Triple Therapy Slows Glioblastoma Growth and Extends Survival in Preclinical Study

BUFFALO, NY - April 4, 2025 – A new #research paper was #published in Oncotarget, Volume 16, on March 27, 2025, titled “Imipridones ONC201/ONC206 + RT/TMZ triple (IRT) therapy reduces intracranial tumor burden, prolongs survival in orthotopic IDH-WT GBM mouse model, and suppresses MGMT." Researchers from Brown University, led by first author Lanlan Zhou and corresponding author Wafik S. El-Deiry, have shown that combining a new class of drugs called imipridones with standard glioblastoma treatments significantly improves outcomes in mice. The study tested ONC201 and its analog ONC206 in combination with radiation therapy and the chemotherapy drug temozolomide (TMZ), a regimen referred to as IRT. This triple therapy slowed tumor growth and extended survival in a mouse model of glioblastoma, offering a potential new strategy for one of the most aggressive and treatment-resistant brain cancers. Glioblastoma is a fast-growing brain tumor with a poor prognosis and limited treatment options. Standard care typically includes surgery, radiation, and TMZ, but most patients still face a short life expectancy. While ONC201 and ONC206 are currently being studied in clinical trials as single agents, there has been limited information on how they interact with standard therapies. This study is the first to show that both drugs work synergistically with radiation and TMZ, strengthening their overall effects. The results showed that in both laboratory-grown tumor cells and mice, the triple therapy significantly slowed cancer cell growth, reduced tumor size, and prolonged survival compared to using any single or double treatment. Mice treated with IRT lived an average of 123 days, with some surviving more than 200 days—far longer than the 44 to 103 days observed with other treatment combinations. In addition to directly killing tumor cells, ONC201 and ONC206 lowered the expression of MGMT, a protein that helps tumors resist chemotherapy, making the treatment more effective. The researchers also found that the triple therapy reshaped the tumor environment. It decreased levels of harmful molecules that promote tumor growth and immune evasion while increasing signals that activate the immune system. This dual action—directly attacking tumors and boosting immune responses—adds to the potential impact of this treatment approach. “Overall, our preclinical findings support further exploration of the ONC201 and ONC206 IRT regimen as a potential treatment for GBM and diffuse gliomas with H3K27M mutations.” While these findings are based on preclinical mouse models, they offer strong support for advancing this triple therapy to clinical trials. ONC201 and ONC206 are promising due to their ability to cross the blood-brain barrier and enhance the effects of standard treatment. You'll know if there's a catastrophic noise being baked into the recording (electrical hum, static noise, buzzing sounds). 2. You won't be off-mic half of the time. (You can hear when you're in the sweet spot of your microphone, and when you're not). 3. Increased awareness of Plosives (hard P'S and B's), Mouth-noises (YUCK!), and Proximity Effect (overemphasised and potentially muddy bass in the voice). 4. Awareness of rubbing, zipping and scratching of clothing, jewellery, furniture, mic stand, and the microphone itself. 5. You can get more consistent levels between speakers on the recording, which can reduce post-production time and lessen the need for automated and potentially expensive levelling tools. 6. You can make choices about what environmental noises are actually making it into the recording. (You won't waste time continually referencing things that are barely audible on the actual recording). 7. You'll know if you're baking-in destructive or bad sounding audio effects into the recording (overly aggressive noise-gates, reverb effects, or too much compression and/or bass and treble boost). 8. When Benjamin Hanna assembled the schedule for his first season as artistic director of the Indiana Repertory Theatre, he placed “Little Shop of Horrors” as the closing production for the 2023-24 season. The IRT, founded in 1972, had not previously staged “Little Shop of Horrors”—which debuted in New York in 1982 and was adapted for a 1986 film. Perhaps more noteworthy is that “Little Shop of Horrors” is a musical. The IRT last staged a traditional Broadway musical in 2013, when Stephen Sondheim's “A Little Night Music” was part of the schedule. For this edition of the podcast, IBJ arts reporter Dave Lindquist talks with Benjamin Hanna and “Little Shop of Horrors” cast member Rob Johansen, who preview the production that opens April 17th. Rob has appeared in more than 50 productions at the IRT. This time, he's the puppeteer controlling Audrey Two, the otherworldly plant that requires human blood to thrive. He is currently serving as Senic Designer for IRT's season opener, Mary Shelley's Frankenstein. I get so real in this episode and share the struggles I have faced with burnout.  I thought I had it all together, but that always changes with life. The only constant in life is change.  However, as we all know, it is HARD AF to accept that we struggled to implement tools for a better life, have been promoting them, and now they are NOT working for us.  That is where I found myself.  I had to RE-CREATE everything for myself once the exhaustion set in.  And now, I am passing on ALL THE THINGS I am learning to you - IRT.  Because you deserve to learn the tools as quickly as I do.  They might even save you from getting as mentally exhausted as I am. ✅It's funny, Alexa tells me the synonyms for burnout include : neurosis, mentally exhausted, and collapse. Lethargy, weakness, passive, sluggish, frazzled, listless, drained, worn out.  And it is "funny" because people called me all these things.  BUT I did NOT see it as burnout.  I thought straight up - "Yeah, I AM EXHAUSTED!!" I should sleep ALL DAY.  When really, my body needed a break from work and to just be taken care of by the beach

Helplessness, Trauma, and Nightmares with Dr. Greg Mahr

Can we predict who will develop PTSD? Our guest, Dr. Greg Mahr, studies patients with acute trauma, and he discovered that their dreams in the days and weeks after their injury suggest who will recover fully and who will be mired in PTSD months later. Post Traumatic Stress Disorder is a condition that occurs after trauma marked by intense nightmares and increased risk of suicide, but only about 30% of those traumatized go on to develop PTSD. In Dr. Mahr's acute trauma center in Detroit, he interviews victims of car accidents and assaults and asks about their dreams in the days immediately after the trauma and finds that experiences of helplessness in their dreams and during their trauma correlate with the development of PTSD. After the traumatic event, nightmares tend to replicate the event exactly, but changes soon develop in those who recover in which the person finds some agency and the dreams eventually start to have happier endings. This mimics the process of IRT, Image Rehearsal Therapy, in which people can deliberately change their nightmares by imagining new versions of the troubling dreams. Dr. Mahr speculates that one of the purposes of dreaming is to encourage us to talk about our traumas and that they help us develop a narrative about the event. We take a question from Amy about how to bring up the topic of nightmares with a doctor and end by talking about post-traumatic growth and the idea that dreams are messages from ourselves.
