In this episode of the Epigenetics Podcast, we talked with Bas van Steensel from the Netherlands Cancer Institute about his work on characterizing chromatin at the Nuclear Lamina. The Interview starts with discussing Bas van Steensel's significant contributions to understanding genome-nuclear lamina interactions. Bas detailed the development of the DAM-ID technique during his postdoctoral studies, which provided a novel way to map genome-wide occupancy and identify Lamina-Associated Domains (LADs). He elaborated on how LADs reveal a distinct domain architecture, often correlating with gene expression levels. This prompted an exploration of the dynamics of these domains during differentiation processes, allowing insights into how gene activation and repression are influenced by their positioning relative to the nuclear lamina. The conversation highlighted the intricate relationship between chromatin dynamics and gene regulation, with Bas sharing compelling findings on how LADs behave during cell differentiation. The research indicated that regions moving away from the lamina often correlated with increased gene expression, revealing a complex interplay of spatial genome organization and transcriptional activity. Additionally, we ventured into the significance of outreach and transparency in scientific research. Bas shared his philosophy regarding collaboration and the ethical responsibility of scientists to share knowledge and resources openly. He emphasized that making lab notebooks and research processes accessible can greatly enhance reproducibility and understanding in the scientific community. Welcome to The Nonlinear Library, where we use Text-to-Speech software to convert the best writing from the Rationalist and EA communities into audio. This is: Can Generalized Adversarial Testing Enable More Rigorous LLM Safety Evals?, published by Stephen Casper on July 30, 2024 on The AI Alignment Forum. Thanks to Zora Che, Michael Chen, Andi Peng, Lev McKinney, Bilal Chughtai, Shashwat Goel, Domenic Rosati, and Rohit Gandikota. TL;DR In contrast to evaluating AI systems under normal "input-space" attacks, using "generalized," attacks, which allow an attacker to manipulate weights or activations, might be able to help us better evaluate LLMs for risks - even if they are deployed as black boxes. Here, I outline the rationale for "generalized" adversarial testing and overview current work related to it. See also prior work in Casper et al. (2024), Casper et al. (2024), and Sheshadri et al. (2024). Even when AI systems perform well in typical circumstances, they sometimes fail in adversarial/anomalous ones. This is a persistent problem. State-of-the-art AI systems tend to retain undesirable latent capabilities that can pose risks if they resurface. My favorite example of this is the most cliche one many recent papers have demonstrated diverse attack techniques that can be used to elicit instructions for making a bomb from state-of-the-art LLMs. There is an emerging consensus that, even when LLMs are fine-tuned to be harmless, they can retain latent harmful capabilities that can and do cause harm when they resurface ( Qi et al., 2024). A growing body of work on red-teaming ( Shayegani et al., 2023, Carlini et al., 2023, Geiping et al., 2024, Longpre et al., 2024), interpretability ( Juneja et al., 2022, Lubana et al., 2022, Jain et al., 2023, Patil et al., 2023, Prakash et al., 2024, Lee et al., 2024), representation editing ( Wei et al., 2024, Schwinn et al., 2024), continual learning ( Dyer et al., 2022, Cossu et al., 2022, Li et al., 2022, Scialom et al., 2022, Luo et al., 2023, Kotha et al., 2023, Shi et al., 2023, Schwarzchild et al., 2024), and fine-tuning ( Jain et al., 2023, Yang et al., 2023, Qi et al., 2023, Bhardwaj et al., 2023, Lermen et al., 2023, Zhan et al., 2023, Ji et al., 2024, Hu et al., 2024, Halawi et al., 2024) suggests that fine-tuning struggles to make fundamental changes to an LLM's inner knowledge and capabilities. For example, Jain et al. (2023) likened fine-tuning in LLMs to merely modifying a "wrapper" around a stable, general-purpose set of latent capabilities. Even if they are generally inactive, harmful latent capabilities can pose harm if they resurface due to an attack, anomaly, or post-deployment modification ( Hendrycks et al., 2021, Carlini et al., 2023). We can frame the problem as such: There are hyper-astronomically many inputs for modern LLMs (e.g. there are vastly more 20-token strings than particles in the observable universe), so we can't brute-force-search over the input space to make sure they are safe. So unless we are able to make provably safe advanced AI systems (we won't soon and probably never will), there will always be a challenge with ensuring safety - the gap between the set of failure modes that developers identify, and unforeseen ones that they don't. This is a big challenge because of the inherent unknown-unknown nature of the problem. However, it is possible to try to infer how large this gap might be. Taking a page from the safety engineering textbook -- when stakes are high, we should train and evaluate LLMs under threats that are at least as strong as, and ideally stronger than, ones that they will face in deployment. First, imagine that an LLM is going to be deployed open-source (or if it could be leaked). Then, of course, the system's safety depends on what it can be modified to do. So it should be evaluated not as a black-box but as a general asset to malicious users who might enhance it through finetuning or other means. This seems obvious, but there's preced...

B. Cox is joined once again by Van Everette of The Scenario Radio Show to review 90s R&B duo Zhané's debut album Pronounced Jah-Nay as it turns 30. The two vocalists, Renee Neuville and Jean Norris, met each other while students at Temple University and while they had two different backgrounds when it came to singing, they would find a medium that would make them a cohesive act. After linking up with power players in Philly DJ Jazzy Jeff and The Fresh Prince (Will Smith) and doing a showcase that caught some eyes and ears, they were taken under the tutelage of Naughty By Nature producer KayGee and were signed to Motown Records.Combining their many musical inspirations and influences, the group put out a very fun, yet mature sounding album. It was backed by their intriguing and smooth harmonies and deft songwriting over a variety of beats crossing many genres. The album was boosted three singles: the classic party jam "Hey Mr. DJ", "Groove Thing" and "Sending My Love".The album was certified platinum and things appeared to be on the up and up for the duo. However, they released only one other album, 1997's Saturday Night, to less acclaim and fanfare before splitting up. Nonetheless, they made their mark on 90s R&B with their debut which continues to be brought this day as a highlight in 1994.Check out Van Everette and The Scenario Radio Showhttps://www.thescenarioradioshow.com/Visit The Vault Classic Music Reviews Onlinewww.vaultclassicpod.comBuy Exclusive The Vault Podcast Merchandise!www.vaultclassicpod.com/storeSupport The Vault Classic Music Reviews on Buy Me A Coffeehttps://www.buymeacoffee.com/vaultclassicpodBuild Your Own Amazing Podcast Website In Less Than 5 Minutes!https://www.podpage.com/?via=ivecre8Show NotesUDiscoverMusic: Pronounced Jah-Nay: How Zhané Made A Name For Themselveshttps://www.udiscovermusic.com/stories/pronounced-jah-nay-zhane-album/Grammys: 'Pronounced Jah-Nay at 25': Zhané's Renee Neuville and Jean Norris look back at their classic R&B debut https://www.grammy.com/news/pronounced-jah-nay-25-zhanes-renee-neufville-jean-norris-look-back-their-classic-rbAlbumism: Zhané's Debut Album "Prounounced Jah-Nay" Turns 25 | Anniversary Retrospectivehttps://albumism.com/features/zhane-debut-album-pronounced-jah-nay-turns-25-anniversary-retrospectiveSupport this podcast at — https://redcircle.com/the-vault-classic-music-reviews-podcast/donationsAdvertising Inquiries: https://redcircle.com/brandsPrivacy & Opt-Out: https://redcircle.com/privacy
In his weekly Steelers mock draft tracker video, host Adam Bittner welcomes Post-Gazette Steelers insider Ray Fittipaldo to analyze the week in NFL draft predictions for the team from across the web. How does Illinois DT Jer'Zahn Newton fit with coach Mike Tomlin's defense as one of the few defensive linemen expected to go in the first round? Is MIssouri's Darius Robinson a potential Day 2 option? Will the Steelers value Alabama CB Kool-Aid McKinstry's profile the way they valued Joey Porter Jr.'s last year? And is Michigan CB Mike Sainristil a Day 2 corner who could fill a need on the inside? And is NC State's Payton Wilson a linebacker to watch? Our duo tackles those topics, and studies the center market as well. Is Jackson Powers-Johnson now a bonafide Day 1 pick? Welcome to The Nonlinear Library, where we use Text-to-Speech software to convert the best writing from the Rationalist and EA communities into audio. This is: Deep Forgetting & Unlearning for Safely-Scoped LLMs, published by Stephen Casper on December 5, 2023 on The AI Alignment Forum. Thanks to Phillip Christoffersen, Adam Gleave, Anjali Gopal, Soroush Pour, and Fabien Roger for useful discussions and feedback. TL;DR This post overviews a research agenda for avoiding unwanted latent capabilities in LLMs. It argues that "deep" forgetting and unlearning may be important, tractable, and neglected for AI safety. I discuss five things. The practical problems posed when undesired latent capabilities resurface. How scoping models down to avoid or deeply remove unwanted capabilities can make them safer. The shortcomings of standard training methods for scoping. A variety of methods can be used to better scope models. These can either involve passively forgetting out-of-distribution knowledge or actively unlearning knowledge in some specific undesirable domain. Desiderata for scoping methods and ways to move forward with research on them. There has been a lot of recent interest from the AI safety community in topics related to this agenda. I hope that this helps to provide a clarifying framework and a useful reference for people working on these goals. The problem: LLMs are sometimes good at things we try to make them bad at Back in 2021, I remember laughing at this tweet. At the time, I didn't anticipate that this type of thing would become a big alignment challenge. Robust alignment is hard. Today's LLMs are sometimes frustratingly good at doing things that we try very hard to make them not good at. There are two ways in which hidden capabilities in models have been demonstrated to exist and cause problems. Jailbreaks (and other attacks) elicit harmful capabilities Until a few months ago, I used to keep notes with all of the papers on jailbreaking state-of-the-art LLMs that I was aware of. But recently, too many have surfaced for me to care to keep track of anymore. Jailbreaking LLMs is becoming a cottage industry. However, a few notable papers are Wei et al. (2023), Zou et al. (2023a), Shah et al. (2023), and Mu et al. (2023). A variety of methods are now being used to subvert the safety training of SOTA LLMs by making them enter an unrestricted chat mode where they are willing to say things that go against their safety training. Shah et al. (2023) were even able to get instructions for making a bomb from GPT-4. Attacks come in many varieties: manual v. automated, black-box v. transferrable-white-box, unrestricted v. plain-English, etc. Adding to the concerns from empirical findings, Wolf et al. (2023) provide a theoretical argument as to why jailbreaks might be a persistent problem for LLMs. Finetuning can rapidly undo safety training Recently a surge of complementary papers on this suddenly came out. Each of which demonstrates that state-of-the-art safety-finetuned LLMs can have their safety training undone by finetuning ( Yang et al.. 2023; Qi et al., 2023; Lermen et al., 2023; Zhan et al., 2023). The ability to misalign models with finetuning seems to be consistent and has shown to work with LoRA ( Lermen et al., 2023), on GPT-4 ( Zhan et al., 2023), with as few as 10 examples ( Qi et al., 2023), and with benign data ( Qi et al., 2023). Conclusion: the alignment of state-of-the-art safety-finetuned LLMs is brittle Evidently, LLMs persistently retain harmful capabilities that can resurface at inopportune times. This poses risks from both misalignment and misuse. This seems concerning for AI safety because if highly advanced AI systems are deployed in high-stakes applications, they should be robustly aligned. A need for safely-scoped models LLMs should only know only what they need to One good way to avoid liabilities from unwanted capabilities is to make advanced AI systems in high-stakes settings know what they need to kno...

How they "mass-reported" to Chinese authorities, a fan fiction about him and fellow actor Wang Yibo. Synopsis: Can I Tell You Something Crazy is a new scripted series under the #PopVultures banner, which examines with fresh eyes, events that shook Asian media and entertainment. In 2019, Chinese actor Xiao Zhan was easily one of the hottest stars in China. The previously little-known star had broken out that year in China and across the world with the mega-hit Chinese fantasy period series The Untamed, where he acted opposite fellow Chinese star Wang Yibo. Adapted from a hugely popular boys' love novel named Grandmaster Of Demonic Cultivation, The Untamed attracted a large, ardent fan base that shot both its leads to A-list status. But less than a year after Xiao found fame, his loyal fan base became his undoing. In a controversy termed the 227 (Feb 27, 2020) incident, Xiao's fans following reported a piece of fan fiction that they felt portrayed their idol in an unflattering light and it caused one of the world's largest fan fiction sites - Archive Of Our Own - to be banned in China. In response, fan fiction readers, writers and more, targeted Xiao Zhan, calling for the boycott of brands he works with and review-bombing projects he had previously been in. The boycott was so widespread and overwhelming that Xiao eventually apologised for his fans' actions. In this episode of Can I Tell You Something Crazy, #PopVultures host Jan Lee takes another look at the 227 incident - a fight between two factions of fandom that blew up beyond anyone's imagination. It is a story of para-social relationships, toxic fandoms, the murky politics of boys' love in China, censorship, reporting culture and also, a story about stories - the value, power and refuge that fan fiction often provides. In this episode of the Epigenetics Podcast, we caught up with Luca Giorgetti from the Friedrich Miescher Institute to hear about his work on long-range transcriptional control by 3D chromosome structure. Luca Giorgetti's research focuses on chromosomal interactions, transcriptional output, and the dynamics of enhancer-promoter relationships. His lab investigated the causal relationship between chromosome interactions and transcriptional events. They've found that by manipulating the contact probabilities between an enhancer and a promoter by changing their distance, these changes had a substantial effect on transcription levels. This project was an experiment that Luca Giorgetti was eager to do, and it allowed him to establish a smooth functional relationship between contact probabilities and changes in transcription levels. In this episode of the Epigenetics Podcast, we caught up with Jop Kind from Hubrecht Institute to talk about his work on single cell DamID, EpiDamID, and Lamina Associated Domains (LADs). Jop Kind started out developing single cell DamID (scDamID), based on the DamID technique. First, this technique was adapted to a microscopic readout which enabled them to follow the localisation of chromatin domains after cell division. Next, the lab expanded this technique into the NGS space and created genome-wide maps of nuclear lamina Interactions in single human cells. Since LADs are in a heterochromatic chromatin context, the lab expanded scDamID into the epigenetic space. They first combined it with a transcriptional readout. Later-on they developed EpiDamID, a method to target a diverse set of chromatin types by taking advantage of the binding specificities of single-chain variable fragment antibodies, engineered chromatin reader domains, and endogenous chromatin-binding proteins.
