Abstract: Cargo traffic through the Golgi apparatus is mediated by cisternal maturation, but it remains largely unclear how the cis-cisternae, the earliest Golgi sub-compartment, is generated. Here, we use high-speed, high-resolution confocal microscopy to analyze the spatiotemporal dynamics of a diverse set of proteins that reside in and around the Golgi in budding yeast. We find many mobile punctate structures that harbor yeast counterparts of mammalian endoplasmic reticulum (ER)-Golgi intermediate compartment (ERGIC) proteins, which we term "yeast ERGIC". It occasionally attaches onto the ER exit sites and gradually matures into the cis-Golgi. Upon treatment with the Golgi-disrupting agent brefeldin A, the ERGIC proteins form larger aggregates corresponding to the Golgi entry core compartment in plants, while cis- and medial-Golgi proteins are absorbed into the ER. We further analyze the dynamics of several late Golgi proteins. Together with our previous studies, we demonstrate a detailed spatiotemporal profile of the cisternal maturation process from ERGIC to Golgi and further to the trans-Golgi network. Abstract: Gametogenesis in budding yeast involves large-scale rearrangement of membrane traffic to allow de novo formation of a membrane, called the prospore membrane (PSM). However, the mechanism underlying this event is not fully elucidated. Here, we show that the number of endoplasmic reticulum exit sites (ERES) per cell fluctuates and switches from decreasing to increasing upon the onset of PSM formation. Reduction in ERES number is accompanied by a transient stall in membrane traffic, resulting in loss of the preexisting Golgi apparatus from the cell, as well as local ERES regeneration, leading to Golgi reassembly in nascent spores. We have revealed that protein phosphatase-1 (PP-1) and its development-specific subunit, Gip1, promote ERES regeneration through Sec16 foci formation. Furthermore, a mutant with impaired ERES formation showed defects in PSM growth and spore formation. Thus, ERES regeneration in nascent spores facilitates the segregation of membrane traffic organelles, leading to PSM growth. Copy rights belong to original authors. Dans la famille des smartoons, voici Piccoma, deuxième opérateur francophone après Naver-Webtoon, présente en France depuis mars 2022, filiale européenne de Kakao Piccoma Corp., la branche japonaise de Kakao, N°1 du manga numérique dans l'archipel. Piccoma se veut une plateforme ouverte à tous, un peu comme Netflix, des smartoons (terme générique qui englobe webtoons et mangas numériques) où toutes les créations du monde sont présentes de diffusion. On y trouve ainsi la version numérique légale des mangas publiés par Kurokawa, Pika ou Glénat. La structure est dirigée par Kim Hyung-Rae qui travailla longtemps pour Delitoon avant de prendre la direction de Piccoma Europe voici un an. Today I talked to Chris McMorran about his new book Ryokan: Mobilizing Hospitality in Rural Japan (U Hawaii Press, 2022). Amid the decline of many of Japan's rural communities, the hot springs village resort of Kurokawa Onsen is a rare, bright spot. Its two dozen traditional inns, or ryokan, draw nearly a million tourists a year eager to admire its landscape, experience its hospitality, and soak in its hot springs. As a result, these ryokan have enticed village youth to return home to take over successful family businesses and revive the community.  Chris McMorran spent nearly two decades researching ryokan in Kurokawa, including a full year of welcoming guests, carrying luggage, scrubbing baths, cleaning rooms, washing dishes, and talking with co-workers and owners about their jobs, relationships, concerns, and aspirations. He presents the realities of ryokan work—celebrated, messy, ignored, exploitative, and liberating—and introduces the people who keep the inns running by making guests feel at home. McMorran explores how Kurokawa's ryokan mobilize hospitality to create a rural escape from the globalized dimensions of everyday life in urban Japan. Ryokan do this by fusing a romanticized notion of the countryside with an enduring notion of the hospitable woman embodied by nakai, the hired female staff who welcome guests, serve meals, and clean rooms. These women are the face of the ryokan. But hospitality often hides a harsh reality. McMorran found numerous nakai in their 50s, 60s, and 70s who escaped violent or unhappy marriages by finding employment in ryokan. Yet, despite years of experience, nakai remain socially and economically vulnerable. Through this intimate and inventive ethnography of a year in a ryokan, McMorran highlights the importance of both the generational work of ryokan owners and the daily work of their employees, while emphasizing the gulf between them. With its focus on small, family-owned businesses and a mobile, vulnerable workforce, Ryokan makes an invaluable contribution to scholarship on the Japanese workplace. It also will interest students and scholars in geography, mobility studies, and women's studies and anyone who has ever stayed at a ryokan and is curious about the work that takes place behind the scenes. Today I talked to Chris McMorran about his new book Ryokan: Mobilizing Hospitality in Rural Japan (U Hawaii Press, 2022). Amid the decline of many of Japan's rural communities, the hot springs village resort of Kurokawa Onsen is a rare, bright spot. Its two dozen traditional inns, or ryokan, draw nearly a million tourists a year eager to admire its landscape, experience its hospitality, and soak in its hot springs. As a result, these ryokan have enticed village youth to return home to take over successful family businesses and revive the community.  Chris McMorran spent nearly two decades researching ryokan in Kurokawa, including a full year of welcoming guests, carrying luggage, scrubbing baths, cleaning rooms, washing dishes, and talking with co-workers and owners about their jobs, relationships, concerns, and aspirations. He presents the realities of ryokan work—celebrated, messy, ignored, exploitative, and liberating—and introduces the people who keep the inns running by making guests feel at home. McMorran explores how Kurokawa's ryokan mobilize hospitality to create a rural escape from the globalized dimensions of everyday life in urban Japan. Ryokan do this by fusing a romanticized notion of the countryside with an enduring notion of the hospitable woman embodied by nakai, the hired female staff who welcome guests, serve meals, and clean rooms. These women are the face of the ryokan. But hospitality often hides a harsh reality. McMorran found numerous nakai in their 50s, 60s, and 70s who escaped violent or unhappy marriages by finding employment in ryokan. Yet, despite years of experience, nakai remain socially and economically vulnerable. Through this intimate and inventive ethnography of a year in a ryokan, McMorran highlights the importance of both the generational work of ryokan owners and the daily work of their employees, while emphasizing the gulf between them. Momokichi Momoi, the mob boss owner of a professional baseball team known as "the Peach Twisters", has a unique way of dealing with ineffective players: killing them. His daughter, Momomi, is twice as twisted as her father. Mikura and the Danger Service Agency are hired by a mysterious old man to kidnap Momokichi. But during that attempt they run afoul of Momomi. Before the resulting firefight, Momokichi dies when his head falls on a ventilation pipe. Now Mikura and the D.S.A. have to come up with a new plan before they end up as Momomi's next target. Mikura, Kurokawa, and Harada are the three members of the Danger Service Agency (DSA). Mikura is the brawn of the group, Harada is the brains, and Kurokawa is just a bitter ex-cop who likes to think he's in charge. They'll take on any job as long as it involves much danger and, of course, money. Their biggest case, however, could prove to be finding out why someone wants Kurokawa assassinated. The DSA's headquarters is an old red British double-decker bus that has been converted into living quarters for the three main characters. It has been placed on the rooftop of a two-story building which houses Mugiyama's barber shop on the ground floor. Chihiro Kurokawa started multifamily investing in 2017 and has been a principal in four multifamily transactions totaling 479 units and $28.9 million in purchase volume. He has also invested as a limited partner in mobile home parks, development, multifamily, retail and self-storage. He was previously an analytics professional and holds an MBA from UCLA Anderson. He is training to qualify for the Boston Marathon and enjoys alpine skiing.  In this episode Chihiro talks about leveraging the power of relationships and mentorship to enter the multifamily space and manage his first syndication deal in 2019 for a 128 unit property. Vì sao Pháp trở thành thị trường lớn thứ hai của manga Nhật Bản?

Tại Pháp, « manga » – truyện tranh Nhật Bản, giờ là một trong những món quà không thể thiếu trong các dịp lễ Noel, Tết mừng năm mới. Năm 2021, số lượng manga bán ra ở Pháp đã đạt con số kỷ lục : 50% truyện tranh bán ra là các tập manga Nhật Bản. Vì sao người Pháp lại mê truyện tranh Nhật Bản đến thế ? Ngày 08/12/2021, tập thứ 100 của One Piece – bộ truyện tranh Nhật Bản bán chạy nhất tính từ năm 1997 (25 triệu bản tại Pháp) – đã ra mắt giới hâm mộ với 250.000 ấn bản. Một con số tương đương với một tác phẩm văn học được trao giải Goncourt. Đây chỉ là một phần trong thị trường manga đang nở rộ tại Pháp. Khủng hoảng dịch tễ không thể ngăn cản mức tăng trưởng với ba con số ấn tượng trong thị trường truyện tranh ở Pháp : 124%. Năm 2021, lần đầu tiên số đầu sách bán ra đạt mức kỷ lục 28 triệu bản trong giai đoạn từ tháng 1 đến tháng 8/2021, so với con số 20 triệu bản truyện tranh truyền thống, theo số liệu từ Viện GfK. Manga : Truyện ba xu đổ bộ đến lãnh địa của Tintin Đối với các nhà phát hành tại Pháp, « thị trường manga đang trong thời kỳ hoàng kim ». Xuất hiện lần đầu ở Nhật Bản trong những thập niên 1930, « Manga » - còn có nghĩa là mạn họa, du nhập vào Pháp vào những năm 1990. Vào thời điểm đó, thể loại truyện tranh Nhật Bản này bị phản đối, từng bị ví là loại truyện tranh ba xu, rẻ tiền, ngôn từ nghèo nàn, bạo lực. Đến mức tại nước láng giềng, đích thân vua Bỉ tìm cách cấm đoán loại truyện tranh mà ông cũng cho là bạo lực và vô bổ. Nhưng bất chấp những chỉ trích mạnh mẽ, thị phần manga Nhật Bản không ngừng tăng. Mười lăm năm sau ngày « đổ bộ », manga chiếm đến 38% thị trường truyện tranh ở Pháp. Tính đến 31/12/2005, đã có đến 628 đầu sách của 231 tác giả được dịch sang tiếng Pháp. Và 15 năm tiếp theo, năm 2021, thị phần manga Nhật Bản lên đến 50%. Làm thế nào mà manga, một thể loại truyện tranh dân gian dài tập, chủ yếu dựa trên những hình thức thái quá, đối đầu, và khoái cảm nhục dục, lại có thể gây được tiếng vang ở Pháp ? Trước hết, giới chuyên ngành đều nhìn nhận công đầu thuộc về Jacques Glénat và Dorothée. Người thứ nhất đã cho du nhập lọai truyện tranh từ Nhật Bản. Người thứ hai đã biết cách thổi bùng ngọn lửa nhu cầu. Để rồi từ đó, lần lượt các bộ One Piece, Naruto, Dragon Ball – 7 viên ngọc rồng… lần lượt đổ bộ lên mảnh đất của Tintin, Gaston Lagaffe hay Asterix. Trả lời báo Les Echos, bà Marion Glénat, Corveler, con gái Jacques Glénathồi tưởng : « Vào cuối những năm 1980, cha tôi có một chuyến đi đến Tokyo. Trong va-li của mình, ông mang theo bộ truyện tranh "Les passagers du vent" với hy vọng bán được cho các nhà xuất bản Nhật. Họ nghe ông ấy một cách rất lịch sự, nhưng lại nói với cha tôi rằng họ cũng có vài thứ để đề nghị. Cuối cùng, cha tôi đi về với chiếc va-li chất đầy manga và tự nhủ : Thử xem sao ! » Nhưng nhà nghiên cứu Jean-Marie Bouissou, trong một bài viết đăng trang mạng CAIRN.INFO hồi năm 2006, từng lưu ý rằng công nghệ văn hóa Nhật Bản là nhà vô địch về « truyền thông hỗn hợp », chia nhỏ lợi nhuận và mở nhiều kênh tuyên truyền để có thể luồn lách các chính sách bảo hộ tại nhiều nước. Tại Pháp, đất nước mà manga vấp phải thái độ thù nghịch từ các bậc phụ huynh, các nhà giáo dục, giới truyền thông, các cơ quan công quyền và nhiều nhà xuất bản truyện tranh lớn, việc thâm nhập được tiến hành thông qua các loạt phim hoạt hình nhiều tập, tiếp đến là các loại trò chơi điện tử và các lá bài như Pokemon, Yu Gi Oh. Nghiên cứu của Jean-Marie Bouissou nhận thấy 95% những người được hỏi cho biết họ đã đến với manga đều thông qua các phương tiện trên. Về điểm này, Grégoire Hellot, giám đốc sưu tập nhà xuất bản Kurokawa, trong một chương trình của đài France Inter hồi năm 2018, có nhớ lại như sau : «  Điều này cũng tương tự cho tất cả các nước khác Tây Ban Nha, Ý, bắt đầu từ loạt phim Goldorak, Candy, Albator, đổ vào Pháp cuối những năm 1970. Trẻ em thời kỳ đó rất thích những bộ phim này. Rồi sau đó, còn có chương trình truyền hình câu lạc bộ Dorothe kết hợp với các loạt phim hoạt hình như Những hiệp sĩ của cung hoàng đạo, Những kẻ sống sót, v.v… Phim hoạt hình Nhật Bản cung cấp một phong cách kể chuyện và những câu chuyện sâu sắc hơn nhiều so với phim hoạt hình Mỹ thời bấy giờ, phải nói là quá nghiêm túc. Trong khi đó, phim hoạt hình Nhật Bản nói đủ chuyện, người ta có thể nói là họ cố gắng không đánh lừa trẻ em. Họ giới thiệu nhiều câu chuyện giàu cảm xúc, khiến trẻ em cảm động. Những câu chuyện đó đi theo trẻ cho đến khi chúng trở thành thanh thiếu niên vào cuối những năm 90. Nhưng nghị sĩ thời kỳ đó là Ségolène Royal đã làm đảo lộn mọi thứ khi cho rằng phim hoạt hình Nhật Bản làm cho người ta trở nên điên rồ và thế là không còn phim hoạt hình Nhật Bản trên truyền hình nữa. Do vậy, có một nhu cầu và nhờ vậy mà vào thời điểm đó những bộ truyện tranh giấy đầu tiên được xuất bản. Những người trẻ tuổi, thanh thiếu niên đã từng được xem phim hoạt hình và không còn xem phim hoạt hình nữa, nhưng vẫn tiếp tục yêu thích chúng đã đắm mình vào truyện tranh và rồi từ đó mà truyền tai nhau. » Manga và những phân khúc thị trường Một điểm khác khiến manga dễ dàng chinh phục thị trường truyện tranh Pháp đó là sự đa dạng phân khúc thị trường, theo giới tính, đủ mọi lứa tuổi… Trong thể loại này, người ta có « shônen manga » (dành cho nam thiếu niên) và « shôjo manga » (dành cho thiếu nữ). Rồi còn có những loại manga dành cho các đối tượng trưởng thành hơn như « seinen manga » (dành cho người lớn trẻ tuổi), loạt truyện dài tập lịch sử, rồi còn có « shakai manga » (chuyên nói về những vấn đề xã hội) hay như « gekiga » (chuỗi tập truyện cho người lớn thường bạo lực và u tối). Đối với anh Grégoire Hellot, đây chính là thế mạnh để manga Nhật Bản có thể cạnh tranh trực diện với các thể loại « Comics » của Mỹ và truyện tranh truyền thống Pháp – Bỉ. « Manga Nhật Bản bắt đầu từ cho trẻ nhỏ và có rất nhiều phân khúc thị trường. Có những tập truyện dành cho nam giới, nhưng cũng có loại truyện dành cho nữ giới. Truyện cho phụ nữ đi làm, cũng như là truyện cho các bà nội trợ. Nhưng đối tượng chính của manga vẫn là lứa tuổi thanh thiếu niên. Truyện tranh phương Tây, bất kể là Mỹ hay Pháp người ta thường có xu hướng xấu hổ hay bối rối khi nói về những câu chuyện yêu đương trong lĩnh vực giải trí. Trong khi đó manga Nhật Bản không ngần ngại đi sâu vào tâm lý của các nhân vật. Nghĩa là quý vị có thể sẽ thấy những nhân vật siêu anh hùng có thể đấm vỡ một bức tường, hay một cô gái xuất hiện nói ấp a ấp úng. Những nhân vật mà một thanh thiếu niên có thể dễ dàng tìm thấy chính mình. »  Điểm cuối cùng làm nên lợi thế cho manga Nhật Bản là có kích cỡ nhỏ, dễ dàng bỏ túi để có thể đọc trên tầu điện ngầm. Giá một tập manga cũng « mềm » chỉ tầm từ 6-10 euro, thay vì là từ 15-19 euro đối với một tập truyện tranh có kích cỡ lớn hơn một chút một tờ giấy khổ A4. Manga : Một phần di sản văn học Với hơn 28 triệu ấn bản (40 triệu theo Les Echos) trong năm 2021, nước Pháp được ví như là « tổ quốc » thứ hai của manga. Truyện tranh Nhật Bản gần như là sản phẩm văn hóa không thể thiếu tại các hiệu sách, bất kể quy mô lớn nhỏ. Tại Montpellier, hiệu sách Sauramps, lớn nhất của thành phố, từ 15 năm qua đã dành hẳn một gian riêng cho manga Nhật Bản. Trả lời RFI Tiếng Việt, Leslie Bonnet, một nhân viên phụ trách gian sách manga cho biết, mỗi tháng hiệu sách bán ra khoảng 16.000 quyển manga cho khoảng 300 đầu sách, cho đủ mọi lứa tuổi từ 8 đến trên 50. Theo lời cô Bonnet, nhu cầu về sách tăng vọt đến mức hiệu sách phải tính đến chuyện mở rộng thêm gian hàng trong sắp tới : « Từ hai năm nay, nhu cầu về truyện tranh Nhật Bản bất ngờ bùng phát. Đến mức hiện tại chúng tôi thiếu cả chỗ để sách. Nếu muốn đáp ứng được nhu cầu thị trường, sớm hay muộn gì chúng tôi cũng sẽ phải nới rộng gian hàng. Đúng là trong vòng có hai năm, gian hàng này đã được cơi nới thêm gần như một gian rưỡi nữa. » Cũng theo Leslie Bonnet, doanh thu từ manga hiện nay là bằng một nửa của truyện tranh. « Về mặt doanh số, đúng là doanh số bán ra của manga là bằng một nửa truyện tranh. Điều đó có nghĩa là để có thể bằng với doanh số của truyện tranh truyền thống, số đầu sách manga bán ra phải nhiều hơn gấp 3 lần. Bình thường một tập truyện tranh giá khoảng 15 euro, trong khi một tập manga chỉ có 6 đồng. » Trước nhu cầu ngày càng lớn và thế hệ « phim hoạt hình Nhật Bản » nay cũng đã trưởng thành, một thế hệ nhà bán sách mới, đam mê manga, ra đời. Một lợi thế cho nhiều nhà sách nhỏ để có thể cạnh tranh, đồng thời cho phép nhiều người hâm mộ được chìm đắm trong thế giới nhân vật ưa thích của mình. « Đúng vậy, cần phải hiểu rõ lĩnh vực này để có thể hướng dẫn khách hàng. Điểm lợi hiệu sách manga ở đây thường chúng tôi là những fan hâm mộ, rất thích đọc manga nữa. » Để vinh danh nền nghệ thuật thứ 9 này, một bức ảnh cỡ lớn (XXL) nhân vật chính Kaiju trong bộ truyện cùng tên đã được dán một bên tường kính của BNF. Một cách thức để nhìn nhận manga Nhật Bản giờ cũng là một phần của di sản văn học, như lời nhận định của bà Marine Planche, Trung tâm Văn học Quốc gia dành cho giới trẻ, thuộc Thư Viện Quốc Gia Pháp (BNF), trên đài TV5Monde. « Đó cũng là một phần của những điều tạo nên ngành xuất bản sách, một phần của di sản văn học Pháp, có thể sánh ngang vai cùng với bao thể loại khác. Ở đây có một lĩnh vực đang ngày càng được công nhận một cách hợp pháp. »

How do we think about our customers?  How authentic are our communications as a result?  Are we telling enough stories about them?  In this week's show, we will see how a master of communication does it.   I am sure you have you seen notices explaining that this location is going to close while the building is being reconstructed and that it will reopen at a specified day in the future?  One notice however has become much talked about locally amongst Japanese retailers.  Toraya are a famous traditional Japanese sweets manufacturer and retailer.  Mr. Mitsuhiro Kurokawa is the 17th generation of his family to lead the business and his “we are rebuilding” notice is considered outstanding, even in a country where omotenashi or unparalleled service is renowned.   Most such notices tell facts, supply relevant data, maps and provide the obligatory greetings about serving us again when they reopen.  Kurokawa san did all of that but much more.  He put the current change in historical perspective, noting the business started in Kyoto in 1586, moving to Tokyo in 1869 and to this location in 1964.  By doing this he is assuring us of their long traditions, longevity and capacity to change with the times when needed.   He then started to tell some stories about the customers they have had at this shop on Aoyama Street in Akasaka, over the last 51 years.  What stories are you currently telling to your clients?   He mentioned that every three days, a male customer visited the shop to enjoy oshiruko     (bean paste sweet soup with grilled mochi).  Another customer, a kindergarten aged boy came with his mother to the shop every day and bought a bite sized yookan   (sweet bean paste block).  A 100 year old lady regularly came by wheelchair to the shop.  She later became hospitalized and her family came to buy namagashi   (fresh Japanese sweets) and higashi    (a dried sugar sweet), to take to the hospital for her.  Even after she couldn't eat anything anymore, they found if they crushed the dried sugar sweet she could still enjoy it.   He mentioned that he couldn't include all of the episodes they have shared over these 51 years with their customers, but he said he and the staff keep them, one by one, in their hearts forever.   Telling customer stories is powerful.  Kurokawa san made the customers experiences come alive and he subtly linked them to the products they enjoyed.  The feeling of the notice is that there is a special bond they feel with all of their customers and even though they won't reopen on that site for another three years, they won't have forgotten them and look forward to serving them forever.   Are we communicating we feel a special bond with our customers?  Are we weaving enough customer stories into our communications?   Kurokawa san's notice gets attention in Japan because of the sincerity in the message.  He is regarded as really epitomising the spirit of a family that has served customers for 17 generations.   We may not be the 17th generation in our business, but we can bring more heart into the service we provide our customers.      Action Steps   Are we really thinking about creating an emotional connection with our clients Are we telling enough happy client stories in our communications Are we fully aware of the content of all the touch points we have with our buyers Are we serving from the heart or just the head Are we instilling the right frame of reference into our staff, regarding how to properly serve the client.

Malheureusement pour lui, il a grandit dans une famille de Shaman et il a le pouvoir de voir les fantômes et à cause de ça il va devoir participer au Shaman Fight. Le tournoi qui a lieu tout les 500 ans qui désigne qui va devenir le prochain Shaman King.c'est le pitch du manga de Hiroyuki Takei dont on vous parle cette semaine dans MangaDiscovery, c'est un pur shonen nekketsu avec tout les clichés du genre : le tournoi, les différentes évolutions de pouvoirs, le sacrosaint pouvoir de l'amitié ou les rivaux qui vont devenir amis. Par certains cotés, il change un peu des autres manga du Shonen Jump, avec un héros plus calme ou une philosophie du zen qui est distillée tout le long de Shaman king mais surtout des personnages haut en couleurs. On finit l'émission avec un petit débat sur les fins des mangas dans le Shonen Jump, car Shaman King a marqué par une fin un peu brouillonne que l'auteur a eu la chance de pouvoir réécrire après la fin dans le Jump. Starting his career as medical practitioner, specializing in nephrology (kidney disease) , Kiyoshi's field of activities took a big leap beyond the medical practice.   At 84 years strong, Kiyoshi is one of the most globally recognized opinion leaders from Japan.    He was the chairman of the Fukushima Nuclear Accident Independent Investigation Commission.  “How could this happen in Japan, a country with reputation for excellence in engineering and manufacturing,” questions Kiyoshi.  "Japan has to show leadership to learn from failure and disaster." Kiyoshi encourages the young people of Japan to “take a leap!” Getting college credit or not is irrelevant.  It is the experience that should be valued, gaining new perspectives about Japan. For the world, Kiyoshi sends his greetings.  “There are many young eager stars in Japan.  Come work with us, come study with us.”  We live in a connected world."

I begin by talking about the how the microbiome is one of the five natural defense systems in the body and ways that it is impacted by the foods we eat and how we digest them. I also highlight the impact of an imbalanced microbiome on a host of mental and physical health problems in ourselves, as well as its impact intergenerationally.  Throughout this episode I highlight scientific research that demonstrates the connection between our gut and our brain – emotionally and physiologically - and weave in evidence-based principles and strategies drawn from Eastern and Western medicine for taking care of this gut-microbiome-axis in order to promote longevity, holistic wellness, and healing. We proposed a study on whether Malaysian architects, urban designers and planners refer to precedents of urban shophouses in the inner city streets of Malacca and George Town when coming up with concept and ideas in their work. Based on Michael Gibert's doctoral thesis, Kisho Kurokawa said that: "the difference between Western and Japanese concepts of space is the difference between spatial confrontation and spatial continuity", so do we Malaysian architectural designers think about these theories and put into practice and further that "the Japanese concept of space, on the contrary, reaches out to embrace nature, and to achieve unity and harmony with it, with always a conscious effort made to allow inner and outer space to inter-penetrate", so is that similar to our streets? I believe that the Malaysian precedents mentioned provided this sense of scale, intimacy, sense of proportion and materiality when I walked on those five-foot way and entering in and out those shophouses. Kurokawa also said: Asian public space is the street (no spatial or functional clear demarcations), and I felt that too when walking around in, it's close up and personal, but does that inspire Malaysian designers at all?© 2021 Talk Architecture, Author: Naziaty Mohd Yaacob

Abstract: Exciting developments in both in vitro and in silico technologies have led to new ways to identify patient specific cardiac mechanisms. The development of induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs) has been a critical in vitro advance in the study of patient-specific physiology, pathophysiology and response to drugs. However, the iPSC-CM methodology is limited by the low throughput and high variability of resulting electrophysiological measurements. Moreover, the iPSC-CMs generate immature action potentials, and it is not clear if observations in the iPSC-CM model system can be confidently interpreted to reflect impact in human adults. There has been no demonstrated method to allow reliable translation of results from the iPSC-CM to a mature adult cardiac response. Here, we demonstrate a new computational approach intended to address the current shortcomings of the iPSC-CM platform by developing and deploying a multitask network that was trained and tested using simulated data and then applied to experimental data. We showed that a deep learning network can be applied to classify cells into the drugged and drug free categories and can be used to predict the impact of electrophysiological perturbation across the continuum of aging from the immature iPSC-CM action potential to the adult ventricular myocyte action potential. We validated the output of the model with experimental data. The method can be applied broadly across a spectrum of aging, but also to translate data between species. I am sure you have you seen notices explaining that this location is going to close while the building is being reconstructed.   One notice however has become much talked about amongst Japanese retailers.  Toraya are a famous traditional Japanese sweets manufacturer and retailer.  Mr. Mitsuhiro Kurokawa is the 17th generation of his family to lead the business and his “we are rebuilding” notice is considered outstanding, even in a country where omotenashi is renowned. Most such notices tell facts, supply relevant data and provide the obligatory greetings about serving us again when they reopen.  Kurokawa san did all of that but much more.  He put the current change in historical perspective, noting the business started in Kyoto in 1586, moving to Tokyo in 1869 and to this location in 1964.  He then started to tell some stories about the customers they have had at this shop on Aoyama Street in Akasaka, over the last 50 plus years. He mentioned that every three days, a male customer visited the shop to enjoy oshiroko (bean paste sweet soup with grilled mochi).  Another customer, a kindergarten aged boy came with his mother to the shop every day and bought a bite sized yookan (sweet bean paste block).  A 100 year old lady regularly came by wheelchair to the shop.  She later became hospitalized and her family came to buy namagashi (fresh Japanese sweets) and higashi (a dried sugar sweet), to take to the hospital for her.  Telling customer stories is powerful.  Kurokawa san made the customers experiences come alive and he linked them to the products they enjoyed.  The feeling of the notice is that there is a special bond they feel with all of their customers. Are we communicating we feel a special bond with our customers?  Are we weaving enough customer stories into our communications, real episodes that the reader can visualise in their mind's eye?  We know the human touch is so important but often we can get tied up with the spec, the data, the details and lose sight of how we can differentiate ourselves in the market.  Is your communication all about you and what you can do and nothing about your customers? We may not be the 17th generation in our business, but we can bring more heart into the service we provide our customers.  We can start right now with the service we provide and how we communicate that service.

Before we get into this week's topic, here is what caught my attention lately. Japan's agricultural workforce has shrunk by fifty percent in the last ten years.  In 2007 there were three point one two million farmworkers. The shortage of workers is a result of the aging of the farmer population. In 2017 it had become one point eight one million.  The solution – robots.  The Toyohashi University of Technology has created a one meter high handcart type robot, equipped with three cameras and two infrared radar devices to follow flower pickers, pick up the flowers and deliver them to designated collection points. They are extending the scope to picking fruit, and vegetables and delivering components in factories. This is episode number 43and we are talking about Old School Japanese ServiceSoredewa ikimasho, so let's get going. I am sure you have you seen notices explaining that this location is going to close while the building is being reconstructed and that it will reopen at a specified day in the future?  Given the increasingly stringent earthquake code here in Tokyo, we are seeing many businesses opting to re-build their premises.  One notice however has become much talked about amongst Japanese retailers. Toraya are a famous traditional Japanese sweets manufacturer and retailer.  Mr. Mitsuhiro Kurokawa is the seventeenth generation of his family to lead the business and his “we are rebuilding” notice is considered outstanding, even in a country where omotenashi is renowned. Most such notices tell facts, supply relevant data and provide the obligatory greetings about serving us again when they reopen.  Kurokawa san did all of that but much more.  He put the current change in historical perspective, noting the business started in Kyoto in Fifteen eight six toward the end of the Muromachi period which was thirteen thirty eight to fifteen seventy three, moving to Tokyo in eighteen sixty nine and to this location in nineteen sixty four.  By doing this he is assuring us of their long traditions, longevity and capacity to change with the times when needed.  He then started to tell some stories about the customers they have had at this shop on Aoyama Street in Akasaka, over the last fifty one years. He mentioned that every three days, a male customer visited the shop to enjoy oshiroko (bean paste sweet soup with grilled mochi).  This is considered a bit unusual in Japan, because men don't normally have such a sweet tooth, so this customer stood out from others. Another customer, a kindergarten aged boy came with his mother to the shop every day and bought a bite sized yookan (sweet bean paste block).  One day he came by himself to shop.  The staff were worried about him and so they went out with him and found that the mother was secretly hiding and watching that he was OK. He mentioned that he couldn't include all of the episodes they have shared over these fifty one years with their customers, but he said he and the staff keep them, one by one, in their hearts forever. Telling customer stories is powerful.  Kurokawa san made the customers experiences come alive and he linked them to the products they enjoyed.  Rather than just a cold statement of the facts, he crafted a statement of love for their customers.  The feeling of the notice is that there is a special bond they feel with all of their customers and even though they won't reopen on that site for another three years, they won't have forgotten them and look forward to serving them forever. Are we communicating we feel a special bond with our customers?  Often, corporate communications becomes machine like and wrapped up in what can sound like marketing department dross.  Kurokawa san conveys a lot of heart felt feelings in this simple notice about the main store being rebuilt.  Are we weaving enough customer stories into our communications? I don't mean fake propaganda stories, but real episodes that the reader can visualise in their mind's eye? Kurokawa san's notice get attention in Japan because of the sincerity in the message.  He is regarded as really epitomising the spirit of a family that has served customers for seventeen generations. We may not be the seventeenth generation in our business, but we can bring more heart into the service we provide our customers.  We can start right now with the service we provide and how we communicate that service. In this episode, we head back for a quick stop over in the Kumamoto prefecture. Kumamoto is located in the center of Kyushu along the western coast. Among the attractions of Kumamoto are Mount Aso, one of the largest active volcanoes in the world, Kumamoto Castle in the capital city of Kumamoto, and Kurokawa Onsen (or hot spring baths). Our adventures took us through Kumamoto twice; once to visit Mt. Aso and Kurokawa, and another trip to visit Kumamoto castle. Highly recommend Kurokawa – such an amazing town with charming ryokan inns in a delightfully natural setting. Definitely a must stop to experience traditional Japanese hospitality and rejuvenate your spirit. Next we stopped over in Kumamoto city for a quick visit to the castle. Unfortunately, the castle was damaged by two powerful earthquakes in 2016 so the inner grounds were closed to the public but we were still able walk around the outside of the castle to get an idea of the sheer size of the complex as well as view the restoration in progress. In addition, the shops at the entrance to the castle offer a great spot to get some refreshments or souvenirs. And don't forget to check out our pics as well. Enjoy!

Omotenashi: Real Japanese Customer Service   I am sure you have you seen notices explaining that this store location is going to close while the building is being reconstructed and that it will reopen at a specified date in the future? Given the increasingly stringent earthquake code here in Tokyo after 2011, we are seeing many businesses opting to re-build their premises. One notice however has become much talked about locally amongst Japanese retailers. Toraya are a famous traditional Japanese sweets manufacturer and retailer. Mr. Mitsuhiro Kurokawa is the 17th generation of his family to lead the business and his “we are rebuilding” notice is considered outstanding, even in a country where omotenashi or customer first service is renowned.   Most such notices tell facts, supply relevant data and provide the obligatory greetings about serving us again when they reopen. Kurokawa san did all of that but much, much more. He put the current change in historical perspective, noting the business started in Kyoto in 1586 toward the end of the Muromachi (1338-1573) period, moved to Tokyo in 1869 and to this location in 1964. By doing this he is assuring us of their long traditions, longevity and capacity to change with the times when needed. He then started to tell some stories about the customers they have had at this shop on Aoyama Street in Akasaka, over the last 50 plus years.   He mentioned that every three days, a male customer visited the shop to enjoy oshiruko (bean paste sweet soup with grilled mocha or pounded rice). This is considered a bit unusual in Japan, because men don't normally have such a sweet tooth, so this customer stood out from others.   Another customer, a kindergarten aged boy came with his mother to the shop every day and bought a bite sized yookan (sweet bean paste block). One day he came by himself to shop. The staff were worried about him and so they went out with him and found that the mother was secretly hiding and watching that he was okay.   A 100 year old lady regularly came by wheelchair to the shop. She later became hospitalised and her family came to buy namagashi (fresh Japanese sweets) and higashi (a dried sugar sweet), to take to the hospital for her. Even after she couldn't eat anything anymore, they found if they crushed the dried sugar sweet she could still enjoy it.   He mentioned that he couldn't include all of the episodes they have shared over these 50 plus years with their customers, but he said he and the staff keep them, one by one, in their hearts forever.   Telling customer stories is powerful. Kurokawa san made the customers the centerpiece and their experiences come alive. He linked the customers to the products they enjoyed. Rather than just a cold statement of the facts, he crafted a statement of love for their customers. The feeling of the notice is that there is a special bond they feel with all of their customers and even though they won't reopen on that site for another three years, they won't have forgotten them and look forward to serving them forever.   What can we learn from this excellent customer focus? Are we communicating we feel a special bond with our customers? Often, corporate communications becomes machine like and wrapped up in what can sound like marketing department dross. Kurokawa san conveys a lot of heart felt feelings in this simple notice about the main store being rebuilt. Are we weaving enough customer stories into our communications? I don't mean fake propaganda stories or plastic stories that politicians love to use these day, but real episodes that the reader can visualise in their mind's eye? Even in a country like Japan, with such high levels of customer focus, Kurokawa san's notice gets attention because of the sincerity of his message. He is regarded as really epitomising the spirit of a family running a retail business, that has served customers for 17 generations.   We may not be the 17th generation in our business, but we can bring more heart into the service we provide our customers. We can start right now with the service we provide and how we communicate that service. What does your current customer communication say about you? Can it be improved?     Action Steps   Are we really thinking about creating an emotional connection with our clients Are we telling enough happy client stories in our communications Are we fully aware of the content of all the touch points we have with our buyers Are we serving from the heart or just the head Are we instilling the right frame of reference into our staff, regarding how to properly serve the client.     Engaged employees are self-motivated.
