The innate immune system, which is the immune system humans are born with, features signaling cells known as neutrophils, monocytes, and macrophages. These cells are heavily influenced by circadian rhythms. Maria hypothesizes that cancer cells have their own internal clocks that are able to tune into and mimic the circadian rhythms of the tissues they infect. Is this how they evade the human immune system? By synchronizing the internal clocks of both cancer cells and immune system cells, can we teach our immune systems to eliminate cancer? Parasitic diseases in European aquaculture continue to pose economic and ecological threats to farmed and wild fish populations. The primary infection with ectoparasites can open the door to secondary or super- infections caused by bacterial and viral pathogens. Specifically, the infection with Argulus foliaceus (fish louse) in freshwater cyprinid fish such as common carp (Cyprinuscarpio) can increase production losses in affected aquaculture operations, or stocked water bodies. However, current veterinary drug use regulations in Germany limit the range of medications approved as treatment against ectoparasites in freshwater fish. A comparable parasite Lepeoptheirussalmonis (salmon louse) treatment with Diflubenzuron, Ivermectin and Doramectin (pesticides), has been approved in Salmon aquaculture in various countries. In order to control parasite populations, fish farmers frequently reach for non-approved but available treatments without veterinary supervision, possibly leading to consequences such as environmental damage, emergence of drug resistance or unwanted side effects on cultured and wild fish. The focus of this study is to investigate possible side effects of Diflubenzuron, Ivermectin, and Doramectin treatments on the innate immune system of a cyprinide fish. The overall research objective is to determine which antiparasitic drug and in what concentration and dose would present an effective treatment of the A. foliaceus without causing measurable side effects on fish neutrophil function. The first specific aim was to determine in vitro effects of commercial formulations of diflubenzuron, ivermectin and doramectin on fathead minnow (Pimephales promelas) neutrophil function. Treatment effects in vitro on oxidative burst, degranulation, and neutrophil extracellular trap (NETs) release were studied. Application of ivermectin and doramectin (in estimated plasma concentrations of 308 and 125 ng/ml respectively) caused no significant stimulation of oxidative burst, degranulation of primary granules and NETs release. However, application of diflubenzuron (in estimated plasma concentrations of 200 ng/ml) caused a significant stimulation of oxidative burst. Diflubenzuron treated neutrophils showed up to three time’s higher activity than the non-treated control. The second specific aim of the study was establishing a cell culture protocol of fathead minnow liver, spleen and kidney cells. In order to examine whether an incubation of the cells in culture of 48 hours with Diflubenzuron would alter the RNA expression in these organs, no significant change in expression of the tested genes (glutathione reductase, glutathione synthase, catalase, glutathione-s-transferase, catalase and CYP A1) compared to a control could be proven. The observed effect indicates that the tested antiparasitic compounds have the potential to interfere with disease resistance in fish populations by modulating immune responses during treatment. Therefore, further study is required to find optimal therapeutic dose for effective and safe treatment against ectoparasites in cyprinide fish. The following conclusions can be drawn from the presented findings: In vitro studies on the innate immune system as the here presented neutrophil studies can give reliable first answers to questions concerning in vivo diseases and treatments. With studies on the gene expression, far-reaching and profound negative effects can be ruled out, when such investigations remain without significant results, as shown here.

The accumulation of plastic debris in the environment has been recognized as a matter of international concern. A minimum of 5.25 trillion plastic particles were estimated to float on the surface of the ocean which is due to a low amount of recycling (37 %) and mismanaged waste that enters into the environment. Large plastic debris can lead to suffocation and death after entanglement or ingestion. Small plastic items like microplastic (< 5 mm) and nanoplastics (< 100 nm) can harm organisms on the cellular level since their small size enables them to pass cell boundaries and enter surrounding tissues. The amount of microplastic, and potentially nanoplastic, is increasing due to their release to the environment and the breakdown of larger plastic items in aquatic ecosystems. Microplastic has been observed to enter into the circulatory system of mussels inducing inflammatory reactions and granulocytoma formations. A facilitated uptake of nanoparticles into the cells and an interaction with the immune system is enhanced due to their small size and their specific surface characteristics. Therefore, the health hazards of nanoplastics on aquatic organisms that are potentially chronically exposed to these compounds were assessed. The hypothesis tested here is that that polycarbonate (158.7 nm) and polystyrene (41.0 nm) nanoparticles impact the immune system of the freshwater fish fathead minnow (Pimephales promelas, Rafinesque 1820) which is as a model fish organism frequently used for toxicology tests. The size of the particles was measured in plasma and in vitro neutrophil function assays were used to assess the effect of polystyrene and polycarbonate nanoparticles (0.1 µg/µl) on the innate immune system of fathead minnows. The exposure of neutrophils to the particles revealed a significant increase in oxidative burst activity, release of primary granules and release of extracellular nets release (NETs) compared to a non-treated control. The results underline the stress response of the fish innate immune system to polystyrene and polycarbonate nanoparticles and highlight their potential to interfere with the disease resistance in fish populations.

Dr Jan Rehwinkel talks about the role of our innate immune system in our fight against flu virus. The innate immune response is critical for a successful defence against viral infection. The first arm of our immune response is triggered by the detection of the presence of the virus: cell surface receptors detect the presence of viral genomic information. Dr Jan Rehwinkel is working to dissect the molecular biology of nucleic acid sensors. A better understanding of these mechanisms might help us develop better vaccination strategies.

In 1994, in a prospective control trial in cyclosporine-treated, kidneytransplant patients, we observed that treatment of a non-specificallograft injury (postischemic reperfusion injury) leads to asignificant reduction in the incidence of both specificalloimmune-mediated allograft rejection and chronic allograft failure. From these convincing clinical data, we concluded in terms of an‘argumentum e contrario’: it is the tissue injury that induces immunity. As from where we stand today in innate immunity research, these earlyclinical observations can be regarded as the discovery of the existence of a human innate immune system activated by tissue injury and precedingadaptive immunity.

Christine Biron is the chair of the Department of Molecular Microbiology and Immunology at Brown University in Providence, and she focuses her research program on the mechanisms of the innate immune system – the body’s system of non-specific munitions for fighting off pathogens. Dr. Biron is also a newly elected fellow of the American Academy of Microbiology. When a pathogen gets on or in your body, your innate immune system is on the front lines, working against the pathogen is a non-specific manner. In research, the innate immune system got short shrift for a long time, and only in the last 10 or 20 years has the field picked up momentum. Dr. Biron says back when she was in graduate school “the innate immune system wasn’t thought to be very cool”, but she says the field is fast-moving today, in part because of some major discoveries involving Type-1 interferons, natural killer cells, and an increased appreciation of a wider range of antigen processing cells that link the innate and adaptive immune responses. In this interview, I talked with Dr. Biron about our increasing awareness of the innate immune system, why it’s important to bring microbiologists and immunologists together under one big tent, and why it’s best that a battle between a virus and a host ends not in victory for one and defeat for the other, but in détente.
