38-year-old Dyshan Best was asked a simple question. To step out of the vehicle for a pat search. Officer Erin Perotta of the Bridgeport Police Department was looking for a gun after responding to a call about a big fight. Best agreed to step out of the vehicle and be searched, but instead blasted past Perotta. She pursued on foot, joined by officer Yoon Heo. When Best made he way down an alley, he displayed the firearm that the officers had been looking for. A dangerous situation escalating and a weapon in Hand, Heo had to use lethal force to stop the threat. Now people unfamiliar with the danger and the weight of carrying a badge want to weigh in and say Best was taken down unnecessarily, and from behind, rather than admitting compliance was his safest option. Joining me today is Air Force SMSgt (Ret) Juan Lopez. Juan grew up in California both in Los Angeles and later Vacaville. He attended Vacaville High School and graduated in 1999. He joined the Air Force in 2000 and went to Basic Training in 2001. His AFSC was 1C6 and he attended tech school at Vandenberg AFB, CA. Juan's first assignment was to 11 SWS (ALERT) at Schriever AFB, CO. In 2002, he was reassigned to 2 SWS at Buckley AFB, CO. After that tour, his next assignment was to work HEO with 460th Det 1 back at Schriever AFB in 2006. Three years later, Juan was chosen to be an instructor with the 533rd Training Squadron back at Vandenberg AFB. In 2013 he was assigned to 13 SWS at Clear AFB, AK for a one year remote tour. He was then assigned back to Buckley with the 2 SWS to work the SBIRS Block 10 program. In 2017, he was promoted to MSgt and then converted to the Reserves through Palace Front. He was assigned to 4 SWS at Buckley as an Active Guard/Reservist. Later, he was moved to the 710th Operations Group and was promoted to SMSgt in 2019. Juan retired on 1 Nov 2021.

Abstract: Dendritic spines are structural correlates of excitatory synapses maintaining stable synaptic communications. However, this strong spine-synapse relationship was mainly characterized in excitatory pyramidal neurons (PyNs), raising a possibility that inferring synaptic density from dendritic spine number may not be universally applied to all neuronal types. Here we found that the ectopic expression of H-Ras increased dendritic spine numbers regardless of cortical cell types such as layer 2/3 pyramidal neurons (PyNs), parvalbumin (PV)- and vasoactive intestinal peptide (VIP)-positive interneurons (INs) in the primary motor cortex (M1). The probability of detecting dendritic spines was positively correlated with the magnitude of H-Ras activity, suggesting elevated local H-Ras activity is involved in the process of dendritic spine formation. H-Ras overexpression caused high spine turnover rate via adding more spines rather than eliminating them. Two-photon photolysis of glutamate triggered de novo dendritic spine formation in mature neurons, suggesting H-Ras induced spine formation is not restricted to the early development. In PyNs and PV-INs, but not VIP-INs, we observed a shift in average spine neck length towards longer filopodia-like phenotypes. The portion of dendritic spines lacking key excitatory synaptic proteins were significantly increased in H-Ras transfected neurons, suggesting that these increased spines have other distinct functions. High spine density caused by H-Ras did not result in change in the frequency or the amplitude of miniature excitatory postsynaptic currents (mEPSCs). Thus, our results propose that dendritic spines possess more multifaceted functions beyond the morphological proxy of excitatory synapse. Copy rights belong to original authors. Well, it was educational. I learned something because Yeah. I, I at least remember that it was concrete that he suggested not cement. Not cement. Did you go fix it? Fuck, no. I moved onto the next thing. You know what I did today, right? So Right when I, when I got this house going right, I, I was tossing up about what trees I should put out the front. And I really wanted something like, my house is like, it's like a Minecraft house. It's real brutal from the front. Yep. Wood and concrete, like, um, and I wanted something to soen that. So I've found this tree called a pink trumpet tree, whereas living in Varsity Lakes, you saw them everywhere. You see them everywhere. They're like a, they're not like a especially fancy tree. If you see a tree with like pink leaves, it's probably one of those. Yes. Um, and have my heart set on this. So like, for like six months, every time I saw one, I'm like, oh, there's a pink trumpet tree. Like, it was a big thing. So anyway, when I got the house, I finally bought one, went out to the nursery of Mount Tambourine. I bought an established one. It cost me like, I can't remember how much, maybe a thousand bucks, maybe more, I don't even know. Was like an establish tree? Yes. Got delivered and it's like, it was 150 liter, um, pot. So it's, it's big. It's like, it's like the size of a, a barrel, basically a half a barrel. Mm-hmm. Um, so I got delivered in the, in the plastic pot thing, dug a hole, chucked it in there. Mm-hmm. And then realized that you can't put in there with the plastic on cuz it's so heavy. You can't get the fucking plastic on. By the way, I didn't read instructions. Okay. I just bit of a pa, bit of a theme going on here and there. I'll, I'll get to. So I put it in there and then when it was in, when it was in there, I realized, fuck, I need to cut the plastic off. So I've got my arm under there trying to cut the plastic off. Finally got it in there. The roots are going everywhere. Dirt's going everywhere. It's a complete disaster. I put it in, it's not straight, and I'm like, fuck it. That'll do, because it'll probably straighten up. I don't know how trees work. Anyway, I've been staring at it every day for like six months. Okay. At one point I was like, I've gotta straighten this fucking tree because it's ridiculous. It goes like that leaning tower side and push it around. Tried to straighten it. That didn't help. So the tree was it again? Pink trumpet tree. They're really pink. Trumpet tree. If you, if you've ever been to Sanctuary Cove on the Gold Coast, they've got like really full established ones. They're huge. They look like they're looking up what it looks like. Pink trumpet tree. Here we go. A tab Taboo Rose. That's it. That's the one. Oh yeah, they look sick. Well, mine didn't look like that. This one, the one I see on the internet leaning. Oh, okay. Leaning. That could be my one. Does it have any leaves or, or, uh, flowers on it. Pink flowers. Pink flowers mate. And a cop car under it. Oh, it could be mine. Alright. So anyway, anyway, so I had this tree, the leaves started dying. I, I put in, in the Facebook group about landscaping. I'm like, what did I do about this tree? Didn't get anything useful. Eventually I'm like, fuck this tree. I don't like it. It's leaning. The leaves are dying. It's not working. So I ordered cuz I was always tossing up between that and Fran pan tree because Fran like Fran trees, I remember from when I was a kid. They're everywhere around here. They're pretty, they smell and they grow fast and they're pretty hardy fast. They, you can replant them and they're put in your ear when you put one in your ear and all, all those sort of things. I like doing that. Yeah. And when I knocked this house down, I had two amazing grand pan. That got destroyed when the house got knocked down, which I was really bummed about. I wanted to move one of them, but it's too hard. And anyway, so I'm like, fuck it, I'm gonna replace this tree with a fringe APA tree and I'm not gonna make the same mistake. So I ordered the fringe APA tree, and today the guy's like, it's coming. Um, you might need a couple of guys to lift it cuz this is a 200 liter one, which is even bigger. Yes. So it's probably like a 300 liter barrel, like half of that. Right. Um, so this thing, this thing was fucking huge. And it's a two, it's a two and a half meter tree. How do you lift this by yourself? Well, I'll get to that. So she rung me up and she said, all, all right. Um, get some neighbors around cause you need like four or five people to lift this thing. Okay. Like this, this, I can't do that. I'm just here. I'm here by myself. There's no one, no one's gonna help me with it. So this guy rocks up by himself with his massive tree. And wait, before then, I'm like, I've gotta get rid of the other tree first. So I dug the other tree out, I chopped the whole thing up, put it in my willie bin. So my, so my willy bin has this entire tree in it, which was about, it was like a two meter tree. So chopped all that up. Got it. In the willy bin. I'll mate rocks up with this gigantic tree and then we drag it onto the back of the truck and he is got a lift thing to get it to the ground, which is good. And then we drag it up to the hole because I only paid to get it delivered to the curb, but it's out the front of the house. I'm like, if I don't get him to do it now, I'm not gonna even be able to get it in. Okay. I want him to drag it there. And I left it right on the side of the hole. And then I, I thought about the podcast and thought what I should do is watch a video on instructions. Right. On how to plan the trees. So I did that. Yes. And the instructions basically said you take it out of the plastic thing first. Right. And you sit it there in the roots and then you kind of, you know, trim roots if necessary and kind of maneuver into positions. Yes. That kinda thing. Yep. I didn't do that though. Did you? Same thing. I just did the same thing that I did last time, which is I got it close to the edge and then I dragged it in and dumped it in the hole again. Hang on a second. So you actually went and got instruction on how to do something on YouTube? I did. And then you completely ignored it. I, I did. I in the moment, yes, I did ignore it. Yes. Out of expediency, you just wanted to get the job done or impatience, I think. And also like, I just had it so close to the edge and I cut a few little bits of it. I literally did, I don't know, like it just, I just did exactly the same thing that I did last time. I don't even know why I just made a mistake. So now the tree is in the bloody hole with plastic all over it. The other trees in the wheelie bin. This tree is in the hole with plastic all over. The roots are already starting to fall apart because I'd like smudged it in there and then I start cutting the plastic off and I'm trying to rip it out and I calm, my back's fucked and I managed to get under it. Finally rip the thing out. The roots are gone everywhere. I'd push it up and it's like wobbling. It doesn't even stand up. So I've got, I pull all the stakes out of the other parts of the garden that were holding my other trees up to put around this tree to hold this tree up. And I've only got little bits of string so I get it all up like reasonably in line. And the other thing that the video said was like that every tree has like a nice angle to look at it from. Mm-hmm. Like a face. Sure. So I was like, I need to line up this angle facing the road so that when you look at the house, you've got a nice Yep. Tree. Yeah. But then when I looked at it, all the spinning and shit, it's like facing me and it looks pretty average from the street. Okay. Anyway, I finally got it up there and I got it vertical, filled it in, and then I'm just sitting there, my back's fucked. I'm like, this, I've just cut. Totally fucked this whole thing up. Um, but I was happy it was in there. And then some lady walks past and she like stopped. She had her dog and she looks up at the tree. She goes, I don't like these trees. They put their leaves everywhere, all over the ground after you put in all that effort. Oh my God. Did you tell her to get stuffed? My blood started to boil. Yeah. God, I blame you. And I, and I don't, and I don't believe in violence against women or any person for that matter. Yes, yes. So I murdered her dog. It was one of those fluffy white ones. It was. All right. No, no harm done. All right. That's my dancing update. How's the scooter going? The scooters got terrible. Guess what I've gotta do on the weekend? I buy another, no, I'm gonna buy a motorbike. Oh, for fuck sake. What do you mean? You've already got a motorbike? Yeah, I've gotta buy another one. Why? Um, cause I've been looking at, uh, just cause of the motorbike that I've got, right? The, the, the CB 900 Hornet, which I absolutely love to ride. And I've had it since 2015. Right? And, um, it's 2002. It's not getting, it's nothing wrong with this not getting, like, it's rough around the edges, but it's super reliable and I can work on it and all that sort of thing. Uh, years ago when I was living in Geelong, you know, back in 10, 10 years ago, I had a, I think it was a 2002 or 2004 Honda VFR 800. So it's a v4, right? Yeah. And just sounded so fucking beautiful. Like it's half a V eight mate, so just, it's a motorbike that sounded like that. And um, and just over the last few months I've just sort of been started looking at 'em again on like Facebook oh nine Marketplace and stuff like that. That's where it all fucking starts. Exactly. This Facebook marketplace has been the bane of my existence in the last fucking, so bad 12 months. It's fucked. I've just spent so much money on, it's just, just keeping the economy going anyway, so I know exactly what under VFR 800 I want, I want the last of the sixth. Sixth generation. Sixth generation? I think so. Um, wait, so this is the bike you used to have? Yeah, I had a 2002. Okay. Um, but, um, um, but I, but they sort of didn't really change the, they sort of changed the shape in 2001 and they didn't change it again until 2014. Why do you, why did you get rid of that? To get the current bike bought? The Commodore trade it in for the Commodore. Oh, you got, oh, okay. And then you went without a bike for a while. For, for about three years and that sort of thing. So, uh, sorry, is it sixth generation? Yeah, sixth generation Honda bfr 800 and, um, 2013. So it's the last of the good ones because the, the, the seventh generation was just that, not that great. Sorry. The eighth generation just wasn't that, that fucking, it was just boring. Right. And so vfr, what is it? VFR 800. VFR 800. Do you wanna pull up a photo? Yeah. Um, might be able to pull up your Facebook. Um, pull up a photo, mate. Oh, that's sick. That's like a, uh, like a sports kind bike. Sports touring bike. Yeah, exactly. Oh yeah, yeah. Oh, he's the one that I want, here's the one that I want and lemme share my screen. Oh. Need to let me share a screen mate. Good. Can I go? Can I go? You can go like that is exactly what I want. Handicap red is is the beer, is the vtr. The half baring Vtr is the thousand CCV twin. Yeah. And it's like a half bear sort of, but it looks similar to that, doesn't it? Oh, that's more upright. Yeah, they little. They do. Yeah. But this is, but that, that's, that's the bike that I'm looking at at going and buying. So basically handy up this bike that I've found, it's up in Nua that I'm gonna have it dry right up there on Saturday. Handy Apple, red gold rim, gold rims, and a stain tune exhaust system on it. And it's just, I saw it pop up on Buddy Facebook and it was, And it was a good, really sharp price and the pays weren't too bad on it and that sort of thing. And it's like being sold by a dealer, which I don't know if that's a good thing or not a good thing. Um, but that, look at it, it's fucking beautiful. Yeah, that's pretty sick. And so the, I, so when I own mine, um, uh, the, the last one that I owned, I really enjoyed riding it. It just handled so well, you know, it was just so comfortable to ride. And I actually rode it from, well, I tried to ride it from Melbourne or Geelong when I was in Geelong, up to Queensland for a holidays in 2011. And the old ones, the pre 2000 eights actually had a problem where the regulator rectifier the thing that that charges the battery, not the generator of the stator, but the, the regulator rectifier would burn, would burn out the stator. So I was riding, I was just outside 50 Ks outside of Kuni Bar brand going up the new highway. And it just clapped out. Oh. And died. Had to get towed to Kobrand. Um, and this was right when we had that big cyclone that just hummed, um, you know, Gunda windy and all that sort of stuff, right? Yes. And I was heading to my brother's place. And he lives in Anor. Dan thought. Yes. And, and, um, and so he, he drove, he jumped in his ute, drove six hours, picked me in the bike up. I drove the six hours back, and we were two kilometers away from, um, his house. And the, the, the New England Highway had flooded and we couldn't get home. Oh no. Yeah. It was the last bridge. It was the last bridge that we had to cross before his place. And it was flooded. Oh my God. And he had his full drive and he was, we looked at it, we, uh, should we do it? Uh, and he said, no, no. We'll just, he called up a friend who was on that side of the creek. And, um, and, and so we went round to the friend's place and we just smashed whiskey. Like fucking, absolutely. Just, just pounding. Just, just whiskey rocks, right? Oh yeah. That's what my brother likes to do. And, um, and so, um, we've got, um, we stayed a couple of hours and then we'd heard that the, that the, um, um, that the creek, the creek had subsided a little bit. And so, um, we just changed up to the ute and went, and the creek had subsided and the bridge would become visible again. And we're so lucky we didn't attempt to cross the creek because half the road that we were gonna drive across had literally washed out and we would've, oh no. Gone in and fucked Ute and the bike so that the moral of that story is if it's flooded, forget it. And that is for reel. I actually did, I actually did cross one of those little rivers in Dan Thorpe that was flooded, but it was, it was pr reasonably obvious that it wasn't too bad, although I don't fucking have a clue what I'm doing, but it was kind of like, you could sort of tell when it was high and when it was low and it was kind of getting closer. Yeah. But yeah, it happens all the time in it, like every time. Yeah. Yeah. Ex. Yeah, exactly. So, um, so yeah. But anyway, it turns out that was a known problem with that year model, but this 2013 or 2008 onwards doesn't have this problem. And I reluctantly sold the bike in, you know, or traded it in actually on, on the Commod in 2013. So I didn't really have much of a choice because I was, I, I was a, I just just became an owner of a brewing company that didn't make any beer. Yeah. I, my old, my old 1991 Toyota Camry, I crashed. And, um, on, on the way into Meredith Music Festival, just fucking, just, I was fucking just driving, driving down the road into the Meredith Music Festival. The fucking sunflowers were out. It was a beautiful day, and I'm just pumped that I'm, you know, going to go to music. Mar Meredith Music Festival, have a amazing time and that sort of thing. And I'm like, ah, this is amazing. And look in front of me. Like, fuck. Cause the cars had all stopped and I, and I smashed into the car, um, in front of me and, and then a car smashed into me, behind me. Oh, the car in front was one of the people that we were convoying into camp with, and the car behind was my ex-girlfriend, uh, Erica. And, um, and so she crashed into me and she was in a borrowed car because she crashed her car like the week prior. It was just a complete and utter cluster fuck. And so I wound up with my, my Camry was still drivable, her borrowed car was not. And um, uh, and so I had this dodgy fucking, couldn't open the boot, couldn't open the bonnet Toyota Camry that I had to get rid of. So I just sold it to the records for 250 bucks. And I had to buy a new car. And I didn't have any money, but all I had was the VFR and I had to trade it in to buy a car. Oh, okay. And, um, I'll also have like a web app for, you know, because if you, I, I think a lot of people will still use on the computer and we will wanna actually type. But yes, once you can use the AI to generate a lot of content, you'd be surprised how much you can actually create on the phone. Yes. That's pretty nifty, mate. I think that's got some potential. Um, makes writing SOPs very easy, at least getting 80% of the way there. And that's the biggest challenge that I face when I work with my clients is they lack the time to write SRPs. Yeah. Mm-hmm. And everyone goes, Ooh. And then like, and then, and then, oh, we've got this thing called M S N. And so they basically built their own internet. Yeah. And, and it was built into Windows 95 and it locked. And I remember going to the Microsoft Conference, I think it was 96 9 19 96, like they literally the following year. And they were just like, internet, internet, internet. Cause they knew that they'd fucked up and they were just like, we have to do internet. And that was where it all sort of took off. And there an internet explorer became a thing and all that sort of stuff. And you know, and that's, that's kind of what, what, what sort of happened, you know, they make these decisions, they get on the back foot. They, they, they take a step back in technology. And I think that's what Google is about to experience is, is going to be behind the eight ball. And it could last a decade, you know. You know, the other thing, the other thing Windows fucked up was Skype. Like, can you, yeah. Even imagine like, Skype was so popular, it was like the only tour people used. Cause I was like, um, um, I mean, you, you're in craft beer professionals on Facebook Aren. Yeah. Yeah. So there was a guy who, they did, they had a session from this guy called, uh, called a, his, this YouTube channel called Adam Makes Bid Year. And I'd had his CVP thing just added to my watch later and I actually watched it this afternoon finally, three weeks later. And he's very interesting. So he, he's, he's like a content creator that sort of does. He's, he, I actually wanna meet him cause he's very fascinating guy, right? Because he's a teacher as well, or ex-teacher. Mm-hmm. And he's a, he's been a brewer for about 12 years and he does YouTube videos of stuff that he does in the brewery and how he makes beer and all that sort of stuff. Yeah. And then he does like live streams. I'm like, fuck, I wanted to do that. And so I watched his presentation, his CBP presentation, and he mentioned his Instagram and I've just followed him on Instagram and he's just written me back an Instagram message going, oh man, you're the reason I started my YouTube channel. Oh, no way. So, um, you could do lots on, so, okay, so your channel, do you, is that like, is that like old videos I was looking at there or are they like, oh, there's heap. So I, I released four in January and um, I did three part series on yeast propagator, which fucking blew up the internet because people think you can't propagate yeast and plastic. And um, and then I did one about dissolved oxygen and stuff like that. And, you know, I remember when I had come around to yours in like fucking 2017 or 2018 or something like that. Shot, shot down at HQ and yeah. And Kazi was in it and all that sort of thing. And, and um, uh, and it, over the years, like I've really neglected the YouTube channel and it's just such a great way to build trust with potential clients and that sort of thing. And I enjoyed doing it, but it's really hard to do because if you wanna make a 10 minute YouTube video, the amount of work you've gotta put in to write, um, shoot, edit, and publish. Yeah, that's a lot of work. It's a full-time job, I think if you to to be a content creator. Absolutely. Yeah. Um, but you can actually really that 10-minute YouTube video then just have a, a video like, There must be a huge opportunity in like TikTok and Instagram reels and like short form videos, correct? Yeah. And so one of the things I learned from, from this guy Adam, makes beer today, was like, um, he, he said, well, what I do is I'm just very natural. I do, I shoot some stuff in the, in the um, uh, in the, um, in the brewery. And then I'd do a live stream once a month where I just do a q and a and people just ask questions. Sometimes you get home brewers, sometimes you get professional brewers and then just chop out the video from that and share. And what he does, he chops out the bit. But the thing is, mate, is that I've got, and three and a half years of back catalog live coaching call content that's all recorded and can be repurposed and ready to go. And I'm thinking about are they one that that's, that's one to many. That's not like one-on-one is it? It's one to many, but it's basically people asking questions and me giving an answer off the cuff. I'm thinking you can pay someone to just rip all that out and turn it into Correct. Yes, yes. Yeah, exactly. So, and I've got like easily a hundred, hundred 50 hours of content there. And do you have like a path, like a, cuz you said on the o podcast we did the other day, you made like a fair bit of money out of the the um, ye propagator one. Did you, did you Yeah, look, it's um, you know, it the, the prop. Sorry, but actually I just remembered that wasn't a podcast, that was a chat after. Yeah, it was like the, the yeast propagator video series. It was actually something I kind of was interested in and I just went and released those three videos, um, you know, to make a Ye cuz no one had ever, cuz no one had created it. And I was like, fuck, you can just go and create a yeast propagator and save money on yeast and make better beer and stuff like that. And it was just something I was genuinely interested in. And I think that that sort of content is. I feel better about it because with professional brewers, they can really act like fucking home brewers sometimes. Mm-hmm. You come up with an I idea and, and you know, like the, the amount of hate that is propagated series came up with, or, or you know, generated people just going, oh, you can't propagate him plastic and ah, this won't work. That's fine. You have to skin, that's fine. Yeah. It just didn't bother me. I've spoken about that before and it's like, um, but the thing is right, is that there were so many more people who just went, fuck, this is really good. Oh, you've got a coaching program. It's like I just happen to be in the places where people happen to be looking. I don't tend to be on social media. It's a different fucking thing. Here we show that Perk acts primarily through Activating Transcription Factor-4 (Atf4) to stimulate not only pro-apoptotic but also pro-regenerative responses following optic nerve injury. Using conditional knockout mice, we find an extensive Perk/Atf4-dependent transcriptional response that includes canonical Atf4 target genes and modest contributions by C/ebp homologous protein (Chop). Overlap with c-Jun- dependent transcription suggests interplay with a parallel stress pathway that couples regenerative and apoptotic responses. Accordingly, neuronal knockout of Atf4 recapitulates the neuroprotection afforded by Perk deficiency, and Perk or Atf4 knockout impairs optic axon regeneration enabled by disrupting the tumor suppressor Pten. These findings contrast with the transcriptional and functional consequences reported for CRISPR targeting of Atf4 or Chop and reveal an integral role for Perk/Atf4 in coordinating neurodegenerative and regenerative responses to CNS axon injury. Abstract: Stiffened arteries are a pathology of atherosclerosis, hypertension, and coronary artery disease and a key risk factor for cardiovascular disease events. The increased stiffness of arteries triggers the hypermigration and hyperproliferation of vascular smooth muscle cells (VSMCs), leading to neointimal hyperplasia and accelerated neointima formation, but the mechanism of this trigger is not known. Our analyses of whole-transcriptome microarray data sets from mouse VSMCs cultured on stiff hydrogels simulating arterial pathology and from injured mouse femoral arteries revealed 80 genes that were differentially regulated (74 upregulated and 6 downregulated) relative to expression in control VSMCs cultured on soft hydrogels and in uninjured femoral arteries. A functional enrichment analysis revealed that these stiffness-sensitive genes are linked to cell cycle progression and proliferation. Furthermore, we found that survivin, a member of the inhibitor of apoptosis protein family, mediates stiffness-sensitive cell cycling and proliferation in vivo and in vitro as determined by gene network and pathway analyses, RT-qPCR, and immunoblotting. The stiffness signal is mechanotransduced via FAK and Rac signaling to regulate survivin expression, establishing a regulatory pathway for how the stiffness of the cellular microenvironment affects VSMC behaviors. Our findings indicate that survivin is necessary for VSMC cycling and proliferation and regulates stiffness-responsive phenotypes. Copy rights belong to original authors. Abstract: Vascular dysfunction is a common cause of cardiovascular diseases characterized by the narrowing and stiffening of arteries, such as atherosclerosis, restenosis, and hypertension. Arterial narrowing results from the aberrant proliferation of vascular smooth muscle cells (VSMCs) and their increased synthesis and deposition of extracellular matrix (ECM) proteins. These, in turn, are modulated by arterial stiffness, but the mechanism for this is not fully understood. We found that survivin (an inhibitor of apoptosis) is an important regulator of stiffness-mediated ECM synthesis and intracellular stiffness in VSMCs. Whole-transcriptome analysis and cell culture experiments showed that survivin expression is upregulated in injured femoral arteries in mice and in human VSMCs cultured on stiff fibronectin-coated hydrogels. Suppressed expression of survivin in human VSMCs and mouse embryonic fibroblasts decreased the stiffness-mediated expression of ECM components implicated in arterial stiffness, namely, collagen-I, fibronectin, and lysyl oxidase. By contrast, expression of these proteins was upregulated by the overexpression of survivin in human VSMCs cultured on soft hydrogels. Atomic force microscopy analysis showed that suppressed or enhanced expression of survivin decreases or increases intracellular stiffness, respectively. Antariksh Matters: Let’s Not Destroy Satellites in Peacetime — Aditya RamanathanIs it in India’s interests to support a ban on destructive anti-satellite tests? It’s a question Delhi may have to find an answer to in the near future. The trigger for this question is Russia’s ASAT missile test on 15 November 2021. The broader context is a series of moves that could eventually lead to substantial talks on space security. On 15 November, Russia apparently used an A-235 PL-19 “Nudol” Anti Ballistic Missile Interceptor to destroy a defunct Celina-D electronic intelligence satellite at an altitude of 480 kilometres. The collision unleashed 1,500 pieces of debris that could potentially threaten both satellites and inhabited craft such as the International Space Station and the Chinese space station Tiangong. A little over a month after the test, the UN General Assembly passed resolution 76/231 on 24 December, committing itself to convening an open-ended working group that will meet multiple times through 2022 and 2023 to discuss ways to reduce threats in space “through norms, rules and principles of responsible behaviours”. While the working group is likely to consider a range of space-related matters, destructive ASAT missile tests are likely to figure prominently. Partly this is because the resumption of destructive ASAT missile testing since 2007 has created growing concern about space debris. The other reason is that the idea of a ban on destructive tests appears to be gaining ground. A number of prominent authorities on space are calling for a ban. These include Takshashila’s own director, Nitin Pai, as well as scholars from the US-based Secure World Foundation (SWF), a researcher at the Stockholm International Peace Research Institute (SIPRI), and academics. The Return of ASAT MissilesDuring the Cold War, the USSR and the US considered banning ASAT missiles entirely as a class of weapons. These efforts culminated in negotiations in 1978-79 that eventually failed as both states prioritized nuclear arms control and as their relations began to sour over the Soviet invasion of Afghanistan. However, from December 1985, the US and the USSR effectively upheld a voluntary moratorium on destructive tests. The moratorium was shattered on 11 January 2007 by the most destructive kinetic ASAT test in history. On that day, China struck one of its own satellites with an SC-19 missile at an altitude of 865 kilometres. The test left behind at least 3,000 pieces of tracked debris along with perhaps 32,000 pieces of untracked flotsam. In 2008, an American sea-based SM-3 ballistic missile interceptor struck a malfunctioning satellite at an altitude of 370 kilometres. In 2019, an Indian Prithvi Delivery Vehicle Mark-II was tested against a target satellite at an altitude of 282 kilometres. This was followed, most recently, by the Russian test.While states have also continued to carry out both non-destructive tests of ASAT weapons and tests of missile interceptors, it is the destructive ASAT tests that have sparked the greatest concern because of the dangerous debris they generate. The Case for a BanThe upside of a ban is easy to see. Orbital debris from such tests can pose a serious threat to satellites and space stations, especially those in low Earth orbit (LEO). By eliminating this source of debris, spacefaring states make the-already crowded orbits safer for themselves and others. The second (and more uncertain) advantage of a ban is that it could slow down other states pursuing direct ascent ASAT capabilities. However, this advantage is, at best, notional, since ballistic missile defence systems capable of high altitude interceptions, can be easily repurposed into direct ascent ASAT missiles. The potential downside of a ban is that it might affect the development of India’s own ASAT capabilities. This downside is also more notional than real: having already unambiguously demonstrated its ability to intercept and destroy a satellite in LEO, India has no real need to perform destructive tests. While DRDO may consider it necessary to conduct more tests to validate India’s direct-ascent ASAT capabilities, these do not require actual kinetic interception. Indeed, the challenge of intercepting a long-range ballistic missile is far more challenging than that of intercepting an LEO satellite on a known trajectory. What about intercepting satellites at higher altitudes? Here, the limitations of kinetic ASAT weapons become evident. For one, striking satellites in high Earth orbit (HEO) requires purpose-built missiles of much greater range. Furthermore, such missiles would take so long to reach their targets that it would make it much easier for the adversary to take evasive action. Finally, creating debris fields at such altitudes would cause much greater and indiscriminate destruction, endangering India’s own satellites. At higher altitudes, India, like many other states, would be better off employing electromagnetic radiation to disrupt or destroy satellites. In summary, a ban on destructive testing would not constrain India’s own ASAT capabilities and would make the orbits safer for everyone’s satellites. Such a ban is also a low-hanging fruit that could open the door for further negotiations with other states on specific space weapons and space operations more generally. Click here to know moreCyberpolitik: Deplatforming or Unplatforming a country— Prateek WaghreUkraine’s appealsUkraine’s Vice Prime Minister and Minister of Digital Transformation Mykhailo Fedorov’s Twitter account has made appeals to various technology companies to act against Russia by stopping services, providing information, etc. I’ve compiled some of these in a work-in-progress mega note about internet and information ecosystem governance-related aspects of this situation [SochMuch]. Another set of significant appeals was made through letters to ICANN and RIPE NCC to [Pastebin, via Internet Governance Project]:Revoke, permanently or temporarily, the domains “.ru”, “.рф” and “.su”. This list is not exhaustive and may also include other domains issued in the Russian Federation.Contribute to the revoking for SSL certificates for the abovementioned domains.Shut down DNS root servers situated in the Russian Federation(to RIPE NCC) Withdraw the right to use all IPv4 and IPv6 addresses by all Russian members of RIPE NCC (LIRs - Local Internet Registries), and to block the DNS root servers that it is operating.The European Union, for its part, called for an EU-wide ban on Russian state media and announced that it was building tools the block their disinformation in Europe [Politico.EU]. However, there were questions around whether it was legally allowed to do so. We’ll get to some responses, but it is worth noting that these appeals cover large swathes of the internet stack from social media platforms, web services, IP intelligence services, CRM services to Internet Registries. And if you look at the dates, there is something of a pattern here. The appeals start at the user-facing end of the stack and then extend to the more infrastructural parts of it. Now, Ukraine is well within its right to make the appeals it thinks will protect its interests. How other countries, private corporations and people, in general, respond will set some precedents. Company ResponsesSince this is a developing space, I will not try to put down a comprehensive list, but the following resources should give you a sense (note that this is mainly for technology companies):A long thread by Anna Rogers.RestofWorld’s compilation (which does not look like it has been updated for a few days).Techmeme filtered for Russia and Ukraine (utility may drop as the news cycle shifts). Social media platforms started off with limiting ads/monetisation capabilities, more labelling/fact-checking - and, in response to the EU’s calls, restricted Russian state media in Europe. However, evelyn douek is right when she says there seems to be no normative framework. In this instance, social media platforms may have done what many wanted, and a number of interests aligned, as Rasmus Kleis Nielsen stated. But that doesn’t take away from the reality that these actions were arbitrary (note, I am not arguing at this stage, whether they were necessary or not).And as Mike Masnick points out - it won’t always be this way. Because precedent is wielded by whoever thinks they can take advantage of it. Or, if you assume that bad-faith actors will do certain things anyway (which is not an unreasonable assumption, to be fair), then also consider that they will use said precedent as a pretext or justification or weave it into their whataboutery. As Ben Thompson noted [Stratechery (potential paywall)]:Given this, Facebook being available in Russia seems like a net win, and, by the way, I would question exactly what effect banning Russian state media in the E.U. will actually have on this conflict; it seems clear that Russia is losing the battle of public opinion to a degree that no number of pro-Russia articles could undo. It doesn’t matter either way in the short term, but I do worry about the long-term: if Facebook is clearly following the government’s lead in the E.U., it is going to be difficult to see how the company stands up to other governments in the future, even if their requests are more problematic to the readers of this newsletter.Note that not all technology companies acted in response to direct appeals from Ukraine or demands/pressure from EU countries. Many acted of their own accord to stop operations, sales, software updates, close offices, etc., in Russia.Hammers and SplintersOne aspect common to Ukraine’s appeals and actions that were taken by various technology companies is that they move in the direction of effectively deplatforming Russia and Russia-based users. And while I can’t speak to the complete nature of support/opposition for the war against Ukraine among large sections of the Russian population and whether that should serve as justification for attempts to ‘cease the means of communication’ - one has to wonder about the long term ramifications of such actions.Mahsa Alimardani recounts that it can be counterproductive based on the Iranian experience. The unintended consequences of such actions are, in the short term, leaving domestic populations at the risk of greater control, exposure to lower quality information and propaganda, resulting in further isolation. And in the medium-long term, further splintering (perhaps even accelerating) of the internet as many sovereign states will want to avoid being in a similar situation. And this may not stay limited to the realm of the internet but extend to any ‘foreign’ firms. Some take solace in the fact that the internet, as we know it, today does not understand international boundaries. I would caution that it does not mean it never will. Here’s an extract from a conversation between Cloudflare’s CEO Mathew Prince and Ben Thompson [Stratechery (paywall)]Right. But given the nature of the internet, isn’t that the whole problem? Because, anyone in Germany can go to any website outside of Germany.MP: That’s the way it used to be, I’m not sure that’s going to be the way it’s going to be in the future. Because, there’s a lot of atoms under all these bits and there’s an ISP somewhere, or there’s a network provider somewhere that’s controlling how that flows and so I think that, that we have to follow the law in all the places that are around the world and then we have to hold governments responsible to the rule of law, which is transparency, consistency, accountability. And so, it’s not okay to just say something disappears from the internet, but it is okay to say due to German law it disappeared from the internet. And if you don’t like it, here’s who you complain to, or here’s who you kick out of office so you do whatever you do. And if we can hold that, we can let every country have their own rules inside of that, I think that’s what keeps us from slipping to the lowest common denominatorUnplatformingLet’s look at the Russian Government’s response in this limited context (over the last 5-6 days):It has throttled and reportedly blocked the likes of Facebook and Twitter as well as the websites of many western news outlets. There are also question marks over whether it will try to disconnect itself from the internet. As per Oleg Shakirov, that does not appear to be the plan as of now (stress on the as of now).It has updated its criminal code to add a possible 15-year prison term for spreading ‘fake news’ [Reuters].Lawmakers passed amendments to the criminal code making the spread of "fake" information an offence punishable with fines or jail terms. They also imposed fines for public calls for sanctions against Russia."If the fakes lead to serious consequences then imprisonment of up to 15 years threatens," the lower house of parliament, known as the Duma in Russian, said in a statement.In response to these changes, TikTok restricted live streams and new videos in Russia [BBC] and news outlets such as BBC, CNN, Bloomberg have suspended reporting from the country [Hindustan Times].There is a possibility that the Russian government may ‘unplatform’ itself and its citizens from the global internet before it gets deplatformed.An India-related subplot (but not the one you think, i.e. the U.N. votes)In a December 2021 edition of The Information Ecologist (54: Committee Reports), I was critical of certain aspects of the Parliamentary Standing Committee on Communications and Information Technology reports on:Suspension of Telecom Services/Internet and its impact relating to the Ministry of Communications (Department of Telecommunications)Ethical Standards in Media Coverage relating to the Ministry of Information and BroadcastingThere were suggestions to explore “banning of selective services, such as Facebook, WhatsApp, Telegram, etc.” in Report 1, and “develop some legal provisions to counter as big a challenge as fake news” in Report 2. Russia’s own anti-disinformation law, when passed in 2019, included a provision for a 15-day prison term in case of repeated offences [The Moscow Times]. The Russian Government’s actions over the last few months and recent responses tell us that once you have a hammer… Basically, be careful which hammer you give to whom. This also holds for the EU’s as-yet-unspecified plans for ‘anti-disinformation’ tools.Cyberpolitik #2 : The conflicts question for DCNs— Sapni G KThis is adapted from Sapni’s OpEd in the Hindu. Read the full piece here. Through the previous editions of this newsletter, we have discussed DCNs at length. Per our definition, social media platforms such as Facebook, Instagram, Twitter, etc., are examples of DCNs. As we study this space, the evolving conflict in the Russian aggression towards Ukraine raises more significant questions around DCNs. This time, predominantly civilian technology has become a realm of geopolitical show of power, as against the earlier instances where military power and natural resources were the most intertwined with conflict. In the absence of norms around how social media platforms should function as a technology and how these corporations should act, we continue to be a gray area. The challenges specific to the case now are not new. Armed conflicts within and between states have played out in cyberspace for years. It is no surprise then to see the same dynamics play out on social media platforms. The world outside Europe and North America has seen more than its share of conflicts, materialising and exacerbating the troubles of kinetic conflict through cyberspace. Social media platforms have gone by the mantra of “tech neutrality” to avoid taking decisions that may be considered political for too long. The years that have passed have seen an active ignoring of the concerns around social media platforms during a conflict. The lack of clear systems within social media companies that claim to connect the world is appalling. It is time that they should have learned from multiple instances, as recent as the Israeli use of force in Palestine.There was no unpredictability over conflicts in the information age spilling over to social media platforms. It did not even require pre-emption, since these have been recurrent events in the past decade. The international community and the liberal world order had to be proactive but failed to do so. We have missed the chance to have established a clear protocol on balancing the business interests of social media platforms and their intersection with global public life in critical situations. Though late, it would be valuable to have insights and clear frameworks to guide the behaviour of states and these corporations in cases of conflict, which will inevitably spill over to social media platforms in today’s information age. This space appears to be one where India could pursue a diplomatic course. Once these tensions abate, India could initiate conversations on setting norms on responses by social media platforms while conflicts are underway. It would be an opportunity to regain some of India's lost currency in the global order by attempting to establish a rule-based system in a fairly gray area. It is in our national interest and that of a rule-based global polity that social media platforms be dealt with more attention across spheres than with a range of reactionary measures addressing immediate concerns alone. Abstract: Our goal is to learn kinetic rates from single molecule FRET (smFRET) data even if these exceed the data acquisition rate. To achieve this, we develop a variant of our recently proposed hidden Markov jump process (HMJP) with which we learn transition kinetics from parallel measurements in donor and acceptor channels. Our HMJP generalizes the hidden Markov model (HMM) paradigm in two critical ways: (1) it deals with physical smFRET systems as they switch between conformational states in continuous time; (2) it estimates the transition rates between conformational states directly without having recourse to transition probabilities or assuming slow dynamics (as is necessary of the HMM). Our continuous time treatment learns transition kinetics and photon emission rates for dynamical regimes inaccessible to the HMM which treats system kinetics in discrete time. We validate the robustness of our framework on simulated data and demonstrate its performance on experimental data from FRET labeled Holliday junctions. Abstract: Lifetimes of chemical species is typically estimated, on a pixel-by-pixel basis by either fitting time correlated single photon counting (TCSPC) histograms or, more recently, through phasor analysis from time-resolved photon arrivals. While both methods yield lifetimes in a computationally efficient manner, they have limitations. First, both TCSPC and phasor analysis set the number of chemical species by hand before lifetimes can be determined. Yet the number of species itself is encoded in photon arrival times and need not be set by hand a priori. Next, even to determine lifetimes under the assumption of a known number of species, both methods rely on heavy data post-processing of the signal thereby requiring large amounts of data to retrieve lifetimes. As a result, the sample is exposed to orders of magnitude more light than is required and the ability to resolve fast processes is compromised. Here we propose a direct photo-by-photon analysis of data drawn from pulsed excitation experiments to infer, simultaneously and self-consistently, the number of species and their associated lifetimes from as little as a few thousand photons for two species. We do so by leveraging new mathematical tools within the Bayesian nonparametric (BNP) paradigm that we have previously exploited in the analysis of single photon arrivals from single spot confocal microscopy. We benchmark our method on simulated as well as experimental data for one, two, three, and four species with data sets from both immobilized and freely diffusing molecules. The “band” is back together to review some breaking news on the launch date for NASA’s upcoming Mars 2020 Perseverance Rover. We talk about the activities on the International Space Station, where NASA Astronauts Chris Cassidy performed a 6-hour 7-minute spacewalk to replace a set of lithium-ion batteries on the facility’s S6 truss. Completing this work will leave the ISS in an exemplary power configuration for the remainder of its operational life. Attention turns to a Pre-spacewalk briefing NASA’s Kenny Todd, and Steve Stich had good words on how well the SpaceX Crew Dragon is performing for its first-time on-orbit and information on when perhaps the Crew-1 mission could fly. Also included was a status on where Boeing was with remediation work on the CST-100 Starliner spacecraft. There was an abrupt “changing of the guard” at NASA’s Human Exploration and Operations office. Kathy Lueders, the Director of the Commercial Crew Program, was promoted to Associate Administrator for the Human Operations and Exploration Directorate replacing Douglas Loverro. He resigned under a ‘dark cloud’ for what he called ‘a mistake’ in his final letter to the HEO organization. That “mistake” is now under the microscope of the NASA Inspector General’s office. The NASA Headquarters Building in Washington DC has a new name; we tell you who it is and why that honor was bestowed, plus give you a little hint about another historical figure of note we’re going to discuss on a future show. Brian is an agency owner, author of Raise Your Voice, and podcast host with a background in design. In fact, Brian went to school for graphic design before entering the design field.His career evolved from there, though, and he eventually moved in a more marketing-oriented direction, became a solopreneur, brought others into his business, and moved into a mainly strategic role for his agency. The agency, called Aespire, works with businesses and entrepreneurs to improve their messaging, bringing design methods to bear on marketing. Brian's aim is to use words and other design elements to produce clear messaging.Steve and Brian discuss various aspects of the transition from working as a solopreneur to having a team. Brian talks about empowering both workers and business change, developing processes, determining who the company should serve (with either a niche or generalist character), and the reinvention process. Brian started out wanting to solve problems through design, but then specialized further and looked for a vertical sector to which he could specifically cater his work. This brought him to his current work, and his company as it stands now is most suited to companies with about 25 or 30 to about 1,000 employees; leaders of such companies tend to be approachable, want to see growth, and have much to gain from help with communication and the clarity of their message. Brian tries to help them to change their approach, find their voice, and think about their company in the long term. At the present time, as Steve points out, the long-term view necessitates taking into account the COVID-19 pandemic and resulting move of business toward digital platforms. He and Brian discuss their use and the future of such means. They also consider the delivery of value to customers, which Brian sees as a major focus of his 25-year-old company. Aespire helps businesses to unlock their full potential, works with a diverse assortment of brands, and tries to be of service even beyond profit to non-customers. Brian also sees the focus on providing value come into play in conversations with his clients; he works to help them determine their purpose, brand, and story, and so works to help them see their value. Illustrated with several grainy cockpit videos posted to the Times' website, the scoop was captivating. ABC News called it a “bombshell.” Brett Baier of Fox News said, “a lot of people are taking this revelation seriously.” A lot of journalists, that is. Many news consumers did lap up the story like a new X-Files episode but nobody ran for the hills, and nobody in Congress called for hearings about the “revelation” and its seemingly huge implications for civilization. As one observer on Twitter noted: “I mean the New York Times literally just gave us proof of UFOs and the world didn't skip a beat.” The latest installment documents other “close encounters” between Navy pilots and “unidentified aerial vehicles” that date back to 2013 and 2014. The takeaway from this story and the others has been unmistakable: Some kind of unknown, super-advanced form of aviation technology is encroaching on US military air space and outmaneuvering top-gun pilots. If true, this would appear to expose an alarming national defense vulnerability. To understand the Times' sustained interest in UFOs requires an appreciation of Elizondo's central role in its coverage. His willingness to resign from the Pentagon in 2017 and then reveal the existence and details of the UFO program he'd purportedly run, supplied the gravitas at the center of the initial Times account. Without him, it's likely the story would not have gotten so much traction. In appearances last year with Tucker Carlson, Elizondo said that he and the private company that now employs him were studying pieces of a possible extraterrestrial craft. Elizondo also told Carlson that he believed the US government was in possession of an actual UFO. In Episode 6 of Hidden Forces, host Demetri Kofinas speaks with Joan Johnson-Freese about space warfare and the weaponization of outer space. Joan Freese is a professor of National Security Affairs at the US Naval War College. She’s been a faculty member at the Asia-Pacific Center for Security Studies and the Air War College, which emphasizes the employment of air, space, and cyberspace in joint operations. Professor Freese has also served on the Space Studies Board of the National Academies of Science and has often testified before Congress on matters of space warfare and space security. She is the author of multiple books on space warfare and space security, among them, Heavenly Ambitions: America’s Quest to Dominate Space, The Chinese Space Program: A Mystery Within a Maze, and her latest book, Space Warfare in the 21st Century. In this episode, we go into outer space. We don’t just stay in the low earth orbit (LEO) of the international space station, but move all the way to high (HEO), geostationary orbits (GEO) more than twenty-six thousand miles (35,786 km) above the Earth’s equator, where some of our most valuable and vulnerable satellites operate. We look at what the United States, China, and Russia are doing in the area of space warfare. We look at what our militaries are doing to weaponization outer space. We learn about ASAT’s, Kinetic Orbital Strikes, and Kinetic Kill Vehicles. We learn that any significant use of anti-satellite technologies could create a wall of space debris around earth orbit so thick, that we would be unable to launch anything into space including satellites, our space station, and any space missions to Mars and beyond for generations. Any and all satellite communications would go dark. Global distribution networks – including our food supplies, energy, and transportation – would grind to a halt. The global banking and financial system would collapse. Our communication infrastructure would be devastated. The implications of space warfare are catastrophic, and yet, the public is largely unaware of the dangers orbiting right above our heads. It’s high time we take notice.
