Abstract: REM sleep has been hypothesized to promote emotional resilience, but any neuronal circuits mediating this have not been identified. We find that in mice, somatostatin (Som) neurons in the entopeduncular nucleus (EPSom)/internal globus pallidus are predominantly active selectively during REM sleep. This unique REM activity is necessary and sufficient for maintaining normal REM sleep. Inhibiting or exciting EPSom neurons reduced or increased REM sleep duration, respectively. Activation of the sole downstream target of EPSom neurons, Vglut2 cells in the lateral habenula (LHb), increased sleep via the ventral tegmental area (VTA). A simple chemogenetic scheme to periodically inhibit the LHb over 4 days selectively removed a significant amount of cumulative REM sleep. Chronic REM reduction correlated with mice becoming anxious and more sensitive to aversive stimuli. Therefore, we suggest that REM sleep, in part generated by the EP-LHb-VTA circuit identified here, could contribute to stabilizing reactions to habitual aversive stimuli. Abstract: Chronic stress and chronic pain are two major predisposing factors to trigger depression. Enhanced excitatory input to the lateral habenula (LHb) has been implicated in pathophysiology of depression. However, the contribution of inhibitory transmission remains elusive. Here, we dissect an inhibitory projection from sensory thalamic reticular nucleus (sTRN) to LHb, which is activated by acute aversive stimuli. Repeated activation of the LHb-projecting sTRN neurons prevents CRS-induced depression onset and hyperexcitability of LHb neurons. Moreover, acute inhibition of sTRN-LHb circuit induces depressive-like behaviors in healthy mice, while enhancing this circuit relieves depression induced by both chronic stress and chronic pain. Intriguingly, both neuropathic pain and comorbid pain in chronic stress are unaffected by this pathway. Finally, we dissected sTRN upstream brain regions associated with stress. Together, our study demonstrates an inhibitory sTRN-LHb circuit in establishing and modulating depression, thus shedding light on potential therapeutic targets for preventing or managing depression. Abstract: Glutamatergic projection neurons of the lateral habenula (LHb) drive behavioral state modulation by regulating the activity of midbrain monoaminergic neurons. Identifying circuit mechanisms that modulate LHb output is of interest for understanding control of motivated behaviors. A small population of neurons within the medial subnucleus of the mouse LHb express the GABAergic synthesizing enzyme GAD2, and they can inhibit nearby LHb projection neurons; however, these neurons lack markers of classic inhibitory interneurons and they co-express the vesicular glutamate transporter VGLUT2. To determine the molecular phenotype of these neurons, we genetically tagged the nuclei of GAD2-positive cells and used fluorescence-activated nuclear sorting to isolate and enrich these nuclei for single nuclear RNA sequencing (FANS-snRNAseq). Our data confirm that GAD2+/VGLUT2+ neurons intrinsic to the LHb co-express markers of both glutamatergic and GABAergic transmission and that they are transcriptionally distinct from either GABAergic interneurons or habenular glutamatergic neurons. We identify gene expression programs within these cells that show sex-specific differences in expression and that are implicated in major depressive disorder (MDD), which has been linked to LHb hyperactivity. Finally, we identify the Ntng2 gene encoding the cell adhesion protein Netrin-G2 as a marker of LHb GAD2+/VGLUT+ neurons and a gene product that may contribute to their target projections. These data show the value of using genetic enrichment of rare cell types for transcriptome studies, and they advance understanding of the molecular composition of a functionally important class of GAD2+ neurons in the LHb. Abstract: Appropriate evaluation of risk is essential for survival in complex, uncertain environments. Confronted with choosing between a certain (safe) and an uncertain (risky) option, animals of various species show strong preferential traits which are stable across extended periods of time1-8. How such risk preference is encoded in the neural circuitry of the brain remains poorly understood. A candidate brain region is the lateral habenula (LHb), which has been shown to be prominently involved in various value-guided behaviors9-12. Here, using a balanced two-alternative choice task involving risk of loss, we find that neuronal activity in the LHb prior to action selection reflects risk preference and is governed by distinct inputs from hypothalamic subregions. Specifically, by employing multi-fiber photometry and targeted optogenetic perturbations, we identified glutamatergic LHb projections from both lateral and medial hypothalamus (LH/MH) that provide functionally distinct synaptic inputs to the LHb before action selection. Microendoscopic two-photon calcium imaging revealed risk-preference-selective LHb neurons that decreased their selectivity upon chemogenetic silencing of MH but not LH inputs. Finally, optogenetic stimulation of MH- greater than LHb axons evoked both excitatory and inhibitory postsynaptic responses in LHb neurons (indicative of glutamate/GABA co-release for fine-tuned gain control13), whereas LH- greater than LHb projections were purely excitatory. Our results thus reveal functionally distinct hypothalamus-habenula circuits that govern risk preference in situations of economic decision-making. Copy rights belong to original authors. Abstract: Chronic social stress in mice causes behavioral and physiological changes resulting in perturbed rhythms of body temperature, activity and sleep-wake cycle. To further understand the link between mood disorders and temperature rhythmicity, we measured core body temperature (Tcore) in mice before and after exposure to the chronic social defeat stress (CSDS) paradigm. Our results revealed that Tcore amplitudes of stress resilient and susceptible mice are dampened during exposure to CSDS. However, after exposure to social stress, the temperature amplitude of resilient mice recovered faster than the susceptible mice. There were minimal changes in locomotor activity after stress exposure which correlates with regular rhythmic expression of Prok2, an output signal of the SCN (Suprachiasmatic nucleus). We also investigated the molecular changes associated with these dampened temperature rhythms, and determined the expression levels of thermosensitive genes Cirbp, Rbm3 and Hsf1. Expression of Rbm3 and Cirbp in the LHb (Lateral habenula) were blunted 1 day after CSDS. These molecular rhythms recovered 10 days later such that daytime expression was higher compared to nighttime, similar to stress-naive controls. Abstract: The available treatments for depression have substantial limitations, including low response rates and substantial lag time before a response is achieved. We applied deep brain stimulation (DBS) to the lateral habenula (LHb) of two rat models of depression (Wistar Kyoto rats and lipopolysaccharide-treated rats) and observed an immediate (within seconds to minutes) alleviation of depressive-like symptoms with a high response rate. Simultaneous functional magnetic resonance imaging (fMRI) conducted on the same sets of depressive rats used in behavioral tests revealed DBS-induced activation of multiple regions in afferent and efferent circuitry of the LHb. The activation levels of brain regions connected to the medial LHb (M-LHb) was correlated with the extent of behavioral improvements. Rats with more medial stimulation sites in the LHb exhibited greater antidepressant effects than those with more lateral stimulation sites. These results indicated that the antidromic activation of the limbic system and orthodromic activation of the monoaminergic systems connected to the M-LHb played a critical role in the rapid antidepressant effects of LHb-DBS. This study indicates that M-LHb-DBS might act as a valuable, rapid-acting antidepressant therapeutic strategy for treatment-resistant depression and demonstrates the potential of using fMRI activation of specific brain regions as biomarkers to predict and evaluate antidepressant efficacy. Abstract: Excitatory projections from the lateral hypothalamic area (LHA) to the lateral habenula (LHb) drive aversive responses. We used Patch-seq guided multimodal classification to define the structural and functional heterogeneity of the LHA- LHb pathway. Our classification identified six glutamatergic neuron types with unique electrophysiological properties, molecular profiles, and projection patterns. We found that genetically-defined LHA-LHb neurons signal distinct aspects of emotional or naturalistic behaviors: Esr1+ LHA-LHb neurons induce aversion, whereas Npy+ LHA-LHb neurons control rearing behavior. Repeated optogenetic drive of Esr1+ LHA-LHb neurons induces a behaviorally persistent aversive state, and large-scale recordings showed a region-specific neural representation of the aversive state in the prelimbic region of the prefrontal cortex. We further found that exposure to unpredictable mild shocks induced a sex- specific sensitivity to develop a stress state in female mice, which was associated with a specific shift in the intrinsic properties of bursting-type Esr1+ LHA-LHb neurons. In summary, we describe the diversity of LHA-LHb neuron types, and provide evidence for the role of Esr1+ neurons in aversion and sexually dimorphic stress sensitivity. Abstract: Serotonin (5-HT) neurons in the dorsal raphe nucleus (DRN) receive a diverse constellation of long-range synaptic inputs, yet unifying principles of local circuitry and its dynamics are largely unknown - a crucial component of understanding how 5-HT output controls behavior. Here, we developed a formalism bridging optogenetic, electrophysiological, computational and behavioral strategies to reveal how the dynamics of local circuitry in DRN control the expression of reward associations. Using long-range input from lateral habenula (LHb) to interrogate functional DRN circuitry, we uncover 5-HT1A receptor-mediated local recurrent connections between 5-HT neurons, refuting classical theories of autoinhibition by 5-HT1A receptors. These inhibitory 5-HT connections were slow, stochastic, strongly facilitating, and gated spike output of 5-HT neurons. Targeted physiology and modeling approaches revealed that these functional connectivity features collectively support the emergence of a paradoxical excitation-driven inhibition in response to high frequency LHb activation, and of a winner-take-all computation over protracted timescales. In vivo, we found that optogenetic activation of LHb inputs to DRN transiently disrupted expression of a reward-conditioned response in an auditory conditioning task. In accordance with quantitative model predictions, this disruption occurred exclusively at the conjunction of high frequency LHb activation and high predicted reward value, and was not due to a modulation of the underlying reward association. Thus, we propose that recurrent dynamics in the DRN support a contextual value computation, where stable learned associations are integrated with dynamic environmental inputs to support sharp behavioral state transitions in changing environments. In het kader van Rotterdam Pride Week gaan we het hebben over transgenders en de reis naar hun transitie. Wij willen alle taboes rondom transgenders verbreken. Waarom is het zo belangrijk dat wij dit bespreekbaar maken?Transgenders hebben vaak te maken met ongelijke behandeling. In theorie hebben transgenders dezelfde rechten als een niet-transgender, maar in de praktijk is dat vaak anders. Zo wordt 43% van de transgenderjongeren die naar school gaan slachtoffer van verbaal geweld. 1/5 van de transgenderjongeren wordt slachtoffer van lichamelijk geweld. Zo werd laatst de 14-jarige Fréderique in elkaar geslagen toen ze niet wilde vertellen of ze een jongen of meisje is. Ervaringsdeskundige Iris Kay In gesprek met ervaringsdeskundigeOnze chick Izzy die zelf in transitie is gaat in gesprek met Iris Kay. Iris is een ervaringsdeskundige op het gebied van transitie en gender. Iris was enkele jaren activist en organiseert nog steeds verschillende activiteiten voor de LHB en Trans gemeenschap. Ze ontving begin dit jaar een prijs van de Stichting Jillis Bruggeman voor het oprichten van een zelforganisatie LHBTI Nissewaard. Landscape architect Lydia Major recently received a career achievement award from the Minnesota Chapter of the American Society of Landscape Architects. Major is LHB's landscape architecture and planning studio leader, where she oversees a team of mainly women in the firm's Minneapolis and Duluth offices. Her leadership and mentorship of this team — which is unique in the industry — helped Major attain the award, she says. Major began at LHB in 2007, after a former co-worker asked her to join the Minneapolis-based firm. She started her career at Westwood Professional Services after graduating with a master of landscape architecture from the University of Minnesota and a bachelor of arts in English from the University of Wisconsin-Madison. “I sometimes feel like I use my English major more just in daily communication and how we kind of describe [design] to people,” she says. Her work has spanned public, residential and commercial spaces, including the Promenade of Wayzata, the Minnehaha Creek Watershed District Comprehensive Master Plan Document and the Central Riverfront Regional Park Master Plan. In this episode, Major speaks with Reporter Kelly Busche about her career, and how over the past year, she has seen the public's expectations for public spaces like sidewalks, streets and parks, change due to the pandemic and social justice transformation.

In deze aflevering praat ik met LHBTI's over depressie. Die afkorting staat voor lesbische vrouwen, homoseksuele mannen, biseksuelen, transgender- en intersekse personen. Bijna de helft van de Nederlandse LHBTI-volwassenen heeft ooit zelfmoordgedachten gehad, ruim 5x meer dan de gemiddelde bevolking. Is het dan zo slecht gesteld met de acceptatie in Nederland? Wat betekent het om je steeds een ander te voelen? En hoe leidt dat gevoel tot depressies? Ik praat erover met Laura Baams, onderzoeker en docent aan de Rijksuniversiteit Groningen en met Liza Sonneveld, kind- en jeugdpsychiater bij de Care-Express, Opvoedpoli Amsterdam. Daarnaast spreek ik met ervaringsdeskundige Kris van der Veen. Hij groeide op in een klein dorpje in Friesland en werd daar jarenlang gepest. Ze scholden hem al uit voor 'flikker', toen hij nog niet eens wist wat dat betekende. Op een dag, tijdens de gymles, liep het helemaal uit de hand. Hij stond voor zijn gevoel toen voor de keuze: naar de decaan gaan om zijn verhaal te vertellen of een eind aan zijn leven maken. Hij koos het eerste, de school ging aan zijn kant staan en hielp hem weer perspectief te zien voor zijn toekomst. Abstract: Survival depends on the ability to adaptively react or execute actions based on previous aversive salient experiences. Although lateral habenula (LHb) activity has been broadly implicated in the regulation of aversively motivated responses, it is not clear under which conditions this brain structure is necessary to regulate defensive responses to a threat. To address this issue, we combined pharmacological inactivations with behavioral tasks that involve aversive and appetitive events and evaluated defensive responses in rats. We found that LHb pharmacological inactivation did not affect cued threat and extinction learning and memory, anxiety-like or reward-seeking behaviors. Surprisingly, we found that LHb inactivation abolished reactive defensive responses (tone-elicited freezing) only when threat (conditioning) and safety memories (extinction and latent inhibition) compete. Consistently, we found that LHb inactivation impaired active defensive responses (platform-mediated avoidance), thereby biasing choice behavior (between avoiding a threat or approaching food) towards reward-seeking responses. Together, our findings suggest that LHb activity mediates defensive responses only when guided by competing threat and safety memories, consequently revealing a previously uncharacterized role for LHb in experience-dependent emotional conflict. Recently, bright light stimulation (BLS) was shown effective in non-seasonal depression but its mode of action remains elusive. We demonstrate here, using a new mouse model of depression resistant to ADs including ketamine, that chemogenetic activation of lateral habenula (LHb) astroglia prevented the potentiating effect of BLS on the AD response. Additionally, the beneficial action of BLS was associated with upregulation of a specific part of the prefrontal cortex opioid system. Abstract: The lateral habenula (LHb) is an epithalamic brain region associated with value-based decision making and stress evasion through its modulation of dopamine (DA)-mediated reward circuitry. Specifically, increased activity of the LHb is associated with drug addiction, schizophrenia and stress-related disorders such as depression, anxiety and posttraumatic stress disorder. Dynorphin (Dyn)/Kappa opioid receptor (KOR) signaling is a mediator of stress response in reward circuitry. Previously, we have shown that maternal deprivation (MD), a severe early life stress, increases LHb intrinsic excitability while blunting the response of LHb neurons to extra hypothalamic corticotropin-releasing factor (CRF) signaling, another stress mediator. CRF pathways also interact with Dyn/KOR signaling. Surprisingly, there has been little study of direct KOR regulation of the LHb despite its distinct role in stress, reward and aversion processing. To test the functional role of Dyn-KOR signaling in the LHb, we utilized ex-vivo electrophysiology combined with pharmacological tools in rat LHb slices. We show that activation of KORs by a KOR agonist (U50,488) exerts differential effects on the excitability of two distinct sub-populations of LHb neurons that differ in their expression of hyperpolarization-activated cation currents (HCN, Ih). Specifically, KOR stimulation increases neuronal excitability in LHb neurons with large Ih currents (Ih+) while decreases neuronal excitability in small/negative Ih (Ih-) neurons. Additionally, we found that an intact fast-synaptic transmission is required for the effects of U50,488 on the excitability of both Ih- and Ih+ LHb neuronal subpopulations. Consistently, KOR activation also altered both glutamatergic and GABAergic synaptic transmission. While stimulation of presynaptic KORs uniformly suppressed glutamate release onto LHb neurons, we found that U50, 488 either increased or decreased GABA release. We also found that MD significantly increased immunolabeled Dyn (the endogenous KOR agonist) labeling in neuronal fibers in LHb while significantly decreased mRNA levels of KORs in LHb tissues compared to those from non-maternally deprived (non-MD) control rats. While total p38 MAPK (a downstream signaling pathway driven by KOR activation) expression was elevated in the LHb of MD rats compared to non-MD controls, we found that application of KOR-specific agonist, U50,488, onto LHb slices was still able to alter phosphorylated p38 MAPK (ph-p38) expression in MD rats similar to non-MD controls. Moreover, we found that the U50,488-mediated increase in LHb neuronal firing observed in non-MD rats was absent following MD. Altogether, this is the first demonstration of the existence of the functional Dyn/KOR signaling in the LHb that can be modulated in response to severe early life stressors such as MD. However, there is little understanding of how the information provided by the LHb influences learning and memory processes. Fear conditioning (FC) is probably the best understood model of aversive associative learning. Thus, FC constitutes a good behavioral paradigm to analyze LHb function in associative learning. In the present work we studied in rats the effect of interfering with neuronal activity of the LHb on FC using optogenetics and pharmacological tools. We found that exciting or inhibiting the LHb during training, abolishes independent expression of contextual and cued memories, yet memory is normally expressed when the cue is played in the training context. However, we recently observed that optogenetic STN excitation induced a strong jumping/escaping behavior. We hypothesized that STN activation is aversive. To test this, place preference was assessed. Optogenetic excitation of the STN caused potent place aversion. Causality between STN activation and aversion has not been demonstrated previously. The lateral habenula (LHb) is a critical hub for aversion. Optogenetic stimulation of the STN indeed caused firing of LHb neurons, but with delay, suggesting the involvement of a polysynaptic circuit. To unravel a putative pathway, the ventral pallidum (VP) was investigated. VP receives projections from the STN and in turn projects to the LHb. Optogenetic excitation of STN-VP terminals caused firing of VP neurons and induced aversive behavior. Abstract: The lateral habenula (LHb) integrates critical information regarding aversive stimuli that shapes decision making and behavioral responses. The three major LHb outputs innervate dorsal raphe nucleus (DRN), ventral tegmental area (VTA), and the rostromedial tegmental nucleus (RMTg). LHb neurons that project to these targets are segregated and nonoverlapping, and this led us to consider whether they have distinct molecular phenotypes and adaptations to stress exposure. In order to capture a time-locked profile of gene expression after repeated forced swim stress, we used intersectional expression of RiboTag in rat LHb neurons and next-gen RNA sequencing to interrogate the RNAs actively undergoing translation from each of these pathways. The "translatome" in the neurons comprising these pathways was similar at baseline, but diverged after stress, especially in the neurons projecting to the RMTg. Using weighted gene co-expression network analysis, we found one module comprising genes downregulated after stress in the RMTg-projecting LHb neurons; there was an overrepresentation of genes associated with phosphoinositide 3 kinase (PI3K) signaling in this module. Reduced PI3K signaling in RMTg-projecting LHb neurons may be a compensatory adaptation that alters the functional balance of LHb outputs to GABAergic vs. monoaminergic neurons following repeated stress exposure. Abstract: The lateral habenula (LHb) inhibits midbrain monoaminergic neurons, thereby regulating emotion/cognition. Abnormally high activity in the LHb causes behavioral disorders, but how stressful experiences affect neuronal circuits underlying emotion remains poorly understood. Here, we report the effects of chronic stress on the LHb in postnatal day (P)1-9, P10-20, and P36-45 mice in the pre-, early, and late stages of LHb maturation. At P60, only mice exposed during P10-20 exhibited LHb-specific changes: abnormally high-stress reactivity shown by the expression of the immediate-early gene product (Zif268/Egr1) with insufficient number of parvalbumin (PV) neurons containing GABA. Furthermore, these mice showed anxiety/depression-like behaviors in the light-dark box test/ forced swim test. Thus, experiences in early-life are essential for the maturation of neuronal circuits underlying emotion. Early-life stress is thought to have caused anxiety/depression in adulthood by disrupting the maturation of inhibitory PV neurons in the LHb in a period-specific manner. MIDCENTURY FOOTBALL (starts at 1:00) From T-Formations to the Korean War and replacing rosters of war heroes with regular ol' sophomores, we look at football as it stood in 1950. Southern football is on the map, and programs are spending insane (for that time) amounts of G.I. Bill money on building projects, especially for their football teams. Recruiting is a thing, but not at Michigan. New facilities are a thing, but not at Michigan. Revamped passing games, War babies, bowl games, the new Coaches Poll, and your fists have to be in on your chest now. 2. THE TEAM THE TEAM THE TEAM (starts at 20:41) BACKS: Bill “ol' One-Play" Putich for his 1 TD pass vs MSU. Not a great QB but smart. Was later moved to HB and was captain in 1951. VERY young--he turned 18 this season, as a junior. Real concern that Army was going to draft him ahead of the game. FB: Don Dufek: Chicago. Dad of Don, Bill, and Joe. Team MVP. They know him at Grand Valley as the AD who turned around the department. 702 yards on 174 carries (4 YPC) and 8 TDs--really he was averaging 5 YPC except that OSU game when he had 14 carries for 9 yards. LHB Chuck Ortmann: Star in ‘49 until he was concussed against Army. Hampered by back injury in ‘50 (could hardly bend over). RHB Leo Koceski: Was more of a slot role, see interview. Backups: HB Don Peterson (5’10/175) charging for Leo's job from the start. LHB: Super sophomores: “mighty mites” Frank Howell (5’8/155) (also 3rd string LHB), Wes Bradford (5’6/155) and Don Oldham (5’8/166). Oldham in for Ortmann but awful fumble.  Also Dan Hill who was compared to the shifty return specialist Gene Derricotte. QB: Ted Topor, blocking type, converted fullback. Bill Billings (Flint), passer, go-to signal caller, good punter. Tom Witherspoon is a defensive specialist. ENDS: Sophs expected to take over. Nervous about this position though there’s hope for Perry. LE Lowell Perry (Ypsilanti, teammate of Hill), Fastest guy on the team, end-around maven, quick passing games, and makes guys miss. Also an excellent safety. RE Harry Allis: Longtime starter, stalwart. LE on defense is Ozzie Clark who's also a good TE, a big quasi-tackle. Big game against Ohio State. Backups: Sophs Fred Pickard, a combo TE, and Merrit Green on defense. LINE: Offensive C Carl Kreager. 6’4/220. Won’t wear gloves. Backup: John Padjen 5’9/180, plays C and G. Started football program at TF South in Chicago, his son Tom was HC for 40 years, and grandson John is now HC. RT Al “Brick” Wahl. Star of the team, captain, All-American. Nephew of Herb Stager. He came to Michigan in 1945 from Chicago but joined the Army that summer and missed the ‘46 and ‘47 seasons so he’s now 23 years old and a vet. LT Tom “Big Gun” Johnson born in Chicago but from Muskegon heights. Washed dishes, competed in football and track. Was Alvin Wistert’s backup but by the time he graduated he was one of the greatest tackles in Michigan history (more on defense than offense). Second African-American to play for the Packers, after former teammate Bob Mann a year prior. LT Bill Ohlenroth: Plays when Johnson needs a breather. Defensive RG Tom Kelsey (a converted end) and later soph Jim Wolter are playing a hybrid DL/LB position. Swing OL John Hess Sprained ankle vs Minneosta. OL Ralph Stribe moved to G this year because T was strong. Defensive LG Dick McWilliams: best MG in the country. 6’3/248 and immovable. LINEBACKERS: C Roger Zatkoff (Hamtramck), super sophomore, Meyer Morton Award, has to replace Dick Kempthorn “a 215-pound converted fullback, is a bone-crusher of the Kempthorn school. FB Tony Momsen, moved over from center, punted, blocked OSU punt and recovered it. Backups: Ted Topor (6’0/215) the blocking fullback, and Dick Strowzewski, who also plays T. 3. INTERVIEW WITH HALFBACK LEO KOCESKI (starts at 38:10) Recruiting in the late '40s, the training table rules, coming up under the Mad Magicians, Fritz Crisler's retirement, the Korean War, Dan Dworsky on the piano. The time he tried kicking. The '49 team and the comedown as the next wave takes over. 4. THE GAMES, THE GAMES, THE GAMES (starts at 1:17:33) Toughest season you can get, with most of the Top 10 on the docket, starting with #9 Michigan State College, "The Football Factory" which had recently been admitted to the Big Ten and immediately placed on some kind of double-secret probation, but beat us . Dartmouth was a good team in '49 but lost a lot of guys, Michigan wins 27-7. #2 Army at Yankee Stadium began the cascade of injuries, beat Michigan 6-27. #16 Wisconsin was a nice homecoming game after the 1-2 start, a 26-13 win. But then a trip to #14 Minnesota ends in a 7-7 tie with Michigan routinely stuck on the doorstep, and an 0-7 loss to #10 Illinois in the snow is a season-killer. Or is it? Indiana goes away 20-7, and Michigan defeats a tough Northwestern team 34-23, the only giant not to lose to them. And then there's the snow bowl.   5. ROSE BOWL (1:51:01) Michigan takes the train to Pasadena and beats #5 Cal. Why yes of course Sap has video of it!   Cal controls play in the first half but has their first huge run called back for an illegal formation, and misses an extra point try. Michigan figures out the trick to a team that cut blocks all the time: step back a yard at the snap then go over them. Michigan also learns the short passing game and takes it down the field to deep in Bear territory. Dufek took over from there. 6. We talked about everything from bee haulers to bringing back respect in the trucking industry. The bandits are on a mission to better the trucking industry by helping educate those that have not learned about respect. One driver at a time they induct them into the LHB or the Long Haul Bandits Trucker Club. Entrance is not just a signature and you're in. The gate keepers are Sancho and Grump, the Prez and VP. They take the Bandits very serious and they are on guard. If you want to be in the club there are a few things you will have to do and have. The have of course if the "desire". You must really want to be a part of this growing group of truck drivers that want to do things the right way and make the brotherhood of trucking a thing of respect. The second thing you will need to have is a great attitude. You will want to be positive because thats what the industry is in desperate need of. Your Tasks 75 Chrome Shop & The Long Haul Bandits. Your assignment? Just picture "mission impossible" music playing as you're reading this! Your assignment? Help people. Thats right you must record yourself twice a month helping another trucker either back in a door or maybe helping him or her disconnect a trailer but it must be recorded. The second thing you will need to be recorded doing is (Music just got louder Haha) pick up garbage for 15 minutes at a truck stop twice a month. Dent Dent Dent da dent dent dent dent (Music is in dents lol! But seriously this is what you must do to earn your stripes into the club. If your attitude says this sounds dumb then this group is not for you. Think about a new future and is this what you want to be a part of and then move forward. Long Haul Banditz Long Haul Banditz interview. We sat down with the President Sancho Gonzalez and VP Grumpy of the Long Haul Banitz to chat. Who are the Long Haul Banditz? They are a group of men and woman that are trying to reestablish the "Old School" Trucker mentality. The Brotherhood of trucking if you will. This group is on a mission and it doesn't look like they will let anything get In their way! They believe in honor and respect! Grumpy said you got to give respect to get respect. Recently the Banditz showed up at a truck stop as part as their mission. They didn't show up to pass out literature or to protest and make waves. No, instead they showed up armed with trash bags and a great attitude of leading by example. Thats right, including the president of the organization Sancho himself picking up trash. Sancho was filmed picking up "trucker bombs" AKA piss bottles thrown out of semi trucks parked in the lot. This group is trying to change the trucking mentality one mind and one place at a time and its working.https://www.longhaulbandit.com Mission Statement Long Haul Banditz have a mission statement. This statement is like a code or a way of life. They suggest if you want to join the Banditz, to just read the statement. If you practice good and safe driving around semi trucks then you are a bandit. Sancho told us in the interview "If you are going to throw garbage out your window at a truck stop. If you park in the fuel island and stay there for extended periods because your too lazy to park. Just take the Long Haul Banditz shirt off and throw it away. Rick Carter is a Senior Vice President in LHB's Minneapolis office where he utilizes his 30 years of experience to lead the firm's Integrative Design Team, a group of 100 architects, mechanical and electrical engineers, landscape architects, planners and interior designers serving five market sectors. An early adopter of healthy building principles, Rick has led LHB's focus on high-performance design since the early 1990s. He is recognized as an expert in sustainable design and speaks regularly at regional and national conferences on this topic. Rick Carter-Sustainability Minded During Career When Rick was younger he was fascinated by the underground space center.  That center has evolved to the Center for Sustainable Building Research. Sustainability has always been in the forefront of his mind but didn't really come to fruition until he started working.  Rick works extensively on design for efficient resource use and occupant well-being, leading efforts for LHB to be visible experts in regenerative design and research. His projects range from the building to regional scales with the goal of contributing to achieving Minnesota's greenhouse gas emission reduction goals. Rick helped develop the Hennepin County/Minnesota Sustainable Design Guidelines and currently co-manages the Buildings, Benchmarks and Beyond (B3) Project including execution of sustainable guidelines for the State of Minnesota. "We know it's something that we don't know enough about, so we are going to try and learn about it.  We also know that it's okay to go out and tell people we are good at it." – Rick Carter   Proudest Achievements Rick's proudest achievements include his family and two children. The long head of biceps tendon and superior labrum can get a hard time in throwing athletes and patients that fall on their arm or shoulder. This can result in long head of biceps tendon pathology or SLAP tears, and cause ongoing shoulder pain. A lot of our shoulder tests are non-specific, and are unable to identify particular structures that are irritated or painful in the shoulder. In the case of the long head of biceps (LHB) tendon and slap tears, there are a few useful tests that in combination with a good history can help you identify when the structures are involved in your patient's shoulder pain. Backround: The treatment of rotator cuff tears is still challenging. Tendon tissue engineering (TTE) might be an alternative in future. Tenocytes seem to be the most suitable cell type as they are easy to obtain and no differentiation in vitro is necessary. The aim of this study was to examine, if the long head of the biceps tendon (LHB) can deliver viable tenocytes for TTE. In this context, different isolation methods, such as enzymatic digestion (ED) and cell migration (CM), are investigated on differences in gene expression and cell morphology. Methods: Samples of the LHB were obtained from patients, who underwent surgery for primary shoulder arthroplasty. Using ED as isolation method, 0.2% collagenase I solution was used. Using CM as isolation method, small pieces of minced tendon were put into petri-dishes. After cell cultivation, RT-PCR was performed for collagen type I, collagen type III, decorin, tenascin-C, fibronectin, Scleraxis, tenomodulin, osteopontin and agreccan. Results: The total number of isolated cells, in relation to 1 g of native tissue, was 14 times higher using ED. The time interval for cell isolation was about 17 hours using ED and approximately 50 days using CM. Cell morphology in vitro was similar for both isolation techniques.
