We employed rapid GABAergic neuron viral labeling plus unbiased Patch-seq sampling in brain slices to define the signature morpho-electric properties of GABAergic neurons in the human neocortex. Viral targeting greatly facilitated sampling of the SST subclass, including primate specialized double bouquet cells which mapped to two SST transcriptomic types. Multimodal analysis uncovered an SST neuron type with properties inconsistent with original subclass assignment; we instead propose reclassification into PVALB subclass. Our findings provide novel insights about functional properties of human cortical GABAergic neuron subclasses and types and highlight the essential role of multimodal annotation for refinement of emerging transcriptomic cell type taxonomies. Evolutionary expansion of human neocortex is marked by distinctive pyramidal neuron types with extensive branching in L1, but whether L1 interneurons are similarly diverse is underexplored. Using patch-seq recordings from human neurosurgically resected tissues, we identified four transcriptomically defined subclasses, unique subtypes within those subclasses and additional types with no mouse L1 homologue. Compared with mouse, human subclasses were more strongly distinct from each other across all modalities. Accompanied by higher neuron density and more variable cell sizes compared with mouse, these findings suggest L1 is an evolutionary hotspot, reflecting the increasing demands of regulating the expanding human neocortical circuit. This week in Airplane News: The AirCar has been issued an Airworthiness Certificate, Enstrom Helicopters goes bankrupt, Stowaway survives an 11-hour flight and the final update on FlyZolo. The AirCar has been issued a certificate of airworthiness by the Slovak Transport Authority. This certification included more than 70 hours of flight testing and 200 take-offs and landings. Doug Simons represents the third-generation of Enstrom's Colorado-based, family-owned business. Having worked part time at Enstrom's while attending Mesa College in the mid 70's, Simons first exposure to the industry was mucking-out butter fat that had been tracked into the freezers. He later married Jamee Enstrom and joined the business full time in 1979. Doug spent his formative years in the confectionery industry learning the art of candy making under the watchful tutelage of his father-in-law, Emil Enstrom and company founder, Chet Enstrom. He then worked his way through every aspect of the business, was named president of the company in 1986 and purchased the business in 1993. Simons was also the first in the family to have any formal confectionery training attending the Retail Confectioners International (RCI)'s Candy School in 1982. Simons is credited with turning a “Mom and Pop” business into a competitive, high-quality producer of confections. Under his leadership, two plant expansions have been constructed and he has brought innovation through technological advancements to both mail-order and manufacturing. Sometimes change sparks up innovation, and that's certainly what it did for Enstrom Candies during ECRM's recent Candy Planning: Easter & Valentine's Day Virtual Session. Jim Simons, the fourth-generation owner and VP of Sales for the manufacturer of all-natural almond toffee products, has been participating in ECRM in-person Candy Planning sessions for years, ever since they decided to expand from direct-to-consumer onto the store shelves. With our pivot to virtual sessions, Jim and his team wanted to create an experience that was as close to the in-person ECRM meetings as possible, and found some interesting ways to do it, building a set up in their conference room that enable them to share their catalog digitally and provide a view of their product and displays – all while keeping the face-to-face component of the meeting. In fact, they even plan to bring some of their learnings from their virtual presentation to ECRM's in-person sessions one we can all get together again! During this interview with ECRM VP of Content Joseph Tarnowski, Jim gives an overview of his company and products, discusses how they have adapted to the new normal of the pandemic, and relates his successful experience on ECRM's recent Virtual Session.

Teaching self-defense to young women in her late twenties, Rosalind Wiseman saw a world she never expected; one where girls and young women would destroy each other. Not physically, but emotionally and socially. And, it wasn't okay with her. So, she stepped in, started to listen - really listen - not to adults, but to the young women, then all kids, and began to build coalitions in an effort to create better ways to be with each other. That eventually led her to write Queen Bees and Wannabees, a book that stormed onto the scene, rattled people to their core and started not just a conversation, but a global movement.Wiseman has since devoted herself to helping communities shift the way we think about children and teens' emotional and physical wellbeing. Through her teaching, speaking, curricula which is now taught in schools around the world, and media appearances on relational aggression, ethical leadership, the use of social media, and media literacy, she works with educators, parents, children, and teens to help them navigate the power dynamics that influence their lives and relationships. This week Karen talks with her newest friend, Alicia Enstrom! Alicia is a Nashville based Violinist and Composer with an interesting and exciting career that has taken her all over the world. After attending Vanderbilt University for college, Alicia launched her career performing with Cirque Du Soleil and numerous other groups that involved everything from dancing while performing to kicking giant inflatable balls into the audience. Following several years on the road, living out of a suitcase, and exploring the cultures and cuisines of numerous countries, Alicia felt that it was time to “settle down” and finally unpack and put down some roots, and it only made sense that she would return to “Music City” to launch the new stage of her life. She saw Nashville as a chance to explore new playing opportunities and genres and discover everything that was there to be discovered. As the child of farming family parents, Alicia found herself involved in a wide range of extracurriculars that her parents believed would allow her and her siblings to have more opportunities than they did. Being involved in things like sports (including golf!), dance, and music, Alicia became a Jack of all trades and felt that it prepared her to be willing to take chances in her life and not be afraid to try new things. She strongly believes in being willing to fail and faking it until you make it. Since relocating to Nashville, Alicia has been keeping busy subbing with the Symphony, playing for recording sessions, composing, and most recently, working on producing her new EP. When it comes to maintaining her sanity and wellness as a musician, composer, and overall person, Alicia depends on quiet/alone time, self reflection, and exercise to remain centered and at peace. With such an interesting background and history filled with travel and other adventures, Alicia has experienced a great deal and had many ups and downs. If she were to provide one bit of advice for young aspiring musicians, it would be “Remember to Fail”. Alicia feels that it is her failures that have led to all of the bigger and better things in her life. It's important to remember that we are all human and that things happen! In 2004 Dale Thacker got involved as a 1/3 owner in Rangeland Helicopters with Larry Elford and Ian McLeod. Their wish was to bring effective helicopter EMS to the southeastern part of Alberta. They recognized that STARS Air Ambulance was available, but from a trauma perspective considering the “golden hour”, it wasn't sufficient, and wanted better coverage for the people who call this southern region home. The Southern Alberta MedicAir Society had also come to similar conclusions about STARS coverage. The combined synergy of these two groups allowed a new project to take flight in the summer of 2007. Dale sold his shares in Rangeland Helicopters at that time and became a volunteer on the fundraising side of the operation now known as HALO (Helicopter Air Lift Operation) He has been a board member for 13 years and Chairman for the last three. HALO has recently become an Alberta Health Services approved helicopter EMS provider, allowing them a chance to work together and demonstrate ways to help reduce costs of medical emergency trauma services. He will speak on his journey and experiences helping this service in southeastern Alberta become a reality. Speaker: Dale Thacker Dale was born in Taber and other than a spell at SAIT College studying Aircraft Maintenance Engineering, he has lived his entire life on a multi-generational farm 2 miles west of Bow Island. It's an innovative farming operation that grows Spearmint and Peppermint Essential Oils for Wrigley's, Colgate and other companies and produces dill oil for Vlasics and Oscar Meyer. They have held the contract to supply the flavoring for the dill pickle at McDonalds for the last 11 years. They also grow wheat, along with Hybrid Seed canola, peas, beans, lentils, hemp and are now even trying out hops. Dale is the Chairman of the Mint Industry Research Councils “distillation committee” and is tasked with improving on-farm distillation across North America. Dale got his fixed wing pilots license in 1975 and then upgraded to a helicopter license in 1999. This is where he discovered the helicopters unique ability to get to remote locations quickly. He currently flies an Enstrom helicopter on his farm and it allows for crop surveillance and irrigation scheduling using infra-red photography as well as weed scouting and crop maturity checks. The Southern Alberta MedicAir Society had also come to similar conclusions about STARS coverage. The combined synergy of these two groups allowed a new project to take flight in the summer of 2007. Dale sold his shares in Rangeland Helicopters at that time and became a volunteer on the fundraising side of the operation now known as HALO (Helicopter Air Lift Operation) He has been a board member for 13 years and Chairman for the last three. HALO has recently become an Alberta Health Services approved helicopter EMS provider, allowing them a chance to work together and demonstrate ways to help reduce costs of medical emergency trauma services. He will speak on his journey and experiences helping this service in southeastern Alberta become a reality. Speaker: Dale Thacker Dale was born in Taber and other than a spell at SAIT College studying Aircraft Maintenance Engineering, he has lived his entire life on a multi-generational farm 2 miles west of Bow Island. It's an innovative farming operation that grows Spearmint and Peppermint Essential Oils for Wrigley's, Colgate and other companies and produces dill oil for Vlasics and Oscar Meyer. They have held the contract to supply the flavoring for the dill pickle at McDonalds for the last 11 years. They also grow wheat, along with Hybrid Seed canola, peas, beans, lentils, hemp and are now even trying out hops. Dale is the Chairman of the Mint Industry Research Councils “distillation committee” and is tasked with improving on-farm distillation across North America. Dale got his fixed wing pilots license in 1975 and then upgraded to a helicopter license in 1999. This is where he discovered the helicopters unique ability to get to remote locations quickly. He currently flies an Enstrom helicopter on his farm and it allows for crop surveillance and irrigation scheduling using infra-red photography as well as weed scouting and crop maturity checks.
