Those who study neurological diseases and their underlying cellular and molecular mechanisms have a number of scientific models at their disposal. In vivo animal models, particularly those bearing targeted genetic modifications, remain the gold standard, especially when it comes to assessment of behavioral readouts and neurobiological disease mechanisms in vivo. Historically, animal models have been widely used for preclinical validation of drug efficacy and safety.  Increasingly there is a move away from this approach, and human cellular models using induced pluripotent stem cells and their derivatives such as three-dimensional organoid models have recently provided unprecedented genetic and mechanistic insights into disease mechanisms. Human/animal chimeras made by xenografting human cells into the animal brain have unparalleled potential to study human cells in the context of the whole organism. “All I will say is that my scientific opinion is that there is something real going on in New Brunswick that absolutely cannot be explained by the bias or personal agenda of an individual neurologist.” Those are the words of Dr. Michael Coulthart, a microbiologist employed by our federal government, specifically by Public Health Canada. He's talking about the mysterious neurological disease that is striking down people in New Brunswick. We were not supposed to read his words. They are from a leaked email obtained by Canadaland, first published by the newspaper The Guardian.Then there was another leaked email. Doctor Samuel Weiss, a neuroscientist and the scientific director of the Canadian Institutes of Health Research. In his leaked email, he states, “I felt incredibly optimistic that an all out government effort to unravel the mystery was in the cards. However, in short order, the scientific effort was shut down at the request of the governments.”It's nine years since the first case was documented. The numbers, by some reports, have grown to over 300 suspected cases. And we still have no answers. A brain researcher says anyone who's experienced repeated head injuries, or knocks, is at risk of developing chronic traumatic encephalopathy, or CTE.  The family of Billy Guyton, a former Blues and New Zealand Māori halfback who died last year aged 33, received a report from the Brain Bank this week after extensive testing, confirming a diagnosis of CTE. New Zealand Rugby says it is concerned about the possibility that repeated head impacts in rugby may contribute to neurodegenerative diseases in later life. Zach Rhoads - Zach is a behavior specialist in Vermont schools and his school's PBiS (Positive Behavior Interventions and Supports) coordinator. Zach is a counselor and family systems developer for the online Life Process Program - an addiction coaching program developed by psychologist Dr. Stanton Peele. He also writes articles about health and drug policy for Filter Magazine and is coauthor (with Dr. Stanton Peele) of the book, Outgrowing Addiction. (He is currently working on a forthcoming book about student-led education in the United States). Zach believes that community collaboration and cohesion is a bulwark against many forms of mental illness and addiction, and helps people find these resources in ways that makes sense in their own lives. In the mornings, Sonia Vallabh and Eric Minikel's first job is to get their two garrulous kids awake, fed and out the door to daycare and kindergarten. They then reconvene at the office and turn their focus to their all-consuming mission: to cure, treat, or prevent genetic prion disease. Prions are self-replicating proteins that can cause fatal brain disease. For a decade, Sonia Vallabh has been living with the knowledge that she has a genetic mutation that will likely cause in her the same disease that claimed her mother's life in 2010. Upon discovering she had the mutation, Sonia and her husband made a massive pivot: They went from careers in law and urban planning to earning their Ph.D.s, and founding a prion research lab at the Broad Institute of MIT and Harvard. On today's episode, Sonia and Eric talk with Short Wave's Gabriel Spitzer about what it's like to run a lab with one's spouse, cope with the ticking clock in Sonia's genes, and find hope in a bleak diagnosis.
