In this episode of the Epigenetics Podcast, we talked with Yadira Soto-Feliciano from MIT about her work on the Menin-MLL complex and the effect of small molecules on its stability in leukemia. We explore the pivotal moments that led her to cancer biology during her graduate studies, where her work included ground-breaking research on the role of the plant homeodomain Finger protein-6 (PHF-6) in leukemia. This work bridged the realms of chromatin accessibility, transcription factors, and cancer cell lineage, providing critical evidence for the concept of lineage plasticity in cancer biology—a topic that has gained significant traction in recent years. Dr. Soto-Feliciano discusses how advances in techniques like CRISPR and ChIP-sequencing have shaped her research, enabling deeper insights into the mechanisms underlying cancer cell identity. As our discussion transitions, Dr. Soto-Feliciano shares her experience in David Allis's lab, illustrating how the collaboration across diverse scientific disciplines enhanced her understanding of chromatin biology and generated significant insights into the mechanics of epigenetic regulation. Highlighting a recent 2023 publication, we unpack her findings related to the conserved molecular switch between MLL1 and MLL3 complexes. These discoveries revealed how the application of small-molecule inhibitors of the menin-MLL interaction can alter gene expression and affect leukemia cells' responses to treatments. We also touch on the operational dynamics within her lab at MIT, established during challenging times marked by the pandemic. Yadira is dedicated to fostering a collaborative and respectful environment among her team, comprised of PhD candidates and research technicians, all sharing a commitment to unraveling the complexities of chromatin regulation. She emphasizes the significance of understanding chromatin scaffold proteins and their role in regulating gene expression and genome organization.

Eric Perner spent most of his early life and career on horseback learning the craft and trade of equine athletic competitions at the highest level. After competing nationwide as a professional journeymen jockey for 10 years he retired and completed his Mechanical Engineering degree at the University of Tulsa, he has since received his Professional Engineering license and served in multiple executive roles in the O&G industry. Later he moved on from the extractive O&G industry and became a full time cattle rancher, Co-Founder/CEO of REP Provisions, a direct to consumer supplier of locally raised regenerative meats from local family farms, and COO of Rebellion Energy Solutions, a company focusing on the plugging of leaking O&G wells and the restoration of these lands to regenerative landscapes. He uses his diverse skill sets to develop unique environmental solutions in the Energy Industry and for various problems facing agriculture. Eric is an educator on the topic of regenerative agriculture and his unique insights show why cattle can be an intricate and necessary part of our grassland ecosystems; however, the improper management of cattle can lead to environmental degradation. Eric owns the Double P Ranch in Mounds, Oklahoma where they produce grass-fed beef, goat, lamb and pastured pork as part of the supply chain for REP Provisions. He operates his ranch as a Savory Global Business Hub to provide restoration grazing education and guidance for other local ranchers in the methods of holistic management to create the conditions that lead to the regeneration of our native grasslands. In this episode, Eric explains what's so special about regenerative agriculture and why it's the only way to move forward. Eric Perner is the founder of Rep Provisions, a regenerative ranch in Oklahoma that's making incredible strides in this space. That's because Eric and his team actively work to regenerate America's grasslands to produce clean foods that benefit people, animals, and our planet. And he comes on the show to share all of the fascinating things he's up to with regards to that. So you'll hear about how Eric took over a ranch in Oklahoma and completely transformed it in the years he's had it. You'll also discover his regenerative agriculture methods, why they work so well, and the powerful results he's seen since implementing his practices. Eric also chats with Dr. Anthony Gustin about some of the problems regenerative agriculture farmers face, how monocrop farms are destroying our planet, and what it's going to take to fix our food system. They also dive into how regenerative agriculture can transform the omega 6 to omega 3 ratios in pork, how regenerative farms follow natural cycles found in nature and work seamlessly as a result, and so much more. As you'll learn in this episode, Eric's dad taught him the importance of using farming practices that align with nature and work with it to make it better than it was before. He adopted this style of farming and took it further by optimizing the farm he took over where he uses regenerative agriculture to maximize land usage and promote a healthier environment. When you tune in, you'll be amazed and impressed with how much Eric's been able to accomplish in such a short amount of time and how it benefits people, animals, and the environment as a result. If you're into the regenerative agriculture space or you're looking to learn the benefits that come with this powerful farming method, this is an episode you don't want to miss. Tune in now to learn more!

He is the chairman of the Scandinavian Critical Care Trials Group and the strategic research program “New resuscitation strategies in patients with severe sepsis’. The contents of this talk are based on the findings of the TRISS trial - Transfusion Requirements in Septic Shock. This trial, Lower versus Higher Hemoglobin Threshold for Transfusion in Septic Shock was published in the NEJM in October 2014. The aim was to evaluate the recommendations from the Surviving Sepsis Campaign regarding transfusion in septic shock. The recommendation is that after the first 6 hours, transfusion threshold should be a Hb

Large randomized trials on critically ill patients have related the use of hydroxyethyl starch (HES) to negative outcomes. In a recent comment we explained in detail why, from our point of view, transferring the results of VISEP, 6S and CHEST into daily ICU practice is as difficult as their extrapolation to perioperative treatment. Haase, Muller and Perner lately challenged this analysis. However, after having carefully read their letter to the editor we are happy to demonstrate that all points we made were absolutely correct. We agree with Haase et al. that a debate on HES safety is important, but has to be based on facts. The difference might be that we like to thoroughly discuss all of them, including the main one: VISEP, 6S and CHEST do not capture the initial stabilization of their hemodynamically instable patients. The vast majority, including those patients later assigned to the "crystalloid" groups, had been stabilized with colloids before study onset. This is not a big problem, but has to be discussed carefully and honestly to prevent the data from being misinterpreted by users and official authorities.

Theory of Mind is a mental process which can be critical to the understanding of the maintenance of psychic disorders and can have implications for their treatment. The idea of a conceptual organisation of the Theory of Mind is supported by current research from Developmental Psychology (see Perner, Mauer & Hildenbrand, 2011). We could show that Theory of Mind seems to develop with the understanding of identity and is based on a common conceptual basis: the understanding of the relationship between reference and representation. Theory of Mind means the ability to ascribe and reflect own mental states and mental states of others. The concept "Theory of Mind" is used very heterogeneousl and is defined very broadly in the research with adults. The aim of this work is to describe the theoretical and conceptual basis of the Theory of Mind and to show its importance for the Posttraumatic Stress Disorder and Chronic Depression with regards to aetiology concept as well as for its implications in the specific therapy methods [Dialectic Trauma Therapy of (Butollo and Karl, 2012) and the Cognitive Behavioral Analysis System for Psychotherapy (McCullough, 2000, 2006)]. I will take into account the attachment theory, the ability of perspective-taking and the integration of paradoxical information as well as the meaning of trauma as a determining factor in the organisation of the Theory of Mind. The differentiation between reference and representation forms the basis of the ability of perspective-taking as well as understanding identity. This work shows related concepts as well as established assumptions about the organisation of the Theory of Mind and Mentalizing and introduces a meta-representational concept to distinguish between reference and representation. The environment is seen as an external source of supply to the formation of mental representation. This work intends a systematic processing of the conceptual fundament of the Theory of Mind as a basis for the use in the clinical-therapeutic area.
