January 22, 1989. Temecula, California. The housemate of 30-year old Marine Captain Jeffrey Digman returns to their residence and discovers that Jeffrey has been shot to death inside his bedroom. Since a gun Jeffrey owns is lying next to him, his death is ruled to be a suicide and investigators theorize that he had become despondent over a recent transfer to Puerto Rico. However, Jeffrey's parents discover some suspicious discrepancies to make them suspect he was murdered and while his cause of death is eventually changed from “suicide” to “undetermined”, there are no conclusive answers about how he was killed. February 20, 1992. South Korea. 30-year old Army Staff Sergeant Billy Ray Hargrove is found hanging by a parachute cord from a tree outside his residence and his death is ruled to be a suicide. Billy Ray's best friend, 37-year old Army Sergeant First Class Mike Carmichael becomes convinced that Billy Ray was murdered and vows to perform his own independent investigation. However, six weeks later, Mike is discovered hanging by a parachute cord from a locker handle inside his barracks office. Even though Mike's death is also ruled to be a suicide, his family believes that he was murdered in order to cover up the truth about Billy Ray's death. Episode 6: The DepartedRelease Date: 2006In this week's episode, we discuss one of our favorite Martin Scorsese films - the only film that won him his first and to date only Oscar - The 2006 multi-starrer, the mobster crime drama - The Departed. The Departed is a remake of the HK film Infernal Affairs with an all-star cast of Leonardo DiCaprio, Matt Damon, Jack Nicholson, Martin Sheen, Mark Wahlberg, and Alec Baldwin; Vera Farmiga plays the love interest of both Damon and DiCaprio's characters. The Departed was a critical and commercial success, receiving acclaim for its direction, performances, screenplay, and editing. It won several accolades, including four Oscars at the 79th Academy Awards: for Best Picture, Best Director, Best Adapted Screenplay, and Best Film Editing. We both watched it in the theatres when it was released and 17 years on when we rewatched it for this podcast, we still rated it high on entertainment but we did find a few loopholes in the script - however, the acting by Leo & Matt still remains the highlight of the movie along with the shocking deaths of the main characters. We did conclude that Scorsese loved Wahlberg or his character Digman over the rest of the cast - tune into the episode to find out why! Synopsis: South Boston cop Billy Costigan (Leonardo DiCaprio) goes undercover to infiltrate the organization of gangland chief Frank Costello (Jack Nicholson). As Billy gains the mobster's trust and climbs up the ranks, Frank implants his mole Colin Sullivan (Matt Damon) to infiltrate the police department and report its activities to him. Abstract: Chronic exposure to manganese (Mn) can lead to manganism, a neurological disorder sharing symptoms with those of Parkinson's disease (PD). Although the mechanisms of Mn toxicity are not fully understood, studies have shown that Mn increased the expression and activity of leucine-rich repeat kinase 2 (LRRK2), leading to inflammation and toxicity in microglia. LRRK2 G2019S mutation is the most common LRRK2 mutant associated with genetic PD and exhibits elevated LRRK2 kinase activity. Thus, we investigated whether Mn-increased microglial LRRK2 kinase function is responsible for Mn-induced toxicity and if it is exacerbated in G2019S using WT and LRRK2 G2019S knock-in mice as well as BV2 microglia cells. Results showed that Mn (MnCl2 30 mg/kg, nostril instillation, daily for 3 weeks) caused motor deficits, cognitive impairments, and dopaminergic dysfunction in WT mice, which were exacerbated in G2019S mice. Mn increased proapoptotic Bax, NLRP3 inflammasome, and inflammatory cytokines IL-1{beta} and TNF- in the striatum and midbrain of WT mice, which were exacerbated in G2019S mice. In addition, BV2 cells were transfected with either human LRRK2 WT or G2019S followed by exposure to Mn (250 uM) to compare Mn toxicity in two genotypes and gain more mechanistic insights. Results showed that Mn increased TNF-, IL-1{beta}, and NLRP3 inflammasome activation in BV2 cells expressing WT LRRK2, which was exacerbated in G2019S-expressing cells, while pharmacological inhibition of LRRK2 mitigated these toxicities in both genotypes. Moreover, the media from Mn-treated BV2 microglia expressing G2019S caused greater toxicity to CAD neuronal cells compared to media from microglia expressing WT. Mn-LRRK2 activated RAB10, an LRRK2 substrate, which was exacerbated in G2019S, and played a critical role in LRRK2-mediated Mn toxicity by dysregulating the autophagy-lysosome pathway, and NLRP3 inflammasome in microglia. Taken together, our findings suggest that microglial LRRK2 via RAB10 plays a critical role in Mn-induced neuroinflammation, and inhibition of LRRK2 kinase activity can be a potential target to mitigate Mn's inflammatory neurotoxicity. A number of Hollywood stars are joining the cast of the upcoming adult animated series Digman! Maya Rudolph, Danielle Radcliffe, and Edgar Wright are among a few stars who will be part of the show, with Brooklyn Nine-Nine star Andy Samberg in the lead. Samberg is stepping in as the voice of the titular character, Rip Digman. He will also serve as co-creator along with Neil Campbell. His Brooklyn Nine-Nine co-star Melissa Fumero will also play an important role in the show, and actors Jane Lynch, Kyle Mooney, Cole Escola, Lauren Lapkus, and Paul Rust are also joining the cast. This earthly life does not consist in accumulating a bunch of possessions and then losing everything when you die. So Jesus directs our attention away from all kinds of greed. Instead, we set our hearts and minds on things above and treasure the real life that we will enjoy with our Savior forever.
Series: Usually the more powerful something is, the harder it is to control. Not so with God. After Jesus returned to heaven, God unleashed his power in the world through his Holy Spirit. At the same time, he tied up that power to his Word so that we would always know where we can find it and how we can use it. Today: The more complicated something is, the more likely people are to avoid speaking of it. Why try to explain astrophysics or computer software to a three-year-old? Yet the triune God does the exact opposite with his divine nature. In the Scriptures God clearly communicates the majesty and mystery of the Trinity. Why? Not so that we would comprehend it with complete understanding but so that we would apprehend it with hearts of faith. The one eternal God -- Father, Son, and Holy Spirit -- makes himself known to each of us in order to bless us and keep us now and forever.
The Holy Spirit's life-giving Word removes the burden of trying to justify ourselves before God. We can't do that. Yet we don't have to, because Jesus Christ already has done that for us by his holy life and sacrificial death. Our deeds of love don't make us right with God. Christ alone does that. In fact, now that a “not guilty” verdict in God's courtroom is a certainty, we have been blessed with freedom. Freedom to produce the fruits of the Spirit. Freedom to love our neighbor.

Radio Rackham spørger Digman og Krarup om formens attraktioner og faldgruber, om grænser og om faren for at spille offerkort og dyrke usund narcissisme, og samtalen kommer så vidt, at Johan Krarup undervejs erklærer han har fortrudt alt have kastet sin tid og kærlighed over tegneserieformen.
