CHICAGO - Chicago health officials confirmed Wednesday that there is a 'small number' of tuberculosis (TB) cases among migrants in the city, and their medical teams are ramping up contact tracing. The Chicago Department of Public Health (CDPH) said the cases were reported in "a few different shelters" in the city; however, officials did not disclose the exact number of confirmed cases, or which shelter locations they originated from. "These outbreaks happen in close quarters, people who are living close to one another," said Dr. Aniruddha Hazra, associate professor of medicine, Infectious Diseases and Global Health at the University of Chicago. Abstract: The map of synaptic connectivity among neurons in the brain shapes the computations that neural circuits may perform. Inferring the design principles of neural connectomes is, therefore, fundamental for understanding brain development and architecture, neural computations, learning, and behavior. Here, we learn probabilistic generative models for the connectomes of the olfactory bulb of zebrafish, part of the mouse visual cortex, and of C. elegans. We show that, in all cases, models that rely on a surprisingly small number of simple biological and physical features are highly accurate in replicating a wide range of properties of the measured circuits. Specifically, they accurately predict the existence of individual synapses and their strength, distributions of synaptic indegree and outdegree of the neurons, frequency of sub-network motifs, and more. Furthermore, we simulate synthetic circuits generated by our model for the olfactory bulb of zebrafish and show that they replicate the computation that the real circuit performs in response to olfactory cues. Finally, we show that specific failures of our models reflect missing design features that we uncover by adding latent features to the model. Thus, our results reflect surprisingly simple design principles of real connectomes in three different systems and species, and offer a novel general computational framework for analyzing connectomes and linking structure and function in neural circuits. We are on Prosperity Principle number 10, which is I Create Daily Rituals. In order for you to be successful you must have habits. Successful people create daily habits that will guide them in their life, even when they don't feel like doing things that they need to do. Habits take over when their motivation runs out. You must create daily habits in order to drive your success, and you need to commit to your rituals on a daily basis. Be specific with what your intent is. Don't just go through the motions of what you read in books. Do things that are going to create specific intent (like we talked about yesterday) to help you reach your goals. Over time, your rituals are going to establish basic patterns of discipline in your life. So that when you don't feel motivated, when you don't feel like doing what you need to do, they carry you through. Let's be honest. None of us feel like doing what we need to do all of the time. The difference between successful people and unsuccessful people is that successful people create habits, rituals, and daily activities for success. So, how do you Set Yourself Up with Successful Rituals? Here are some strategies and things that you can do in order to set yourself up for success and create daily rituals that will guide you where you need to be. TIME OF DAY I would highly recommend that you analyze the time of day that's going to help you the most. I really like starting the day in the morning with a couple of habits that are going to get my day started right. If you're like most entrepreneurs, myself included, when you start your day out right, it tends to go in a great positive direction. When you don't start out right, it's hard for you to be able to catch up. Sometimes you feel like you're behind, and you feel like you're just playing catch-up. So, the time of day is important! I highly recommend the morning. Now, there are a lot of individuals out there that will try to tell you to get up at three, four, or o'clock in the morning in order to make this work. Don't listen to that if you are not a morning person. Just start where it works for you. Just start with something you know you will be consistent with. Analyze the best time of day for you and start setting yourself up for success. If it's going to be in the evenings or the afternoon, do that! But start today. SMALL NUMBER of ITEMS Second, start with a limited number of practices that you could do EVERY day. Because here's the problem: most of us overload ourselves. We create this big, long list. I've done it many times. You probably have too: Motivation Inspiration Meditation Affirmations Do this Do that Work out Pray Meditate Whatever it is Forget doing all of it! Set yourself up for success by just creating two or three things that you can commit to, and you can follow through with. If you don't commit and follow through with them, it's just going to be something that's going to set you back and make you feel like a failure. So be limited. Be specific. Be Simple with yourself to start. SET SPECIFIC TARGETS and GOALS Target the RITUALS that are going to help you make progress with growth in your life. Of course, we want everyone to have affirmations. We would love for everyone to work out. We would love for everybody to read or to meditate. We would love to know that everyone is getting some type of personal accountability. But what is most important for you. Target the thing that is what you need for your results. Also, to ensure that you're growing, don't target things that are always easy for you to do. Get outside your comfort zone as well. And it goes without saying that you need to set yourself up for success… If you say to yourself, “I'm going to work out,” and you don't have a gym membership ... Or you're going to work out, but you don't have anything laid out the night before ... Remember, when you get up in the morning you might be really tired as all of us are, and you'll just fall right back into bed. So set yourself up for success the night before. Let's face it, most of us don't feel like doing things we need to do “right when we're starting to do them”. So, you need to line yourself up for success. How do you do that? If it's working out, that's laying the clothes out the night before. If it's accountability, then get a partner get a mentor. If it's reading, have those books next to your bed or wherever you can get to them. Have the stuff ready so that you are lined up for success. RITUALS THAT CREATE RESULTS FOR YOU Use what works best for you. You know reading through books is great, but if you are better at listening to the audio books, listen to the audio books. If you feel like music motivates you more than a podcast, create a playlist of songs you love and that motivate you. If you need guided meditation vs. open meditation, do the guided meditation. It's better that you follow what's most effective for you, so you will be consistent…but more importantly so you get RESULTS. TRACK AND MONITOR YOUR RITUALS. Lastly track and monitor your daily rituals and your habits. If you don't track and monitor your rituals, then you're just going about it without any specific intent, accountability or direction. We are on Prosperity Principle number 10, which is I Create Daily Rituals. In order for you to be successful you must have habits. Successful people create daily habits that will guide them in their life, even when they don't feel like doing things that they need to do. Habits take over when their motivation runs out. You must create daily habits in order to drive your success, and you need to commit to your rituals on a daily basis. Be specific with what your intent is. Don't just go through the motions of what you read in books. Do things that are going to create specific intent (like we talked about yesterday) to help you reach your goals. Over time, your rituals are going to establish basic patterns of discipline in your life. So that when you don't feel motivated, when you don't feel like doing what you need to do, they carry you through. Let's be honest. None of us feel like doing what we need to do all of the time. The difference between successful people and unsuccessful people is that successful people create habits, rituals, and daily activities for success. So, how do you Set Yourself Up with Successful Rituals? Here are some strategies and things that you can do in order to set yourself up for success and create daily rituals that will guide you where you need to be. TIME OF DAY I would highly recommend that you analyze the time of day that's going to help you the most. I really like starting the day in the morning with a couple of habits that are going to get my day started right. If you're like most entrepreneurs, myself included, when you start your day out right, it tends to go in a great positive direction. When you don't start out right, it's hard for you to be able to catch up. Sometimes you feel like you're behind, and you feel like you're just playing catch-up. So, the time of day is important! I highly recommend the morning. Now, there are a lot of individuals out there that will try to tell you to get up at three, four, or o'clock in the morning in order to make this work. Don't listen to that if you are not a morning person. Just start where it works for you. Just start with something you know you will be consistent with. Analyze the best time of day for you and start setting yourself up for success. If it's going to be in the evenings or the afternoon, do that! But start today. SMALL NUMBER of ITEMS Second, start with a limited number of practices that you could do EVERY day. Because here's the problem: most of us overload ourselves. We create this big, long list. I've done it many times. You probably have too: Motivation Inspiration Meditation Affirmations Do this Do that Work out Pray Meditate Whatever it is Forget doing all of it! Set yourself up for success by just creating two or three things that you can commit to, and you can follow through with. If you don't commit and follow through with them, it's just going to be something that's going to set you back and make you feel like a failure. So be limited. Be specific. Be Simple with yourself to start. SET SPECIFIC TARGETS and GOALS Target the RITUALS that are going to help you make progress with growth in your life. Of course, we want everyone to have affirmations. We would love for everyone to work out. We would love for everybody to read or to meditate. We would love to know that everyone is getting some type of personal accountability. But what is most important for you. Target the thing that is what you need for your results. Also, to ensure that you're growing, don't target things that are always easy for you to do. Get outside your comfort zone as well. And it goes without saying that you need to set yourself up for success… If you say to yourself, “I'm going to work out,” and you don't have a gym membership ... Or you're going to work out, but you don't have anything laid out the night before ... Remember, when you get up in the morning you might be really tired as all of us are, and you'll just fall right back into bed. So set yourself up for success the night before. Let's face it, most of us don't feel like doing things we need to do “right when we're starting to do them”. So, you need to line yourself up for success. How do you do that? If it's working out, that's laying the clothes out the night before. If it's accountability, then get a partner get a mentor. If it's reading, have those books next to your bed or wherever you can get to them. Have the stuff ready so that you are lined up for success. RITUALS THAT CREATE RESULTS FOR YOU Use what works best for you. You know reading through books is great, but if you are better at listening to the audio books, listen to the audio books. If you feel like music motivates you more than a podcast, create a playlist of songs you love and that motivate you. If you need guided meditation vs. open meditation, do the guided meditation. It's better that you follow what's most effective for you, so you will be consistent…but more importantly so you get RESULTS. TRACK AND MONITOR YOUR RITUALS. Lastly track and monitor your daily rituals and your habits. If you don't track and monitor your rituals, then you're just going about it without any specific intent, accountability or direction. The Commerce Commission wants a 'please explain' after timber giant Carter Holt Harvey abruptly cut supplies to a number of building retail chains prompting concerns that costs will rise and construction could slow. The company has stopped selling structural timber to ITM and Mitre 10 but continues to supply to some of the bigger operators. The Commission says it was aware of supply issues in the building industry, however Carter Holt had not specifically advised the commission of its intention to cease supply to some customers. And it says it will speak to Carter Holt to understand what is happening and the potential ongoing implications for competition. Meanwhile the supply squeeze will end up hurting smaller building operators according to one industry insider. Marty Verry is chief executive of Red Stag - a privately owned independent timber company based in Rotorua. He says if the Kawerau mill gets up to speed, the supply should not be a major problem. Carter Holt Harvey has released a statement saying there have been some short-term industry-wide supply issues. It goes on to say it's continuing to meet its contractual obligations and the curtailment of supply relates to around 10 percent of the company's structural timber volume. It says there is no issue related to log supply, and it's not exporting structural timber.

Google alone accounts for nearly half of the external referral traffic—traffic, that is, that comes from platforms, apps, and other outside sources— to news sites. Together with the fact that Facebook referral traffic is on the wane, this means that Google’s search algorithm is now perhaps the most powerful mediator of online attention to the news. Google announced tons of new ad features yesterday - two of the more controversial ones within the SEO community are the discover ads that show up on the Google mobile home page and the gallery ads that make some of the search ads on the search results page massively. This week, WhatsApp began urging its 1.5 billion or so users to update their apps to get the latest security patch. The Facebook-owned company — which touts its "simple, secure" messaging — said it discovered spyware had been installed remotely on "dozens" of smartphones through the app. The hack affected both Apple and Android users. Without naming the company, WhatsApp described the hackers as "a private company that has been known to work with governments to deliver spyware." In this you will learn how to create more opportunities for yourself... albeit in a slightly counter-intuitive way. The Canvas Strategy In the book, Ego is the Enemy, Ryan Holiday introduced a stand out concept known as the “Canvas Strategy”. It stems back to his first job as a Hollywood assistant when he was given the advice, "make other people look good" if you want to succeed. He describes it as decent, yet fairly inadequate advice. He now believes he wouldn't have succeeded as he did if he had stuck to that approach. Instead, he chose to help people actually BE good rather than simply trying to make them look good. He wanted to build space so that people can become great at what they do. Not simply making them look better, but giving them what they need to actually BE better. His advice instead became: "find canvases for other people to paint on". What he describes as "The Canvas Strategy". A Counter-Intuitive Approach This is a startling difference. It can be applied in many areas of your daily life. In many ways it can feel counter-productive, but Ryan insists that the way to create more opportunities and to succeed without being driven by ego is: give away ideas, share your networks, produce abundance, and give it away. These are key concepts when it comes to being a gentle rebel, similar to what we talked about in Episode 217. Let go of those ego-driven feelings that eye others with suspicion and as competition. Remove the instinct to hoard opportunities and stop others from receiving them. And instead, to be frivolous, and actively build canvasses for others to do what they do best in ways that they are unrestricted in their ability to do great things. You Have Been Given Canvasses Everyone has experiences of being given a canvas to paint on. Whether from a teacher, a parent, a member of your family, or a stranger who saw potential in you and wanted you to give it a go reaching it. These experiences are ones in which the other person gave you the opportunity to shine, without any obvious desire for anything in return. They are people who you remember with fondness, and feel a deep bond towards. Background: Clinical chemical blood analysis including plasma electrolytes is routinely carried out for the diagnosis of various organ diseases. Phenotype-driven N-ethyl-N-nitrosourea (ENU) mouse mutagenesis projects used plasma electrolytes as parameters for the generation of novel animal models for human diseases. Methods: Here, we retrospectively evaluated the use of the plasma electrolytes calcium, chloride, inorganic phosphorus, potassium and sodium in the Munich ENU mouse mutagenesis project where clinical chemical blood analysis was carried out on more than 20,000 G1 and G3 offspring of chemically mutagenized inbred C3H mice to detect dominant and recessive mutations leading to deviations in various plasma parameter levels. Results: We identified a small number of animals consistently exhibiting altered plasma electrolyte values. Transmission of the phenotypic deviations to the subsequent generations led to the successful establishment of mutant lines for the parameters calcium and potassium. Published data from other phenotype-driven ENU projects also included only a small number of mutant lines which were generated according to altered plasma electrolyte levels. Conclusion: Thus, use of plasma electrolytes detected few mouse mutants in ENU projects compared to other clinical chemical blood parameters.
