any of the genetic risk variants at the three loci was observed (P>0.13). This study confirms that variants at CFH, C3, and ARMS2 confer significant risks for GA due to AMD. In contrast, our data indicate no association of these variants with disease progression which may have important implications for future treatment strategies. Other, as yet unknown susceptibilities may influence disease progression.
