Ep 89 – George Floyd's Murder Was A Hit I realized that the facts did not add up, especially when I learned that the two men knew each other and had worked together at the same nightclub. Listed below are my notes for the podcast. Please do your homework and see if you come to the same conclusion. SUMMARY 1. George Floyd, a black man whose death Monday sparked ongoing protests, and Derek Chauvin, identified by local media as the officer who was videotaped kneeling on Floyd's neck for minutes shortly before his death, worked at the same nightclub Maya Santamaria owned El Nuevo Rodeo Club in Minneapolis' south side until selling it just months ago, and confirmed to local media outlets that both Floyd and Chauvin worked security at the establishment. Chauvin worked outside the club while security guards, like Floyd, worked inside. They had overlapping shifts. They had to have known each other. El Nuevo Rodeo Club, where Chauvin and Floyd both worked, is just down the street from Minneapolis' Third Precinct. 2. Chauvin married a Hmong woman, and two other arresting officers are of Hmong descent. 3. Hmong Gangs are very active in the Northeast, including the gang Menace of Destruction (MOD). MOD is known for prostitution, drugs, and counterfeiting. 4. Despite having Chauvin as off-duty security outside the club, the police could never solve the crimes or connect the suspects to El Nuevo Rodeo Club activity. 5. In 2012, the FBI busted a drug ring that involved the Mexican gang La Familia (also known for drugs and counterfeiting) and the Hmong gang Menace of Destruction. The ring involved the selling of methamphetamine, a “Club Drug” being pushed by Mexican cartels. EL NUEVO RODEO CLUB – MAY HAVE BEEN GROUND ZERO FOR COUNTERFEITING. Owned by Maya Santamaria, a singer, waitress, and music promoter with a degree in anthropology. The city is pushing to shut down the Rodeo. The nightclub's future lies in the hands of a judge who is deciding whether to recommend that the City Council yank its liquor license. In 2006, the club got slapped with fines for violating city code: It had hosted more people than the fire code allowed, had advertised as a nightclub when it was licensed as a restaurant/nightclub, and was Questioned about selling more alcohol than food (the liquor license requires that 60 percent of sales be food). That December, city officials convened to determine whether El Nuevo Rodeo's liquor license should be renewed. Santamaria agreed to submit her food and alcohol receipts for the next year. In April 2008, three people were shot outside of Denny's, right across the street from the nightclub. A story in the Longfellow/Nokomis Messenger connected the shooting to nightclub patrons, though police were never able to prove it. On November 14, 2008 there were multiple police calls associated with the nightclub. Incidents ranged from shots fired (almost an officer-involved shooting), to several felony assaults (on patrons and staff), to numerous misdemeanors (obstructing, disorderly conduct, assault, theft The city compiled its case against the nightclub: a stack of 28 police call reports from 2008 and six from 2009. The city attorney on the case described the business as having "an inordinately high number of incidents of criminal behavior." DEREK CHAUVIN – MAY HAVE BEEN IN HMONG GANG Records show that the 44-year-old Chauvin initially studied cooking before taking courses in law enforcement and doing two stints in the Army as a military police officer in the late 1990s, serving at Fort Benning, Georgia, and in Germany. Chauvin became a Minneapolis police officer in 2001. He had 17 complaints against him, including one for pulling a woman out of her car during a speeding stop. He won two medals of valor, one in 2006 for being part of a group of officers who opened fire on a stabbing suspect who pointed a shotgun at them and another in 2008 for a domestic violence incident in which Chauvin broke down a bathroom door and shot a suspect in the stomach. Married to Kellie Chauvin, a Laotian immigrant who became the first Hmong winner of the Mrs. Minnesota pageant. In 2009, he single-handedly apprehended a group of gang members while working as an off-duty security guard at the El Nuevo Rodeo, a Minneapolis nightclub. (Chauvin shoots most suspects but apprehends an entire gang BY HIMSELF. This does not add up). HMONG GANGS IN MINNEAPOLIS - MENACE OF DESTRUCTION" (MOD), FORMERLY KNOWN AS "MASTERS OF DESTRUCTION – LINKED TO “CLUB DRUGS” AND COUNTERFEITING is a Hmong street gang created in 1988. Today, it is active in every state, with a large Hmong community in California, the Midwestern United States, known for prostitution, money laundering, the distribution of narcotics, and counterfeiting. A national drug ring based in Twin Cities was busted after months of surveillance and wiretaps in March 2012. The cartel La Familia smuggled millions of dollars worth of methamphetamine from Mexico to Minnesota. The drugs then went through a local member of the cartel and the Menace of Destruction gang. La Familia is also known for counterfeiting. Eleven-year veteran and native Hmong speaker Tou Thao began as a community service officer and was the subject of six complaints. No longer chiefly made by "cooks" in makeshift labs in the U.S., methamphetamine is now the domain of Mexican drug cartels that are mass-producing high-quality quantities of the drug and pushing it into markets where it was previously unknown. While methamphetamine remains a street drug in many parts of the country, it functions as a “club drug” in the Northeast United States. While diffused to the Northeast, certain elements of the drug economy and networks of use have allowed it to retain its status as a club drug. Methamphetamine use has been found in a range of club subcultures, including electronic dance music clubs, raves, and circuit parties (Mansergh et al., 2001; McCaughan, Carlson, Falck, & Siegal, 2005; Yacoubian et al., 2004). THE ARREST – LOOKED LIKE AN EXECUTION 46-year-old George Floyd was arrested on suspicion of forgery on May 25. In a value-based care environment, clinical NLP can augment manual work by nurses, clinical coders, and clinicians to identify relevant, often unstructured information at the right time for specific patients, and payers to undergo chart review for risk adjustment. Calum explains, "Within healthcare, data and information are hugely fragmented. There was a HIMSS report in 2020 that said the average healthcare system in the U.S. has 15 disparate EMR systems it is pulling data from. Going back to your first question, the 360-degree view of a patient is about using as much data as available to understand patients and the populations they exist in. If it's difficult to access and make the most of the data. That's the first and probably one of the biggest barriers that we have in healthcare, this disparate data that exists in the ecosystem, and the multiple different formats that it exists in." "In the healthcare setting, huge amounts of clinical information is in text form, either dictated and transcribed or typed directly by clinicians into the free text boxes. These have become the kind of dumping ground for much clinically relevant information in a clinical setting. Ce 4ème mini-épisode hors-série entend combiner l'identité arabo-musulmane en France et la séropositivité au VIH. C'est un sujet qui me tient très à cœur et pour lequel il est indispensable d'effacer les stéréotypes. Selon l'ONU, 1,7 million de personnes ont contracté le VIH l'an dernier et plus de 40 millions de personnes vivent avec actuellement. Je ne vais pas vous faire un exposé sur les trithérapies, les sarcomes de Kaposi, l'inhibiteur de protéase ou l'épidémiologie historique de la maladie. Je vais plutôt vous montrer en quoi il est insoutenable de vivre avec son symptôme le plus persistant : la sérophobie. On m'a dit « elle je la touche pas avec un bâton, ça s'voit elle a l'DAS ». On a dit « regarde sa gueule à lui, il prend tellement cher qu'on dirait un zombie sidatique ». On m'a dit que « si j'avais le SIDA, ce serait de ma faute et qu'on viendra pas me soutenir à l'hôpital. ». On dit « si t'as le SIDA, c'est que t'es sûrement pédé ». Toutes ces remarques odieuses, c'est purement et simplement de la sérophobie, c'est-à-dire le rejet et la discrimination des personnes séropositives. Il existe une réelle discrimination sociale de la part de l'entourage, de l'employeur, du corps médical et des institutions… comme c'est bizarre, on dirait qu'on parle de ce que subissent certaines personnes musulmanes ou racisées ! Rappelons que Jean-Marie Le Pen voulait créer des « sidatoriums » pour isoler les porteurs du VIH et les traiter hors du cadre médico-hospitalier, des camps qui rappellent d'autres camps de la mort. En Égypte, plusieurs personnes porteuses du virus ont été incarcérées arbitrairement depuis 2007 pour la simple raison qu'elles étaient séropositives… Alors oui, il faut parler de l'intersectionnalité entre islam, culture arabe et cette sérophobie latente dont souffre les personnes qui vivent avec le VIH ou le SIDA. En France, 173 000 personnes vivent avec le VIH. 24 000 ignorent qu'ils sont porteurs. 1/4 découvrent leur séropositivité trop tard. Un quart des 15-24 ans (24%) pensent encore que le virus du Sida peut se transmettre par un baiser (ce qui est faux), selon un sondage mené en 2021 par l'Ifop. Bon alors, comment déconstruire la peur du VIH ? Comment éviter les conséquences psychologiques de la sérophobie sur les personnes concernées : rejet, exclusion, moqueries, discriminations ? Est-ce qu'il y a des similitudes entre sérophobie, islamophobie et racisme ? Est-ce Allah qui nous punit de nos actes de fornication ? Si vous voulez bouquiner

Dans notre épisode du jour, je vais parler de corps et de sexualité. On va essayer ensemble de comprendre comment braver l'interdit de la sacralisation absolue du corps, qui n'est scientifiquement qu'un tas d'os et d'organes, voué à périr. Contrairement à l'esprit, qui tisse notre rapport à la spiritualité, le corps est immanent, temporaire, fugitif, changeant et éphémère. Il est amené à se modeler, à s'éprouver, à se vivre avant de s'étioler dans une mort certaine. Il est donc profane et non pas sacré. Et si le corps était si sacré, pourquoi alors avoir rendu sale, bestial et vicieux le sexe qu'il porte ? Pourquoi avoir fait de la sexualité un péché originel ? Entre deux personnes éprouvant du désir l'un envers l'autre, il y a une limite spatiale (hududu Allah) qui tracent une frontière empêchant l'introduction du désir. Le corps est un territoire qui appartiennent à soi et à Dieu. En arabe, il y a plusieurs mots pour indiquer le corps. Cette frontière s'adresse au jassad جسد et non pas au jism جسم (qui est un corps masse, forme et substance) et au badn بدن (qui n'est que l'enveloppe, la cuirasse corporelle dans ses transformations physiques). Le jassad tend à désigner le corps dans sa dimension sensorielle, esthétique et affective. C'est le terme utilisé pour désigner le corps dans l'amour, où le sensible serait le siège de la volupté et de la jouissance. NB : on va aussi parler de TDS, c'est-à-dire de Travail Du Sexe, ou des travailleur·euse·s du sexe. Mon invitée du jour est connue sur Instagram elle se nomme @thepainproofpriestess, mais son vrai nom, celui qu'elle a décidé d'incarner, c'est Bebe MELKOR-KADIOR. Bebe est une afroféministe pro-sexe queer. Elle se définit aussi comme une femme noire, autrice, tantôt performeuse pornographique, tantôt fakir, parfois artiste & modèle, souvent comme travailleuse du sexe et fièrement comme « salope ». Oui, Bebe scande haut et fort la nécessaire réappropriation des corps des femmes et de leurs sexualités, départis des injonctions du patriarcat et des masculinités toxiques. C'est ce qu'elle écrit notamment dans son manifeste Balance ton corps, publié en 2020 aux éditions La Musardine. « Personne d'autre que moi ne peut me dicter comment vivre ma relation avec mon corps. Aujourd'hui, on va aborder une question complexe mais qui tend, comme d’habitude dans JINS, à réfléchir sur l’inclusion. Mais avant toute chose, je préfère déjà préciser que tous les mots insultants, argotiques et péjoratifs employés auparavant sont à oublier, je leur préférerais toujours le terme « travailleur·euse·s du sexe ». Car oui, il s’agit d’un travail. Au Maroc, l'expression « filles qui sortent » désigne celles qui fréquentent les night-clubs et les bars la nuit pour gagner leur vie. Au-delà du fait prostitutionnel, le « sortir » renvoie aussi plus largement aux distances qu’une partie de la jeunesse féminine des classes populaires prend avec les normes, la moralité et la respectabilité. Loin d’être confiné à la marginalité, le sortir joue sa partition dans les métamorphoses de l’ordre sexuel et intime dans un contexte d’essor de l’économie du divertissement et d’accroissement des inégalités. Y émergent de nouvelles valeurs qui remettent en cause les régimes moraux et juridiques tout en réaffirmant l’ordre hétérosexuel. Pour tenter d’y voir plus clair sur la très épineuse question de la prostitution, dans un contexte arabo-musulman, colonial et postcolonial, j’ai réussi à parler à une très grande pointure, spécialiste de ce sujet, il s’agit de Mériam CHEIKH. Membre du département des études arabes de l’INALCO, Mériam est une anthropologue spécialisée dans la dissidence morale des jeunes des classes populaires au Maroc. Son travail se concentre sur les processus de transformation de la sexualité, de l'intimité et des rapports de genre à l'œuvre au sein des contre-cultures juvéniles marocaines. Cohabitant avec une dizaine de jeunes femmes engagées dans le sortir à Tanger, Mériam Cheikh a mené une ethnographie longitudinale sur sept ans. À l’intersection de la génération, du sexe et de la classe, elle analyse des trajectoires où se succèdent socialisations familiale et scolaire, élaboration de la sexualité et insertions professionnelle et matrimoniale. Les Filles qui sortent. Abstract: Parkinson's disease and Dementia with Lewy Bodies are two common neurodegenerative disorders marked by proteinaceous aggregates composed primarily of the protein -synuclein. -Synuclein is hypothesized to have prion-like properties, by which misfolded -synuclein induces the pathological aggregation of endogenous -synuclein and neuronal loss. Rab27a and Rab27b are two highly homologous Rab GTPases that regulate -synuclein secretion, clearance, and toxicity in vitro. In this study, we tested the impact of Rab27a/b on the transmission of pathogenic -synuclein. Double knockout of both Rab27 isoforms eliminated -synuclein aggregation and neuronal toxicity in primary cultured neurons exposed to fibrillary -synuclein. In vivo, Rab27 double knockout mice lacked fibril-induced -synuclein inclusions, dopaminergic neuron loss, and behavioral deficits seen in wildtype mice with fibril-induced inclusions. Studies using AlexaFluor488-labeled -synuclein fibrils revealed that Rab27a/b knockout prevented -synuclein internalization without affecting bulk endocytosis. Rab27a/b knockout also blocked the cell-to-cell spread of -synuclein pathology in multifluidic, multichambered devices. This study provides critical insight into the role of Rab GTPases in Parkinson's disease and identifies Rab27s as key players in the progression of synucleinopathies. Abstract: Alpha-synuclein (syn) is the key component of proteinaceous aggregates termed Lewy Bodies (LBs) that pathologically define a group of disorders known as synucleinopathies, including Parkinsons Disease (PD) and Dementia with Lewy Bodies (DLB). Syn is hypothesized to misfold and spread throughout the brain in a prion-like fashion. Transmission of syn necessitates the release of misfolded syn from one cell and the uptake of that syn by another, in which it can template the misfolding of endogenous syn upon cell internalization. 14-3-3 proteins are a family of highly expressed brain proteins that are neuroprotective in multiple PD models. We have previously shown that 14-3-3{theta} acts as a chaperone to reduce syn aggregation, cell-to-cell transmission, and neurotoxicity in the in vitro pre-formed fibril (PFF) model. In this study, we expanded our studies to test the impact of 14-3-3s on syn toxicity in the in vivo syn PFF model. We used both transgenic expression models and adenovirus associated virus (AAV)-mediated expression to examine whether 14-3-3 manipulation impacts behavioral deficits, syn aggregation, and neuronal loss in the PFF model. 14-3-3{theta} transgene overexpression in cortical and amygdala regions rescued social dominance deficits induced by PFFs at 6 months post injection, whereas 14-3-3 inhibition by transgene expression of the competitive 14-3-3 peptide inhibitor difopein in the cortex and amygdala accelerated social dominance deficits. The behavioral rescue by 14-3-3{theta} overexpression was associated with delayed syn aggregation induced by PFFs in these brain regions. Conversely, 14-3-3 inhibition by difopein in the cortex and amygdala accelerated syn aggregation and cortical pyramidal neuron loss induced by PFFs. 14-3-3{theta} overexpression by AAV in the substantia nigra (SN) also delayed syn aggregation in the SN and partially rescued PFF-induced dopaminergic cell loss in the SN. 14-3-3 inhibition in the SN accelerated nigral syn aggregation and increased PFF-induced dopaminergic cell loss. Abstract: Nearly all human multi-exonic genes undergo alternative splicing (AS) via regulation by RNA-binding proteins (RBPs), but few studies have examined the temporal dynamics of AS and its regulation during cell differentiation in the bone niche. We sought to evaluate how AS, under the control of RBPs, affects cell fate commitment during induced osteogenic differentiation of human bone marrow-derived multipotent stem/stromal progenitor cells (MSPCs). We generated a time-course RNA sequencing (RNA-seq) dataset representative of induced MSPC differentiation to osteoblasts. Our analysis revealed widespread AS changes, coordinated with differential RBP expression, at multiple time points, including many AS changes in non-differentially expressed genes. We also developed a computational approach to profile the dynamics and regulation of AS by RBPs using time-course RNA-seq data, by combining temporal patterns of exon skipping and RBP expression with RBP binding sites in the vicinity of regulated exons. In total we identified nine RBPs as potential key splicing regulators during MSPC osteogenic differentiation. Perturbation of one candidate, KHDRBS3, inhibited osteogenesis and bone formation in vitro, validating our computational prediction of "driver" RBPs. Overall, our work highlights a high degree of complexity in the splicing regulation of MSPC osteogenic differentiation. Our computational approach may be applied to other time-course data to explore dynamic AS changes and associated regulatory mechanisms in other biological processes or disease trajectories. Interview - Gary Yacoubian President of SVS Speakers From the SVS Website: SVS was founded in 1998 by four audiophile/engineers who noted that customers were paying too much for lackluster subwoofer performance because of a manufacturing to sales process that was stacked against the consumer. The conventional model, where just good enough subs were sold at high markups did a disservice to customers who wanted great audio experiences but didn't have unlimited funds. To fix a broken system, SVS pioneered a disruptive, future-facing model by investing heavily in product engineering and performance and reducing operational costs by selling direct to its customers over the Internet. Customers could demo world-class subwoofers and speakers in their homes exactly where the products would be installed, allowing them to make the most informed purchase decision possible, without risk. This strategy allowed SVS to establish a global presence and continues to earn acclaim from professional and amateur reviewers, in forums, and at audio shows, while continuing to grow retail and direct distribution. UHD Blu-ray Many of you either have or will soon have a new UHD TV that will be begging you to throw some UHD content at it. Right now you can stream UHD content from Netflix and Amazon but while good, its not nearly as good as you can get from disc. There aren't any discs, or players, out there that can support UHD. But there will be in the near future. The Blu-ray Disc Association has released a new specification that may help you justify the purchase of a shiny new UHD TV. So what do the new players and disc give you? Better Color Simply put, the new specification allows your TV to display more colors than your current HDTV. You probably are thinking that your TV already does a good job with this but it can only display about 30% of what your eye can see. The new specification will display about 75%. At this writing we don't know of UHD TV that supports this spec nor do we know when we will see content that does. But its nice to know that your player will be ready when the content is there! High Dynamic Range (HDR) If you have a relatively new smartphone you may have heard of this term. On your phone your camera takes two (or more) shots of the same image, one with the darkest exposure and one with the lightest. Then it combines the best of the images to create one that shows great detail in all areas, nothing washed out and blacks looking black. This will provide depth and greater contrast on screen. 10-bit Bottom line on this one is that it makes better color and HDR possible. Higher Frame Rates Until Hollywood shoots at 60 frames a second, think of this as future proofing the standard. This will help 3D as well. Better Compression There is a lot of data that needs to be stored on a disc for 4K so a more efficient way of compressing it is required. That's where H.265 comes into play. The High Efficiency Video Codec (HVEC) is twice as efficient as H.264 (mpeg4). As a side note, HVEC is what makes streaming 4K from Netflix and Amazon possible at 7 to 10Mbps. Now imagine how good it would be at a little more than 100Mbps. Yes it will be a while before streaming catches up to fixed media. Larger Capacity All those bits need to be stored someplace. The new discs will have capacities of 66GB or 100GB Audio TBD
