In episode 116, we take some time to look at the work of Schulz friend and fellow cartoonist Jim Sasseville. From ghosting for the Peanuts comic books to trying to launch his onw strip, where did Sasseville's style match up with Schulz and where did it diverge? Also, what's up with the nude models? (Yep. You read that correctly.) I'm Stacey Simms and these are the top diabetes stories and headlines of the past seven days. XX In the news is brought to you by T1D Exchange! T1D Exchange is a nonprofit organization dedicated to improving outcomes for the entire T1D population. https://t1dexchange.org/stacey/ XX Interesting new study looking at preventing type 1. The drug, currently known as IMCY-0098, blocks the immune system response that is thought to be the trigger. Very early on here, the Oxford scientists who are part of the study say it will likely be at least ten years before they'll be able to bring it to market, if it works. One of the most promising aspects is that the injection seems to only target production of cells that damage the immune system, so it doesn't seem to affect the body's ability to protect itself against other infections. https://www.dailymail.co.uk/health/article-10952755/Type-1-diabetes-stopped-strikes-thanks-simple-jab.html XX No surprise but glad this is being looked at. Older adults with type 1 diabetes using CGMs for the first time had lower A1Cs AND fewer lows. This study called WISDM - Wireless Innovation for Seniors with Diabetes Mellitus (WISDM) followed adults over 60 for six months. These researchers say the benefits of CGM combined with recently improved Medicare coverage should serve to increase adoption of CGM as standard of care in all adult populations, especially older people who are at increased risk of hypoglycemia. https://www.healio.com/news/endocrinology/20220627/cgm-improves-time-in-range-reduces-hba1c-for-older-adults-with-type-1-diabetes XX New resource thanks to the Helmsley Trust. This is for pharmacists and it's hoped it will expand patient access to CGMs. The release says Involving pharmacists in providing access is especially important for rural communities where they may be the most reachable healthcare professionals given the lack of diabetes specialists across the U.S. The guide was developed by the American Pharmacists Association and its Foundation http://ow.ly/zZ3c50Jz20f XX Looks like the Lilly Diabetes Journey Award Medals are getting a second life. As we reported back in January, after nearly 50 years, Lilly decided to end the program which gives a small medal for 10,25, 50 and 75 years of living with diabetes. The group Children with Diabetes – best known for putting on the Friends for Life Conference has taken it over. They'll now be called the CWD Journey Awards. You don't have to attend one of their conferences to get one and there is no cost. I'll give you the link to learn more and order one. XX The vets and keepers at the Royal Zoological Society of Scotland (RZSS) diagnosed the first reported case of diabetes in a giant anteater. Stephanie Mota, resident veterinary surgeon at RZSS said: “Keepers first discovered something was wrong when Nala was losing weight despite eating the same amount, or sometimes even more, than usual. “We carried out a full health check under general anaesthetic, running lots of tests and found that Nala has type 1 diabetes.” While the condition is known to occur in domestic cats, dogs and in tamanduas (Tuh-man-doo-uhs) in the wild, no other cases have been reported in giant anteaters. BTW a Tu-man-doo-uh is a smaller anteater and I spent far too much time today watching videos of them because I had never head of them and they are amazing. Dexcom donated the CGM to the Zoo's charity. The story I read here says. Due to her lovely personality, Nala is the ideal candidate for this technology which helps us, and her amazing team of keepers, manage her condition in the best possible way. https://www.edinburghnews.scotsman.com/news/people/edinburgh-zoo-monitor-uks-first-diabetic-giant-anteater-with-blood-glucose-monitor-usually-used-on-humans-3749422 XX Right back to the news in a moment but first As I mentioned, The T1D Exchange Registry is an online research study, designed to harness the power of individuals with type 1 diabetes. It's a research study conducted online over time, designed to foster innovation and improve the lives of people with T1D. Personal information remains confidential and participation is fully voluntary. By sharing opinions, experiences and data, patients can help advance meaningful T1D treatment, care and policy Sign up at T1DExchange.org slash Stacey (that's S-T-A-C-E-Y). XX Some great stories from our amazing community. First, Sebastien Sassevile completed the world's toughest ultra-cycling race, the Race Across America. It took 12 days and over 3-thousand miles. Sasseville who lives with type 1 basically spent 20 hours a day on his bike. He's the first person with type 1 to finish this grueling race. -- Don Muchow told us he was going to do it and the 60 year old has finished a 10k swim around Key West. I spoke to Muchow just after his 2021 Disney to Disney run – that's Disneyland in California to Disneyworld in Florida and he alluded to doing this race next. I've linked up his race report blog.. it's a very interesting read, especially how he treated a high blood glucose with more carbs. He came in second overall and first male in any age group. https://swimswam.com/60-year-old-iron-man-don-muchow-swims-around-key-west-for-diabetes-awareness/ https://t1determined.org/event-report-swim-around-key-west-10k/?fbclid=IwAR1U7XdI4_uw9OSe994TKuu7IuYjWC8FwHhby0Pm-RRzDIYAd9pGCMj6AD0 XX And finally, another former guest Dr. Nat Strand who works at the Mayo Clinic debuted a film she made with colleagues. It tells the story of what it is to be Black at Mayo Clinic, in healthcare, in America. Dr. Strand says, “By providing a platform for 12 courageous individuals to share their realities, we hope to move viewers toward active antiracism and start the conversation for many to continue.”   XX Next week you'll hear from Katie who started a podcast when she didn't hear the diabetes show she wanted.. that sounds familiar. You may know her online as Sugarmama Podcast. The long format episode out right now is all about the new documentary regarding stem cell research, talking to the director of “The Human Trial” i Listen wherever you get your podcasts That's In the News for this week.. if you like it, please share it! XX In the news is brought to you by T1D Exchange! T1D Exchange is a nonprofit organization dedicated to improving outcomes for the entire T1D population. https://t1dexchange.org/stacey/ XX Tandem's mobile bolus feature is in limited release. Approved earlier this year, they've released the feature to a very small number of users and are expecting a wider, still limited, release in the next few weeks. This is the feature that allows you to bolus by phone and use the t-connect app to see pump data on your phone as well. It's not full pump control via phone, but it's a big step forward in terms of convenience for users and for Tandem' next product, the MOBI pump – which won't have a screen and will have full phone control. I'm asking Tandem to come on the show and share more about the features and how it works. It's interesting that you can't dismiss alerts and alarms from the app and must do so on the pump itself. I'll link up the site you need to sign up for the limited release. You will need to update the software on your tslim x2 pump and take some additional online training. This is US only. https://www.tandemdiabetes.com/landing-pages/remote-bolus XX The Eversense E3 gets European approval. This is a partnership between Ascensia diabetes care and Senseonics. The E3 is the six month version of the implantable CGM system. It's also approved for insulin treatment decisions, which is a switch from the XL version already available in Europe. The E3 was approved in the US earlier this year and should be distributed in Europe in the fall. https://finance.yahoo.com/news/ascensia-diabetes-care-announces-european-060000125.html XX XX New guidelines for treating diabetes in the hospital. This is from the Endocrine Society, which last updated their guidelines ten years ago. New this time around, hospital use of continuous glucose monitoring and insulin pump therapy, providing inpatient diabetes education as part of a comprehensive diabetes discharge-planning process, use of noninsulin glucose-lowering therapies, and more. Adult patients with diabetes or newly recognized hyperglycemia account for greater than 30% of noncritically ill hospitalized patients. https://www.clinicaladvisor.com/home/topics/endocrinology-information-center/endocrine-society-issues-new-guidelines-for-hospitalized-patients-with-diabetes/ XX Works just as well and costs less.. that's the upshot of a new study on Basaglar, the copycat insulin to Lantus when it comes to type 2 diabetes. The findings come from 14 commercial health plans and Medicare Advantage plans. Basaglar was approved as a biosimilar insulin glargine by the US Food and Drug Administration in 2015. This was a large study of thousands of patients and also showed that there was better adherence to Basaglar, no reason for that was given, but it could be the lower cost. https://www.medscape.com/viewarticle/975440 XX Right back to the news in a moment but first As I mentioned, The T1D Exchange Registry is an online research study, designed to harness the power of individuals with type 1 diabetes. XX Very early but Swedish researchers have now identified a molecule that helps stimulate the growth of new insulin-producing cells, and uncovered how it works. These researchers looked at a molecule known as CID661578 and found that it binds to a protein called MNK2. In doing so, it allows two other proteins to interact at higher levels, which ultimately leads to greater beta cell regeneration. The team tested their molecule in zebrafish, and found that it lowered blood glucose levels when compared to a control group. In pig pancreas cells grown in the lab, the molecule was shown to trigger the formation of new beta cells, while human pancreas organoids given the molecule produced more insulin. Long way to go, but still interesting. XX Type 1 Endurance athlete Sebastien Sasseville is taking part in the race across American this week. Called the world's premier ultra endurance race, it's literally a cycling event from the West Coast to the East Coast with a maximum lenthg of 12 days. That mean they have to cover about 275 miles every day. The original concept runs all the way back to 1887 when newspaperman George Nellis rode across the country via railroad routes in 80 days. The more modern version began in the 1970s, when John Marino decided to see how fast he could get across the country on a bicycle. The first head-to-head race came in 1982, and there were four starters. It's been called a brutal version of the tour de France. Sasseville has been on the show before and I'll catch up with him after he recovers here. He's been up Mt Everest, ran across Canada and did the brutal race across the Sahara Desert. He's sponsored by Tandem. https://cyclingtips.com/2022/06/how-to-watch-race-across-america-raam-route-map-elevation-live-dot-watching/ XX On this week's long format episode, you'll hear my conversation with Sernova's CEO all about their cell therapy and the search for a functional cure for type 1. Next week, a little less technology.. a fun conversation with a woman frustrated with the limits of wearing her pump, especially with skirts, so she found a new solution. I'm Stacey Simms and these are the top diabetes stories and headlines of the past seven days. Check out my new guide with top tips to clear your diabetes clutter. Everything from how to start to where to donate and how to keep it from taking over your house. Head over to diabetes-connections dot com to organize your diabetes supplies! XX Some new information about Dexcom's upcoming G7, which has been submitted to the US FDA and in Europe. Latest clinical study show the MARD of the G7 is 8.2 for adults, compared to 9 for the G6. MARD is the Mean absolute relative difference and the lower the better here. G7 was even lower, 8.1 for kids. This is close to the same results they talked about last summer, but the group in the trial was bigger. G7 is expected to get approval in Europe this quarter and likely in the US much later this year. Our last longer format interview episode is with Dexcom's CEO and we go in depth on this. https://www.drugdeliverybusiness.com/dexcom-ceo-touts-unprecedented-performance-of-g7-in-clinical-trial-awaits-fda-decision/?fbclid=IwAR3G5_Fu9fhPfR0M3FzgCNCsFYmo4gDRDy5nJySgxt56mMVJgrpUsVQedis https://investors.dexcom.com/static-files/0c3012e2-40f4-4046-a962-85e6b421d490 XX Two COVID and diabetes studies I want to talk about.. the first showing that more children are being diagnosed with type 1 and type 2 diabetes after getting COVID. This study looked at databases of people under 18 starting in March 2020 and going for 18 months. There are a LOT of questions here.. including whether post-COVID type 2 diabetes will actual be a temporary or chronic condition. Which leads us to the other study.. this one of adults, in almost 4-thousand covid patients, just under half developed high blood sugar levels, including many who were not previously diabetic. These researchers say a lot of the patients here were in their 30s and 40s, no sign of diabetes before COVID.. and the levels of glucose in their blood were incredibly high, sometimes more than twice the level that indicates diabetes. These patients still had high levels of C-peptide, which shows that they were still producing insulin. The theory here is that something is disrupting the fat cells. But the researchers admit they are just at the beginning of figuring this out. It's this latter study and others like it that seem to be showing that there really is something different about COVID and blood glucose. I saw a lot of people dismissing the children's study with, well, any virus can cause T1D.  I'm glad these researchers are digging into what's going on. https://www.sciencenews.org/article/coronavirus-covid-diabetes-fat-cells-blood-sugar https://www.nytimes.com/2022/01/07/health/kids-covid-diabetes-cdc.html XX Abbott unveils plans for a new line of consumer bio-wearable sensors that will collect a broader range of biological readings to help users optimize their exercise and nutrition regimens and overall health. Called “Lingo”- which are still under development and aren't intended for medical use— they are based on the existing Freestyle Libre diabetes monitoring technology. We're talking about glucose, ketones, lactate and alcohol. Interesting to me that these were shown at the Consumer Electronics Show and not a medical conference, but Abbott is up front that these are basically for athletes and not for people with diabetes or those who need to make medical decisions based on the sensors. We'll see which of these makes it off the drawing board. https://www.fiercebiotech.com/medtech/abbott-ceo-ford-unveils-lingo-line-sports-biosensors-based-diabetes-monitoring-tech XX Another divestment for Lilly Diabetes – last week we told you they were doing away with their Journey Medals for diaversaries.. they have since announced that T1D everyday magic is no more. This was a partnership between Lilly and Disney that was a blog and a place for recipes and the home of those Coco books, the cute monkey with diabetes who hangs out with Mickey & Minnie Mouse. As of today you can still get digital versions of the books via the website but we'll see how long that lasts. Full disclosure: I wrote for them a couple of times – and was paid by them – glad I saved those columns. https://www.t1everydaymagic.com/thank-you-for-sharing-the-magic/ XX Interesting news from an amazing athlete with type 1. We told you that Sebastien Sasseville biked across Canada this summer. He previously went up Mt Everest and did a brutal race across the Sahara. Now, all the data on his blood sugar during that recent ride has been published. It's in the Journal of Diabetes, Obesity & Metabolism. Sasseville wore the Tandem and Dexcom Control IQ system during the ride.. the article is about how using that kind of automated insulin delivery system can help ultra athletes with diabetes.   https://dom-pubs.onlinelibrary.wiley.com/doi/10.1111/dom.14629?fbclid=IwAR2JLTn9dGJu43have5EMqXkWlENuDgPFDQq6RrsbY7TJyRJV0aERsDxYbo XX This week marks 100 years since the first person received a shot of insulin. Canadian teenager Leonard Thompson got that life saving injection on January 11, 1922. Of course, this was via Dr. Frederick Banting and his team.. Thompson was drifting in and out of a diabetic coma and weighed only 65 pounds. He was 14 years old. The first shot was found to be impure and didn't work. But they were able to fix the problem and administer a second purer shot. Thompson only lived to age 27 but his actions helped save so many lives. https://www.healthline.com/diabetesmine/diabetes-care-joslin-100-years-first-human-insulin-shot#A-century-of-Joslin XX Before I let you go, as I mentioned earlier, the podcast this week is an update from Dexcom. CEO Kevin Sayer answers your questions about the G7 and lots more. Listen wherever you get your podcasts or if you're listening to this as on a podcast app, just go back an episode. That's In the News for this week.. if you like it, please share it! Thanks for joining me! See you back here soon.

I'm Stacey Simms and these are the top diabetes stories and headlines of the past seven days.  As always, I'm going to link up my sources in the Facebook comments – where we are live – and in the show notes at d-c dot com when this airs as a podcast.. so you can read more when you have the time. XX In the News is brought to you by Inside the Breakthrough. A new history of science podcast full of “Did You Know… Stuff” XX The top story this week.. one step forward, one step back on a lawsuit involving all three insulin makers. A federal judge dropped anti-trust claims – but the case will move forward under federal racketeering laws. That means Eli Lilly, Novo Nordisk and Sanofi face organized crime charges. The Judge says the plaintiffs plausibly alleged schemes of unlawful bribery and mail and wire fraud. The other defendants in this case are pharmacy benefit managers Express Scripts, CVS Health, and UnitedHealth. Lilly, Novo Nordisk, and Sanofi are accused in the lawsuit of inflating the official list price of insulin while the actual prices negotiated by pharmacy benefit managers remained flat due to massive rebates from drug companies. https://news.bloomberglaw.com/pharma-and-life-sciences/sanofi-novo-insulin-price-suit-advances-with-trimmed-claims XX In a separate development, Lilly announced they have spent one billion dollars on Protomer Technologies – a company who's been working on what they call smart insulin. These proteins that can sense concentrations of specific molecules and adjust to create variable doses. This pipeline includes an insulin product that adjusts to different glucose levels in diabetic patients. In 2020 Lilly led an investment round that gave them 14% of the company. This deal gives them the rest. We've heard about glucose-responsive insulin for a while now, JDRF has funded a lot of the research. A spokesperson says this significant milestone brings the promise of the game changing technology one step closer to the clinic. https://www.biospace.com/article/lilly-buys-protomer-for-1-billion-plus-for-next-generation-insulin-products/ XX Mixed news for Provention, the makers of teplizumab. The US FDA says no to the drug shown to delay type 1 diabetes in those most at risk. It made it through an FDA advisory panel earlier this year, but the full FDA issued what's called a Complete Response Letter or CRL which means they've completed the review and won't approve in its current form. However, earlier this week, the company got good news from the UK. Teplizumab was awarded an Innovation Passport – a new designation that is meant to fast track investigational medicines through the regulatory process there. https://www.fiercebiotech.com/biotech/provention-bio-slapped-fda-rejection-for-diabetes-hopeful-teplizumab https://finance.yahoo.com/news/teplizumab-awarded-innovation-passport-united-110000773.html XX An Australian study shows good outcomes with closed loop therapy for older adults with long time type 1. This study looked at 30 healthy adults aged 60 and older who'd previously used an insulin pump. They used the Medtronic 670G either in manual or auto mode for four months at a time then switched to the other mode for another 4 months. They all spent more time in range when in the closed loop stage.. and biggest benefit was seen overnight. Less hypoglycemia was a big benefit for this group. These researchers say they want people to know that older age is not a barrier to closed-loop therapy and closed-loop has important clinical benefits Bit of an editorial here: I would not call the 670G or really any current commercial pump offering a closed loop. To me, if you're still bolusing for meals, that's a hybrid closed loop or partial loop, but potato potahto perhaps. https://www.healio.com/news/endocrinology/20210706/closedloop-system-improves-cgm-metrics-for-older-adults-with-type-1-diabetes XX Pixar released the teaser for it's next movie.. Turning Red and eagle eyed members of the DOC spotted what looks like a CGM and a purple insulin pump – maybe an older model Medtronic? I heard from a source at Pixar with type 1 who says this was absolutely on purpose It is definitely intentional to include better representation of the real world, whether it is wheelchair users, crutches, pumps/cgms, hearing aids, and more…  This initiative is largely credited to our characters art and crowds departments. I'm working with the Pixar PR folks and hope to have more info and an interview as the movie is closer to release. XX A class of drugs widely used to treat heartburn and stomach ulcers, improve blood sugar in patients with diabetes when added to their usual treatment. These drugs are known as proton pump inhibitors – they include Prilosec and omeprazole. They suppress acid and affect certain hormones that are important in glucose regulation. They did not prevent diabetes.. but these researchers say if somebody is already on a PPI and they are doing well, it might also be helpful for their diabetes. Those with higher A1Cs benefited the most. https://www.webmd.com/diabetes/news/20210702/antacids-aid-blood-sugar-control-in-people-with-diabetes XX More to come, but first, I want to tell you about one of our great sponsors who helps make Diabetes Connections possible. Inside the Breakthrough is a podcast that mixes historical wisdom with modern insight – it's a science show that's also entertaining. I love it. They cover everything from accidental discoveries to famous names in science to old myths hand washing to and even a horse poop crisis in New York City – luckily averted. And this all actually relates to diabetes! Listen to Inside the Breakthrough wherever you listen to podcasts.. Back to the news… XX A new attempt to climb K2 – the second highest mountain on earth - something accomplished by only one person with type 1 diabetes before. Right now, Jerry Gore – diagnosed as an adult - is acclimating at base camp. Gore is 60 years old and has been climbing for 40 years. He hopes to raise money for his charity, Action4Diabetes, which provides healthcare and support to disadvantaged young people with Type 1 diabetes in South East Asia. https://www.justgiving.com/fundraising/jerry-gorek2?experiments=b2c_059_donate_to_yourself_v3&successType=StaticDonateButtonClick?utm_source=twitter&utm_medium=socpledgemobile&utm_content=jerry-gorek2?experiments=b2c_059_donate_to_yourself_v3&successType=StaticDonateButtonClick&utm_campaign=post-pledge-mobile&utm_term=MdnpGx3vR XX Congrats to Sebastien Sasseville who just biked across Canada in 15 days. We told you about this trip when he kicked off two weeks ago.. I don't think anyone expected the weather to do what it did – Canada broke several heat records – and there were storms and wind. Sasseville lives with type 1, he did this to raise awareness for access to technology. He's climbed Everest and has completed several grueling extreme races & posted on his Instagram: “This was the hardest thing I have ever done, and could not have done it without the crew. They kept me safe and alive, I will be forever grateful. We did it boys!!! Chase life experiences and create memories with people you love. XX In the News is brought to you by Inside the Breakthrough. This new brand ReliOn Novolog will have vials priced at $72 and a package of pens at $85 – compared to the list price of $289 and $558 respectively Walmart worked directly with Novo Nordisk for the manufacturing of this insulin which will be in regular stores this week and in Sam's Club locations in mid-July. XX Insulet is out with the results of their latest pivotal trial for the Omnipod 5 Automated Insulin Delivery System in very young children. I can tell you from experience, this is a tough age group for many reasons, including the fact that they can't self-treat or really tell you what's going on. Parents and caregivers reported significantly improved sleep quality Another Omipod 5 study with people aged 6 through 70 showed similar improvements. The system is awaiting FDA approval. XX ViaCyte, Inc. released preliminary clinical data from its stem cell-derived islet cell replacement therapy for people with type 1 diabetes. They show the implanted pancreatic cells produced their own insulin – which increase glucose-responsive C-peptide levels, increased time in range, and reduced A1C. This method protects the stem cells in a pouch – that's put very simply – Viactye is working on several different methods toward a functional cure. https://www.prnewswire.com/news-releases/viacyte-reports-compelling-preliminary-clinical-data-from-islet-cell-replacement-therapy-for-patients-with-type-1-diabetes-301320084.html XX Another company on the same track.. Sernova Corp also showing their  Cell Pouch transplanted with insulin-producing islets is safe and effective. One patient has finished the trial and has now remained insulin independent – that means no requirement for injectable insulin - for 14 months with optimal glucose control. This is a person who's lived with type 1 for 47 years.. https://www.thenewswire.com/press-releases/1k3wFKYMv-sernovas-principal-investigator-presents-interim-data-and-positive-patient-outcomes-of-phase-i-ii-t1d-study-at-the-american-diabetes-association-81st-scientific-sessions.html XX More from ADA conference - In two small studies, the ultra-rapid acting insulin Afrezza was found to be safe in children with type 1 and adults with type 2 diabetes. Inhalable insulin is currently only approved for adults so the goal here was to look at safety.. which looks good. But it's also worth noting that . Time in Range increased from 39% at baseline to 62% which is more than four additional hours spent in range each day! https://diatribe.org/top-highlights-day-1-ada-scientific-sessions-2021 XX New info in on Medtronic's Extended-Wear Infusion Set. This is the pump inset that lasts seven days. This study shows that 75% of the time it makes it to the full seven days – Medtronic's 3 day infusion sets have a 67% survival rate. Device failure rates were very low – 8 cases out of more than 3-thousand sessions. Satisfaction was high – with people citing convenience of use, ease of insertion, and comfort of wear. This extended wear set is approved in Europe, available in Finland and has been submitted to the US FDA. https://diatribe.org/day-2-3-highlights-ada-scientific-sessions-2021   XX More to come, including news on Control IQ and an endurance athlete starts an amazing new trek.. but first, I want to tell you about one of our great sponsors who helps make Diabetes Connections possible. Inside the Breakthrough is a podcast that mixes historical wisdom with modern insight – it's a science show that's also entertaining. I love it. They cover everything from snake oil to the actual Eureka moment. There's even an episode about the guy who discovered the importance of hand washing in hospitals and how no one believed him. And this all actually relates to diabetes! Listen to Inside the Breakthrough wherever you listen to podcasts.. Back to the news… XX Control-IQ continues to get high marks. This study looked at adults with an average age of 70 who already had A1Cs around 6.9. Three months after staring Control IQ, time in range increased from 64% to 87% which is 5 and a half hours more each day.  All of these adults had prior experience with continuous glucose monitoring and insulin pumps. XX Former guest of the podcast, Sébastien Sasseville, is off on a mission to cycle across Canada in 14 days! Sasseville is a Canadian endurance athlete – he lives with type 1 -  and it's going to take about 250 miles a day to get to this goal. Sasseville says he's riding to bring attention to the issue of access to technology – he uses the tslim x2 and Dexcom G6 – and to support JDRF Canada's Access for All campaign. He set off on June 28th and is documenting the journey on social media.   Sebastien Sasseville cycles across Canada: https://www.facebook.com/CanadaRuns.   That's Diabetes Connections – In the News.  If you like it, share it. Today's guest is Sebastien Sasseville. Sebastien was diagnosed with Type 1 diabetes in 2002, the most severe form of diabetes. Sebastien vowed that the obstacle would never keep him from living life to the fullest, and today his story is a testament to how challenges can be turned into opportunities. In 2008, Sebastien reached the summit of Mount Everest. He then completed six Ironmans and the mythical Sahara race, a 250 kilometer, self-supported ultra-marathon over five days through the Sahara Desert. In 2014, he faced the biggest challenge of his life: run across Canada from St. John's, Newfoundland, to Vancouver: 7,200 km, or 170 marathons in nine months. His next big adventure is coming up very soon, (hint: it involves lots of cycling), but you'll have to listen to the interview to get the details. Sebastien also spent 10 years in the corporate world and blends that experience with his unique athletic achievements to deliver inspirational and motivational keynotes to large organizations. Per his LinkedIn profile, he works with performing organizations that have reached base camp, but are looking to transform, so as to dictate rhythm, stay at the forefront and get to the top. Some of his clients include Cisco, SAP, and Johnson & Johnson. In this interview, we get into what it's like for Sebastien to live with Type 1 diabetes, his corporate career and transition to keynote speaking, what it means for him to make a real impact, and his many feats of endurance. A., Francis, N. Abstract: Polycomb Group (PcG) proteins organize chromatin at multiple scales to regulate gene expression. A conserved Sterile Alpha Motif (SAM) in the Polycomb Repressive Complex 1 (PRC1) subunit Polyhomeotic (Ph) is important for chromatin compaction and large-scale chromatin organization. Like many SAMs, Ph SAM forms helical head to tail polymers, and SAM-SAM interactions between chromatin-bound Ph/PRC1 are believed to compact chromatin and mediate long-range interactions. To understand mechanistically how this occurs, we analyzed the effects of Ph SAM on chromatin in vitro. We find that incubation of chromatin or DNA with a truncated Ph protein containing the SAM results in formation of concentrated, phase-separated condensates. Condensate formation depends on Ph SAM, and is enhanced by but not strictly dependent on, its polymerization activity. Ph SAM-dependent condensates can recruit PRC1 from extracts and enhance PRC1 ubiquitin ligase activity towards histone H2A. Overexpression of Ph with an intact SAM increases ubiquitylated H2A in cells. Thus, phase separation is an activity of the SAM, which, in the context of Ph, can mediate large-scale compaction of chromatin into biochemical compartments that facilitate histone modification. Danny Sasseville has been with Constant Technologies for 23 years. Originally an architect and structural engineer for the company, he's now the VP of Project Engineering. Sassevillle joined host Lukas Cox to talk about implementing systems for quality control, the benefits of these systems and his experience with them at Constant Technologies. After the company evolved into the AV space, Sasseville moved to AV design. He said that his background is actually a benefit and that the projects he works on have more to do with architecture than engineering. One way that Constant Tech sustains its culture and grows as a company is by being purpose-driven. Making sure everyone is informed and on board with the overarching mission is important. Sasseville said that he and his engineering team developed their quality control system from the ground up. They took in many different perspectives to make sure anyone involved in the process can fully understand and implement their drawings. He said that the company has started a new position for quality control: a Director of Installation services. This position oversees all installations, making sure every action is done the same way for every project and process. The goal of the position, said Sasseville, is to implement and “follow a systematic sense of rules.” Sasseville said that making sure all departments are involved in what's going on in a project is “vital to our success.”

On any project, finishing quickly and efficiently is key, but this is especially true when products are assembled overseas then wired on the job site. Suddenly, it becomes a tremendous expense (and hardship) to try to get the correct pieces to the correct technician when everything could have been avoided by having a detailed plan in place and by going above and beyond in drawings. On this episode, Daniel Litwin, the Voice of B2B, speaks with Danny Sasseville, CTS-D AIA, vice president of project engineering at Constant Technologies, about the importance of detailed and engineered AV system design documents, and why they're crucial for a unique design. For starters, Sasseville mentions the importance of pre-sales communication with the clients' architects and designers. Asking lots of questions helps the provider determine whether they are giving the consumer what he or she wants and needs, and that all the purchased gear will be accurate and efficiently used. The next key to success is detailed drawings. Sasseville explains how, during his time in the AV industry, he noticed there wasn't a standard for the quality of design documents. He decided to change that at Constant Technologies, and singlehandedly introduced the Architectural and AV system design and engineering department. Now at the company, design engineers and installers must have correct specifications, know whether the resolution and cable quality can support the distance limitations, and detail all of the minutia in the drawings. Quality design documents don't just make the process easier; they save time and money. Scaling and testing the client’s hardware will no longer, and should never have been, tested on the load-in day. Proactivity, like applying “cable labels” to each individual piece of gear, is what's helping elevate companies like Constant Technologies to a more efficient future in AV.

Sebastien Sasseville grew up in a small town in Quebec Canada. From an early age, he was raised with a solid work ethic. This key characteristic was developed through the day to day work on his family's farm that Sebastien was responsible for. In our conversation, Sebastien explained that those early experiences taught him that anything is possible, but nothing comes easy.   It wasn't until he was diagnosed with type 1 diabetes at the age of 22, that he started to truly understand his potential. Sebastien initially ignored his symptoms, although he knew he was type 1 without the lab tests. A few years earlier his younger brother was diagnosed with type 1, so Sebastien had intimate knowledge of the disease.   As a result of his diagnosis, Sebastien quickly started to view his new reality as an opportunity to live a better life. He saw type 1 as a deep motivator and a source of energy, which gave him purpose.   In 2008, that purpose led him to realize a childhood dream, and took him to the peak of Mt. Everest.   In 2012, that purpose empowered him to run The Sahara Race, a 5 day, multi-stage, self-sustaining, 250K race across the desert.    And in 2014, he used his purpose and platform to spread more awareness about diabetes by running across Canada in 9 months.   Sebastien is also a six time Ironman competitor.   This is where I had a lightbulb moment with Sebastien. While all of those journeys are quite amazing, it's not about the achievements. Its not about the task of summiting Everest, or running across the Sahara, or even running across the North American continent. It's simply about what they mean.   In this conversation, we talk about the importance of patience, and we talk about the importance of preparation, and the details of training and the actual expeditions. But the key message to pull is Sebastien's message of motivation, of commitment, and the benefits of hard work so we can make our impact on this planet in some way. Ultimately, it's not about the enormity of the challenges each of us takes on necessarily, but it is about what they mean to us, and how those challenges support our mission.
