Abstract: The structure of neural circuitry plays a crucial role in brain function. Previous studies of brain organization generally had to trade off between coarse descriptions at a large scale and fine descriptions on a small scale. Researchers have now reconstructed tens to hundreds of thousands of neurons at synaptic resolution, enabling investigations into the interplay between global, modular organization, and cell type-specific wiring. Analyzing data of this scale, however, presents unique challenges. To address this problem we applied novel community detection methods to analyze the synapse-level reconstruction of an adult fruit fly brain containing over 20 thousand neurons and 10 million synapses. Using a machine-learning algorithm, we find the most densely connected communities of neurons by maximizing a generalized modularity density measure. We resolve the community structure at a range of scales, from large (on the order of thousands of neurons) to small (on the order of tens of neurons). We find that the network is organized hierarchically and larger-scale communities are composed of smaller-scale structures. Our methods identify well-known features of the fly brain, including its sensory pathways. Moreover, focusing on specific brain regions, we are able to identify subnetworks with distinct connectivity types. For example, manual efforts have identified layered structures in the fan-shaped body. Our methods not only automatically recover this layered structure, but also resolve finer connectivity patterns to downstream and upstream areas. We also find a novel modular organization of the superior neuropil, with distinct clusters of upstream and downstream brain regions dividing the neuropil into several pathways. These methods show that the fine-scale, local network reconstruction made possible by modern experimental methods are sufficiently detailed to identify the organization of the brain across scales, and enable novel predictions about the structure and function of its parts. Ich habe diese Woche einen neuen Wartungsvertrag von einem Meerwasseraquarium übernommen. Hierbei wird  das bestehende 800 Liter Aquarium in den nächsten Tagen komplett entleert, auseinander geschnitten und entsorgt. Danach wird das Aquarium durch ein neues Becken und eine komplett neue Aquarientechnik ausgetauscht. Die Aquarientechnik wird in diesem Zuge in den darunterliegenden Keller verlegt, da es den Kunden sehr wichtig ist, dass das neuen Meerwasser-becken absolut leise ist. Damit die Rohre in den Keller verlegt werden können, wurde bereits direkt unter dem Aquarium eine Kernbohrung durch die Betondecke durchge-führt. Der Fischbesatz des Aquarium besteht aus mehreren Doktorfischen, einem Pinzettfisch und einer Zebramuräne. Auffällig bei allen ist Fisch, dass sich die oberen Hautschichten im Kopfbe-reich teilweise komplett abgelöst haben.  Dies zieht sich fast bei allen Fischen bis zum Ansatz der Rückenflosse fort. Dort wo die Fischschleimhaut ursprüngliche bunt gefärbt war sind nur noch große weiße Flecken vorhanden, die von groben Hautkanten umgeben sind. Solche starke Probleme mit der Fischschleimhaut haben häufig ihren Ursprung in einer sehr langen Fehlernährung, deren Folge dann starke Mangelerscheinungen sind. Auch in diesem Fall liegt die Ursache der Fütterung.  Die Fische wurde über Jahre mit Süßwassertrockenfutter ernährt.  Süßwassertrockenfutter wird auf den Bedarf von Süßwasserfischen und nicht von Meerwasserfischen entwickelt. Dadurch fehlen lebenswichtige Inhaltsstoffe für Meerwasserfische. Neben dem genannten Trockenfutter bekamen die Fische noch rote und weiße Mückenlarven. Der Vorteil momentan ist, dass alle Fische sehr gut fressen und keiner das Futter verweigert. Wir haben jetzt im ersten Schritt das Futter sofort auf das Dr. Basslers Bio Fishfood Garlic umgestellt. Das ist die Futtersorte die wir hauptsächlich bei allen unserer Wartungs-kunden verwenden. BASSLEER BIOFISH FOOD GARLIC enthält natürlichen Knoblauch. Der Vorteil ist, dass es auch von bereits erkrankten Zierfischen, die häufig unter Appetitlosigkeit leiden, gern gefressen wird.  Knoblauch enthält den natürlichen Pflanzenwirkstoff Allicin sowie einen sehr hohen Anteil an Mangan, Vitamin B6, außerordentlich viele Spurenelemente (Selen, Jod, Germanium, etc.) und Enzyme. Um die wertvollen Inhaltsstoffe zu schonen wird ausschließlich frischer Knoblauch (kein Granulat) nach dem Granulierprozess zugemischt. Ein weiterer Vorteil von diesem Granulatfutter ist, dass die Körner verschieden schnell absinken und dadurch jeder Fisch, egal in welcher Wasserschicht, oben unten oder in der Mitte aufhält gut versorgt wird. Um die Fische adhoc noch besser mit Vitaminen zu versorgen, sprühen wir das Granulatfutter jeden zweiten Tag mit Power Fish von Dr. Bassler ein.  Power Fish ist enthält Vitaminen und Spurenelementen und reichert das Granulatfutter an. Ich habe selbst noch nie so starke Mangelerscheinungen bei Meerwasser-fischen gesehen. Ich kenne nur die typischen leichten Veränderungen der Schleimhaut bei Doktorfischen im Kopfbereich, jedoch nicht so stark wie in diesem Fall, dass sich diese in ganzen Fetzen abgelöst haben. Terrice Bassler, a leadership coach, says that, when she is coaching someone, “I look for the red thread. I look for that thread that runs through somebody's life story, even if it looks a little disconnected and wild.”Bassler's own red thread may be providing service to others.Raised in the Helderberg Hilltowns, where both sides of her family had lived for generations, she says, “I was the first grandchild on either side of the family so I benefit from a lot of attention and love and teaching, which is with me still today.”She describes, in this week's Enterprise podcast, a series of seemingly small encounters that became life-changing events. “Small opportunities are what big opportunities look like when they're young,” she says.As Bassler has worked with visionaries “at extreme emotional edges,” she said she learned how to keep herself whole and support courageous others to stay whole.Now, she said, international organizations are “more clued up about what needs to be done to support people at extreme emotional edges — and COVID has pushed that even further.”Bassler calls this the mainstreaming of trauma.When she was shaken by the death of her father, she stumbled upon a class in neurogenic shaking, which is used to release trauma and tension.She practiced what is known as TRE — tension or trauma release exercise — that helps her stay regulated. With the shutdown caused by the coronavirus pandemic, Bassler became a certified provider and now teaches TRE online.She has supported health-care workers and first responders. “A lot of folks are struggling …,” she said. “They've gotten to that edge where they can't make meaning of what's happening. There's a continuous state of dread and uncertainty or discomfort.”Bassler says she helps people in a few lessons learn how to keep themselves regulated.Noticing the way society has become fragmented and divided — even in the town where she was raised — Bassler has found it useful to look for the wounded place — for her, that may be despair about the state of democracy — in the person with whom you are trying to connect.“That's going to make it easier to find common ground,” she said.One of the things Bassler remembers about growing up in the Hilltowns was that it was completely normal to drop in, “to just pop by.”“I've lived all over the world. Never anywhere I've lived since is dropping in as normal …. It was a queue of safety. Abstract: Natural products constitute a vast yet largely untapped resource of molecules with therapeutic properties. Computational approaches based on structural similarity offer a scalable approach for evaluating their bioactivity potential. However, this remains challenging due to the immense structural diversity of natural compounds and the complexity of structure-activity relationships. We here assess the bioactivity potential of natural compounds using random forest models utilizing structural fingerprints, maximum common substructure, and molecular descriptors. The models are trained with small-molecule drugs for which the corresponding protein targets are known (1,410 drugs, 0.9 million pairs). Using these models, we evaluated circa 11k natural compounds for functional similarity with therapeutic drugs (1.7 million pairs). The resulting natural compound-drug similarity network consists of several links with support from the published literature as well as links suggestive of unexplored bioactivity of natural compounds. As a proof of concept, we experimentally validated the model-predicted Cox-1 inhibitory activity of 5-methoxysalicylic acid, a compound commonly found in tea, herbs and spices. In contrast, a control compound, with the highest similarity score when using the most weighted fingerprint metric, did not inhibit Cox-1. Our results illustrate the importance of complementing structural similarity with the prior data on molecular interactions, and presents a resource for exploring the therapeutic potential of natural compounds. Well I just look at myself as an ordinary person register your lab work hard to become our guitars and you know I appreciate all the parents out there so much I may not yield to be an honor to have people listen to me naturally like what I'm doing, you still get a kick out of. When people come up to and say are you Steve Lynch yeah absolutely no will of course you know it's it's one of those things where the recognition and respect that you received the excitement from that never goes away. A lot of people get a chance to do some really cool things over the years of me being in radio I had the chance actually broadcast from inside of a bridge over the Mississippi River walk while they were building this bridge. I'm guessing that you had the chance to do some cool things that the average ordinary Joe hadn't had the chance to do is just the traveling yell and just being on stage plates, such are numerous amount of people and it was only autograph I did a lot of traveling. My clinics was promoting my books. I have three books out cultlike touch looks one, two and three of us promoting those like Dr. travel to 20 different countries in Q3 25 cities felt that was great. It's a blessed life. You know it really is. That's where I feel about him very thankful. Steve a lot of times people think of rock 'n' roll they think of sex drugs and rock 'n' roll that can ask about the typical question to most people ask, but I do ask about you know seeing some year friends and some of that you know people that you that you played with over the years, you know, succumb to drug addictions and things like that. It was, it isn't hard to be on you know be on stage and BN on the road and and and Kelly keep a clean and free life. Like the I was just you as I drink too much, but that you know that I was looking thought that I just didn't like okay so so that was that was easy for me to not but I didn't try but it was easy for me to share with you. I was just going through focusing on playing guitar and and just putting on a good performance and I've always just got My mindset that links it was difficult to also see all the people go through the hardships that you know what drugs you will because of our lives and see people go don't go very fast when they do try to come… Same as they were previously so that is. His music business different now than it was in the 80s. Europe your gambling into all these different aspects of the music business and trying to be a musician and songwriter. All the same time. Like with autograph. I was basically the general manager the tour manager and the business manager on the night on the one that was leaked the liaison in between, you know, all different aspects. The endorsements the booking agent. I did most most all the interviews and then I took care of all the banking eye to eye pain. Everybody I get all the business taxes and everything so you know those there is a big burden what I limit SSN because I think that the that the big thing now is that you have quit the band autograph. Steve stepped off from you know from from performing. Are you still a part of the band even though you're not playing with them or if you basically cut ties altogether. Well, I basically cut ties altogether. You know I wanted to move on to something different. And one thing my fiancé later told Ms. that I thought was a really good analogy of you know after your if you have a CD in your car and you play it over and over again for 35 years. Eventually you want to change that CD in your car and go on to different things you want your listener difference because you just got tired of playing the same songs doing the autograph that you know I look at myself like I have a lot more to offer the what I was contributing to autograph and point out is the last job we did get off your ass. That was mainly all written by sign that my writing was going in a completely different direction. Attached as it is now so it just didn't sound like autograph at all. I need not at all. I had the opportunity to interview Steve a few times over the last handful years and always enjoy chatting with them always enjoy seeing you guys live and always enjoy just the person that you are Steve because a lot of times rock stars. A lot of times people in your position. Don't give time to be ordinary person and you know were friends on Facebook. I've tagged you in a couple things and you actually respond to your fans, which I think is is cool. Well I just look at myself as an ordinary person register your lab work hard to become our guitars and you know I appreciate all the parents out there so much I may not yield to be an honor to have people listen to me naturally like what I'm doing, you still get a kick out of. When people come up to and say are you Steve Lynch yeah absolutely no will of course you know it's it's one of those things where the recognition and respect that you received the excitement from that never goes away. A lot of people get a chance to do some really cool things over the years of me being in radio I had the chance actually broadcast from inside of a bridge over the Mississippi River walk while they were building this bridge. I'm guessing that you had the chance to do some cool things that the average ordinary Joe hadn't had the chance to do is just the traveling yell and just being on stage plates, such are numerous amount of people and it was only autograph I did a lot of traveling. My clinics was promoting my books. I have three books out cultlike touch looks one, two and three of us promoting those like Dr. travel to 20 different countries in Q3 25 cities felt that was great. It's a blessed life. You know it really is. That's where I feel about him very thankful. Steve a lot of times people think of rock 'n' roll they think of sex drugs and rock 'n' roll that can ask about the typical question to most people ask, but I do ask about you know seeing some year friends and some of that you know people that you that you played with over the years, you know, succumb to drug addictions and things like that. It was, it isn't hard to be on you know be on stage and BN on the road and and and Kelly keep a clean and free life. Like the I was just you as I drink too much, but that you know that I was looking thought that I just didn't like okay so so that was that was easy for me to not but I didn't try but it was easy for me to share with you. I was just going through focusing on playing guitar and and just putting on a good performance and I've always just got My mindset that links it was difficult to also see all the people go through the hardships that you know what drugs you will because of our lives and see people go don't go very fast when they do try to come… Same as they were previously so that is. His music business different now than it was in the 80s. Europe your gambling into all these different aspects of the music business and trying to be a musician and songwriter. All the same time. Like with autograph. I was basically the general manager the tour manager and the business manager on the night on the one that was leaked the liaison in between, you know, all different aspects. The endorsements the booking agent. I did most most all the interviews and then I took care of all the banking eye to eye pain. Everybody I get all the business taxes and everything so you know those there is a big burden what I limit SSN because I think that the that the big thing now is that you have quit the band autograph. Steve stepped off from you know from from performing. Are you still a part of the band even though you're not playing with them or if you basically cut ties altogether. Well, I basically cut ties altogether. You know I wanted to move on to something different. And one thing my fiancé later told Ms. that I thought was a really good analogy of you know after your if you have a CD in your car and you play it over and over again for 35 years. Eventually you want to change that CD in your car and go on to different things you want your listener difference because you just got tired of playing the same songs doing the autograph that you know I look at myself like I have a lot more to offer the what I was contributing to autograph and point out is the last job we did get off your ass. In 1990 a young Bonnie Bassler, mesmerized by glow-in-the-dark bacteria that could talk to their peers to coordinate light production, wondered whether other bacteria could talk too. The answer, she soon found out, was yes – including all the nasty bacteria that cause disease. Today, Bonnie Bassler is a professor in molecular biology at Princeton University and an authority in the field of bacterial communication. Her findings, that all bacteria can talk, revolutionized the way we think of bacteria and opened the doors to important medical and industrial applications. But the discovery of bacterial communication has given us much more than new drugs. It has shown us how bacteria live in the real world and forced us to reconsider our own human nature – if bacteria are talking, social beings too, how different are we from them after all? To navigate a complex world, microbes use chemical signals to sense and communicate with one another. Filmed live on January 28th, 2013, at ASM's headquarters, catch a glimpse into the fascinating language of bacteria with discussions by Bonnie Bassler, Princeton University, and Steven Lindow, University of California, Berkley. Dr. Bonnie Bassler, Princeton University Bonnie Bassler Ph.D. is a Howard Hughes Medical Institute Investigator and the Squibb Professor of Molecular Biology at Princeton University. The research in her laboratory focuses on the molecular mechanisms that bacteria use for intercellular communication. This process is called quorum sensing. Bassler's research is paving the way to the development of novel therapies for combating bacteria by disrupting quorum-sensing-mediated communication. Dr. Bassler was awarded a MacArthur Foundation Fellowship in 2002. She was elected to the American Academy of Microbiology in 2002 and made a fellow of the American Association for the Advancement of Science in 2004. Dr. Bassler was the President of the American Society for Microbiology in 2010-2011; she is currently the Chair of the American Academy of Microbiology Board of Governors. She is also a member of the National Science Board and was nominated to that position by President Barak Obama. The Board oversees the NSF and prioritizes the nation's research and educational priorities in science, math and engineering. Dr. Steven Lindow, University of California, Berkeley Steven Lindow Ph.D. is a Professor at the University of California, Berkley where his research focuses on various aspects of the interaction of bacteria with the surface and interior of plants. Dr. Lindow' s lab uses a variety of molecular and microscopy-based methods to study the ecology of bacterial epiphytes that live on the surface of plants as well as certain bacteria that are vascular pathogens of plants. They also study bacteria that live in and on plants that are fostered by consumption of the alkaloids produced by endophytic fungi. The longer-term goal of their research is to improve plants' productivity by achieving control of plant diseases through altering the microbial communities in and on plants. Dr. Lindow is a member of the National Academy of Sciences, and was elected to fellowship in both the American Academy of Microbiology and the American Association for the Advancement of Science in 1999.
