Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.03.17.532993v1?rss=1 Authors: Draeger, T. N., Rey, M.-D., Hayta, S., Smedley, M., Alabdullah, A.-K., Moore, G., Martin, A. C. Abstract: Tetraploid and hexaploid wheat have multiple genomes, with successful meiosis and preservation of fertility relying on synapsis and crossover only taking place between homologous chromosomes. In hexaploid wheat, the major meiotic gene TaZIP4-B2 (Ph1) on chromosome 5B, promotes crossover between homologous chromosomes, whilst suppressing crossover between homeologous (related) chromosomes. Tetraploid wheat has three ZIP4 copies: TtZIP4-A1 on chromosome 3A, TtZIP4-B1 on 3B and TtZIP4-B2 on 5B. Previous studies showed that ZIP4 mutations eliminate approximately 85% of crossovers, consistent with loss of the class I crossover pathway. Here, we show that disruption of two ZIP4 gene copies in Ttzip4-A1B1 double mutants, results in a 76-78% reduction in crossovers when compared to wild-type plants. Moreover, when all three copies are disrupted in Ttzip4-A1B1B2 triple mutants, crossover is reduced by over 95%, suggesting that the TtZIP4-B2 copy is also affecting class II crossovers. This implies that, in wheat, the class I and class II crossover pathways may be interlinked. When ZIP4 duplicated and diverged from chromosome 3B on wheat polyploidization, the new 5B copy, TaZIP4-B2, may have acquired an additional function to stabilize both crossover pathways. In plants deficient in all three ZIP4 copies, synapsis is delayed and does not complete, consistent with our previous studies in hexaploid wheat, when a similar delay in synapsis was observed in a 59.3Mb deletion mutant, ph1b, encompassing the TaZIP4-B2 gene on chromosome 5B. These findings confirm the requirement of ZIP4-B2 for efficient synapsis, and suggest that TtZIP4 genes have a stronger effect on synapsis than previously described in Arabidopsis and rice. Thus, ZIP4-B2 accounts for the two major phenotypes reported for Ph1, promotion of homologous synapsis and suppression of homeologous crossover. Copy rights belong to original authors. TRAGEDY OF THE SOLOMONIC ERA As in the case of Israel's possession of the land in Joshua, (PH1-10 after the Northern & Southern Campaign effectively eliminated the 31 kings) so toward the end of narrating the zenith of Israelite kingship in Solomon, 1 Kings 8:56 says that Solomon's reign and building of the temple happened "according to all that He promised; not one word ... failed of all His good promise, which He promised through Moses His servant." “You will bring them in and plant them in the mountain of your inheritance, in the place you made for your residence, (Palace) O Lord, the sanctuary, (Temple) O Lord, that your hands have established.” Exodus 15:17 This is not mere fulfillment of prophecies about a kingdom and a temple; in the light of the links to Gen. 1-2, this is also the realization of a kingdom and a temple in what appeared to be an emerging new creation (X2M-99-). And, as with Joshua, although there was an escalated fulfillment of prophecies of the Israelite kingship and of the temple (and even more so with Solomon), these fulfillments were not consummated. Subsequent history reveals this lack of consummation in that sin by both Solomon and Israel occurred, resulting not in elevation of conditions into a new creation (X2M99-) where perfect righteousness dwells but rather in the deleterious consequences of the splintering of Israelite kingship and kingdom, destruction of the temple, and dismantling of the nation itself in exile. Also, both Moses's prophecy in Exod. 15:17-18 and God's promise in 2 Sam. 7:10-16 (an allusive development itself of the Exod. 15 passage) affirm that the kingdom and the temple will be established forever, which did not happen with Solomon. Lack of complete fulfillment is indicated also by the conditional way that the fulfillment is expressed (1 Kings 2:4; 8:25; 9:1-9)… “and the Lord will fulfill his promise to me, ‘If your descendants watch their step and live faithfully in my presence with all their heart and being, then,' he promised, ‘you will not fail to have a successor on the throne of Israel.'” 1 Kings 2:4 NET …as well as by the observation, as in Joshua, that left in Israel were some of the people remaining from the enemy nations,"whom the sons of Israel were unable to destroy utterly" (1 Kings 9:21). THE WHY OF ZECHARIAH - see notes in book The essence is the restoration | Tikkun and consummation | Olam of God's kingdom. SIX DAYS Invitation Initiation Designation Probation Conferral Confirmation SEVENTH DAY Consummation The consummation which would have been typified in Solomon was only completed in Jesus that the fullness of Christ would be in us in this end-time generation by transfiguration leading to glorification... “After Solomon finished building the Lord's temple, the royal palace, and all the other construction projects he had planned, the Lord appeared to Solomon a second time… Jesus fulfills the complete conditions of the covenant with one sacrifice Beginning next week, X2M-114 we will start with the book of Zechariah detailing through 7 visions/aspects of our King of Glory… X. Introduction to Structure 1. Governor of the Nations 2. Avenger of the Afflicted 3. Builder of God's City 4. Guardian of God's Courts 5. Anointer of God's Temple 6. Desolator of the Apostates 7. Judge of the World Much of the church usually camps out around 1, 2 or a few of the above 7 attributes of Jesus…we need a robust multifaceted view of Him in consummation….that He's the Governor, Avenger, Builder, Guardian, Anointer, Desolator of the apostates, Judge…the King of Glory! Our aperture of Him will not then be 1, 2 or few sides but multifaceted and our vision will be reality in our midst within and without as Collider comes into fruition. That even as we behold (vision) Him in His consummation…the consummation of the ages we will become like Him.

The effect of a synthetically produced pheromone (PAP) on the well-being of pigs during transport to the slaughter house Even though numerous attempts to reduce stress during the transport of slaughter pigs, the losses due to transportation induced stress are between 0.03% and 0.5% in the EU and 0.4% in Germany. PSE is seen in 10 to 20% of the animals. A naturally occurring pheromone is secreted by the mammary glands of the sow, which have a calming and stress reducing effect on the piglets as well as on adult pigs. In this placebo controlled double blind study, the possible stress and aggression reducing potential of the synthetically produced `Porcine Appeasing Pheromone (PAP)´ (SSNA Sanofi Santé Nutrition Animale) was examined in slaughter pigs during their transport to the slaughter house and the possibility of improved meat quality. Eleven groups, each containing 24 pigs (n=264 Pigs; Breed Pi x DL) weighing 102 ± 10.6 kg were observed. Each group was transported in a livestock trailer, traveling the same route with the same driver for 40 to 45 minutes from the finishing barn to the slaughterhouse. Before and after the pigs were transported, a number of parameters were recorded. For the experiment, 6 ml of the PAP was sprayed along the back of the neck in five of the groups (n=150 pigs) while in the other six groups (n=144 pigs) a placebo was applied the same way. The cortisol level in the saliva of the live animals was determined in eight animals of each group (1. base level at the finishing barn, 2. after transport, 3. 30 minutes after arriving at the slaughter house in a holding pen). The animals‘ behavior was also observed using video surveillance in the holding pen for 25 minutes (scan sampling every 30 seconds). The heart rates of eight animals per group were recorded using a Polar® sport tester; the surface body temperature was measured using infrared thermography and the animals were weighed. Post mortem, the pH1 and pH24, the ability to conduct in the ham and in the chops were determined and the carcasses were examined for injuries due to fighting. In addition, the air humidity and ambient air temperature on the outside and inside of the livestock trailer was measured. The results show a definite stress- or transport-induced activation of the Hypothalamic-hypophyseal-adrenal axis (HPA) in all animals. A significant increase of the cortisol levels were evident in the saliva samples taken from all the pigs after transport versus the base level samples taken in the finishing barn. The animals treated with the pheromone had a significantly lower saliva cortisol concentration (p=0.02) compared to the pigs who were given the placebo (placebo 22.55 ± 6,4 nmol/l, pheromone 19,15 ± 7,26 nmol/l). Analyzing the heart rates revealed no significant difference between the two groups. The highest heart rates were reached during loading (170 - 173 bpm) and the rate slowly decreased to 118 - 120 bpm while in the holding pen. The severe strain the animals are under becomes evident when one looks at the high heart rates. The body temperature taken at the base of the ear also showed no significant difference between the control and PAP group. The base temperature, although dependent on the ambient air temperature and weather conditions, remained steady in its increase during transport on cold or warm days (4.5 ± 2.5 °C on cold and 4.6 ± 1.9 °C on warm days). Even the parameters weight loss and behavior showed no differences between the PAP group and the control group. The carcasses had few minor injuries, which were evenly spread between both groups. This can be attributed to the fact that the groups were transported individually by pens without mixing unfamiliar animals. On the other hand, the measurement pertaining to the quality of the meat did show a possible influence by the application of the PAP. The pH1 in the longissimus dorsi muscle of the pheromone treated pigs is significantly higher 45 minutes after slaughter (p
