After completing a PhD in cancer biology at the University of Chicago, Illinois, in 2017, Tim Fessenden moved to a laboratory at the Massachusetts Institute of Technology in Cambridge to focus on immunology.Fessenden, who is now an editor at the Journal of Cell Biology in New York City, says that alongside adjusting to a new lab culture, he needed to learn new techniques, adding: “I am a lifelong student, someone who always wants to be the dumbest person in the room.”Fessenden is joined by physician-scientist Ken Kosik, and Jennifer Pursley, a particle physicist-turned-medical physicist.Kosik's neuroscience research and collaborations are influenced by his close working proximity to physical scientists. Abstract: Abnormal intracellular accumulation of Tau aggregates is a hallmark of Alzheimer's disease (AD) and other Tauopathies, such as Frontotemporal dementia (FTD), which can be caused by mutations of Tau. Mutated and pathological Tau can undergo a range of post-translational modifications (PTMs) that might trigger or modulate disease pathology. Recent studies indicate that modification of wild type Tau by Small ubiquitin-like modifier SUMO isoform 1 (SUMO1) controls Tau hyperphosphorylation and aggregation, suggesting that SUMOylation acts as a central regulator of Tau's biochemical properties. Besides SUMO1, Tau is modified by SUMO2/3, however the consequences of this modification have not been investigated. Here, using viral approaches on primary hippocampal neurons, transgenic mice expressing mutant Tau and SUMO2, and iPSC-derived neurons from FTD patients, we evaluated whether SUMO2/3 conjugation modifies the neurodegenerative disease pathology associated with the aggregation-prone mutant Tau P301L, P301S, and R406W variants. We found that mutant forms of Tau are targets of SUMO2/3, and SUMO2/3 conjugation is neuroprotective. Importantly, expression of mutant Tau is accompanied by a significant reduction of SUMO2/3 conjugation levels, and restoring levels of SUMO2 reduces mutant Tau aggregation and phosphorylation in all model systems Furthermore, overexpression of SUMO2 restores levels of pre- and post-synaptic markers, associated with a complete rescue of the LTP and memory deficits in transgenic mice expressing mutant Tau. These findings bring to light the potential therapeutic implication of manipulating SUMO conjugation to detoxify Tau through PTM-based approaches. Copy rights belong to original authors. We are humbled to host Kenneth Kosik, professor of Neuroscience at UC Santa Barbara on this week's Colombia Calling podcast, the final for 2021. Kenneth S. Kosik, M.D. is an American neurologist, author, researcher and professor in neuroscience at the University of California, Santa Barbara. Kosik provided much of the original data on the largest family in the world with a genetic form of Alzheimer's disease located in Antioquia, Colombia. And this is where our interest lies. We discuss the "paisa mutation," which has appeared in Antioquia and certain genetic mutations which cause early onset Alzheimer's disease in the region and that have been traced all the way back to the Iberian peninsula and the year 1500! This is a revealing and fascinating episode, one which we feel is fitting to end 2021.

The final announcement is that my cat Poe is finally home and recovering from a scary illness. He developed what’s called pyothorax, which is an infection in the chest, and in Poe’s case we still don’t know what caused it. After a week in the veterinary intensive care unit, he’s finally home and getting better all the time. That’s why last week’s episode was so short, and if you messaged me this week about something and I seemed impatient when I replied, that’s why. I just haven’t had any mental energy to concentrate on anything but Poe. Thank you to everyone at the Animal Emergency and Specialty Center of Knoxville for taking such good care of him. We’ve got something fun and a little different this time, inspired by two things. First, I saw a tweet about a captive beluga whale who had apparently learned to mimic human speech and one night told a diver in his pool to get out. Then the awesome podcast BewilderBeasts had a segment about a harbor seal in Maine who was rescued by a fisherman as a pup, which reminded me of a similar situation with another harbor seal in Maine, Hoover the Talking Seal. That’s right, it’s an episode about mammals that can talk, including one of my favorite cryptozoological mysteries ever. Before we learn about talking animals, we need to learn a little bit about how humans talk. Humans produce most vocal sounds using our larynx, which is sometimes called a voicebox. The human larynx is situated at the top of the throat, and it helps us breathe, helps keep food from going down the wrong tube and into the lungs, and enables us to make sounds. It consists of cartilage, small muscles, and flaps of tissue called vocal folds or vocal cords. There are two kinds of vocal folds: the true vocal folds that are connected to muscles and actually produce sound, and the false vocal folds that don’t have any connected muscles and just help with resonance. Usually resonance just makes the sound louder, but humans have learned to do amazing things with our voices. Some cultures use the false vocal folds to create a secondary tone. It’s called overtone singing,

Amino acids can give clues to the next step in solving Alzheimer's. Finding the intermediary steps in Alzheimer's is very difficult, however, as only the end stage is realistically viewable. New research on the Colombian population may show a higher concentration of pre-Alzheimer's mutations and may help researchers in the future. Abstract: Amyloid aggregation of tau protein is implicated in neurodegenerative diseases, yet its facilitating factors are poorly understood. Recently, tau has been shown to undergo liquid liquid phase separation (LLPS) both in vivo and in vitro. LLPS was shown to facilitate tau amyloid aggregation in certain cases, while independent of aggregation in other cases. It is therefore important to understand the differentiating properties that resolve this apparent conflict. We report on a model system of hydrophobically driven LLPS induced by high salt concentration (LLPS-HS), and compare it to electrostatically driven LLPS represented by tau-RNA/heparin complex coacervation (LLPS-ED). We show that LLPS-HS promotes tau protein dehydration, undergoes maturation and directly leads to canonical tau fibrils, while LLPS-ED is reversible, remains hydrated and does not promote amyloid aggregation. We show that the nature of the interaction driving tau condensation is the differentiating factor between aggregation-prone and aggregation-independent LLPS. Copy rights belong to original authors. Z., Shea, J.-E., Han, S., Kosik, K. Abstract: Tau protein in vitro can undergo liquid liquid phase separation (LLPS); however, observations of this phase transition in living cells are limited. To investigate protein state transitions in living cells we found that Cry2 can optogentically increase the association of full lengh tau with microtubules. To probe this mechanism, we identified tau domains that drive tau clustering on microtubules in living cells. The polyproline rich domain (PRD) drives LLPS and does so under the control of phosphorylation. These readily observable cytoplasmic condensates underwent fusion and fluorescence recovery after photobleaching consistent with the ability of the PRD to undergo LLPS in vitro. In absence of the MTBD, the tau PRD co-condensed with EB1, a regulator of plus-end microtubule dynamic instability. The specific domain properties of the MTBD and PRD serve distinct but mutually complementary roles that utilize LLPS in a cellular context to implement emergent functionalities that scale their relationship from binding alpha-beta tubulin heterodimers to the larger proportions of microtubules. Over 5 million Americans are afflicted with Alzheimer’s dementia. A medical case has arisen from a family in Columbia, where a woman with a genetic mutation which virtually guaranteed that she would develop early onset Alzheimers did not develop the disease until her 70s. Juliana Acosta-Uribe, a graduate student in the Kosik Lab, along with several other co-researchers, further investigated what could have caused this exception as well as what this could mean for Alzheimer therapy. A recent story in the New York Times suggested that abrupt changes in personality may be early warning signs for Alzheimer's disease. These changes in behavior can range from - agitation - to depression - to suddenly developing an overactive sexual appetite. It can set in even before the person starts to lose their memory. To find out more - I spoke with Dr. Kenneth Kosik, co-director of the Neuroscience Research Institute at UC Santa Barbara. Dr. Kosik is the author of Outsmarting Alzheimer's, What You Can Do to Reduce Your Risk.
More than 5 million people in the USA are suffering from Alzheimer’s Disease, and that number is expected to triple by 2050. While there is no cure for this common form of dementia, experts say there are things you can do to help reduce the risk of suffering from Alzheimer’s later in life. Neurologist Dr. Kenneth S. Kosik has been researching Alzheimer’s Disease for 35 years. The Harriman Professor of Neuroscience Research at UC Santa Barbara, and co-director of the Neuroscience Research Institute, shares these important tips to help ward off Alzheimer’s. “Know your Numbers” “Know your blood pressure, know your glucose or sugar level, know your cholesterol or lipid levels,” said Dr. Kosik. If any of these numbers are abnormal, a person is at greater risk of Alzheimer’s. If your numbers are off, talk to your doctor about what you can do to get them back on track. Keeping these numbers in check can also reduce the risk of diabetes, stroke and heart disease.
