Abstract: Stiffened arteries are a pathology of atherosclerosis, hypertension, and coronary artery disease and a key risk factor for cardiovascular disease events. The increased stiffness of arteries triggers the hypermigration and hyperproliferation of vascular smooth muscle cells (VSMCs), leading to neointimal hyperplasia and accelerated neointima formation, but the mechanism of this trigger is not known. Our analyses of whole-transcriptome microarray data sets from mouse VSMCs cultured on stiff hydrogels simulating arterial pathology and from injured mouse femoral arteries revealed 80 genes that were differentially regulated (74 upregulated and 6 downregulated) relative to expression in control VSMCs cultured on soft hydrogels and in uninjured femoral arteries. A functional enrichment analysis revealed that these stiffness-sensitive genes are linked to cell cycle progression and proliferation. Furthermore, we found that survivin, a member of the inhibitor of apoptosis protein family, mediates stiffness-sensitive cell cycling and proliferation in vivo and in vitro as determined by gene network and pathway analyses, RT-qPCR, and immunoblotting. The stiffness signal is mechanotransduced via FAK and Rac signaling to regulate survivin expression, establishing a regulatory pathway for how the stiffness of the cellular microenvironment affects VSMC behaviors. Our findings indicate that survivin is necessary for VSMC cycling and proliferation and regulates stiffness-responsive phenotypes. Copy rights belong to original authors. Abstract: Vascular dysfunction is a common cause of cardiovascular diseases characterized by the narrowing and stiffening of arteries, such as atherosclerosis, restenosis, and hypertension. Arterial narrowing results from the aberrant proliferation of vascular smooth muscle cells (VSMCs) and their increased synthesis and deposition of extracellular matrix (ECM) proteins. These, in turn, are modulated by arterial stiffness, but the mechanism for this is not fully understood. We found that survivin (an inhibitor of apoptosis) is an important regulator of stiffness-mediated ECM synthesis and intracellular stiffness in VSMCs. Whole-transcriptome analysis and cell culture experiments showed that survivin expression is upregulated in injured femoral arteries in mice and in human VSMCs cultured on stiff fibronectin-coated hydrogels. Suppressed expression of survivin in human VSMCs and mouse embryonic fibroblasts decreased the stiffness-mediated expression of ECM components implicated in arterial stiffness, namely, collagen-I, fibronectin, and lysyl oxidase. By contrast, expression of these proteins was upregulated by the overexpression of survivin in human VSMCs cultured on soft hydrogels. Atomic force microscopy analysis showed that suppressed or enhanced expression of survivin decreases or increases intracellular stiffness, respectively. I think it's interesting that a protein in our bodies, that we develop when we are young and shuts off as we get older, that's labeled "Survivin" can actually be used to fight Glioblastoma and potentially other rare cancers. This month's Survivor guest is Robyn Bloom. A REAL ONE, to say the least. Robyn is a life coach practitioner, author, mentor, and mother to a group of awesome young women. She like to acknowledge that she is also a former victim of her own bullshit (her words, not mine LOL). She likes to use the word former because she now takes time to hold herself accountable for any chaos and toxic energy she brings. In 2016, while in the hospital dying, her journey began. In that hospital bed, she started forgiving herself for some of her decisions and found a little peace. For her, it was that peace that saved her life. During her search for self, she became a life coach. And in 2020, she was blessed to rehab a house that she affectionately named The Healing House. This is where she works with women and girls doing healing, bonding, and meditation. Robyn believes that we all have the power to heal but it takes hard work, dedication, and a strong support system. In this episode of the Epigenetics Podcast, we caught up with Professor Bill Earnshaw, Wellcome Trust Principal Research Fellow at the University of Edinburgh, to talk about his work on the role of non-histone proteins in chromosome structure and function during mitosis. In the beginning of Bill Earnshaw's research career little was known about the structure that holds the two individual sister chromatids together. This led to Bill pioneering in the use of autoantibodies for the identification and cloning of key chromosomal proteins. He used serum from a scleroderma patient to identify and clone human centromeric proteins, which paved the way for the molecular characterization of the metazoan kinetochore. Later the chromosomal passenger complex (CPC) was identifies in his lab using biochemical studies. This complex contains Aurora B kinase plus its targeting and regulatory subunits INCENP, Survivin, and Borealin/Dasra B. More recently, he teamed up with the laboratories of Job Dekker and Leonid Mirny. In this collaboration they used a system for synchronous mitotic entry developed by Kumiko Samejima.These studies used a combination of chemical biology, gene targeting, Hi-C genomics, and polymer modeling to explore the roles of condensin I and condensin II in mitotic chromosome formation. This week, we’re talking about The Four Stages of Your Financial Life. In your financial life there are 15, yes count’em, 15 things that you need to do. 15 sounds daunting, doesn’t it? If you write them all down that’s not a quick post-it note, it is a full blown and hardy list. That’s why I’ve broken up your financial life into four stages for you to accomplish those 15 things in. Because how much nicer of a number does four sound than 15. Your financial life starts between the age of 18 to 22 and goes all the way until you retire around the age of 67-ish. In your financial life, you are going to work through four different stages. I like to call those: Survivin’, Drivin’, Arrivin’ and Thrivin’ (yes, I know those rhymed). Within each of those four stages I have broken up those 15 things you need to accomplish appropriately in each. When you have completed the designated set of “to-do’s” in each category, you can move onto the next the stage. Before we dive into all you need to get done, we’re going to do a little self-evaluation. If you’ve been following my blog or podcast for a while you know I’m big on the self-evaluation. It gives us a starting point. A great inside to not only where you are in your financial life, but it will tell us the stage you’re in and whether there are things you need to catch up on or plan for. In this episode I will break down each stage and the tasks they include for you to successfully, at your own pace, make it to the next stage of your financial life. You may be sitting there reading this and asking yourself, “How long does it take to make it through each stage?” Excellent question. It will be different for each individual, and that’s the beautiful thing about it. It may take a fresh college graduate who started a respectable salaried job right after school only two or three years to get out of survivin’. While it may take someone who has restarted their financial life 10 years to accomplish all the tasks in the first stage. This is where the 15 things come into role as: goals. Goals are apart of why I created these stages, to help focus on individual tasks to be accomplished. If you aren’t setting and working towards a goal(s), then you could be survivin’ for the rest of your life. You don’t want that, and I don’t want that. Focus on working towards your goals. The more your focus on them, the sooner you’ll make it through to the next. I’ll be here to help you through each and every stage along the way.

The study is subdivided into two different parts: the first part deals with the development of a method to gain uterus milk in vivo during the preimplantation periode in cattle for the investigation of regulatory factors. The second part investigates different proteases in bovine follicles 20 hours after GnRH (Gonadotropin releasing hormone) injection (shortly bevor ovulation) for comparable as well as in the corpus luteum (CL) during oestrous cycle and induced luteolysis. In addition apoptotic as well as anti-apoptotic factors were evaluated in the CL during oestrous cycle and induced luteolysis. For the development of a method for gaining uterus milk in vivo during the first 24 days of gravidity in cattle, nine heifers were cycle synchronised using the Ovsynch method and artificially inseminated. Before flushing an epiduralanaesthesia was given and both uterus horns were flushed with 13ml 0.9% NaCl using a balloon embryo transfer catheter at day 5, 7, 12, 17 and 24 of gravidity. The catheter was placed 1cm cranial to the bifurcatio uteri in both horns. It was possible to retrive between 3ml and 13ml of the used flushing fluid. The uterus milk from the ipsilateral horn was inspected for an embryo and an EDTA-stabilisator was given to the uterus milk of both horns. An infection of the uterus occured in three heifers after the second and in five heifers after the third flushing. In one heifer no infection was found. Between day 17 and day 24 all heifers showed clear signs of oestrus. It was possible to detect progesterone, oestradiol-17-beta, PGF2alpha and VEGF via enzyme immunoassay (EIA) and radio immunoassay (RIA), respectively. Durch den Transfer tumorspezifischer T-Zellen können solide Tumore nicht nur in Tieren, sondern auch im Menschen erfolgreich behandelt werden. Dudley et al. konnten kürzlich nach dem autologen Transfer stimulierter tumorinfiltrierender Lymphozyten (TIL) nach vorheriger non-myeloablativer Konditionierung in zwei von zwölf Patienten mit Melanomen eine Tumorregression erreichen (Dudley et al. 2002). Änliche Erfolge konnten Chang et al. bei der Behandlung von fortgeschrittenen Nierenkarzinomzellen erzielen (Chang et al. 2003). Sie vakzinierten Patienten mit Calmette-Guérin Bacillus versetzten Tumorzellen und ernteten anschließend die T-Zellen aus den angrenzenden Lymphknoten der Vakzinierungsregion. Nach autologem Transfer der vorwiegend CD8+ T-Zellen, welche in vitro mit anti-CD3 stimuliert wurden, sprach die Therapie bei neun von 34 Patienten an. Die Mechanismen der T-zellvermittelten Tumorregression sind aber bis heute noch unklar. Gegenstand dieser Arbeit war es daher Faktoren zu untersuchen, welche eine Rolle bei der Regression von D5-Melanomlungenmetastasen durch den adoptiven Transfer CD4+/CD8+ T-Zellen aus Tumorvakzin drainierenden Lymphknoten (TVDLN) nach Vakzinierung mit D5G6 (einem GM-CSF transduzieren D5-Subklon) spielen. Hierzu wurden D5-Zellen in vitro mit Zytokinen, welche von Effektor T-Zellen (TE) produziert werden, stimuliert. Wir konnten zeigen, dass D5-Tumorzellen die als proapoptotisch geltenden Rezeptoren TNF-R I sowie Fas nach IFN-g-Stimulation exprimieren. Die Inkubation mit den Liganden dieser Rezeptoren TNF-a, LT-a und FasAb bewirkt keine Apoptoseinduktion. Gleichzeitige Inkubation dieser Liganden führt nur zu einer Apoptoserate von 25% in D5. Mögliche Gründe der relativen Apoptoseresistenz von D5 könnten die intrazellulären Apoptoseinhibitionsproteine c-IAP I, c-IAP II und Survivin sein, welche wir in D5 nachweisen konnten. Da nach adoptivem Transfer tumorspezifischer TE Makrophagen die pulmonalen Tumormetastasen infiltrieren, untersuchten wir, ob TE Chemokine exprimieren. Unsere Untersuchungen ergaben, dass TE die Chemokine MIP-1a, MIP-1b und RANTES tumorspezifisch exprimieren. Nach Stimulation von D5-Tumorzellen mit den Zytokinen TNF-a und IFN-g, welche von TE tumorspezifisch exprimiert werden, wird die Chemokin-mRNA von KC, MCP-1, IP-10 und Mig in D5 exprimiert. Auch die Koinkubation von D5 mit TE induziert die Expression der mRNA von KC, MCP-1, IP-10 und Mig. Deutlich erhöhte KC und MCP-1 Proteinkonzentrationen wurden im Überstand von D5-Tumorzellen mittels ELISA nach TNF-a Stimulation gegenüber unstimulierten und IFN-g stimulierten D5-Zellen festgestellt. Um zu prüfen, ob die von D5 gebildeten Substanzen auch tatsächlich chemotaktisch auf Makrophagen wirken, führten wir einen chemotaktischen Assay durch. Wir konnten zeigen, dass es zu einer signifikanten Steigerung der Anzahl migrierter Makrophagen in Richtung D5- Tumorzellen nach deren Stimulation mit IFN-g und TNF-a kommt. Da wir in TE die Expression von TNF-a-mRNA und die Sekretion von IFN-g tumorspezifisch nachweisen konnten, besteht Grund zur Annahme, dass in vivo TE in der Lage sind Tumorzellen zur Chemokinbildung anzuregen, was zu einer Invasion von Makrophagen in die Tumorregionen führt. Dass Makrophagen möglicherweise eine entscheidende Rolle bei der Tumorregression spielen, deckt sich auch mit der Beobachtung von Feinmesser et. al., welche mit einer peritumorösen Injektion von Multikinen (Zytokin-/Chemokinmischung) Kopf und Halstumore behandelten (Feinmesser et al. 2003). Nach dieser Therapie zeigte sich in den Tumorregionen der erfolgreich behandelten Patienten im Gegensatz zu Non-Respondern eine hohe Anzahl migrierter und aktivierter Makrophagen. Wir fanden mit den Ergebnissen unserer Studie erstmals Hinweise darauf, dass im murinen Melanommodell B16BL6-D5 nicht ausschließlich die direkte T-zellabhängige Zytotoxizität zur Tumorregression führt, sondern dabei weitere Zellarten wie Makrophagen/Monozyten involviert sind, welche mittels tumorspezifischer Chemokine rekrutiert werden. Die vorliegende Arbeit liefert einen Beitrag zur Erforschung der Mechanismen T-zellvermittelter Tumorregression. Die Kenntnisse dieser Mechanismen haben entscheidende Bedeutung bei der Entwicklung neuer, effektiver Behandlungskonzepte gegen solide Tumore.
