In part because there are over 10,000 known human diseases and symptoms thereof may have numerous possible explanations, frequently diagnostic tests can be in-determinative or less informative than observing (termed: watchful waiting) a suspected disease's clinical course over time.  Because of diagnostic complexities population level diagnostic errors represent a significant public health problem.  Nevertheless, despite the progress made in treating cancer - as Ropeik writes in his introduction two-thirds of nearly 200 types of cancer are either treatable as chronic diseases or entirely curable - cancer today remains the emperor of nosophobias that in turn leads to over-screening, over diagnosis and false positives, over treatment, potentially harmful side effects, death and excessive healthcare budgeting and wasteful spending.  As one reviewer of the book wrote, “Ropeik details how the gravity force of cancerphobia warps risk perception, leading to personal and societal harms and legislative misdirection.”  During this interview Mr. Ropeik begins by clarifying the book's discussion is at the population level, disputes the belief cancer always needs be diagnosed as soon as possible and describes the US Preventive Services Taskforce's (USPSFT's) work upon which his book is based. He next discusses USPSTF's (evolving) mammography screening recommendations for breast cancer, the prevalence of associated false positive diagnoses particularly related to DCIS (ductal carcinoma in situ). He discusses his chapter regarding environmental agents as cancer agents or contributing to the fear of cancer, the lack of a relationship between federal funding and the burden of disease, e.g., breast v pancreatic cancer funding. He concludes by discussing policy solutions that can simultaneously reduce cancer phobia and improve the effectiveness and cost cancer care. Mr. David P. Ropeik is a retired Harvard University Instructor, author, and international consultant on risk perception, risk communication, and risk management. He worked as a television reporter for WCVB-TV in Boston from 1978 – 2000 specializing on environment and science issues, wrote a science column for The Boston Globe, taught journalism at Boston University, Tufts University, and MIT, was a Knight Science Journalism Fellow at MIT and a member of the Board of Directors of the Society of Environmental Journalists. Mr. Ropeik previously published “How Risky Is It, Really? Why Our Fears Don't Always Match the Facts,” (2010, McGraw Hill), and co-author of “RISK, A Practical Guide for Deciding What's Really Safe and What's Really Dangerous in the World Around You,” (2002, Houghton Mifflin). Mr. Ropeik has also authored more than 50 articles, book chapters, and other essays on risk perception and risk communication published in The New York Times, The Washington Post, The Los Angeles Times, USA Today, Scientific American, The Atlantic Monthly, Health Affairs, Issues in Science and Technology and elsewhere. He writes a blog for Psychology Today and blogged at Big Think and The Huffington Post. Gábor Somlyai is a molecular biologist and world's leading expert in the area of deuterium in biology. He has been working for over 30 years to research and publish his findings on the role of deuterium depletion in cancer.Gábor graduated as a biologist from the University of Szeged in 1982. He received a scholarship from the Hungarian Academy of Sciences while studying for his PhD and defended his thesis in molecular biology in 1988.-In the wake of the Hungarian Nobel-prize winning scientist Albert Szent-Györgyi – who said that the true cause of cancer should be looked for at the sub-molecular level – Gábor began his examining the biological importance of naturally occurring deuterium as a senior research fellow at the Hungarian Institute of Oncology in 1990.-In 1993 Gábor established HYD LLC. for Cancer Research and Drug Development to carry out cancer research and drug development based on the proprietary procedure called deuterium depletion. NK Cells and Curing Cancer with Nina Horowitz

Nina is one of the up-and-coming superstars in biotech. Having just earned her PhD in bioengineering at Stanford in the Sunwoo Lab this summer, she has the scientific horsepower and storytelling ability to make a large impact on drug development, business, and the lives of patients. At the age of 8, Nina was diagnosed with an ovarian teratoma. In some ways, that shifted Nina towards a career in science and a mission to cure cancer. But while she was always interested in science, growing up in the suburbs of New York City enabled her to become world-class oboe and bassoon player as well. But in her words, she chose the research path rather than joining a conservatory because "science is a way to help other people [while] music is fun." At Williams College, she studied mathematics and biology and became interested in the progress going on in the cell therapy space, particularly CAR-T. This is around 2014 when companies like Juno Therapeutics, Novartis, and Kite Pharma were engineering patient T-cells to hone in on lymphomas. With a growing interest in bioengineering and the early clinical successes in CAR-T, Nina was compelled to join the LEAP program at Boston University to study bioengineering with a goal of engineering immune cells to target cancer. After applying to a number of graduate programs, Nina ended up at Stanford. She rotated through 5 labs: Irv Weissman, Garry Nolan, Crystal Mackall, Edgar Engleman, and John Sunwoo. These experiences ended up having a major impact on her research where she was able to have the flexibility to pursue the projects she was excited by in cell therapies and use various tools/methods she picked up from the other 4 groups in her graduate research. Early-on in grad school, her cancer relapsed and she was treated again. One thing about Nina is her grit. She was able to still move forward with her work despite all of this. Truly amazing. Her first project ended up not working out perfectly but taught Nina how to choose problems to work on. Michael Fischbach helps teach a class at Stanford, BioE 395, on this topic that Nina and I would recommend checking out. Her next project focused on studying subtypes of NK cells in tumors and generating functional data to figure out if they are a unique class of immune cells or an artifact of single-cell sequencing. After figuring out how to run an experiment with 1 month of prep, around 4 days of work, and then 3 weeks of data acquisition and analysis (shout-out to Gail for the help), Nina was able to discover a novel functionality of a new NK cell type that resembles ieILC1s, which have potent anti-tumor activity. Once she was able to show the experiment worked, Nina did the harrowing experiment another five times to verify her discovery and did an incredible job to combine various tools to map out new NK cell biology. Today's guest is Dr. Mary Lou Jepsen – serial entrepreneur, inventor, pioneer in VR, medical imaging and telepathic technology, former professor at MIT Media Lab and currently, founder of Openwater.Mary Lou spent many of her childhood years unwell before she was finally diagnosed with a brain tumour in her twenties. And going through that had a huge impact on her life as you might expect. For the many years she was quite literally dying, she decided she only wanted to work on really interesting projects - as she says to me in this interview, “I never really thought I would live very long and so I wanted to find really interesting things to do with my life for however long I might live.”Mary Lou became fascinated with holography and optics whilst at school and spent her career pioneering in VR and in the optics space as an engineering executive at Intuit, Google, Facebook, Oculus, and with her own four startups which included multi-billion dollar non-profit One Laptop Per Child.She is now using her decades long experience in this space and her experience with a brain tumour to spearhead Openwater where the goal is to create the technology to be able to see deep into the body with the detail of a 3D camera. When Jeff's wife got cancer six years ago, this diagnosis woke him up to the fact that healthcare is not designed to heal but to make money. So, Jeff and his wife took the journey to try to save her life. They went to Germany, healed her cancer naturally, and when he started talking about it on social media, was told that he should make a film about it.That film, Cancer Can Be Killed, Natural Ways of Curing Cancer – got made and then got taken down. We talk about censorship and how we have to find our own answers.Jeff had to do something bigger than he'd ever been as he saw the misinformation and suffering out there. He created a film to discuss the mind/body connection and how trauma creates disease. Making it even more special, Jeff decided to allow himself to be in front of the camera. Dr. Varani's current research on the gut, supplements, and inflammation ABOUT THE INSTRUCTOR Dr. Varani is an internationally recognized expert in human cell and organ culture, particularly models of skin. He has worked with a variety of biotechnology and large pharmaceutical companies, including Johnson and Johnson, Genentech, and Amgen. Utilizing in vitro and in vivo models he has supported retinoid, anti-inflammatory, immunomodulatory, and anti-cancer drug research. Dr. Varani's research focuses on the processes of skin homeostasis, aging, atrophy, and fibrotic skin disease. His studies examine regulation of fibroblast proliferation, signaling pathways controlling growth, extracellular matrix production, elaboration of matrix metalloproteinases, and their inhibitors. His group utilizes chemically-induced skin irritation to understand mechanisms of inflammatory skin disease such as psoriasis, drug-induced injury, and Rosacea. Dr. Varani has developed human colon organ models to understand the process of growth and differentiation in this tissue. These studies have implications for colon cancer and inflammatory bowel disease. Dr. Varani received his undergraduate degree from St. John's University in Collegeville, Minnesota and his Ph.D. at the University of North Dakota in 1974. After Post-Doctoral work at the University of Connecticut, he came to the University of Michigan as an Assistant Professor in 1980. He was promoted to Associate Professor in 1985 and became a Full Professor in 1991. Currently, Dr. Varani serves as professor emeritus at The University of Michigan.
