To date, measuring aspartate concentrations in live cells was deemed impossible. We present iAspSnFR, a genetically-encoded biosensor for intracellular aspartate, engineered by displaying and screening biosensor libraries in HEK293 cells. In live cells, iAspSnFR exhibits a dynamic range of 130% fluorescence change and detects reduced aspartate levels upon glutamine deprivation or glutaminase inhibition. Furthermore, iAspSnFR tracks aspartate uptake by excitatory amino acid transporters, or of asparagine after co-expression of an asparaginase. Importantly, iAspSnFR reports aspartate depletions upon electron transport chain inhibition, and therefore it can serve as a proxy for mitochondrial respiration. Consequently, iAspSnFR can dissect the major cellular pathways of aspartate production, offering immediate applications, particularly in cancer biology, such as screening small molecules targeting aspartate and glutamine metabolism. They claim it "began as an icy planetesimal that was irradiated at low temperatures by cosmic rays during its interstellar journey, and experienced warming during its passage through the Solar System. This explanation is supported by a large body of experimental work." But Avi Loeb disagrees: “In a new paper published recently in Nature -- https://www.nature.com/articles/s41586-022-05687-w Acceleration of 1I/‘Oumuamua from radiolytically produced H2 in H2O ice -- Jennifer Bergner and Darryl Seligman suggest that the peculiar acceleration observed by the first known interstellar object `Oumuamua, can be explained if `Oumuamua was made of water ice which was partly dissociated into hydrogen by cosmic-rays along its interstellar journey. In the abstract of the paper, the authors admit that past models involving a pure hydrogen iceberg or a pure nitrogen iceberg do not work due to theoretical or observational inconsistencies. In fact, the hydrogen iceberg model was proposed in a previous 2020 paper by Darryl Seligman himself. Grace Cathedral, San Francisco, CA 2D14 1 Lent (Year A) 11:00 a.m. Eucharist Sunday 26 February 2023 Genesis 2:15-17, 3:1-7 Psalm 32 Romans 5:12-19 Matthew 4:1-11 Often I start with a question but today I want to finish with one. Sixty years ago President John F. Kennedy, full of confidence in modern science, made two predictions. First, that within a decade a human being would walk on the moon. And second, that science would, “make the remote reaches of the human mind accessible.” At that time few people might have guessed that traveling 239,000 miles through oxygen-less -455 degree Fahrenheit outer space would be far, far easier than discovering psychiatric cures for mental illness. In David Bergner's book The Mind and the Moon, he shares the story of his younger brother Bob who was diagnosed with bi-polar at age twenty-one, institutionalized and medicated with drugs that had debilitating side-effects. Bergner also follows Caroline a woman from Indiana who started hearing voices as a child and whose drug treatments, starting in elementary school, led to obesity and losing control of her forearms and hands. He introduces us to a civil rights lawyer named David whose severe depression during the Trump administration could not be mitigated by either the drugs his doctor prescribed or the psychedelics he turned to afterwards. Bergner points out how little we understand the mind. He writes about the damage that can be caused by a psychological diagnosis (which puts us in a kind of box and separates us from other people) and of many drug treatments which have questionable efficacy. Forty million American adults and millions more children are on psychiatric drugs. In one ten year period the number of children diagnosed with bi-polar increased fortyfold. Thirty to forty percent of our students are treated with psychiatric medication at some point in their college years. I'm not trying to make a point about how we treat mental illness. I just want to remind us how much we are a mystery to ourselves. Perhaps the saddest part of Bergner's book for me came when he quoted the prominent neuroscientist Eric Nestler. Nestler said that unquestionably fewer Americans should be on psychotropics for depression and anxiety. He went on, “Exercise. Better sleep. Mindfulness. The belief in something bigger than yourself. Religion if you are religious.” Nestler said, “People with religious beliefs benefit greatly from them.” He wondered if they fostered a, “capacity to bring order and meaning into one's life.” And then the sad part. He said that religion was not part of his own life. He tragically explained, “The thing about religion is, I can't know whether Jesus is the Son of God or whether Allah rose to heaven on a winged horse. Those are not scientifically knowable.” This is an insurmountable barrier for him, and many others. Theology has failed our generation when ordinary people think that they have to believe something contrary to science in order to be religious. Christians like us have a lot of work to do in explaining this to the people around us. The stakes are high. On Tuesday one of our daughters' friends took his own life only months before graduating from college. From his social media posts you would never have realized that there was anything wrong. Did he understand that he is a child of God? There is no way for us to know. This morning, the first Sunday in Lent, we have before us central biblical texts: the temptation of Jesus in the wilderness and the Garden of Eden. How do these ancient stories help modern people to understand God? How do we interpret them? What meaning should they have in our life? For Matthew wilderness is equivocal. On the one hand it has no structure and is void. On the other hand it represents limitless possibility, a context for encountering God. In ancient scripture the number forty represents a long time. This connects Jesus with other figures who persevered over time. My friend Matt Boulton proposes an alternative to the way we usually interpret this story. For him it is not about a hero bravely resisting temptations to comfort, security and glory with admirable self-control. It is not about the devil offering something that Jesus deeply desires and Jesus gritting his teeth and responding like someone on a diet knowing that he should not have another plate of cookies. This is not about sacrifice but about trust. Similarly the story of Eden's forbidden fruit is not primarily about sin and disobedience or temptation, but rather about fear, and the failure to trust. Let's begin by seeing the connection between Jesus' temptation in the wilderness and Moses.

Und Karlfried Bergner nutzt dafür die Kraft aus der Mitte der Organisation. Er kooperiert mit Graswurzelbewegungen, die es, wie wir lernen, auch in seiner Behörde gibt. Und sorgt mit einem Multiplikatorenprogramm für die Verbreitung frischer Ideen für agile Zusammenarbeit im Haus. Der Blick auf eine Behörde wie das Auswärtige Amt hat mindestens zwei Perspektiven. Die eine Perspektive zeigt eine Behörde klassischen Zuschnitts mit traditionellem Aufbau und festgelegten Entscheidungs- und Kommunikationsstrukturen. Die andere Perspektive fällt allerdings auch auf ein Haus, dessen Mitarbeiter im Ausland mit Unsicherheit und permanentem Wandel umgehen müssen, die unter bestimmten Umständen schnell handeln müssen auch ohne direkten Draht nach Berlin und ohne Ansage "von oben". In diesem Spannungsfeld widmet sich Karlfried Bergner der Frage, wie neue Formen der Zusammenarbeit im Auswärtigen Amt etabliert werden können. "Die Frage ist, ob die Rahmenbedingungen im öffentlichen Dienst eigentlich Agilität zulassen" erklärt Bergner und stellt fest, dass aus seiner Sicht bei dem, was derzeit an neuen Arbeitsformen auch im öffentlichen Dienst probiert wird, noch viel Spielraum gibt: "Wir sind noch weit weg von irgendwelchen roten Linien", erzählt Bergner. Und schnell sind wir uns einig: Auch in Behörden sind die Freiräume oft viel größer, als die Mitarbeiter denken - oft dienen vermeintliche Grenzen als Ausrede, das Neue nicht anzupacken. "Man kann schon noch eine ganze Menge ausprobieren", so Bergner. Um diese Zusammenarbeit in der Behörde in das 21. Jahrhundert zu holen, geht es im Projekt "Agiles Arbeiten in der Auslandsvertretung" um die Verbesserung der Zusammenarbeit der Zentrale mit den Auslandsvertretungen. Über die Kooperation mit "DigitalServices4Germany" kommen in den Fellowship-Programmen "Tech4Germany" und "Work4Germany" junge Studien-Absolventen in das Auswärtige Amt, die zu einer verbesserten Digitalisierung im öffentlichen Dienst beitragen wollen. Gerade dieTeilnahme in agilen Workshops mit den jungen Fellows inspirierte Bergner, sich tiefer mit dem Thema neue Zusammenarbeit auseinanderzusetzen. Ein zweiter Auslöser führt Bergner dann zu einer weiteren Säule für die Etablierung neuer Arbeitweisen: Die Corona Krise stellt das Auswärtige Amt wie viele andere Unternehmen vor die Herausforderung: Wie bewältigen wir die Umstellung auf mobiles Arbeiten? In dieser Zeit stößt Bergner eine selbstorganisierte Gruppe, eine Graswurzel in der Behörde: Sie nennen sich "Friends of Digi", und sie sind den Kollegen bei der Kenntnis und Nutzung digitaler Technologien um Längen voraus. Im ersten Schritt helfen sie bei der Dokumentation neuer Lösungen für die Remote Worker, und bieten Hilfe für Kollegen im Umgang mit digitalen Werkzeugen von Zoom über Webex bis hin zu Messenger Diensten. Sie probieren gemeinsam Tools wie Slack als Gruppenchat oder Mural und Miro als Whiteboard aus - alles Lösungen, die im Kernbereich des Auswärtigen Amts sicher nicht zugelassen werden würden, aber zum einen vielleicht die Peripherie stärken könnten und zum anderen erstmals für ein Grundverständnis sorgen, wie Zusammenarbeit heute mit zeitgemäßen Werkzeugen organisiert werden kann. Denn einer der größten Schmerzen ist, so Bergner, das Fehlen einer Plattform für digitale Zusammenarbeit. Die Art und Weise, wie er mit den jüngeren Kollegen schnell das Auswärtige Amt in einer Krisensituation unterstützen konnte, beeindruckt ihn nachhaltig. "Veränderungen geht nicht nur Top Down oder Bottom up, es braucht beides".

That sounds like deep euphemism to me, but it's worth asking what this vague and sinister term actually means before asking whether it exists. Jeffrey Bergner, author of The Vanishing Congress, is a long-time Washington insider who has found the roots of something like a deep state in the legislative branch's abdication of its one job: to make the laws. This might sound like a dream come true for libertarians, but since nature abhors a vacuum, this vital function ends up getting usurped by unelected bureaucrats and unaccountable district judges.The Guest - Jeffrey Bergner served as Assistant Secretary of State for Legislative Affairs from 2005 until July 2008. He is the President and Managing Financial Partner of Bergner Bockorny, Inc., as well as an adjunct professor at the National Security Studies Program at Georgetown University.The Problem - Bergner says that Congress has ceded too much of its authority to the executive branch — with its mutant bureaucratic army — and broken down the checks and balances that the Founders designed to ensure that government represents the people and not its own entrenched interests. When the executive branch is given such broad leverage to implement laws without express authorization of Congress, presidents and executive agencies are encouraged to legislate by fiat. Then come the executive orders…The Solution - For starters, Bergner suggests that Congress would be 20% more efficient with 20% fewer staff. I'm conflicted, since I like the idea of shrinking government, but I'm not so sure about increasing congressional “efficiency.” Is getting more stuff done always a good thing? I'll question Bergner on this idea, as well as his proposal to reduce debate on cabinet nominations from 30 hours to 2 hours, and how direct election of senators forever changed the political landscape in America.
