Paula Cilleros-Holgado from Pablo de Olavide University discusses a #research paper she co-authored that was #published in Volume 17, Issue 2 of Aging (Aging-US), entitled “Mitochondrial dysfunction, iron accumulation, lipid peroxidation, and inflammasome activation in cellular models derived from patients with multiple sclerosis.” DOI - https://doi.org/10.18632/aging.206198 Corresponding author - José Antonio Sánchez-Alcázar - jasanalc@upo.es Video interview - https://www.youtube.com/watch?v=wIV0lAHPA_M Abstract Multiple sclerosis (MS) is an inflammatory demyelinating disease of the central nervous system (CNS). Despite advancements in managing relapsing active illness, effective treatments for the irreversible progressive decline in MS remain limited. Research employing skin fibroblasts obtained from patients with neurological disorders revealed modifications in cellular stress pathways and bioenergetics. However, research using MS patient-derived cellular models is scarce. In this study, we collected fibroblasts from two MS patients to investigate cellular pathological alterations. We observed that MS fibroblasts showed a senescent morphology associated with iron/lipofuscin accumulation and altered expression of iron metabolism proteins. In addition, we found increased lipid peroxidation and downregulation of antioxidant enzymes expression levels in MS fibroblasts. When challenged against erastin, a ferroptosis inducer, MS fibroblasts showed decreased viability, suggesting increased sensitivity to ferroptosis. Furthermore, MS fibroblasts presented alterations in the expression levels of autophagy-related proteins. Interestingly, these alterations were associated with mitochondrial dysfunction and inflammasome activation. These findings were validated in 7 additional patient-derived cell lines. Our findings suggest that the underlying stress phenotype of MS fibroblasts may be disease-specific and recapitulate the main cellular pathological alterations found in the disease such as mitochondrial dysfunction, iron accumulation, lipid peroxidation, inflammasome activation, and pro-inflammatory cytokine production. Methylene blue (MB) has gained some massive exposure the last couple of months and is seemingly one of the hottest topics; and not just in biohacking and longevity circles, but even amongst general health and wellness enthusiasts. This exponential rise in MB is largely due to a handful of viral social media posts the last couple of months, which then prompted many influencers and/or medical professionals to voice their opinions and/or concern surrounding MB. There is some great info being circulated, but, needless to say, there is also a lot of misinformation being propagated.I wanted to take to the mic and address some of this misinformation and concern head-on. One issue that I see arise is around the quality of MB and, more specifically, the fact that MB can be derived from petroleum substances. Notice how I said, "can be." Yes, different industrial use cases may obtain their MB from petroleum-based sources, but is not the same MB that would be considered safe for human consumption. Also, there is a notion going around that you can make "homemade" leucomethyene blue simply be adding vitamin C to MB. I explain why this is a half-truth and does not lead to the same quality, purity, stability, bioavailability or effectiveness in mitochondria.The study covered in today's episode demonstrates how the mitochondria are being recognized as an integral therapeutic target for traumatic brain injuries (TBIs). That would mean a.) mitochondrial dysfunction is a large part of the brain damage (and, thus, recovery) associated with TBIs and, presumably, other head trauma, such as concussions; and b.) anything that can improve mitochondrial function, particularly things that can positively affect the brain, has a large potential role to play in the recovery from TBIs and other head trauma. The Mitochondrial Triad ticks all of those boxes. Last month I was invited to speak at a functional medicine symposium in Rome, Italy. This is the same conference where I gave an in-depth presentation explicitly on photobiomodulation two years prior. This time around, I spoke on the term/theory I have coined: The Mitochondrial Triad.As many of you know by this point, this Triad is, what I believe to be at this point in time based on the information I have gathered, the three most efficient and efficacious ways to boost your mitochondrial function: red light therapy, methylene blue and carbon 60. The presentation I gave first explained why the mitochondria are the focal point of this Triad and justified why the crux of modern health and disease is predicated on mitochondrial function; or said another way, why mitigating and preventing mitochondrial dysfunction is the ultimate goal of wellness and longevity. Exercise is good for you, except in very specific cases when it isn't. One of the few examples, post-exertional malaise (PEM), is the subject of today's Raise the Line episode in our series on post-acute infection syndromes produced in collaboration with the Cohen Center for Recovery from Complex Chronic Illnesses at Mt. Sinai. “The key to understanding PEM is that it's not just fatigue. It's really the worsening of symptoms or the onset of new symptoms after exercise,” says Dr. Rob Wust of Vrije University Amsterdam. His research group is one of several around the world now focused on this previously obscure phenomenon due to the prevalence of long COVID in which PEM is a nearly universal symptom. And while it's good news that this debilitating affliction is receiving new attention, no clear picture of causes and treatments has yet emerged. In terms of suspects, the body's energy suppliers, mitochondria, are a popular target, but while Wust's team and others attempt to unravel the mysteries of mitochondrial dysfunction, millions of patients suffering with PEM need strategies to avoid exhaustion. In that regard, Dr. David Putrino of the Cohen Center says patients and clinicians alike need to take all use of bodily energy into account, not just obvious activities such as walking. “The brain uses a lot of energy, so cognitive exertion will cause crashes. With the full reveal of The Mitochondrial Triad coming soon, it begs the question: why do I believe the focal point of our health, wellness, and, more precisely, our longevity is centered around or organelle friends, the mitochondria? And with the first two pillars being disclosed, in red light therapy and methylene blue, what is the third pillar? All of these questions are answered in today's solosode!For decades, the primary thesis for why we age was The Free Radical Theory of aging. However, this doesn't hold up in instances where someone consistently exercises (and, thus, produces more ATP that inherently produces more of the natural byproduct of free radicals) that leads to health- and longevity-promoting benefits. That's where The Mitochondrial Theory of Aging emerges and is currently the most respected postulation on why we age. However, the first rendition of this theory still included free radicals as a main culprit for aging; this was remedied in the second version, which removed free radicals as the central causation for aging and suggested the main source was in the numerous mechanisms lying within the mitochondria. Huzzah! Another week, another solosode and more research to delve into and learn from. Just what the doctor ordered...But before we cover research, I thought it would be worth our while to learn from the Finnish biohacking experts — one of which is previous TRLR guest, Dr. Olli Sovijärvi — in their massive tome, Biohacker's Handbook. Specifically, I thought it would be great provide you with some strategies and tactics for your sauna sessions via a sauna protocol they provide in the book.
