Abstract: The ability to prioritize task-relevant inputs enables efficient behavior across the human lifespan. However, contexts in which feature relevance is ambiguous require dynamic exploration rather than stable selectivity. Although both cognitive flexibility and stability generally decline with ageing, it is unknown whether the aging brain differentially adjusts to changing uncertainty. Here, we comprehensively assess the dynamic range of uncertainty adjustments across the adult lifespan (N = 100) via behavioral modelling and a theoretically informed set of human neuroimaging signatures (EEG-, fMRI-, and pupil-based). As a group, older adults show a broadscale dampening of neuro-computational uncertainty adjustments. In support of a 'maintenance' account of brain aging, older individuals with more young-like neural recruitment were better able to select task-relevant features, also in a Stroop task with low perceptual demands. Our results highlight neural mechanisms whose maintenance plausibly enables flexible task set, perception, and decision computations across the adult lifespan. Copy rights belong to original authors. Using computational modelling and human single-neuron recordings, we show that the more precisely hippocampal spiking variability tracks the composite features that comprise each individual stimulus, the better those stimuli are later remembered. We propose that moment-to-moment spiking variability may provide a new window into how the hippocampus constructs memories from the building blocks of our sensory world. C., Lindenberger, U., Kuehn, S., Werkle-Bergner, M., Mather, M. Abstract: Researchers have identified changes in dopaminergic neuromodulation as playing a key role in adult memory decline. Facilitated by technical advancements, recent research has also implicated noradrenergic neuromodulation in shaping late-life memory development. However, it is not yet clear whether these two neuromodulators have distinct roles in age-related cognitive changes. Combining longitudinal high-resolution magnetic resonance imaging of the dopaminergic substantia nigra-ventral tegmental area (SN-VTA) and the noradrenergic locus coeruleus (LC), we found that dopaminergic and noradrenergic integrity are differentially associated with individual differences in younger (n = 69) and older adults' (n = 251) memory performance. While LC integrity was related to better episodic memory across several memory tasks, SN-VTA integrity was linked to working memory. Moreover, consistent with their dense interconnection and a largely shared biosynthesis, dopaminergic and noradrenergic brain regions' integrity were positively related, and correlated with medial temporal lobe volumes. Longitudinally, we found that older age was associated with more-negative change in SN-VTA and LC integrity (time point 1--time point 2; mean delay ~1.9 years). Importantly, changes in LC integrity reliably predicted future episodic memory performance (at time point 3). These findings support the feasibility of in-vivo indices for catecholaminergic integrity with potential clinical utility, given the degeneration of both neuromodulatory systems in several age-associated diseases. Moreover, they highlight differential roles of dopaminergic and noradrenergic neuromodulatory nuclei in late-life cognitive decline. Copy rights belong to original authors. Abstract: The ability to flexibly switch between tasks is key for goal-directed behavior and continues to improve in late childhood. Children's task switching difficulties are thought to reflect less efficient engagement of sustained and transient control processes, resulting in lower performance on blocks that intermix tasks (sustained demand) and trials that require a task switch (transient demand). Sustained and transient control processes are associated with frontoparietal regions, which develop throughout childhood and may contribute to task switching development. We examined age differences in the modulation of frontoparietal regions by sustained and transient control demands in children (8-11 years) and adults. Children showed greater performance costs than adults, especially under sustained demand, along with less upregulation of sustained and transient control activation in a set of frontoparietal regions. Compared to adults, children showed greater increases in connectivity between the inferior frontal junction (IFJ) and lateral prefrontal cortex (lPFC) from single to mixed blocks. For children whose sustained activation was less adult-like, increased IFJ-lPFC connectivity was associated with better performance. Children with more adult-like sustained activation showed the inverse effect. Taken together, these results suggest that the configuration of regions that control task switching effectively undergoes developmental changes in later childhood. Hi, I'm a Global Health Economist. My name is Hyorin Nam, an upcoming college, pre-med student pursuing a major in Life science and Biotechnology. My podcast aims to educate people about urgent humanitarian crises and recent scientific development to increase awareness of global injustice and improve the quality of human health. Today, June 2nd, Wednesday, 2021, we will discuss the Tulsa Race Massacre, one of the single worst incidents of racial violence in American history, and whether teens get vaccinated even with parental disapproval. Today's highlights Tulsa Race Massacre took place on May 31 and June 1, 1921, was the incident wherein White residents attacked Black residents and burned businesses of Greenwood District in Tulsa, Oklahoma, the US, which was considered a Black Wall Street, a commercial center, with 41 grocery stores, 30 restaurants, 11 boarding houses, nine billiard parlors, five hotels, and many other businesses. Most states require a parent's or legal guardian's consent for anyone under 18 to get a vaccine, including the COVID-19 shot. While teens are more susceptible to the more severe multisystem inflammatory syndrome in children (MIS-C)- a serious complication that causes inflammation of internal organs, many parents disapprove of their children getting vaccinated. How can they vaccinate? Why do they want to vaccinated despite disapproval? Teens want to get vaccinated to protect the health of their own and community. “Giving teenagers and young adults the option and the autonomy to make those decisions really helps equip them for moving into adulthood and shows them they have responsibilities to take care of themselves and their community by getting vaccinated,” says Lindenberger, who testified before Congress in 2019 about the dangers of vaccine misinformation. Teens want to get vaccinated to improve their mental health. Research suggests that during the pandemic, teens have suffered from increased anxiety, depression, and loneliness. A recent report from the nonprofit Fair Health found that mental health insurance claims for teens age 13 to 18 doubled in March and April of 2020 compared to those months the previous year.

Abstract: Socio-economic status (SES) has been proposed to have facilitating and protective effects on brain and cognition. Here we show that relationships between SES, brain volumes and general cognitive ability differ significantly across European and US cohorts (4-97 years, N {approx} 500,000; 54,000 with brain imaging). Education was positively related to intracranial (ICV) and total brain gray matter (GM) volume. Income was related to ICV, but not GM. Relationships varied significantly across samples, and SES was more strongly related to brain and cognition in US than European cohorts. Differences in neuroanatomical volumes explained part of the SES-cognition relationships. SES was more strongly related to ICV than to GM, implying that SES-cognition relations in adulthood are less likely grounded in neuroprotective effects on GM volume in aging. Rather, a relationship may be established early in life. The findings underscore that SES has no uniform association with, or impact on, brain and cognition. Copy rights belong to original authors. Abstract: Adaptive human behavior builds on prior knowledge about stimulus relevance. Some environments cue such knowledge more than others. To behave adaptively, observers need to flexibly adjust sensory processing to the degree of contextual uncertainty. We hypothesize that the neural basis for these perceptual adjustments consists in the ability of the cortical network to switch back and forth between a rhythmic state that serves selective processing, and a state of elevated asynchronous neural activity that boosts sensitivity. To test this hypothesis, we recorded non-invasive EEG and fMRI BOLD dynamics while 47 healthy young adults performed a parametric visual attention task with varying numbers of relevant stimulus features. Drift-diffusion modeling of response behavior and electrophysiological signatures revealed that greater contextual uncertainty lowered the rate of evidence accumulation while increasing thalamocortical engagement, with concomitant increments in cortical excitability and pupil dilation. As predicted, uncertainty-related processing adjustments were expressed as switches between a state of phase-dependent excitability modulation in the alpha band and a state of increased irregularity of brain dynamics. We conclude that humans dynamically adjust sensory excitability according to the processing fidelity afforded by an upcoming choice, and that neuromodulatory processes involving the thalamus play a key role in adjusting excitability in the human brain. Abstract: Normal aging and Alzheimer's Disease (AD) are accompanied by large-scale alterations in brain organization that undermine brain function. Although hemispheric asymmetry is a global organizing feature of cortex thought to promote brain efficiency, current descriptions of cortical thinning in aging and AD have largely overlooked cortical asymmetry. Consequently, the foundational question of whether and where the cerebral hemispheres change at different rates in aging and AD remains open. First, applying vertex-wise data-driven clustering in a longitudinal discovery sample (aged 20-89; 2577 observations; 1851 longitudinal) we identified cortical regions exhibiting similar age-trajectories of asymmetry across the adult lifespan. Next, we sought replication in 4 independent longitudinal aging cohorts. We show that higher-order regions of cortex that exhibit pronounced asymmetry at age ~20 also show asymmetry change in aging. Results revealed that both leftward and rightward asymmetry is progressively lost on a similar time-scale across adult life. Hence, faster thinning of the (previously) thicker homotopic hemisphere is a feature of aging. This simple organizational principle showed high consistency across multiple aging cohorts in the Lifebrain consortium, and both the topological patterns and temporal dynamics of asymmetry-loss were markedly similar across replicating samples. Finally, we show that regions exhibiting gradual asymmetry-loss over healthy adult life exhibit faster asymmetry-change in AD. Overall, our results suggest a system-wide breakdown in the adaptive asymmetric organization of cortex across adult life which is further accelerated in AD, and may implicate thickness asymmetry as a viable marker for declining hemispheric specialization in aging and AD. Abstract: While development and aging of the cerebral cortex show a similar topographic organization and are mainly governed by the same genes, it is unclear whether the same is true for subcortical structures, which follow fundamentally different ontogenetic and phylogenetic principles than the cerebral cortex. To test the hypothesis that genetically governed neurodevelopmental processes can be traced in subcortical structures throughout life, we analyzed a longitudinal magnetic resonance imaging dataset (n = 974, age 4-89 years), identifying five clusters of longitudinal change in development. With some exceptions, these clusters followed placement along the cranial axis in embryonic brain development, suggesting continuity in the pattern of change from prenatal stages. Developmental change patterns were conserved through the lifespan and predicted general cognitive function in an age-invariant manner. The results were replicated in longitudinal data from the Lifebrain consortium (n = 756, age 19-83 years). Genetic contributions to longitudinal brain changes were calculated from the Vietnam Era Twin Study of Aging (n = 331 male twins, age 51-60 years), revealing that distinct sets of genes tended to govern change for each developmental cluster. This finding was confirmed with single nucleotide polymorphisms and cross-sectional MRI data from the UK Biobank (n = 20,588, age 40-69), demonstrating significantly higher co-heritability among structures belonging to the same developmental clusters. Together, these results suggest that coordination of subcortical change adheres to fundamental principles of lifespan continuity, genetic organization and age-invariant relationships to cognitive function. Abstract: Background: Older persons with poor sleep are more likely to develop neurodegenerative disease, but the causality underlying this association is unclear. To move towards explanation, we examine whether sleep quality and quantity are similarly associated with brain changes across the adult lifespan. Methods: Associations between self-reported sleep parameters (Pittsburgh Sleep Quality Index;PSQI) and longitudinal cortical change were tested using five samples from the Lifebrain consortium (n=2205, 4363 MRIs, 18-92 years). Analyses were augmented by considering episodic memory change, gene expression from the Allen Human Brain Atlas, and amyloid-beta (A{beta}) accumulation (n=1980). Results: PSQI components sleep problems and low sleep quality were related to thinning of the right lateral temporal cortex. The association with sleep problems emerged after 60 years, especially in regions with high expression of genes related to oligodendrocytes and S1 pyramidal neurons. BMI and symptoms of depression had negligible effects. Sleep problems were neither related to longitudinal change in episodic memory function nor to A{beta} accumulation, suggesting that sleep-related cortical changes were independent of AD neuropathology and cognitive decline. Conclusion: Worse self-reported sleep in later adulthood was associated with more cortical thinning in regions of high expression of genes related to oligodendrocytes and S1 pyramidal neurons, but not to A{beta} accumulation or memory decline. The relationship to cortical brain change suggests that self-reported sleep parameters are relevant in lifespan studies, but small effect sizes, except for a few restricted regions, indicate that self-reported sleep is not a good biomarker of general cortical degeneration in healthy older adults. Abstract: Objective: To test the hypothesis that worse self-reported sleep relates to reduced hippocampal integrity as indexed by increased intra-hippocampal water diffusion, and that this relationship is stronger in the presence of {beta}-amyloid (A{beta}) accumulation, a marker of Alzheimer's disease (AD) pathology. Methods: Two-hundred and fifty-one participants, aged 19-81 years, completed the Pittsburgh Sleep Quality Index, and 2 diffusion tensor imaging sessions, on average 3 years apart, allowing estimates of decline in hippocampal microstructural integrity as indexed by increased mean diffusivity (MD). We used the delayed recall from the California Verbal Learning Test to measure memory change. 18F-Flutemetamol PET, in 108 participants above 44 years of age, yielded 23 A{beta} positive cases. Genotyping enabled controlling for APOE {varepsilon}4 status, and polygenic scores for sleep efficiency and AD. Results: Worse global sleep quality and sleep efficiency related to more rapid reduction in hippocampal microstructural integrity over time. Focusing on sleep efficiency, this relationship was stronger in presence of cortical A{beta} accumulation. Sleep efficiency also related to memory decline indirectly via hippocampal integrity decline. The results were not explained by genetic risk for sleep efficiency and AD. Conclusions: Poor self-reported sleep efficiency related to decline in hippocampal integrity, especially in the presence of A{beta} accumulation. Poor sleep and hippocampal microstructural decline may partly explain memory decline in older adults with A{beta} pathology. The relationships were not explained by genetic risk, and poor self-reported sleep efficiency might constitute a risk factor for AD, although the causal mechanisms driving the of observed associations are unknown.
