C. Abstract: Consciousness is comprised of arousal (i.e., wakefulness) and awareness. Substantial progress has been made in mapping the cortical networks that modulate awareness in the human brain, but knowledge about the subcortical networks that sustain arousal is lacking. We integrated data from ex vivo diffusion MRI, immunohistochemistry, and in vivo 7 Tesla functional MRI to map the connectivity of a subcortical arousal network that we postulate sustains wakefulness in the resting, conscious human brain, analogous to the cortical default mode network (DMN) that is believed to sustain self-awareness. We identified nodes of the proposed default ascending arousal network (dAAN) in the brainstem, hypothalamus, thalamus, and basal forebrain by correlating ex vivo diffusion MRI with immunohistochemistry in three human brain specimens from neurologically normal individuals scanned at 600-750 micron resolution. We performed deterministic and probabilistic tractography analyses of the diffusion MRI data to map dAAN intra-network connections and dAAN-DMN internetwork connections. Using a newly developed network-based autopsy of the human brain that integrates ex vivo MRI and histopathology, we identified projection, association, and commissural pathways linking dAAN nodes with one another and with cortical DMN nodes, providing a structural architecture for the integration of arousal and awareness in human consciousness. We release the ex vivo diffusion MRI data, corresponding immunohistochemistry data, network-based autopsy methods, and a new brainstem dAAN atlas to support efforts to map the connectivity of human consciousness. R., Alzheimer's Disease Neuroimaging Initiative Abstract: INTRODUCTION: Discovery of the associations between brain structural connectivity and clinical and demographic variables can help to better understand the vulnerability and resilience of the brain architecture to neurodegenerative diseases and to discover biomarkers. METHODS: We used four diffusion-MRI databases, three related to Alzheimer's disease, to exploratorily correlate structural connections between 85 brain regions with non-MRI variables, while stringently correcting the significance values for multiple testing and ruling out spurious correlations via careful visual inspection. We repeated the analysis with brain connectivity augmented with multi-synaptic neural pathways. RESULTS: We found 34 and 37 significant relationships with direct and augmented connectivity, respectively, which were generally stronger for augmented connectivity. Age was consistently linked to decreased connectivity, and healthier clinical scores were generally linked to increased connectivity. DISCUSSION: Our findings help to elucidate which structural brain networks are affected in Alzheimer's disease and aging and highlight the importance of including indirect connections. In this episode of the Epigenetics Podcast, we caught up with Jane Mellor from the University of Oxford to talk about her work on H3K4me3, SET proteins, Isw1 and their role in transcription. Since the beginning of the century, Jane Mellor and her team have focused on H3K4 trimethylation and the factors that influence this mark. They discovered that H3K4me3 is an almost universal mark of the first nucleosome in every transcribed unit and all organisms. She could subsequently, together with the Kouzarides lab, identify SetD1, the enzyme that is responsible for writing this modification. Later on, the team characterized Isw1, a chromatin remodeler which “reads” H3K4me3. More recently the lab focuses on how the polymerase transcribes throughout the first nucleosomes of the transcribed region at the +2 nucleosome, with the help of Spt4. Eric Fischl was born in 1948 in New York City and grew up in the suburbs of Long Island. He began his art education in Phoenix, Arizona where his parents had moved in 1967. He attended Phoenix College and earned his B.F.A. from the California Institute for the Arts in 1972. He then spent some time in Chicago, where he worked as a guard at the Museum of Contemporary Art. In 1974, he moved to Halifax, Nova Scotia, to teach painting at the Nova Scotia College of Art and Design. Eric had his first solo show, curated by Bruce W. Ferguson, at Dalhousie Art Gallery in Nova Scotia in 1975 before relocating to New York City in 1978. Eric's paintings, sculptures, drawings and prints have been the subject of numerous solo and major group exhibitions and his work is represented in many museums, as well as prestigious private and corporate collections, including The Metropolitan Museum of Art, The Whitney Museum of American Art, The Museum of Modem Art in New York City, The Museum of Contemporary Art in Los Angeles, St. Louis Art Museum, Louisiana Museum of Art in Denmark, MusÈe Beaubourg in Paris, The Paine Weber Collection, and many others. Fischl has collaborated with other artists and authors, including E.L. Doctorow, Allen Ginsberg, Jamaica Kincaid, Jerry Saltz and Frederic Tuten. He is also the founder, President and lead curator for America: Now and Here. This multi-disciplinary exhibition of 150 of some of Americaís most celebrated visual artists, musicians, poets, playwrights, and filmmakers is designed to spark a national conversation about American identity through the arts. The project launched on May 5th, 2011 in Kansas City before traveling to Detroit and Chicago. Eric is a Fellow at both the American Academy of Arts and Letters and the American Academy of Arts and Science. He lives and works in Sag Harbor, NY with his wife, the painter April Gornik.

V., Greve, D., Fischl, B., Bjornerud, A., Walhovd, K. B., Fjell, A. M. Abstract: An accurate and reliable whole-brain segmentation is a key aspect of longitudinal neuroimaging studies. The ability to measure structural changes reliably is fundamental to detect confidently biological effects, especially when these affects are small. In this work, we undertake a thorough comparative analysis of reliability, bias, sensitivity to detect longitudinal change and diagnostic sensitivity to Alzheimer's disease of two subcortical segmentation methods, Automatic Segmentation (ASEG) and Sequence Adaptive Multimodal Segmentation (SAMSEG). These are provided by the recently released version 7.1 of the open-source neuroimaging package FreeSurfer, with ASEG being the default segmentation method. First, we use a large sample of participants (n = 1629) distributed across the lifespan (age range = 4-93 years) to assess the within-session test-retest reliability in eight bilateral subcortical structures: amygdala, caudate, hippocampus, lateral ventricles, nucleus accumbens, pallidum, putamen and thalamus. We performed the analyses separately for a sub-sample scanned on a 1.5T Siemens Avanto (n = 774) and a sub-sample scanned on a 3T Siemens Skyra (n = 855). The absolute symmetrized percent differences across the lifespan indicated relatively constant reliability trajectories across age except for the younger children in the Avanto dataset for ASEG. Although both methods showed high reliability (ICC > 0.95), SAMSEG yielded significantly lower volumetric differences between repeated measures for all subcortical segmentations (p < 0.05) and higher spatial overlap in all structures except putamen, which had significantly higher spatial overlap for ASEG. Second, we tested how well each method was able to detect neuroanatomic volumetric change using longitudinal follow up scans (n = 491 for Avanto and n = 245 for Skyra; interscan interval = 1-10 years). Both methods showed excellent ability to detect longitudinal change, but yielded age-trajectories with notable differences for most structures, including the hippocampus and the amygdala. For instance, ASEG hippocampal volumes showed a steady age-decline from subjects in their twenties, while SAMSEG hippocampal volumes were stable until their sixties. Finally, we tested sensitivity of each method to clinically relevant change. We compared annual rate of hippocampal atrophy in a group of cognitively normal older adults (n = 20), patients with mild cognitive impairment (n = 20) and patients with Alzheimer's disease (n = 20). SAMSEG was more sensitive to detect differences in atrophy between the groups, demonstrating ability to detect clinically relevant longitudinal changes. Both ASEG and SAMSEG were reliable and led to detection of within-person longitudinal change. This work proposes a novel algorithm to optimize the negative log likelihood of the spatial distribution of speckle. Specifically, the proposed method formulates a penalized negative log likelihood (P-NLL) cost function and proposes a majorize-minimize-based optimization method that removes speckle from OCT images. The optimization reduces to solving an iterative gradient descent problem. We demonstrate the usefulness of the proposed method by removing speckle in OCT images of uniform phantoms with varying scattering coefficients and human brain tissue. Abstract: Structural brain connectivity has been shown to be sensitive to the changes that the brain undergoes during Alzheimer's disease (AD) progression. In this work, we use our recently proposed structural connectivity quantification measure derived from diffusion MRI, which accounts for both direct and indirect pathways, to quantify brain connectivity in dementia. We analyze data from the ADNI-2 and OASIS-3 datasets to derive relevant information for the study of the changes that the brain undergoes in AD. We also compare these datasets to the HCP dataset, as a reference. Our analysis shows expected trends of mean conductance with respect to age and cognitive scores, significant age prediction values in aging data, and regional effects centered among subcortical regions, and cingulate and temporal cortices. Results indicate that the conductance measure has prediction potential, especially for age, that age and cognitive scores largely overlap, and that this measure could be used to study effects such as anti-correlation in structural connections. L., the TRACK-TBI Investigators Abstract: Diffusion tractography MRI can infer changes in network connectivity in patients with traumatic brain injury (TBI), but pathological substrates of disconnected tracts have not been well-defined due to a lack of high-resolution imaging with histopathological validation. We developed an ex vivo MRI protocol to analyze tract terminations at 750 micron resolution, followed by histopathologic evaluation of white matter pathology, and applied these methods to a 60-year-old man who died 26 days after TBI. Analysis of 74 cerebral hemispheric white matter regions revealed a heterogeneous distribution of tract disruptions. Associated histopathology identified variable white matter injury with patchy deposition of amyloid precursor protein and loss of neurofilament-positive axonal processes, myelin dissolution, astrogliosis, microgliosis, and perivascular hemosiderin-laden macrophages. Multiple linear regression revealed that tract disruption strongly correlated with neurofilament loss. Around the globe, April Gornik is known as one of the world’s most celebrated landscape painters, whose name is often sprinkled into conversations, and on museum walls, with the likes of Hockney, Turner, and Monet. But around Sag Harbor, she’s also known as Wonder Woman. Okay, maybe not exactly. But North Haven resident Gornik is involved in myriad causes to protect the environmental and cultural aspects of the area she and her husband, the artist Eric Fischl, choose to call home. She cofounded the Sag Harbor Partnership, which currently is the platform for the renovation of the Sag Harbor Cinema, she and Fischl have preserved acres of wetlands, the Eastville Historical Society, and there is much more. She even came up with a way to fish for sharks without hurting them. For real. Gornik has been the recipient of Guild Hall’s Lifetime Achievement Award in 2003. But back to Sag Harbor. Gornik is passionate about the history of the area – and not just the environmental and artistic history, but the industrial history as well.
In 2001, artist Eric Fischl created a sculpture in response to the 9/11 attacks called, “Tumbling Woman.” The piece initially caused controversy when it was unveiled. Fischl talks about the New York art scene in the 1970s, the process of creating “Tumbling Woman,” and how it felt to see it on display for the first time at the 9/11 Memorial & Museum years after he first created the piece. Join one of the nation’s most prolific researchers on HIV/AIDS to learn more about the journey medical professionals have taken to treat this disease. For more than 20 years Margaret Fischl has been part of the team that built the University of Miami AIDS Clinical Research Unit, the first National Institutes of Health-funded AIDS research center in Florida, a state with the highest number of diagnosed HIV infections. Fischl will share her work as an international expert in designing and conducting clinical studies that have led to groundbreaking treatments for HIV and rapid, dramatic reductions in the number of HIV-related deaths. She will also touch upon the challenges that lie ahead as researchers push to identify a vaccine to prevent HIV infection.
Jack Fischl and Kyle Wiggins studied across the Charles River from each other in Boston, but they didn’t meet until they both became Peace Corps volunteers in Panama. Even then, they were placed in two communities that were a 14-hour bus ride apart. So how did they build a successful Latin American travel marketplace together? It started with a simple Wordpress site they created over several visits to their local internet cafes. After realizing their communities had no way of marketing the unique tours they were offering, and that local tour guides were being ripped off by large corporations, Jack and Kyle came up with Keteka. In this episode, Jack and Kyle explain what they learned from going through Start-Up Chile and the Booking.com Accelerator program, raising a funding round through Latin American angel investors on FounderList, and receiving investment from more traditional VCs like my firm Magma Partners. But it all started with the lessons they learned in the Peace Corps. Tapping into Panama’s tourism dollars: a “dignified economic opportunity” If Jack and Kyle’s communities from the Peace Corps had anything in common, it was that both looked to tourism as a potential economic opportunity for their area. However, traditional tourism would not come easily in a place with no electricity or running water, let alone WiFi and payments systems. Kyle and Jack realized there was a need to bridge the gap between these rural communities, their skilled local tour guides, and a tourism market that operates through online transactions. From this epiphany came Keteka, originally a Wordpress website built in an internet cafe in Panama. When a friend emailed Jack the link to apply for Start-Up Chile, he almost didn’t believe the opportunity existed. Focus on the business first. Fancy tech can always wait until you have validated with real customers. How to fundraise from Latin America Split between Chicago and Santiago, Keteka is already an unconventional company. Their funding rounds, which have included several equity-free grants, as well as syndicate angel investment, are off-the-beaten-path, as well. Andrew Fischl didn’t start fencing until his junior year of high school but achieved a lot very quickly: he was 5th in the country in 2013. Before fencing, Andrew had tried many sports: soccer, tennis, baseball, lacrosse, etc but none of these sports really engaged him the way fencing did. The reason most people know him is because of the videos he creates. Going by the name Cyrus of Chaos (his handle in Super Smash Brothers) his YouTube channel currently houses over 2500 videos of international-level competitions. He started recording bouts at his first Div 1 NAC in Columbus in 2007, when he, in his words, “was a wide-eyed little fencer who got destroyed in my pool leaving me the rest of the day to walk around and watch. I remember seeing so many things that made me think "oh I should be doing it like this" or “I want to try the timing of that next time I am at practice". Then the next day I couldn’t specifically remember anything.” He took his parent’s videocamera to the next tournament and began recording everything he could. He still does that to this day. Andrew’s biggest regret is that he can’t make it to every international tournament to capture footage. Daniel Fischl didn't find working in laboratories particularly appealing, and whilst completing a PhD in Plant Molecular Genetics at UC Davis (California) he was introduced to viticulture and was seduced with what lay outside. It also helped that the viticulture and winemaking students seemed to have more fun. Fast forward many years of experience consulting as a viticulturist and agricultural scientist, he and his winemaker wife Michelle started the Linnaea Vineyards project with a Napa Valley Cabernet Sauvignon. As he got work consulting in vineyards around the world he and Michelle discovered more terroirs and made more wines, across a number of continents and expressions.

Currently, Fischl is working as curator and founder of America: Now and Here, a cross-country traveling exhibition of multi-disciplinary works reflecting differing viewpoints and hopes for America.
