Abstract: Microvillus Inclusion Disease (MVID), caused by loss-of-function mutations in the motor protein Myosin Vb (MYO5B), is a severe infantile disease characterized by diarrhea, malabsorption, and acid-base instability, requiring intensive parenteral support for nutritional and fluid management. Human patient-derived enteroids represent a model for investigation of monogenic epithelial disorders but are a rare resource from MVID patients. We developed human enteroids with different loss-of function MYO5B variants and showed that they recapitulated the structural changes found in native MVID enterocytes. Multiplex Immunofluorescence imaging of patient duodenal tissues revealed patient-specific changes in localization of brush border transporters. Functional analysis of electrolyte transport revealed profound loss of Na+/H+ exchange (NHE) activity in MVID patient enteroids with near-normal chloride secretion. The chloride channel-blocking anti-diarrheal drug, Crofelemer, dose-dependently inhibited agonist-mediated fluid secretion. MVID enteroids exhibited altered differentiation and maturation versus healthy enteroids. Inhibition of Notch signaling with the g-secretase inhibitor, DAPT, recovered apical brush border structure and functional Na+/H+ exchange activity in MVID enteroids. Transcriptomic analysis revealed potential pathways involved in the rescue of MVID cells including serum- and glucocorticoid-induced protein kinase 2 (SGK2), and NHE regulatory factor 3 (NHERF3). These results demonstrate the utility of patient-derived enteroids for developing therapeutic approaches to MVID. Copy rights belong to original authors. Abstract: For cultured meat to succeed at scale, muscle cells from food-relevant species must be expanded in vitro in a rapid and reliable manner to produce millions of metric tons of biomass annually. Toward this goal, genetically immortalized cells offer substantial benefits over primary cells, including rapid growth, escape from cellular senescence, and consistent starting cell populations for production. Here, we develop genetically immortalized bovine satellite cells (iBSCs) via constitutive expression of bovine Telomerase reverse transcriptase (TERT) and Cyclin-dependent kinase 4 (CDK4). These cells achieve over 120 doublings at the time of publication and maintain their capacity for myogenic differentiation. Way back in the early 1800's, several groups of textile factory workers in England took to destroying machinery in cotton and wool mills believing that the arrival of those machines threatened their jobs. They were led by a named Ned Ludd. Very quickly, they became known as Luddites. Now, the term Luddite refers to a person who is opposed to new technology. Currently, there's a group of teenagers in New York City who meet regularly at a local library with this rule: no smartphones. They call themselves the Luddite Club. One member, high school student Lola Shub was like many of her peers, obsessed with her phone. She was constantly snapping selfies and scrolling through social media. But when she saw the freedom a friend experienced when she ditched her smartphone, Shub did the same. She offers this advice to teens: “Spend time getting to know yourself and exploring the world around you, it's so much more fulfilling and real than the one inside your expensive little box.”

Happy Saturday, friends! Here in sunny and cold California, our shelter in place order has turned into a STAY HOME order, which is a little more restrictive. We've been sheltering in place now since March 18 and STAYING HOME starts today. We're still allowed to go to the store for essential needs, and we can go for walks while maintaining #socialdistancing, so it doesn't change things a ton for us. Today we took a nice family walk on a trail through rolling hills near our house, and had a good old time playing in trees and dry river beds and such. Normally, we would focus on 1st Thessalonians for today's pod, but it just so happens I'm preaching on 1st Thessalonians this Sunday, so I didn't want things to be redundant for our church family, many of whom listen to this pod. Which brings us to a fascinating passage in Psalms 7: My shield is with God, who saves the upright in heart. 11 God is a righteous judge and a God who shows his wrath every day. 12 If anyone does not repent, he will sharpen his sword; he has strung his bow and made it ready. 13 He has prepared his deadly weapons; he tips his arrows with fire. Psalms 7:10-13 That's kind of terrifying, right? I have a pretty nice bow, but I'm not a bow hunter. I also have a few nice swords, a couple of which are pretty deadly, but I am not a knight or Samurai warrior. If you got word that I was coming after you with my bow, I guess you should be pretty nervous, but honestly, I'm no Robin Hood, and you'd probably be fine, especially if you are moving. I'm a little better with the sword, but one time I chopped off the tip of my own toe with a sword, so you might escape from that situation pretty well too. However...to have God stringing His bow? That's just terrifying. Not only that, but He apparently has other DEADLY weapons and HE TIPS HIS ARROWS WITH FIRE? What in the world is going on here?! Let's read our Psalms passages and then come back and talk about what is going on. So - really scary stuff. Those who do not repent, God sharpens His sword, strings His bow (and those fiery arrows), and gets His other deadly weapons ready. Let's talk about repenting first. What does it mean? The Hebrew word is שׁוּב/Shub and it means to turn back or return. In the New Testament, the Greek word is Μετανοήσατε (Metanoēsate) and it is the FIRST word out of Peter's mouth when people respond to his message at Pentecost in Acts 2: 37 When they heard this, they were pierced to the heart and said to Peter and the rest of the apostles: “Brothers, what should we do?”38 Peter replied, “Repent and be baptized, each of you, in the name of Jesus Christ for the forgiveness of your sins, and you will receive the gift of the Holy Spirit. 39 For the promise is for you and for your children, and for all who are far off, as many as the Lord our God will call.” Acts 2:37-39 The word there means to change your mind. So both the Old Testament and New Testament words for repentance essentially mean to turn away from one direction and turn towards another direction - in the context of Psalms 7, to turn your mind and thinking away from sinful ways or human ways and to turn towards God's ways. As John Piper notes, the Greek word for “repent” refers to “a change of the mind’s perceptions and dispositions and purposes. . . . Repenting means experiencing a change of mind that now sees God as true and beautiful and worthy of all our praise and all our obedience.” Let's consider some other explanations of repentance from various pastors and theologians: The first principle we see [in learning about repentance] is the putting off and the putting on. We touched on it, but we have to hit it. The putting off and the putting on. Putting off means stop doing something, and putting on means start doing something. Everybody knows that change is two-factored, right? Everybody knows that in order to turn, metanoia (repentance means to turn), you can’t turn toward unless you turn away. You can’t turn away without turning toward something else. It’s two-factored. That seems so obvious. Timothy J. Keller, The Timothy Keller Sermon Archive (New York City: Redeemer Presbyterian Church, 2013). Repentance is a discovery of the evil of sin, a mourning that we have committed it, a resolution to forsake it. It is, in fact, a change of mind of a very deep and practical character, which makes the man love what once he hated, and hate what once he loved. Almost every time I turn to God and His Word and pray in the midst of this pandemic, I get the word to 'repent,' as I've mentioned before. I certainly believe that God is calling me to personally turn away from other things and turn towards Him in this season, I also believe that is call going out to all people in all nations right now, so I am very, very attuned to the call of God in His Word to repent, and I am extremely moved now, more than ever, of the truth of 2nd Chronicles 7: 13 If I shut the sky so there is no rain, or if I command the grasshopper to consume the land, or if I send pestilence on my people, 14 and my people, who bear my name, humble themselves, pray and seek my face, and turn from their evil ways, then I will hear from heaven, forgive their sin, and heal their land. 15 My eyes will now be open and my ears attentive to prayer from this place. 2 Chronicles 7:13-15 Ponder that passage and pray it. Listen to the Word of God in this season and respond to the leadings of His Spirit. Allow me to close with a great partial message from John Piper on Jesus' call to repent: Unless you repent, you will all likewise perish. (Luke 13:3, 5) The first demand of Jesus’ public ministry was, “Repent.” He spoke this command indiscriminately to all who would listen. It was a call for radical inward change toward God and man. Two things show us that repentance is an internal change of mind and heart rather than mere sorrow for sin or mere improvement of behavior. First, the meaning of the Greek word behind the English “repent” (metanoeo) points in this direction. It has two parts: meta and noeo. The second part (noeo) refers to the mind and its thoughts and perceptions and dispositions and purposes. The first part (meta) is a prefix that regularly means movement or change.1 So the basic meaning of repent is to experience a change of the mind’s perceptions and dispositions and purposes. The other factor that points to this meaning of repent is the way Luke 3:8 describes the relationship between repentance and new behavior. It says, “Bear fruits in keeping with repentance.” Then it gives examples of the fruits: “Whoever has two tunics is to share with him who has none, and whoever has food is to do likewise” (Luke 3:11). This means that repenting is what happens inside of us that leads to the fruits of new behavior. Repentance is not the new deeds, but the inward change that bears the fruit of new deeds. Jesus is demanding that we experience this inward change. Why? His answer is that we are sinners. “I have not come to call the righteous but sinners to repentance” (Luke 5:32). What was Jesus’ view of sin? In the parable of the prodigal son, Jesus describes the son’s sin like this: “He squandered his property in reckless living . . . [and] devoured [it] with prostitutes” (Luke 15:13, 30). But when the prodigal repents he says, “Father, I have sinned against heaven and before you. I am no longer worthy to be called your son.” Therefore, throwing your life away on reckless living and prostitutes is not just humanly hurtful; it is an offense against heaven—that is, against God. That’s the essential nature of sin. It’s an assault on God. We see this again in the way Jesus taught his disciples to pray. He said that they should pray, “Forgive us our sins, for we ourselves forgive everyone who is indebted to us” (Luke 11:4). In other words, sins that God forgives are compared to the ones people commit against us, and those are called debts. Therefore, Jesus’ view of sin was that it dishonored God and put us in debt to restore the divine honor we had defamed by our God-belittling behavior or attitudes. That debt is paid by Jesus himself. “The Son of man came . . . to give his life a ransom for many” (Mark 10:45). But for us to enjoy that gift he says we must repent. Repenting means experiencing a change of mind that now sees God as true and beautiful and worthy of all our praise and all our obedience. This change of mind also embraces Jesus in the same way. We know this because Jesus said, “If God were your Father, you would love me, for I came from God.” Seeing God with a new mind includes seeing Jesus with a new mind. No one is excluded from Jesus’ demand to repent. He made this clear when a group of people came to him with news of two calamities. Innocent people had been killed by Pilate’s massacre and by the fall of the tower of Siloam (Luke 13:1-4). Jesus took the occasion to warn even the bearers of the news: “Unless you repent, you will all likewise perish” (Luke 13:5). In other words, don’t think calamities mean that some people are sinners in need of repentance and others aren’t. All need repentance. Just as all need to be born anew because “that which is born of the flesh is [merely] flesh” (John 3:6), so all must repent because all are sinners. When Jesus said, “I have not come to call the righteous but sinners to repentance” (Luke 5:32), he did not mean that some persons are good enough not to need repentance. He meant some think they are (Luke 18:9), and others have already repented and have been set right with God. For example, the rich young ruler desired “to justify himself” (Luke 10:29) while “the tax collector . . . beat his breast, saying, ‘God, be merciful to me, a sinner!’ [and] went down to his house justified [by God!]” (Luke 18:13-14). Therefore, none is excluded. All need repentance. And the need is urgent. Jesus said, “Unless you repent, you will all likewise perish.” What did he mean by perish? He meant that the final judgment of God would fall on those who don’t repent. “The men of Nineveh will rise up at the judgment with this generation and condemn it, for they repented at the preaching of Jonah, and behold, something greater than Jonah is here” (Matthew 12:41). Jesus, the Son of God, is warning people of the judgment to come, and offering escape if we will repent. If we will not repent, Jesus has one word for us, “Woe, to you” (Matthew 11:21). This is why his demand for repentance is part of his central message that the kingdom of God is at hand. “The time is fulfilled, and the kingdom of God is at hand; repent and believe in the gospel” (Mark 1:15). The gospel—the good news—is that the rule of God has arrived in Jesus to save sinners before it arrives at his second coming in judgment. So the demand to repent is based on the gracious offer that is present to forgive, and on the gracious warning that someday those who refuse the offer will perish in God’s judgment. After he had risen from the dead Jesus made sure that his apostles would continue the call for repentance throughout the world. He said, “Thus it is written, that the Christ should suffer and on the third day rise from the dead, and that repentance and forgiveness of sins should be proclaimed in his name to all nations, beginning from Jerusalem” (Luke 24:46-47). So the demand of Jesus to repent goes to all the nations. It comes to us, whoever we are and wherever we are, and lays claim on us. This is the demand of Jesus to every soul: Repent. Be changed deep within. We've been sheltering in place now since March 18 and STAYING HOME starts today. We're still allowed to go to the store for essential needs, and we can go for walks while maintaining #socialdistancing, so it doesn't change things a ton for us. Today we took a nice family walk on a trail through rolling hills near our house, and had a good old time playing in trees and dry river beds and such. Normally, we would focus on 1st Thessalonians for today's pod, but it just so happens I'm preaching on 1st Thessalonians this Sunday, so I didn't want things to be redundant for our church family, many of whom listen to this pod. Which brings us to a fascinating passage in Psalms 7: My shield is with God, who saves the upright in heart. 11 God is a righteous judge and a God who shows his wrath every day. 12 If anyone does not repent, he will sharpen his sword; he has strung his bow and made it ready. 13 He has prepared his deadly weapons; he tips his arrows with fire. Psalms 7:10-13 That's kind of terrifying, right? I have a pretty nice bow, but I'm not a bow hunter. I also have a few nice swords, a couple of which are pretty deadly, but I am not a knight or Samurai warrior. If you got word that I was coming after you with my bow, I guess you should be pretty nervous, but honestly, I'm no Robin Hood, and you'd probably be fine, especially if you are moving. I'm a little better with the sword, but one time I chopped off the tip of my own toe with a sword, so you might escape from that situation pretty well too. However...to have God stringing His bow? That's just terrifying. Not only that, but He apparently has other DEADLY weapons and HE TIPS HIS ARROWS WITH FIRE? What in the world is going on here?! Let's read our Psalms passages and then come back and talk about what is going on. So - really scary stuff. Those who do not repent, God sharpens His sword, strings His bow (and those fiery arrows), and gets His other deadly weapons ready. Let's talk about repenting first. What does it mean? The Hebrew word is שׁוּב/Shub and it means to turn back or return. In the New Testament, the Greek word is Μετανοήσατε (Metanoēsate) and it is the FIRST word out of Peter's mouth when people respond to his message at Pentecost in Acts 2: 37 When they heard this, they were pierced to the heart and said to Peter and the rest of the apostles: “Brothers, what should we do?”38 Peter replied, “Repent and be baptized, each of you, in the name of Jesus Christ for the forgiveness of your sins, and you will receive the gift of the Holy Spirit. 39 For the promise is for you and for your children, and for all who are far off, as many as the Lord our God will call.” Acts 2:37-39 The word there means to change your mind. So both the Old Testament and New Testament words for repentance essentially mean to turn away from one direction and turn towards another direction - in the context of Psalms 7, to turn your mind and thinking away from sinful ways or human ways and to turn towards God's ways. As John Piper notes, the Greek word for “repent” refers to “a change of the mind’s perceptions and dispositions and purposes. . . . Repenting means experiencing a change of mind that now sees God as true and beautiful and worthy of all our praise and all our obedience.” Let's consider some other explanations of repentance from various pastors and theologians: The first principle we see [in learning about repentance] is the putting off and the putting on. We touched on it, but we have to hit it. The putting off and the putting on. Putting off means stop doing something, and putting on means start doing something. Everybody knows that change is two-factored, right? Everybody knows that in order to turn, metanoia (repentance means to turn), you can’t turn toward unless you turn away. You can’t turn away without turning toward something else. It’s two-factored. That seems so obvious. Timothy J. Greetings! Welcome to the show today. Heroes— You know, I think it is very important for each and every one of us to have heroes; men and women of God we look to to inspire us and show us how to truly live for God. One of the heroes of scripture who inspires me the most is the prophet Daniel; a man taken into captivity to Babylon, which was the reigning world empire and forced to live amongst a strange people in a strange land surrounded by all sorts of false gods and various temptations. And yet in the midst of all that, he remembers to whom he really belongs and remains true to God in the face of (what seem at least to me to be) insurmountable difficulties. Backdrop— Now, just to give you a little background, Israel (because of their unfaithfulness to God) had been sent into exile. God had made a covenant promise to His people to be faithful to them; to prosper them; to bless them. Yet that covenant was contingent upon obedience to Him. They were to remain consecrated, set apart for Him. It was their obedience to God that would allow His presence to dwell with them in the temple; and it was that presence that would bless them. But if Israel was unfaithful to God, then God promised judgment on them. We see this clearly in Deuteronomy 28, which says… Deuteronomy 28:15 (ESV)— 15 “But if you will not obey the voice of the Lord your God or be careful to do all his commandments and his statutes that I command you today, then all these curses shall come upon you and overtake you. Deuteronomy 28:36a (ESV)— 36 “The Lord will bring you and your king whom you set over you to a nation that neither you nor your fathers have known…. Deuteronomy 28:64a (ESV)— 64 “And the Lord will scatter you among all peoples, from one end of the earth to the other,… Temple Destroyed— Now Israel, unfortunately, did not pay heed to this. They began worshiping the same gods and engaging in the same practices as the pagan nations. So God had to bring judgment. Israel was attacked and many Israelites were carried away into exile into Babylon. And we know that Daniel was among them. And not long after that the temple in Jerusalem, the center of their worship and the zenith of their faith; the very temple that was built and dedicated by Solomon, was destroyed. What This Did to Them— Now we cannot even begin to imagine what this was like. Jerusalem and particularly the temple were the heart of Jewish worship. In Jerusalem you had the monarchy, which had now ended. The temple in which the presence of God dwelt was destroyed (something they thought could never happen). And with the temple went their ability to function the way that God had intended them to. With the temple went the sacrifices and the priesthood. They lost everything. And eventually that is where all sin leads us; it leaves us destitute. It takes away everything from us. Unpopular Themes— Now we often do not like to talk about the judgment of God on sin. In fact in many churches nowadays if a pastor wants to start talking about judgment or sin, he probably ought to start packing his bags; because he very likely will not be at that church very long. Judgment and sin are very unpopular topics. Yet both are necessary before we can begin to understand God's grace and redemption. Because the message of Grace isn't that God overlooks sin, but that God's righteous judgment on sin was born by Jesus Christ. God's Judgment is Always Redemptive— Furthermore, those who look negatively at this biblical theme of judgment don't understand the true nature of God's judgment, which is always redemptive. God judges us (ultimately) because He wants to preserve us. You see, it is sin that destroys. And therefore God's judgment is always aimed towards liberating us from bondage to sin. It is always redemptive. Many of us love… Jeremiah 29:11 (ESV)— 11 For I know the plans I have for you, declares the Lord, plans for welfare and not for evil, to give you a future and a hope. Written to Those Under God's Judgment— Many of us know this verse by heart. But what many don't realize is that this verse was written to a people who were in exile; right in the center; the very heart of God's judgment. Even Daniel's name means “God is my judge.” The Promise of Restoration— And this reveals something remarkable to us about the way that God judges. Whenever God judges, He always leaves His people with hope; the promise of their restoration if they but turn back to Him. Even in His judgment He already begins to show them His mercy; His promise. And this pattern stretches all the way back to Genesis to the Fall of mankind in the Garden of Eden. Skins of Grace— Now usually when we talk about the Fall in those first chapters of Genesis we focus in on this theme of God's punishment. And I don't want to downplay that. But I think what we often miss is also a deep expression of God's loving provision. Adam and Eve had sinned against God; they had turned their backs on Him; and the Bible says they realized they were naked and hid. And God comes to them and lovingly leads them to acknowledge their sin. And afterwards He must, because He is a Holy God, deal with their sin. They are cast out of the Garden of Eden. Yet God does not send them out of the Garden naked and afraid, covered in nothing but the fig leaves they had tried to manufacture to cover themselves, though He had every right to. Rather, He takes an animal and kills it; He removes the skin of the animal and clothes them with that skin (foreshadowing the whole animal sacrificial system). I just think, what an amazing picture; that even though sin brought upon them shame (justly), God covers that shame with the skin of an animal. The death of that animal removed their shame. Even in the skin of that animal God was giving them hope; because He would eventually allow another to have His flesh torn and pierced; His very Son. And that death would be the death to end all death; to completely remove our shame and unite us again with the presence of God. As the skin of the animal covered them, foreshadowing sacrifice and the whole sacrificial system, so also it foreshadowed the day when we would be covered by the blood of Christ. Satan's Voice vs. God's Voice— When God judges us, He always leaves us with hope. In fact, some time ago I found myself sharing with somebody something my own mentor had told me years ago about how to distinguish in my mind between God's pointing sin out in my life and Satan's trying to condemn me. And that is a real struggle for many Christians; and it may be for you as well; knowing when God is trying to correct you and when it is really Satan trying to tear you down. The acid test is this; if the source is Satan, it will leave you with a feeling of failure and the claim that you will never escape. But if the source of that voice is God, though the failure may be pointed out, He will always leave you with hope; hope that by His power, He can enable you to overcome. Because God always gives (even in His rebuke) the hope of future restoration if only we will repent and seek God (as Jeremiah 29:13 says) with all our hearts. Repentance is the key to redemption— And repentance is key to redemption. And repentance means more than saying I'm sorry. No, the Greek word “repentance” (μετανοέω) means a change of mind; a change in the inner man. And the Biblical Hebrew word for repent is שׁוּב (SHUB ), which means “to turn back, return.” It is directional. So it is more than confession; more than just being sorry and expressing it. No, it means to change your direction. You are going one way and now you begin to go the opposite way. If we want to be saved, we must μετανοέω; we must שׁוּב; we must change in the inner person and we must change our direction. And I want to encourage you today that, no matter how far you may have fallen, if you are willing to repent like that, God will help you to repent and God's promise of restoration will be available to you. The Temple Dedication— When the temple was first completed, Solomon (I think with amazing wisdom and foresight) prays this incredible prayer of dedication. And in it he seems to anticipate a time when Israel might turn from God. Listen to what he prays in… 1 Kings 8:46-51 (ESV)— 46 “If they sin against you—for there is no one who does not sin—and you are angry with them and give them to an enemy, so that they are carried away captive to the land of the enemy, far off or near, 47 yet if they turn their heart in the land to which they have been carried captive, and repent and plead with you in the land of their captors, saying, ‘We have sinned and have acted perversely and wickedly,' 48 if they repent with all their mind and with all their heart in the land of their enemies, who carried them captive, and pray to you toward their land, which you gave to their fathers, the city that you have chosen, and the house that I have built for your name, 49 then hear in heaven your dwelling place their prayer and their plea, and maintain their cause 50 and forgive your people who have sinned against you, and all their transgressions that they have committed against you, and grant them compassion in the sight of those who carried them captive, that they may have compassion on them 51 (for they are your people, and your heritage, which you brought out of Egypt, from the midst of the iron furnace). Daniel turned his eyes toward Jerusalem— Isn't it interesting that the Bible says (when Daniel is taken captive and placed under the charge of the chief of the eunuchs)… Daniel 1:9 (ESV)— 9 And God gave Daniel favor and compassion in the sight of the chief of the eunuchs, What had Solomon prayed for? That when Israel is taken captive, God might… 1 Kings 8:50b (ESV)— grant them compassion in the sight of those who carried them captive, that they may have compassion on them God is answering Solomon's prayer. Why? Because Daniel (who knows what Solomon wrote) is doing exactly what Solomon said would be their visual act of repentance. 1 Kings 8:46-51 (ESV)— 48 if they repent with all their mind and with all their heart in the land of their enemies, who carried them captive, and pray to you toward their land, which you gave to their fathers, the city that you have chosen, and the house that I have built for your name, Remember King Darius' edict that all people in his kingdom (Israel included) should worship no other god except him as king? When Daniel hears that edict, how does Daniel respond? It says in… Daniel 6:10 (ESV)— 10 When Daniel knew that the document had been signed, he went to his house where he had windows in his upper chamber open toward Jerusalem. He got down on his knees three times a day and prayed and gave thanks before his God, as he had done previously. Deliverance through Cyrus— You see, Daniel is modeling what real repentance looks like at that open window. And it is interesting to note that not long after this God does bring deliverance to His people through the hand of Cyrus the Great, the founder of the Persian empire, which we will talk more about in the coming weeks. A feeling of hopelessness— You know, In general I sense this general feeling of hopelessness among Christians. I have even heard it said that America is too far gone to be able to turn things around. Now I understand that viewpoint, though I believe our passages today reveal to us another, more accurate way of seeing our current predicament. Yes I believe the time is fast approaching when God is going to judge this nation. But what I want you to see today is that this is not something we should dread, because (although it is going to be difficult) it is going to be redemptive. And if we individually and collectively repent, God will restore us. God always offers the hope of restoration to those under judgment; if they but turn to Him. And so this week, let us commit to live lives that are characterized by true repentance. May we seek God with all our hearts. And may you experience (in every way) His love, His redemption, and His restoration in your own life. Amen.
