Aaron Corcoran aka Skinner released his debut album New Wave Vaudeville on January 7. He talks about playing open mic nights when he was in his mid-teens, his DIY ethos, his creative process and influences, and discusses making some of the tracks on the album. Buy New Wave Vaudeville: https://skinner97.bandcamp.com/album/new-wave-vaudeville ---- Press release: Skinner is the project of the Dublin-based multi-instrumentalist, singer and producer Aaron Corcoran who was inspired by the New York No wave scene in the late '70s/early '80s for this debut record. The record's lead track "Calling In Sick" perfectly exemplifies this influence as he takes cues from the genre, while also carving his own path on the scene as well. Speaking about "Calling In Sick", he said, "The song is about calling in sick to the world. Life is tough for most people right now and there is no down time for most of us. Some days just feel like you're hurtling towards destruction and I just wanted to write a song that captures that feeling of anger and frustration of having to say enough is enough. Fuck everything I'm just going to do me for today. And I think that's fair." 'New Wave Vaudeville', the song and album title, references the New Wave Vaudeville music variety freak show that took place at the 57 Club in New York from 1978 to 1983. The club was known for its free-form art shows, a debaucherous wild riposte to creeping commerciality of modern culture of the time. "The ethos of the album is to celebrate the weird part of who you are and be proud of it," says Aaron. "You might feel strange in society but you don't when you're listening to your favourite music, and this record is a space for the outsiders to feel comfortable." Skinner's debut album takes inspiration from the outsiders in alternative music who populate the greatest hits of no wave, dance-punk, Latin disco and post-punk genres - Liquid Liquid, A Certain Ratio, Kid Creole And The Coconuts and James Chance And The Contortions to name a few. "I like the idea of mixing genres that shouldn't go together," Aaron adds. "That's probably why I like no wave music. It's just total and utter free expression of music without any constraint or rules. And it's not constrained by any societal ideals like a lot of other music can be. It's pure raw human emotion and expression." Throughout the LP, Corcoran makes use of vocal training by Dara Kiely of Dublin noise-rock Gilla Band, who taught Aaron how to scream effectively while singing without losing your voice, as heard on the tracks "Calling In Sick", "Tell My Ma" and "When You Live In A Shoe". Percussion also plays a big part in the record, with Latin American music and disco cited as an influence on the 10 tracks on the LP. New Wave Vaudeville was recorded and produced by Aaron Corcoran at the Meadow and engineered alongside Rian Trench. The album was mixed by Sean Corcoran, and mastered by Tj Lipple.

Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.01.23.525126v1?rss=1 Authors: Kaizuka, T., Hirouchi, T., Saneyoshi, T., Hayashi, Y., Takumi, T. Abstract: Numerous proteome analyses have been conducted on the postsynaptic density (PSD), a protein condensate beneath the postsynaptic membrane of excitatory synapses. Each has identified several hundred to thousands of proteins. While proteins with predictable functions have been well studied, functionally uncharacterized proteins are mostly overlooked. In this study, we perform a meta-analysis of the 35 PSD proteome datasets, including 5,869 proteins, identifying 97 uncharacterized proteins that appeared in multiple datasets. We focus on the top-ranked protein, FAM81A, renamed DRACC1. DRACC1 is expressed in forebrain neurons and enriched at the synapse. DRACC1 interacts with PSD proteins, including PSD-95, SynGAP, and NMDA receptors, and promotes liquid-liquid phase separation of those proteins. Consistently, the downregulation of DRACC1 in neurons causes a decrease in the size of PSD-95 puncta and the frequency of neuronal firing. Our results characterize DRACC1 as a novel synaptic protein facilitating the assembly of proteins within PSD. It also indicates the effectiveness of a meta-analytic approach of existing proteome datasets in identifying uncharacterized proteins. Copy rights belong to original authors. Announcements Sanctuary Park Defendants Statement A Statement from the Aston Park Defendants in Response to APD's January 11th Press Release -January 14th, 2023: "On Wednesday, January 11th, 2023, Asheville Police Department (APD) issued a widely circulated press release stating that 120,000 lbs of “trash” were removed from two “vacant” homeless encampments in West Asheville. We believe that this press release is part of an ongoing misinformation campaign by the City of Asheville to justify evicting encampments, fracturing communities of care, and broadly criminalizing unsheltered homelessness without creating real solutions. APD claims that the two camps were vacant, but admits that over the course of two weeks, they forced the people living there to leave. “Services” were offered to the people displaced, but were limited to rides, sharing information about local shelters, and helping people register for a housing list with a months-to-years long wait for placement. We question the utility of these services to people who are chronically homeless and unsheltered. According to the city of Asheville's 2022 Point in Time count, 232 (36%) of people without housing were unsheltered, defined as “sleeping outside or in other locations not suitable for human habitation.” These people are criminalized with trespassing laws and ordinances restricting camping. Yet, limited capacity and other barriers to shelter access often leave people with no other choices. When people camp together in larger groups, they are able to share resources and build community. Providing these camps with basic waste disposal and sanitation services would eliminate the health risks used to justify their removal. Instead, people camping are blamed for these conditions, then forced to relocate with only what they can carry. Tents, shelters, and other necessities must be left behind, and are relabeled “trash.” APD, city government and anti–homeless businesses use this, coupled with overblown and misleading claims about violent crime in camps, as justification for displacing unsheltered people again and again. Since the 11th, APD evicted two more camps in East Asheville, just ahead of a cold front bringing ice and snow. Camp evictions in February and December of 2021 under similar conditions sparked widespread public outrage. Despite this, conditions for people living unsheltered have mostly remained unchanged. Misinformation about “litter” and crime, alongside unfulfilled promises of long-term solutions, have redirected public attention from ongoing violence. Not only are camp sweeps violent, they are an ineffective and expensive strategy for managing unsheltered homelessness. There are more humane, longer-term and lower-cost alternatives. More on this, along with other updates, coming soon. In the meantime, check out our website, avlsolidarity.noblogs.org, for more information including links to sources cited for this statement. Please share and spread the word! Sal Principato - Liquid Liquid - in conversation David Eastaugh Liquid Liquid emerged from downtown New York's no wave scene. The group's original records were pressed in very limited quantities on 99 Records, and can now fetch high prices. "Cavern" originally appeared on the EP, Optimo, recorded by Don Hunerberg. Though the pressings were small, the music has had a lasting and far reaching impact. A music video for "Cavern" was produced by Michael Sporn. After "Cavern" was sampled for Grandmaster + Melle Mel's old school rap classic, "White Lines (Don't Don't Do It)," 99 Records took Sugarhill to court over its unauthorized use, and after an expensive court battle, won compensation. Before they could collect, however, Sugarhill went into receivership.[2] The song was also included on the Disco Not Discocompilation album. The first three EPs, plus live material, were reissued in 1997 by Grand Royal (US) and Mo' Wax (UK). After the collapse of both these labels, Domino Recordsreleased the music from all three original 12"s plus extra tracks and early live recordings as Slip In And Out Of Phenomenon in 2008.

APB with Iain Slater in conversation with David Eastaugh APB formed in the small rural town of Ellon, Aberdeenshire, Scotland, and consisted of singer/bassist Iain Slater, guitarist Glenn Roberts, and drummer George Cheyne. Though living in a remote area, they took inspiration from the "DIY" spirit of the punk scene, sounding like bands as Liquid Liquid, adopting a hard-working approach to writing, rehearsing and performing. Meanwhile, in Aberdeen, a small independent record company called Oily had started to release records and they were impressed enough by the band's live shows to release a single in 1981. The song was "Chain Reaction", which had long been a favourite at gigs. It was basically a two and a half minute punk/pop song with fuzzy distorted guitar and Scottish inflected vocals, but already the band was evolving with a sparser and more rhythmical sound. Bootsy Collins, George Clinton, Gang of Four, ESG, Buzzcocks, and The Clash were all filtered through three teenagers from Aberdeenshire to create a catchy, rhythmic sound.

Targeted modulation of protein liquid-liquid phase separation by evolution of amino-acid sequence

Abstract: Rationally and efficiently modifying the amino-acid sequence of proteins to control their ability to undergo liquid-liquid phase separation (LLPS) on demand is not only highly desirable, but can also help to elucidate which protein features are important for LLPS. Here, we propose an innovative computational method that couples a genetic algorithm to a sequence-dependent coarse-grained protein model to evolve the amino-acid sequences of phase-separating intrinsically disordered protein regions (IDRs), and purposely enhance or inhibit their capacity to phase-separate. We apply it to the phase-separating IDRs of three naturally occurring proteins, namely FUS, hnRNPA1 and LAF1, as prototypes of regions that exist in cells and undergo homotypic LLPS driven by different types of intermolecular interaction. We find that the evolution of amino-acid sequences towards enhanced LLPS is driven in these three cases, among other factors, by an increase in the average size of the amino acids. However, the direction of change in the molecular driving forces that enhance LLPS (such as hydrophobicity, aromaticity and charge) depends on the initial amino-acid sequence: the critical temperature can be enhanced by increasing the frequency of hydrophobic and aromatic residues, by changing the charge patterning, or by a combination of both. Finally, we show that the evolution of amino-acid sequences to modulate LLPS is strongly coupled to the composition of the medium (e.g. the presence or absence of RNA), which may have significant implications for our understanding of phase separation within the many-component mixtures of biological systems. Sneaks - "Faith" from the 2020 album Happy Birthday on Merge Records. A new album from Sneaks is definitely 'cause for celebration, and this fourth full-length from Washington, DC’s Eva Moolchan provides a party-perfect groove with electronic dance beats and a Liquid Liquid-influenced bass line.  Like last year's album, Highway Hypnosis, she teams up with mixing engineer Carlos Hernandez and legendary Grammy-Award-winning producer Jacknife Lee (U2, R.E.M., Taylor Swift, to name a few). The production combined with Moolchan's laid-back delivery takes you right back to New York's early '80s no-wave scene. Crank it up and enjoy the festivities. Abstract: Amyloid aggregation of tau protein is implicated in neurodegenerative diseases, yet its facilitating factors are poorly understood. Recently, tau has been shown to undergo liquid liquid phase separation (LLPS) both in vivo and in vitro. LLPS was shown to facilitate tau amyloid aggregation in certain cases, while independent of aggregation in other cases. It is therefore important to understand the differentiating properties that resolve this apparent conflict. We report on a model system of hydrophobically driven LLPS induced by high salt concentration (LLPS-HS), and compare it to electrostatically driven LLPS represented by tau-RNA/heparin complex coacervation (LLPS-ED). We show that LLPS-HS promotes tau protein dehydration, undergoes maturation and directly leads to canonical tau fibrils, while LLPS-ED is reversible, remains hydrated and does not promote amyloid aggregation. We show that the nature of the interaction driving tau condensation is the differentiating factor between aggregation-prone and aggregation-independent LLPS. Copy rights belong to original authors. Abstract: Recent studies show that a single MAPT gene mutation can promote alternative tau misfolding pathways engendering divergent forms of frontotemporal dementia and that under conditions of molecular crowding, the repertoire of tau forms can include liquid-liquid phase separation (LLPS). We show here that following pathogenic seeding, tau condenses on the nuclear envelope (NE) and disrupts nuclear-cytoplasmic transport (NCT). Interestingly, NE fluorescent tau signals and small fluorescent inclusions behaved as demixed liquid droplets in living cells. Thioflavin S-positive intracellular aggregates were prevalent in tau-derived inclusions with a size bigger than 3 m2, indicating that a threshold of critical mass in the liquid state condensation may drive liquid-solid phase transitions. Our findings indicate that tau undergoing LLPS is more toxic amongst a spectrum of alternative conformers; LLPS droplets on the NE that disrupt NCT serve to trigger cell death and can act as nurseries for fibrillar structures abundantly detected in end-stage disease. Z., Shea, J.-E., Han, S., Kosik, K. Abstract: Tau protein in vitro can undergo liquid liquid phase separation (LLPS); however, observations of this phase transition in living cells are limited. To investigate protein state transitions in living cells we found that Cry2 can optogentically increase the association of full lengh tau with microtubules. To probe this mechanism, we identified tau domains that drive tau clustering on microtubules in living cells. The polyproline rich domain (PRD) drives LLPS and does so under the control of phosphorylation. These readily observable cytoplasmic condensates underwent fusion and fluorescence recovery after photobleaching consistent with the ability of the PRD to undergo LLPS in vitro. In absence of the MTBD, the tau PRD co-condensed with EB1, a regulator of plus-end microtubule dynamic instability. The specific domain properties of the MTBD and PRD serve distinct but mutually complementary roles that utilize LLPS in a cellular context to implement emergent functionalities that scale their relationship from binding alpha-beta tubulin heterodimers to the larger proportions of microtubules. EVERYTHING I’VE HEARD Present BILLY BOGUS as 4th guest mix. - Billy (real name Niccolò Bruni), Pizzico Records' founder, started as a DJ in the mid 90s when he was only 18: he was a squatter and he used to arrange unusual performances with endless rotation of FSOL & Klaus Schulze music, filming everybody with a VHS camera and broadcasting everything in the community, creating peculiar vibes and feelings all around. Today his approach to DJing has the same feeling, also with contemporary dance music. Rare vinyls, mp3s, scratchy CDs, the format doesn’t matter, the central themes are what is important to him: italo disco, slow motion funk, trippy techno, every kind of dance music is like a magic mean to a shiny groove and he aims to spread this energy among the people via his own label, Pizzico. Pizzico was founded in 2007, among the others they have discovered and produced artists such as Jonathan Kusuma (Space System), Ichisan, Discodromo, Kid Who, Sare Havlicek, MA Spaventi and many more. After remixing the legendary band The Units and Dennis Young from Liquid Liquid, Billy’s career as a producer began to grow steadily: singles, remixes, three official albums and a jazzy under the moniker of Strata-Gemma. The word about his work was on everyone's lips, including household names like Laurent Garnier and Dimitri From Paris. His tracks have been regularly played by disco legends like Dj Harvey, Stevie Kotey, Gilles Peterson, Ivan Smagghe and Bill Brewster. - More about Billy Bogus : Discogs : http://www.discogs.com/artist/1456389-Billy-Bogus Soundcloud : https://soundcloud.com/billy-bogus Pizzico Records : https://pizzicorecords.bandcamp.com/ - Tracklist : 01. The story of 99 (pronounced “nine nine” and not “ninety-nine”) it's basically the story of Ed Bahlman, a born-and-bred Brooklynite with a strong work ethic and an intense passion for music, especially new and underground music that wasn’t wholly embraced by the mainstream or even alternative establishments. Everything started from a punk clothing store called 99, that later Ed transformed in the infamous record shop, thanks to his incredible selection of obscure and independent rock, reggae and punk music, that later became the ideal launching pad for the homonymous independent record label. The episode features: Y Pants, Liquid Liquid, ESG, Vivien Goldman, Bush Tetras, Maximum Joy, John Cage (in an interview about Glenn Branca) and Glenn Branca.

Italy's I-Robots is one of the most prolific re-editers in the game, putting out a slew of official reworks via his Opilec Music label. With his DJ career stretching back to the early '80s and with a sideline as a second-hand record dealer, his knowledge is impeccable. Uncovering rarities and forgotten gems, he manipulates tracks in both classic chop 'n' splice edit style and also in more complex arrangements. This mix is a journey through his edit suite. *How would you describe your sound?*"Nomad Sound" for sure... Eclectic freestyle without "Taboo" *What and who inspires you and influences your sound?*After a complete life spent in digging and collecting music i could say that i take inspiration from all kind of interesting sound, artist, label, nationality.... African rhythms, Disco beats, Synth mood, Electronic vision, freestyle grooves and around.... for me some seminal influential artists and friends that really REPRESENT are James Brown, Fela Kuti, Kraftwerk, Tangerine Dream, Larry Levan, Daniele Baldelli, Sly & Robbie, John Gibbs, Adrain Sherwood, Lee "Scratch" Perry, Derrick May, Carl Craig, Farley "Jackmaster" Funk, Kevin Saunderson, LTJ Bukem, Klein & M.B.O., Goldie, Liquid Liquid, Ryuichi Sakamoto, YMO, Kool & The Gang, Horace Silver, Gill Scott Heron, Roy Ayers, Many Dibango, Jorge Ben, Milton Nascimento, Airto Moreira & Flora Purim, Herbie Hancok, Francois Kevorkian, Joe Claussell, Ron Trent, Chez Damier, Jus_Ed, Eumir Deodato, Patrick Adams, Sylvester, Giorgio Moroder, Mark Cerrone, Simple Minds, Cabaret Voltaire, Throbbing Gristle, Klaus Schulze, Evangelos Odysseas Papathanassiou-Vangelis, The Human Lague, Brian Eno, Moebius, Connie Plank, Talking Heads, The Units, Schaltkreis Wasserman, Ashra, Manuel Göttsching, Supermax, Yello, ESG, Lil' Louis, Brian Auger and many many more........ *Can you tell us a little about this mix and what it means to you?*It was funny to produce this "freestyle mix" with a live party attitude and no computer - no sync selecting only material that i personally produced or reworked during my career... Konk - Your Life Dj Edit By Petko Turner Formed in 1980 in New York City, KONK was a band that emerged during a period marked by the post-punk/new wave and disco scenes which were prevalent at the time. KONK won the attention of both scenes by playing a hybrid of the two musical styles which involved the blending of a new wave attitude with the carefree nature of disco's dance crowd. KONK's musical influences ranged from Afrobeat, jazz, and funk to hip hop. Given these influence KONK's style is noted as being highly percussive, and containing simple, yet strong, backing bass lines. As part of New York’s post-punk dance scene they regularly played alongside groups such as Liquid Liquid, Bush Tetras, ESG, The Peech Boys and The Lounge Lizards. They have been featured in a recent book by David Byrne and others on the New York post-punk scene. Download for free on The Artist Union

Tag. Viel Spass!
