Abstract: Degeneration of the dopaminergic nigro-striatal pathway and presence of Lewy bodies are pathological hallmarks of Parkinsons disease (PD). Postmortem studies in human tissue have also demonstrated that a decline in mitochondrial number and function is also central to PD pathology. Sirtuin 3 (SIRT3) is a mitochondrial protein deacetylase which has been linked with longevity and cytoprotective effects. SIRT3 serves as a metabolic sensor and regulates mitochondrial homeostasis and oxidative stress, which likely stabilises telomere integrity, delaying senescence. Previously, we have shown that over-expression of SIRT3 rescues motor function and prevents degeneration of dopaminergic neurons in a virally over-expressing mutant (A53T)-alpha-synuclein model of PD. In the present study, we show that in the substantia nigra pars compacta (SNc) of human subjects, SIRT3 levels are negatively correlated with age (p less than 0.05, R=0.6539). In the hippocampus, there was no correlation between SIRT3 levels and age. In human subjects with PD, SIRT3 was reduced by 56.8{+/-}15.5% and 34.0{+/-}5.6% in the SNc and hippocampus respectively regardless of age. Given that age is the primary risk factor for PD, this finding suggests that reduced SIRT3 may be a causative factor contributing to PD pathology. Next in human subjects with PD, we measured whether there was a correlation between the amount of aggregated alpha-synuclein and SIRT3 levels by measuring immunofluorescence of phosphorylated alpha-synuclein (p-syn), which is a marker for Lewy bodies. Interestingly, in the hippocampus, but not SNc, there was a positive correlation between SIRT3 and p-syn levels, despite p-syn being reduced compared to control. Next using an alpha-synuclein seeding rat model of PD, we assessed the disease-modifying effects of viral-mediated SIRT3 infusion. Six months following infusion of alpha-synuclein pre-formed fibrils (PFF) into the SNc, there was 38.8{+/-}4.5% loss of TH-positive neurons, impaired striatal dopamine metabolism and pathological alpha-synuclein throughout the brain. Phosphorylated-alpha-synuclein immunoreactivity was present in the SNc, olfactory tubercle, striatum, amygdala, hippocampus and motor cortex. In PD subjects, synuclein positive aggregates have also been reported in these brain regions. In PFF rats, infusion of rAAV1.SIRT3-myc in the SNc reduced abundance of alpha-synuclein inclusions in the SNc by 30.1{+/-}18.5% which was not seen when deacetylation deficient SIRT3H248Y was transduced. This demonstrates the importance of SIRT3 deacetylation in reducing alpha-synuclein aggregation. However, while SIRT3 transduction reduced aggregation in the SNc, it had no significant effect on phosphorylated-alpha-synuclein levels in other brain regions. These studies confirm that SIRT3 is directly correlated with senescence and aging in humans. We also provide evidence that reduced SIRT3 contributes to the pathology of clinical PD. Finally, by showing that over-expression of SIRT3 prevents alpha-synuclein aggregation through de-acetylation-dependent mechanisms, we further validate AAV1.SIRT3-myc as a potential disease-modifying therapy for PD. I'm your host, Kelly Preece. And every fortnight I talk to a different guest, about researchers development, and everything in between. Hello,   00:33 and thank you for joining us on this online resource. My name is Bisi Obamakin, and I'm a theology PhD student at the University of Exeter. Today, we are joined by an amazing New Testament scholar, and author of many books, including her newest book, creating compassionate campuses, Professor Louise   00:50 Lawrence. Thank you for joining us, I really, really appreciate it like a really, really big   00:57 thanks for inviting me.   00:59 So yeah, a little bit about yourself what you do and what your research passions are.   01:03 Yeah, so my name is Louise Lawrence, as you as you said, I'm Professor of de testament interpretation here in Exeter. My research interests I work in New Testament studies. So particularly sort of cross cultural anthropology, with biblical texts, but latterly, for the last sort of decade, I've been really interested in the ways in which religion and sacred texts sort of sensor bodies and minds and particularly around disability studies, so yeah, so that's, that's my interests,   01:42 for the people that are watching that maybe they've just gotten a PhD or whatever they're doing, right at the beginning of their career, they're not socialized into any institution, what would you say to them? What would you employ to them? With regards to pedagogy and decolonization? And that kind of thing?   01:57 I think, I mean, well, you're a brilliant example of this. And you should probably say a bit about how you're, I'm picking, I'm picking New Testament studies. But, you know, I'll let you talk about you've got more important things to say on this, I think that you must be true to yourself, you know, and in a sense, if it matters to you, it matters. And if you identify in justice, even if other people haven't seemed to be able to have recognized or have sort of been made conscious of that, then call it out. And I think, I think as an early career, academic, you can, there's a very well known thing called imposter syndrome, I shouldn't be here. I don't look like I should be here. I don't sound like I should be here. I'm not clever enough. But everyone goes through those things. Everyone feels those things, it, it's a very natural part of it. And that says probably more about the in hospitality of academia, or the perceived sort of sense of academia than it does about you. And you just have to have the confidence to have that voice. I you say about your, your ways in which you're sort of challenging the Eurocentrism of Biblical Studies. Yeah, and finding a voice that's been lost or not even recognized. It's that that that no curriculum that you've kind of picked up?   03:26 Yeah, I guess it's just kind of coming into the field and not seeing myself and not really knowing where I fit. And thinking, obviously, recognizing that I was born in the UK, and I do have a Western education. But not really fully feeling like I fit into that box. And then, you know, thinking, oh, yeah, I'm African. That's what I am. And they're not fully fit in that box, either. So yeah, I think bringing a kind of Nigerian British kind of hybrid viewpoint, it's been interesting because it has highlighted things that I don't think anyone has really thought about. Things that I've experienced, and I've walked in my life that are just so normal to me that I guess it's kind of almost a bit, that it was a bit kind of weird to think that it's new, like, why is it new to you? You don't know that? Yeah. Yeah, it's quite unique experience. And I think it's been quite quite privileged, bringing that to theology and introducing new ways of knowing and new new lenses of knowledge. Yeah.   04:27 Similar. Yeah, I mean, absolutely. So you're sort of Afro pm perspective as, as brought out so much of the text. And, and so, so much that that of assumptions that have just been accepted as liberal within interpretation, and actually, you know, should they be, I understand what a kind of transformative moment working with the British deaf community because they innocence very much. Made me unlearn things. So I mean, being an academic, you're very bookish i And I'm, I'm working on New Testament interpretation, I, you know, everything's about tanks. And suddenly to be in a context where I should say, You deaf community with a capital D is a cultural group that yeah, that see themselves very much as politicized, you know, death is not deficit death, yes, death is like an ethnic kind of identity marker with traditions, you know, and, and most crucially with their own language that is not to be written or heard. And so, the ways in which they actually modified my understanding of what Bible was, was really interesting. And, you know, in a sense, a gestural performative Bible, actually, for history has probably more of a resonance with with a context, when there was large, low literacy in the ancient world, you know, and so, those kinds of assumptions, but also the way in which they picked up on parts of the text, which I would never have kind of, or interpreters that were very audio centric, would never have picked up on. So one example is there's a story in the New Testament of Jesus healing a deaf man without speech, and the end of the story, it says, And the man spoke plainly, and in the first sort of reactions to this text, one of the group pointed out that that this person could couldn't have been born deaf. And that Jesus wasn't sort of seeing deafness as a bad thing, but needed to be healed or normalized. It was just that this person had been able to hear and then lost his hearing. And I was like, Well, why did you come to that conclusion? He said, Because he speaks plainly. So he's, he's learned spoken language. So you almost like modified that idea that that spoken language is better than being deaf and completely sort of changed it and actually healing narratives for for disability who are themselves, you know, like a colonized group, by the hearing world colonized their language, you know, you must learn this way. There are allied experiences, I think, with the deaf community and, and colonization, and they very much sort of resistant and refigured those those elements, and I think, yeah, and really opened up a whole new sort of avenue for for of understanding of the stories and traditions,   07:45 that is really powerful. I hadn't even thought about that before, like, wow,   07:49 well, most, I don't think I found from looking at common cheese from the 1800s. Right through no one really picks up that at all, most commentators just said, this is a straight healing story, obviously, you know, it's fulfilling prophecy and prophecy is very able that the deaf will hear the blind will see. And, and they completely kind of, yeah, went against that and, and just shows how norms just become complete status quo and accepted. Yeah, how different viewpoints can completely make you unlearn those? Yeah. Wow, I   08:27 think let me through amazing insights and this discussion. Just before we close, like, what exactly are you doing at the moment? What are you working on?   08:37 Yeah. Oh, that's really nice question. I've been doing a lot of marking of exams at the moment. I'm actually working with colleagues in psychology on a big project on student mental health, or this whole institution approaches to student wellbeing. And actually, you know, it may seem rather unconnected to what we're talking about, but I actually, you know, what, the medical model of mental illness I think, you know, has a place there are, of course, students that need that, that that help but actually, the social and cultural model of, of, of mental health and well being, I think, includes a lot about feeling culturally included, about feeling a sense of belonging, about feeling, a sense of being represented. And many of the sort of stories that we've we've we've been shared by students in this institution, and we're working with six other Russell Group institutions, but very much kind of, they are aware of how they curricular you know, their well being isn't something you can just individualize. Actually, it's about the whole student experience. And that includes what they're studying how they're studying how the how they're kind of learning community See makes them feel and I think, kind of cognitive, epistemological justice and feeling that, you know, like you're saying that there is representation that your voice is heard, all of these kinds of elements are things that can really enhance a sense of belonging and, and a sense of, of wellbeing for students. So yeah, so that's, that's kind of the the big project on on my specific New Testament, I'm going to start looking at, and it's only sort of the very beginnings, but I'd like to do a project on age and ageism. In in New Testament texts and interpretation, that too, I think it's very cross culturally constructed. Just to give you an anecdote, but before I finish, talking about global north south norms, what's really interesting is that I've got a postdoc that's working out in Namibia on a project on religion and inclusion in Namibia. And we had to go through all the ethics approval to work with Human Subjects through Exeter. And actually, you know, 18 is kind of the constructed age of adulthood in our context, but in other contexts, that that's a very meaningless kind of number. And it just shows how even up construction of the person or the thinking person or the person that is able to give consent is a construction. And actually, you know, if we're going to be an institution that thinks carefully about how it works with partners in different contexts, it needs to take seriously the ways in which different contexts construct, construct research, and it's sort of, yeah, voices in different sorts of ways. Similarly, anonymity was a really big thing in, in our ethics process in Exeter, as our participants in Namibia and our informants were really, really keen that their name was put, you know, if I'm saying something, I want my name to be in it. And, and we had to fight quite hard that those names were appended to the voices that we were given. A name in is, has also been used in colonial practices, you know, changing names or forced change of names. And so, so names, personhood and identity, you know, again, really important is to be very sensitive to the ways in which people, you know, are attached to names and more names represented. So yeah, so in our publications, we have our informants names, which is very different, I guess, to too many publications that go through ethics approval here.   12:55 I love it, guys. "VACCINATIEPASPOORT OPMAAT NAAR WERELDWIJDE CONTROLEMAATSCHAPPIJ" Vera Sharav is oprichter en voorzitter van de Alliance for Human Research Protection (AHRP), een netwerk van medische professionals en leken met als doel om het recht op medische vrije keuze te waarborgen. AHRP richt zich met name op het naleven van de Eed van Hippocrates, de Code van Neurenberg (https://nl.wikipedia.org/wiki/Code_van_Neurenberg) en de Universele Verklaring over Bio-ethiek en Mensenrechten van UNESCO. De Code van Neurenberg werd na de Tweede Wereldoorlog opgesteld om te voorkomen dat mensen worden blootgesteld aan medische experimenten tegen hun wil, zoals onder de Nazi’s gebeurde. Het eerste punt uit de Code stelt: “De vrijwillige toestemming van de proefpersoon is absoluut noodzakelijk.” Ook dienen mensen die worden blootgesteld aan medisch onderzoek voldoende te zijn geïnformeerd. Deze twee richtlijnen vormen samen het principe van “informed consent”. Volgens Sharav wordt in de huidige vaccinatieprogramma’s niet aan deze voorwaarden voldaan. Zij wijst erop dat de vaccins slechts een tijdelijke goedkeuring hebben, onder noodwetgeving, en dat nog onvoldoende data bekend zijn voor definitieve goedkeuring. Dit betekent dat mensen die nu het vaccin krijgen meedoen aan een medisch experiment, terwijl ze dat niet beseffen. De gevolgen van de vaccins op langere termijn zijn zeer onzeker, stelt Sharav: het gaat om technologieën die nog nooit eerder op mensen zijn toegepast. Bij dierproeven bleek dat dieren die het vaccin hadden gehad bij hernieuwde blootstelling aan het coronavirus extra kwetsbaar waren. In dit interview met consultant en schrijver Elze van Hamelen, wijst Sharav op het gevaar van misbruik van de “openbare gezondheidszorg.” Een individuele arts is, op grond van de Eed van Hippocrates, altijd verantwoordelijk voor zijn handelen tegenover een patiënt. De relatie patiënt-arts is bovendien strikt vertrouwelijk. Maar “wanneer artsen gezondheidszorg-ambtenaren worden, zijn ze niet verantwoordelijk meer,” zegt Sharav. Er ontstaat dan een gevaarlijke situatie: onder het mom van gezondheidszorg kunnen genetische en andere experimenten op mensen worden uitgevoerd. Sharav, die als kind de Nazi’s ontvluchtte, wijst erop dat het uitroeiingsprogramma van de Nazi’s begon met gehandicapten en ouderen, en dat al deze mensen werden vermoord door artsen en ander medisch personeel in medische programma’s. Zij vreest dat de invoering van vaccinatiepaspoorten de opmaat zal vormen naar een nieuwe controlemaatschappij, die dit keer wereldwijd zal worden uitgerold, en die ertoe zou kunnen leiden dat ook nu een groep tweederangsburgers ontstaat die kan worden geïsoleerd of uit de weg kan worden geruimd. On this week's episode of the podcast, we discuss protections in place for human research subjects. To start the conversation, we discuss some infamous examples of past injustices conducted in the name of "research" and the regulations they prompted as a result. We highlight four cases: the Holocaust experiments, the Tuskegee Syphilis Study, the work of James Marion Sims, and the case of Henrietta Lacks. We spend some time detailing historical documents and regulations that were developed to protect human subjects that came out of these horrific historical blunders and atrocities. Next, we discuss in detail what "informed consent" means, the rigorous regulatory process for any research involving humans, and the modern-day Institutional Review Board (IRB). As of May, 2019, only 43 of 50 states require schools to have safety plans and conduct safety drills (Education Commission of the States, 2019). When states mandate schools to have safety plans and conduct safety drills, it is the individual schools left to determine how they will design said plans and drills. In some states, the safety plan must be presented to a school board and submitted to the state Department of Justice. However, without templates and rubrics, there is littler inter-school reliability. What is a “good” safety plan or “effective” safety drill? Nobody knows. STANFORD PRISON EXPERIMENT WAS THE TIPPING POINT FOR PROTECTING HUMAN SUBJECTS. While the Stanford Prison Experiment was originally slated to last 14 days, it had to be stopped after just six due to what was happening to the student participants. The guards became abusive, and the prisoners began to show signs of extreme stress and anxiety. These were university students, assuming assigned roles, as part of an experiment that attempted to investigate the psychological effects of perceived power, focusing on the struggle between prisoners and prison officers. THE MELTDOWN. Similar to the infamous 1963 Milgram shock experiment, Stanford's experiment flew off the rails as subjects in positions of power followed directives that seemingly brought pain or harm to recipients.  Were the subjects acting as prison guards cruel, despicable people? Probably not. But, they were in a university context and a professor was mingling among them. They probably assumed that some measures were in place to prevent harm to recipients - and simultaneously lacked awareness of the endorphin rush they received from exerting their will over others. IRB WAS FORMED. In 1974, universities collectively ratified the Institutional Review Board (IRB) process. The mandate of the IRB is to provide ethical and regulatory oversight of research that involves human subjects by: Protecting the rights, welfare and well-being of human research participants, recruited to participate in research conducted or supported by the university. Psychological and physical welfare are carefully considered and risks are identified and mitigated. WHAT IS COMMON RULE? In 1991, 16 federal agencies formally adopted the core of these regulations in a common Federal Policy for the Protection of Human Subjects also known as the "Common Rule" (Grady, 2015). I propose that adopting the IRB in K-12 settings will increase safety for all drill participants and, through the scientific model, increase efficacy of school safety drills. Common Rule applies, for example, to the Food and Drug Administration and clinical trials for medications. Common Rule was updated in 2019 with greater emphasis placed upon simplifying subject consent forms and clearly explaining potential consequences of participating in studies. IRB or COMMON RULE WOULD ELEVATE SCHOOL SAFETY DRILLS. Schools can establish their own standards for school safety plans and safety drills as long as they are complying with state mandates. In other words, schools IRB or COMMON RULE for these reasons: (1) Corral theatrical intruder drills that might traumatize or physically harm participants. This happens - just do a search on Google for “Intruder Drill Lawsuit”. Also, hyper-realistic drills are not the gold standard. If they were, we would conduct fire drills and tornado drills with similar drama. So, a committee of administrators, teachers, students, parents and board members review each proposed safety drill. (2) Each drill would have at least one learning objective. For example, “If a lockdown is announced during passing time, students would go to a safe location.” You can define “safe” per your site. The DARPA Detection and Computational Analysis of Psychological Signals (DCAPS) program provided initial funding for the Durkheim Project. While DCAPS as a whole addressed PTSD, the Durkheim Project sought to predict military and veteran suicide risk. We developed a clinician's dashboard, which presents suicide risk predictions for the clinician's patients based on analysis of: a) free text portions of VA medical records and, b) opt-in social media postings. Dartmouth's Committee for the Protection of Human Subjects approved our protocol to conduct a Veterans Affairs (VA) medical records study with the White River Junction, Vermont, VA Medical Center. A second protocol has been approved to study opt-in Facebook postings from active duty military personnel and veterans. We have built the software infrastructure to collect these opt-in postings in collaboration with Facebook. A third protocol is currently being developed to support intervention, when high suicide risk is predicted. This talk will describe the Durkheim Project, focusing on privacy issues related to using opt-in social media postings. About the speaker: Paul Thompson's thesis research was on probabilistic information retrieval. He was an assistant professor at Drexel University's College of Information Studies and then a member of PRC, Inc.'s (now part of Northrop Grumman) artificial intelligence development group, where he conducted research in natural language understanding and information retrieval. He later worked for West Publishing Company (now part of Thomson - Reuters). After joining Dartmouth College's Institute for Security Technology Studies in 2001, he became the technical lead for the Semantic Hacking project. He also participated in an Institute for Information Infrastructure Protection (I3P) research project on control system security in the oil and gas industry. He is currently an instructor in the Department of Genetics at the Geisel School of Medicine at Dartmouth. He was the co-principal investigator on the DARPA Detection and Computational Analysis of Psychological Signals program's Durkheim project.

This was a double-blind placebo-controlled study with a cross-over design to examine the effects of scopolamine on cognitive functions in young healthy subjects. Scopolamine hydrobromide was administered subcutaneously to 12 subjects (mean +/- SD age 23.8 +/- 2.2 years) at doses of 0.3 and 0.6 mg in comparison with two placebo conditions. Scopolamine at both doses produced marked sedation as rated by subjects and an observer. In the continuous performance test, vigilance was impaired by both doses of scopolamine. The span of apprehension test showed differing results (only the high dose of scopolamine showed a performance decrement only in the three-character version of the span of apprehension test). Significant impairment by both doses of scopolamine was seen in immediate and delayed free recall, continuous visual recognition, running word recognition and running picture recognition. While scopolamine caused a significant slowing in average reaction times for simultaneous matching as well as for delayed matching, subjects made more errors under scopolamine compared to placebo only in delayed matching, not in simultaneous matching. Also, the main outcome of matching to sample showed significant effects only in delayed matching, not in simultaneous matching. Notable in this study is the incongruity between the simultaneous matching test and the span of apprehension test on the one hand and the other cognitive tests used on the other. These results demonstrated that scopolamine has a greater effect on memory than on attention. Thus, the scopolamine-induced effects in the present study seem to be more relevant to Alzheimer's disease in an advanced phase than to normal aging. Copyright (C) 2003 S. Karger AG, Basel.
