After an unplanned summer break, Scoreography returns to cover the 2024-2025 season, starting with the Challenger Series. We take a look at all the major competitors we've seen so far, specifically from the Nebelhorn and Lombardia Trophy competitions, as well as Cranberry Cup and other small events. Welcome back, Scoreogra-fans! (too much?) It's a new dawn, it's a new day, it's a new SEASON of figure skating — and we're so happy to be back! It was a long summer of far too much adult-ing, and we've missed so much in the world of figure skating. From the ISU's legalization of the backflip (or somersault) in competition to the 2022 Olympic teams from the USA and Japan FINALLY getting their medals at the 2024 games in Paris, we're definitely a bit behind. However, with a new competitive season already in progress, we're moving forward and talking about what we've already seen at early season Challenger Series events including Lombardia Trophy, Nebelhorn Trophy, and Cranberry Cup. Since its still quite early and most skater's programs aren't fully “baked” yet, in this episode we break down which skaters have competed so far by discipline — and dig into who's already looking strong. Women's Division The women of Team USA have come out hot already with some big moments for reigning U.S. Champion, Amber Glenn, and well as two young skaters who are starting to make their mark internationally: Elyce Lin-Gracey and Sarah Everhardt. Amber threw down a personal best short program at Lombardia Trophy — which included a gorgeous triple axel — and skated well enough in the free to earn her first gold at a Challenger Series event. Elyse Lin-Gracey was consistently gaining ground last season, but she looks like she's found her flow and looked incredible at Nebelhorn Trophy. She scored a personal best of 213.33 overall and made the skating world sit up and take notice. Also bringing the heat is Sarah Everhart, who looked fantastic at last year's U.S. Nationals where she landed in 4th overall. She started her season with a victory at Cranberry Cup over the likes of reigning World Silver medalist, Isabeau Levito and Elyse Lin-Gracey. Isabeau Levito, for her part, has perhaps not had the flawless programs and scores she would have liked so far — with a bronze at Cranberry Cup and a silver at Nebelhorn Trophy. However, we are not too concerned, as she's already shown a new short program that taps into an elegant, Audrey Hepburn vibe that looks like a winner to us. Plus, her free skate at Nebelhorn showed significant improvement from Cranberry Cup. We think these are just early competitions and that she looks to be right on track. Team Japan is already on the board with medals as well, with reigning 3-time world champion, Kaori Sakamoto, taking the bronze at Lombardia Trophy with a safe but solid competition. Again, her programs look stellar and we're chalking up the lower-than-usual difficulty and scores to the fact that it's a brand new season. At Nebelhorn Trophy, we saw her Japanese teammate and last season's rookie of the year, Hana Yoshida, take bronze and include triple axels in both her short and long programs. The short program saw a fall on that triple axel, but she scored a nice one in her excellent free skate. Pairs Division Nebelhorn Trophy provided us a fantastic early-season rematch between two of last season's top teams: World Champions Deanna Stelatto-Dudek & Maxime Deschamps and World Bronze medalists, Minerva Hase & Nikita Volodin. Canada's Deanna & Maxime had a great new short program, but drew the short straw, going first at the competition, so were scored lower than they would have been had they skated later. Their new free skate has incredible potential to be one of their best, but a few issues on their side-by-side jumps held them solidly in the silver medal position. Germany's Hase & Volodin looked nearly ready for Worlds in terms of their technical. After having to make some relatively last-minute changes to their music in the offseason, they looked poised and polished — claiming their first victory of this new season. If this represents the promise of what's to come, they will make it an exciting Grand Prix series and beyond. Also looking strong already are Sarah Conti & Niccolo Macii from Italy, who took the gold at Lombardia Trophy with two much better skates than most of what we saw from them last season. It was great to see a solid comeback for this team — and will hopefully lead to more positive outcomes. Reigning world silver medalists, Japan's Riku Miura & Ryuchi Kihara, had a stunning short program at Lombardia Trophy. However, their new long program—while looking like it has the potential to be a gem—saw a scary fall in a lift resulting in what appeared to be at least a minor injury for Ryuchi. Their coach, Bruno Marcotte, later said Ryuchi would recover in a week or two and that no major damage was done, thankfully. Also looking strong so far are Hungary's Maria Pavlova & Alexei Sviatchenko, who were third at Lombardia Trophy, and the U.S.'s Ellie Kam & Danny O'Shea, who had a stunning personal best short program at Nebelhorn Trophy and earned the bronze medal there. Men's Division We were NOT prepared for the Lombardia Trophy rematch of the two top men in the world from last season, Ilia Malinin and Yuma Kagiyama. In what had to be the most exciting moments of the season so far, these two men lit up the arena in Italy — showing programs that look both like they have tons of room to improve and also like they've been skating them for years. Reigning world champ and quad god, Ilia Malinin, has brought some of his exhibition style to his short program with an aggressive music choice and program. His free skate to “I'm Not a Vampire” may be his strongest yet. He did not perform the quad axel here or push to the limits he can on the technical side, but his victory was marked instead by more intensity and connection to the music than we've seen from him before. He's showing growth, and it's exciting to see. Yuma Kagiyama brought his finesse, his edges, his jumps, and his completeness as a skater to Lomabardia and looked absolutely divine as he took silver. As he did last season, his difficulty was somewhat dialed down here (its early season!), but that did not take away from the quality of his programs. In particular, his short program is a masterpiece that — once at its full power — will have audiences in the palm of his hand even more than they already are. A joy! Not too far behind the two of these men was Japan's Shun Sato, who looked better than ever after an off season where he obviously spent time working on his choreography and connection to the music. Worth noting is his score of 285.88, which got him a bronze here but would have taken gold over his countryman, Sota Yamamoto, at Nebelhorn Trophy. Yamamoto had a rough short program at Nebelhorn, but his exceptional free skate took him to victory. Right behind his was a strong performance from Gabriele Frangipani from Italy and an artistic dream from Deniss Vasiļjevs of Latvia. Ice Dance Division Of all the disciplines so far, we've seen the fewest of the top contenders in dance. Reigning world bronze medalists, Guignard & Fabbri of Italy, did make their debut with a fresh and fun rhythm dance. Their free skate seemed a bit less successful, but their scores were still magnificent and took them to an easy gold medal at Lombardia Trophy. The team nipping at their heels all last season, Lilah Fear & Lewis Gibson of Great Britain, looked amazing in their disco-themed rhythm dance. However, their free dance to a Beyonce medley seemed a little more safe than we've seen from them in past seasons. Still, their scores easily took them to victory at Nebelhorn Trophy and sets them up well as the season evolves. Our personal favorites so far this season have to be the two American teams of Christina Carreira & Anthony Ponomarenko, whose free dance to Carmen may well be their best to date, and Emilea Zingas & Vadym Kolesnik. Zingas & Kolesnik looked like a different team in their free dance, showing a new and captivating side to their skating. Just in time for Halloween comes Mimi's Tales of Terror, nine frightful tales illustrated by the manga maestro himself, Junji Ito, and adapted from the original Japanese urban tales collected by Hirokatsu Kihara and Ichiro Nakayama's Shin Mimibukuro (New Earmuffs), a book that's impossible for Japanese fans to find, let alone English readers. A version of Ito's adaptation was originally published back in 2002 as Mimi no Kaidan Kanzenban (Mimi's Ghost Stories), though like Kihara and Nakayama's original, good luck finding a copy these days.

Abstract: Protein-bound ceramides, specialized ceramides covalently bound to corneocyte surface proteins, are essential for skin permeability barrier function. However, their exact structure and target amino acid residues are unknown. Here, we found that epoxy-enone (EE) ceramides, precursors of protein-bound ceramides, as well as their synthetic analog, formed stable conjugates only with Cys among nucleophilic amino acids. NMR spectroscopy revealed that the beta-carbon of the enone was attached by the thiol group of Cys via a Michael addition reaction. We confirmed the presence of Cys-bound EE ceramides in mouse epidermis by mass spectrometry analysis of protease-digested epidermis samples. EE-ceramides were reversibly released from protein-bound ceramides via sulfoxide elimination. We found that protein-bound ceramides with reversible release properties accounted for approximately 60% of total protein-bound ceramides, indicating that Cys-bound EE ceramides are the predominant protein-bound ceramides. Our findings provide clues to the molecular mechanism of skin barrier formation by protein-bound ceramides. Copy rights belong to original authors. Abstract: Maternal behaviours, which are crucial for the survival of mammalian infants, can be learned. How the efficient acquisition of these behaviours is implemented at the neural circuitry level remains poorly understood. Although prevalent models of neural circuits for parental behaviours often assume the frontal cortical network as an integrator of infant-related sensory signals and a controller of decision-making and motivation, these ideas have not been functionally tested. As such, detailed input/output neural circuit organizations of the frontal cortex in relation to parental behaviours remain unknown. Here we show that the orbitofrontal cortex (OFC) promotes efficient learning of maternal behaviours in virgin female mice when they are co-housed with lactating mothers. Chronic microendoscopy in freely behaving animals reveals robust representations of pup-directed anticipatory activities and ongoing sequential motions of pup retrieval that are innately sculpted and largely unaffected by learning. Through viral tracing and manipulations, we functionally identify the submedius thalamus as a prominent presynaptic partner of the OFC that shapes pup-related representations. Optogenetic inactivation of OFC reduces the pup retrieval-related activities of midbrain dopamine neurons that promote maternal behaviours. Collectively, these findings reveal a higher-order cognitive network that connects innately formed, pup-related integrated representation to the top-down control of motivation centers, thus enabling efficient maternal behavioural learning. Makoto Rheault-Kihara leads user acquisition at Hopper, one of the fastest-growing travel apps with over 60 million in sales to date. Makoto was previously the Director of Marketing at Cruisehub, an OTA aiming to modernize the cruise industry and lead search engine marketing at Busbud, a bus travel booking platform in 80 countries. Hopper differentiates its paid social strategy by focusing on user acquisition AND long-term trust through investing in paid social and other mobile ad channels. They use Facebook as the primary focus of their paid social ads with TikTok as a growing secondary channel. Paid social is becoming a lot more abstract and therefore more creative. Makoto believes paid TikTok ads must be kept fresh, platform-focused, and generated at a higher frequency. The pandemic did affect the travel industry but Hopper still achieved a 110% YoY growth last year and is on track to do over $1B of business this year. They did this by following a diversification strategy based on customer needs and expanded into hotels and car rentals. Hopper's customer-obsessed culture and self-contained, autonomous teams have helped them grow multiple product arms simultaneously. A weekly catch-up call and an executive team focused on cross-team communication help these siloed teams at Hopper learn from each other's challenges and successes. Sometimes ads that marketers like don't appeal to actual customers and may end up performing worse than some simple customer-oriented ads. Over time, Hopper moved to these customer-centric ad creatives which helped them reduce expensive failures and triple their RPM (revenue per 1000 impressions). At Hopper, Makoto focuses on maximizing the overall contribution margin of a cohort and on-target rate to check their ability to drive revenue from specific target purchases or user actions. Because Hopper diversified across three generic categories (hotels, flights, car rentals), their ad targeting still focuses on great creatives and has not changed much since IDFA deprecation in iOS 14.5. Known the world over as one of the pioneering forces in the field of Tribal Fusion belly dance, Sharon Kihara is regarded as one of the most seasoned and well-traveled artists in the genre, and is known for her avante-garde artistic sensibility that defies replication. She is both a dance and yoga therapy teacher, 200 RYT certified by Yoga Alliance.In late 2003, at the behest of friend Rachel Brice, Sharon was scouted to perform with the pioneering dance company Bellydance Superstars, created by music mogul and svengali Miles Copeland. Sharon spent over five years on tour as Tribal director, soloist, and choreographer. After leaving the tour to pursue new dreams, she was recruited by Jillina to dance as a principal and choreographer in the unique and critically acclaimed new touring production, Bellydance Evolution. It'll be a year later than planned, and the artist might not be able to attend to install her work or meet the international guests. But Yuki Kihara will be the first Pasifika artist to represent Aotearoa at the 2022 Venice Biennale. Paradise Camp is the name of her work, with themes including climate change and small island ecologies, queer rights, and giving Fa'afafine the attention and respect they have struggled to gain. Yuki has incorporated video filmed in Samoa with a large local cast and crew, as well as photography and archival research. Paradise Camp has a prized location in Venice. It's the Arsenale, one of the two main Biennale hubs. The Arts Council of New Zealand Toi Aotearoa supports our Biennale artists financially and logistically. But they'll have to make big changes because of Covid-19 restrictions. Chair of the Arts Council, Caren Rangi, is the Commissioner of New Zealand's presentation at the Biennale. She tells Lynn Freeman they won't be sending attendants to Venice. Instead they'll use locally-employed people to install and work with visitors on site. Head of User Acquisition @ Hopper: Makoto Rheault-Kihara- Strategic Paid Search Marketing, Massive Growth & The Culture That Empowers It EPISODE SUMMARY: Makoto Rheault-Kihara leads user acquisition at Hopper, one of the fastest-growing travel apps with over 60 million in sales to date. Makoto was previously the Director of Marketing at Cruisehub, an OTA aiming to modernize the cruise industry and lead search engine marketing at Busbud, a bus travel booking platform in 80 countries. Hopper differentiates its paid marketing strategy by focusing on user acquisition AND long-term trust through investing in paid social and other mobile ad channels. They use Facebook as the primary focus of their paid social ads with TikTok as a growing secondary channel. Paid search marketing is becoming a lot more abstract and therefore more creative. Makoto believes paid TikTok ads must be kept fresh, platform-focused, and generated at a higher frequency. The pandemic did affect the travel industry but Hopper still achieved a 110% YoY growth last year and is on track to do over $1B of business this year. They did this by following a diversification strategy based on customer needs and expanded into hotels and car rentals. Hopper's customer-obsessed culture and self-contained, autonomous teams have helped them grow multiple product arms simultaneously. A weekly catch-up call and an executive team focused on cross-team communication help these siloed teams at Hopper learn from each other's challenges and successes. Sometimes ads that marketers like don't appeal to actual customers and may end up performing worse than some simple customer-oriented ads. Over time, Hopper moved to these customer-centric ad creatives which helped them reduce expensive failures and triple their RPM (revenue per 1000 impressions). At Hopper, Makoto focuses on maximizing the overall contribution margin of a cohort and on-target rate to check their ability to drive revenue from specific target purchases or user actions. Because Hopper diversified across three generic categories (hotels, flights, car rentals), their ad targeting still focuses on great creatives and has not changed much since IDFA deprecation in iOS 14.5. Abstract: An increasing number of density maps of macromolecular structures, including proteins and protein and DNA/RNA complexes, have been determined by cryo-electron microscopy (cryo-EM). Although lately maps at a near-atomic resolution are routinely reported, there are still substantial fractions of maps determined at intermediate or low resolutions, where extracting structure information is not trivial. Here, we report a new computational method, Emap2sec+, which identifies DNA or RNA as well as the secondary structures of proteins in cryo-EM maps of 5 to 10 Angstrom resolution. Emap2sec+ employs the deep Residual convolutional neural network. Emap2sec+ assigns structural labels with associated probabilities at each voxel in a cryo-EM map, which will help structure modeling in an EM map. Emap2sec+ showed stable and high assignment accuracy for nucleotides in low resolution maps and improved performance for protein secondary structure assignments than its earlier version when tested on simulated and experimental maps. Yuki is renowned for delving into the complexities of postcolonial histories in the Pacific and interrogating Western misinterpretations from the perspective of Fa’afafine (Samoan for ‘in the manner of a woman’ broadly understood as the LGBTIQ+ in the Western context) community which she belongs to in Sāmoa. May be due to bone mineral decreases or the failure of the microarchitecture. 2:54 Causes: At menopause, the cells that destroy bone become more active than the cells that build bone. These are both natural processes but the balance is disturbed as the sex hormones decrease In the later years of life 80+ the muscles and the bones become more fragile. Without the muscles pulling on the bones (and as they lose strength, this happens naturally), the bones also become weaker Illness and drugs can also affect the bone density. 5:30 No symptoms there is a test called the DEXA test that measures bone density (but not bone microarchitecture) It tests three sites on the body: hip, vertebrae and wrist. Each site receives it’s own score. Normal Score; men and women naturally lose bone density in their fifties but the loss is not enough to increase the risk of fracture Osteopenia: there is bone loss in excess of normal bone loss and the risk of fracture is increased Osteoporosis: there is significant bone loss and the risk of fracture is high. Vertebral fractures can also lead to kyphosis which is 1) a risk factor for more vertebral fractures, it impacts breathing and balance. 9:59 Exercise, especially weight-bearing exercise, is recommended to move the muscles that then pull on the bones to signal the need for increased density. Axial Skeleton – skull, vertebrae, ribs. Protects the core organs and gives structure to the trunk. Women lose more bone (as a percentage) from the axial skeleton than men do so they have an increased risk of vertebral fractures. Need strong core muscles to protect the bones and to help with holding up the trunk. 11:35 Dr. Mersheed Sinaki, has done several studies on osteoporosis at the Mayo Clinic in the US. She recommends more than just weight-bearing exercise but also need to improve a) flexibility, b) core strength, c) cardiovascular fitness, d) axial strength, and e) gait steadiness (to prevent falls from loss of balance). 13:00 Yoga poses to strengthen the axial skeleton (primarily the vertebrae). Laying down: Gentle locust; laying on the stomach, lift the head (in line with the spine) and then release. After building strength with the previous pose, can then transition to clasping the hands behind the back, lifting the head and chest and pulling with the hands towards the feet. Another gentle pose would be to lay on back and lift head and then release, Standing: With one arm on a chair if needed, lift one arm above the head with the inhale and let it come down with the exhale. Do the other arm. With sufficient strength, can use weights or lift both arms up at the same time (with or without weights). Sitting: Bring arms to shoulder height; bend elbows so hand are pointing towards the ceiling and try to press the elbows together and then release. If the student cannot bring the arms to shoulder height, then place the arms along the side and bend the elbows and then press the elbows and release. 16:45 Appendicular Skeleton: limbs, hip and shoulder girdles. Breaks in hip are often at the neck of the femur which causes a fall. Hip fractures are highly correlated with death in older individuals so a hip fracture is quite dangerous. Wrist fractures often occur when trying to stop a fall. And falls increase fracture risk. Rosemarie Chen affectionately known as Pele, is the owner and operator of Naturally Healing Sanctuary a well-being company in Plantation, FL.
