Early Immune Evasion Found in HPV-Related Pre-Cancer Lesions of the Anogenital Region

BUFFALO, NY - April 28, 2025 – A new #research paper was #published in Oncotarget, Volume 16, on April 24, 2025, titled “PD-L1 and FOXP3 expression in high-grade squamous intraepithelial lesions of the anogenital region." Researchers Humberto Carvalho Carneiro, Rodrigo de Andrade Natal, José Vassallo and Fernando Augusto Soares from the Instituto D'Or de Pesquisa e Ensino and Rede D'Or studied early tissue changes caused by human papillomavirus (HPV) in the anal, vulvar, and penile regions. They found that high-grade pre-cancer lesions triggered stronger immune responses and showed higher levels of two immune-related markers, PD-L1 and FOXP3. These findings are important because they help explain how some HPV-related lesions progress to cancer while others heal on their own. High-risk HPV is known to cause several types of anogenital cancers. Before these cancers appear, the virus often leads to abnormal tissue changes known as high-grade squamous intraepithelial lesions. Many of these lesions disappear without treatment, but some become cancer—especially in people with weakened immune systems. This study explored how immune activity may play a role in this progression. The researchers examined tissue from 157 patients—95 males and 55 females—with either high-grade or low-grade HPV-related lesions. They found that T-regulatory cells, marked by the FOXP3 protein, were more common in high-grade lesions. These immune cells are known to suppress immune responses, which can allow infected or abnormal cells to grow. The team also found higher expression of PD-L1, a protein that helps cells evade immune detection, particularly in inflammatory immune cells. "Dense inflammatory infiltrates and high counts of FOXP3+ cells were significantly more frequent in patients with HSILs than in those with LSILsHR (p = 0.04 and 0.02, respectively). HSILs also exhibited higher PD-L1 expression (padj < 0.01 and < 0.01 for the SP142 and 22C3 clones, respectively), based on the Poisson generalized linear model.” These findings suggest that HPV may begin avoiding the immune system early in infection, even before cancer develops. The combination of high FOXP3 and PD-L1 levels may create a protective environment for infected cells, making them harder for the body to eliminate. This immune evasion may allow the lesions to remain and, over time, become cancerous. The study also compared patients with and without HIV to assess whether immune health influenced the results. While those with compromised immune systems had more extensive lesions, PD-L1 and FOXP3 expression was also found in patients with healthy immune systems. This evidence shows that immune evasion by HPV can happen regardless of a person's immune status. Understanding how PD-L1 and FOXP3 function in early HPV-related lesions may help clinicians predict which lesions are more likely to become cancer. These insights could lead to new strategies for monitoring, treating, or preventing HPV-related precancerous lesions and cancer in the anogenital region. The study highlights how early immune system changes can play a key role in the development of HPV-related cancers. Dr. Betsy Young, a physician-scientist from UCSF provides an overview of her funded work: Tumor cGAS-STING repression drives immune evasion in osteosarcoma and is therapeutically targetable via host STING activation. This work was funded by an MIB Agents 2024 OutSmarting Osteosarcoma YI Hope grant Because of Charlotte.Osteosarcoma (OS) has an immunosuppressive macrophage-rich, T-cell-depleted tumor microenvironment (TME). By performing bulk RNA seq of OS cell lines treated with STING agonist, the lab has defined an OS-specific STING activation signature, which demonstrated a significant protective effect on survival in OS patient samples. In immunocompetent OS models, systemic STING agonism shows curative anti-tumor effects, shifts the tumor microenvironment towards a pro-inflammatory phenotype, and induces immunologic memory. Importantly, host STING activation is sufficient to promote this anti-tumor immunity. The lab has demonstrated that STING activation has anti-tumor benefit in animal models and a protective effect in the human disease, nominating this innate immune sensing pathway as an important therapeutic target in OS.As a physician-scientist and a pediatric oncologist, Dr. Young's aim is to advance the field of pediatric oncology in her research career focused on the immunobiology of osteosarcoma. She completed her Pediatric Hematology/Oncology fellowship training at UCSF, receiving strong clinical training in high-risk pediatric solid tumors and early-phase clinical trials. Now, as a faculty member, she is investigating the pathogenesis of osteosarcoma metastasis in the Sweet-Cordero lab at UCSF, with a specific focus on immuno-oncology translational therapeutics.

“Israel built an ‘AI factory' for war. It unleashed it in Gaza,” laments the Washington Post. “Hospitals Are Reporting More Insurance Denials. Is AI Driving Them?,” reports Newsweek. “AI Raising the Rent? San Francisco Could Be the First City to Ban the Practice,” announces San Francisco's KQED. Within the last few years, and particularly the last few months, we've heard this refrain: AI is the reason for an abuse committed by a corporation, military, or other powerful entity. All of a sudden, the argument goes, the adoption of “faulty” or “overly simplified” AI caused a breakdown of normal operations: spikes in health insurance claims denials, the skyrocketing of consumer prices, the deaths of tens of thousands of civilians. If not for AI, it follows, these industries and militaries, in all likelihood, would implement fairer policies and better killing protocols. We'll admit: the narrative seems compelling at first glance. There are major dangers in incorporating AI into corporate and military procedures. But in these cases, the AI isn't the culprit; the people making the decisions are. UnitedHealthcare would deny claims regardless of the tools at its disposal. Landlords would raise rents with or without automated software. The IDF would kill civilians no matter what technology was, or wasn't, available to do so. So why do we keep hearing that AI is the problem? What's the point of this frame and why is it becoming so common as a responsibility-avoidance framing? On today's episode, we'll dissect the genre of “investigative” reporting on the dangers of AI, examining how it serves as a limited hangout, offering controlled criticism while ultimately shifting responsibility toward faceless technologies and away from powerful people. Later on the show, we'll be speaking with Steven Renderos, Executive Director of MediaJustice, a national racial justice organization that advances the media and technology rights of people of color. Allison Black surprised her family by deciding to enlist in the U.S. Air Force after high school. She did it to be part of a mission bigger than herself. After basic training, Black spent the next several years as a Survival, Evasion, Resistance, and Escape - or SERE - instructor, teaching service members what to do if shot down, stuck behind enemy lines, or captured. After several years of that she became an officer in Air Force special operations and served as a navigator aboard an AC-130H gunship. Shortly after 9/11, she and her crew were deployed to Afghanistan.In this edition of Veterans Chronicles, Col. ACTA's Doug Sprei interviews Peter Skerry, professor of political science at Boston College, who has served as a senior fellow at the Brookings Institution and research fellow at the American Enterprise Institute. The Museum was founded in 2009 as a 501(c)3 non-profit corporation with a mission to preserve the rich heritage of NAS Brunswick (NASB) by honoring the tens of thousands of men and women who served here, educate the public about the history of maritime patrol aviation and NASB, and maintain a memorial for those NASB-based airmen who gave their lives in service to their country. The museum is located at 179 Fitch Avenue, Brunswick, ME 04011. Currently, the Museum and Memorial Gardens are open Wednesdays (10:00 am-4:00 pm) and Sundays (12:00 pm-4:00 pm). Aviation News Spirit to Furlough More Than 300 Pilots as Losses Mount Spirit Airlines is experiencing financial difficulty and a second-quarter net loss of $158 million. More red ink is expected for the third quarter. Last month the airline furloughed 186 pilots and plans to do the same for 330 more pilots effective January 31, 2025. (Spirit has 3,500 pilots.) About 120 captains are to be downgraded. Spirit says it will sell 23 of its Airbus A320 and A321 aircraft next year. That's a fleet reduction of about 11 percent. Energy Reserves and Pilot Training For eVTOLs Addressed in FAA's SFAR Rules The FAA issued its final rule for powered-lift vehicles, such as eVTOLS. The Integration of Powered-Lift: Pilot Certification and Operations; Miscellaneous Amendments Related to Rotorcraft and Airplanes - Final Rule adopts permanent amendments and a Special Federal Aviation Regulation (SFAR) for ten years to facilitate the certification of powered-lift pilots, clarify operating rules applicable to operations involving a powered-lift, and finalize other amendments which are necessary to integrate powered-lift into the National Airspace System (NAS). These provide critical command, control, and communication (C3) for the President, the Secretary of Defense, and the Chairs of the Joint Chiefs of Staff. The E-4C will replace the current fleet of E-4B Nightwatch B747-200 aircraft. The contract notice says the award was for $13 Billion. The lawsuit alleges that Bivona, as president of the corporation, made false statements to customs officials and, as a result, avoided paying antidumping duties owed on the imported furniture.
