Giacomo Casanova (1725-1798) fue, además de aventurero, libertino, historiador, escritor, diplomático, jurista, violonchelista, filósofo, matemático, bibliotecario y agente secreto italiano, mucho más que el seductor legendario que sus primeras ediciones censuradas nos hicieron creer. Nacido en Venecia, en el seno de una familia de actores, se formó bajo la tutela del abate Gozzi y destacó por su precocidad intelectual, llegando a graduarse en Derecho Civil y Canónico antes de los dieciocho años. Alejado prontamente de la carrera eclesiástica, Casanova vagó por distintos oficios —entre ellos violinista, secretario y curandero ocasional— y frecuentó las cortes de media Europa, codeándose con personajes tan influyentes como Catalina la Grande en Rusia o el cardenal Acquaviva en Roma. Su estancia en la cárcel de “Los Plomos” de Venecia, de la que consiguió fugarse en 1756, le otorgó una fama de ingenio y audacia que perduraría a lo largo de su vida. Obligado al exilio durante casi dos décadas, recorrió países como Francia, España y Rusia, participando en proyectos financieros, trabajando como diplomático e incluso ejerciendo de espía para diferentes potencias europeas. Gracias a su encanto, su conocimiento de varios idiomas y sus contactos en la alta sociedad, logró sortear numerosos escándalos, aunque su afición al juego y la sátira le granjeó no pocas enemistades. Tras una vida nómada, acabó como bibliotecario en el castillo de Dux (Bohemia), donde redactó la mayor parte de sus memorias, Histoire de ma vie. Es en estos escritos donde emerge un Casanova complejo, con una sexualidad fluida y experiencias no siempre heteronormativas. Ediciones posteriores al siglo XIX censuraron muchos pasajes, ocultando la dimensión más amplia de sus relaciones. Sin embargo, estudiosos contemporáneos han reivindicado su lugar como un referente proto-queer: un hombre de la Ilustración, hedonista y curioso, cuyo legado va más allá de la leyenda de conquistas femeninas. Abstract: Evolutionarily relevant networks have been previously described in several mammalian species using time-averaged analyses of fMRI time-series. However, fMRI network activity is highly dynamic and continually evolves over timescales of seconds. Whether the dynamic principles that govern intrinsic fMRI network fluctuations are conserved across mammalian species remains unclear. Using frame-wise clustering of fMRI time-series, we find that fMRI network dynamics in awake macaques and humans is characterized by recurrent transitions between a set of 4 dominant, neuroanatomically homologous fMRI coactivation modes (C-modes), three of which are also plausibly represented in the rodent brain. Importantly, in all species the identified C-modes exhibit species-invariant dynamic features, including intrinsic infraslow dynamics and preferred occurrence at specific phases of global fMRI signal fluctuations. Moreover, C-modes occurrence rates in awake humans, macaques and mice reflect temporal trajectories of least energy and predicts ranking of corresponding functional connectivity gradients. Our results reveal a set of species-invariant principles underlying the dynamic organization of fMRI networks in mammalian species, and offer novel opportunities to relate fMRI network findings across the phylogenetic tree. Abstract: How the emergent functional connectivity (FC) relates to the underlying anatomy (structural connectivity, SC) is one of the biggest questions of modern neuroscience. At the macro-scale level, no one-to-one correspondence between structural and functional links seems to exist. And we posit that to better understand their coupling, two key aspects should be taken into account: the directionality of the structural connectome and the limitations of describing network functions in terms of FC. Here, we employed an accurate directed SC of the mouse brain obtained by means of viral tracers, and related it with single-subject effective connectivity (EC) matrices computed by applying a recently developed DCM to whole-brain resting-state fMRI data. We analyzed how SC deviates from EC and quantified their couplings by conditioning both on the strongest SC links and EC links. We found that when conditioning on the strongest EC links, the obtained coupling follows the unimodal-transmodal functional hierarchy. Whereas the reverse is not true, as there are strong SC links within high-order cortical areas with no corresponding strong EC links. This mismatch is even more clear across networks. In this episode, Adam and Budi speak with the Head of the Drama Division at Juilliard, Evan Yionoulis.Evan Yionoulis, an Obie award-winning director and nationally-recognized teacher of acting, is  Richard Rodgers dean and director of Juilliard's Drama Division.  Before that, she served twenty years on the faculty of Yale School of Drama, where she was a professor in the practice of acting and directing and a resident director at Yale Repertory Theatre, as well as Lloyd Richards chair of the department of acting from 1998 to 2003. She has directed new plays and classics in New York and across the U.S., enjoying collaborations with major American playwrights, including Adrienne Kennedy and Richard Greenberg. She directed the critically acclaimed world premiere of Kennedy's He Brought Her Heart Back in a Box for Theatre for a New Audience, where she previously directed her Ohio State Murders (Lortel Award for Best Revival) and the Off-Broadway premiere of Howard Brenton's Sore Throats.  She opened Manhattan Theatre Club's Biltmore Theatre (Broadway) with Greenberg's The Violet Hour, directed his Everett Beekin at Lincoln Center Theater, and received an Obie Award for her direction of his Three Days of Rain at Manhattan Theatre Club, having directed the premieres of all three at South Coast Repertory.   At Yale Repertory Theatre, she directed Cymbeline, Richard II, The Master Builder, George F. Walker's Heaven, Brecht's Galileo, Gozzi's The King Stag (which she adapted with her brother, composer Mike Yionoulis and Catherine Sheehy), Caryl Churchill's Owners, the world premiere of Kirsten Greenidge's Bossa Nova, and numerous other productions including Kiss, by Guillermo Calderón. Other credits include productions at the Mark Taper Forum, the Huntington, NY Shakespeare Festival, the Vineyard, Second Stage, Primary Stages, Dallas Theatre Center, Actors Theatre of Louisville, Denver Center, Williamstown Theatre Festival, and many others.  She directed Seven, a documentary theatre piece about extraordinary women from across the globe who work for human rights, in New York, Boston, Washington, Aspen, London, Deauville, and New Delhi. Her short film, Lost and Found, made with Mike Yionoulis, premiered at Cleveland International Film Festival.  Their most recent collaborations are the multi-platform project Redhand Guitar, about five generations of musicians across an American century, and The Dread Pirate Project, about the malleability of identity between the digital and natural worlds.She has received a Princess Grace Foundation Fellowship, Works-in-Progress Grant, and the foundation's prestigious Statuette. Abstract: The macroscale connectome is the network of physical, white-matter tracts between brain areas. The connections are generally weighted and their values interpreted as measures of communication efficacy. In most applications, weights are assigned based on diffusion parameters or inferred using a statistical model, although the model generally scales poorly limiting its applications to relatively small networks. In reality, the ground-truth weights are unknown, motivating the exploration of alternative edge weighted schemes. Here, we explore a simple regression-based, explanatory model that endows reconstructed fiber tracts with directed and signed weights. Benchmarking this method on Human Connectome Project data, we find that the model fits observed data well, outperforming a suite of null models. The estimated weights are subject-specific and highly reliable, even when fit using relatively few training samples. Next, we analyze the resulting network using graph-theoretic tools from network neuroscience, revealing bilaterally symmetric communities that span cerebral hemispheres. These communities exhibit a clear mapping onto known functional systems. We also study the shortest paths structure of this network, discovering that almost every edge participants in at least one shortest path. We also find evidence of asymmetric edge weights, that the network reconfigures in response to naturalistic stimuli, and that estimated edge weights differ with age. In summary, we offer a simple framework for weighting connectome data, demonstrating both its ease of implementation while benchmarking its utility for typical connectome analyses, including graph theoretic modeling and brain-behavior associations. Abstract: Many neurodevelopmental conditions, including autism, affect males more than females. Genomic mechanisms enhancing risk in males may contribute to this sex-bias. The ubiquitin protein ligase E3A gene (Ube3a) exerts pleiotropic effects on cellular homeostasis via control of protein turnover and by acting as transcriptional coactivator with steroid hormone receptors. Overdosage of Ube3a via duplication or triplication of chromosomal region 15q11-13 causes 1-2% of autistic cases. Here, we test the hypothesis that increased dosage of Ube3a may influence autism-relevant phenotypes in a sex-biased manner. We report robust sex-biasing effects on brain connectomics and repetitive behaviors in mice with extra copies of Ube3a. These effects were associated with a profound transcriptional dysregulation of several known autism-associated genes (e.g., FMR1, SCN2A, PTEN, MEF2C, SHANK3, TSC2) as well as differentially-expressed genes identified in human 15q duplication and in autistic patients. Notably, increased Ube3a dosage also affects multiple sex-relevant mechanisms, including genes on the X chromosome, genes influenced by sex steroid hormones, downstream targets of the androgen and estrogen receptors, or genes that are sex-differentially regulated by transcription factors. These results suggest that Ube3a overdosage can critically contribute to sex-bias in neurodevelopmental conditions via influence on sex-differential mechanisms. Abstract: Autism Spectrum Disorder (ASD) is characterized by substantial, yet highly heterogeneous abnormalities in functional brain connectivity. However, the origin and significance of this phenomenon remain unclear. To unravel ASD connectopathy and relate it to underlying etiological heterogeneity, we carried out a bi-center cross-etiological investigation of fMRI-based connectivity in the mouse, in which specific ASD-relevant mutations can be isolated and modelled minimizing environmental contributions. By performing brain-wide connectivity mapping across 16 mouse mutants, we show that different ASD-associated etiologies cause a broad spectrum of connectional abnormalities in which diverse, often diverging, connectivity signatures are recognizable. Despite this heterogeneity, the identified connectivity alterations could be classified into four subtypes characterized by discrete signatures of network dysfunction. Our findings show that etiological variability is a key determinant of connectivity heterogeneity in ASD, hence reconciling conflicting findings in clinical populations. The identification of etiologically-relevant connectivity subtypes could improve diagnostic label accuracy in the non-syndromic ASD population and paves the way for personalized treatment approaches. Abstract: Postmortem studies have revealed increased density of excitatory synapses in the brains of individuals with autism, with a putative link to aberrant mTOR-dependent synaptic pruning. Autism is also characterized by atypical macroscale functional connectivity as measured with resting-state fMRI (rsfMRI). These observations raise the question of whether excess of synapses cause aberrant functional connectivity in autism. Using rsfMRI, electrophysiology and in silico modelling in Tsc2 haploinsufficient mice, we show that mTOR-dependent increased spine density is associated with autism-like stereotypies and cortico-striatal hyperconnectivity. These deficits are completely rescued by pharmacological inhibition of mTOR. Notably, we further demonstrate that children with idiopathic autism exhibit analogous cortical-striatal hyperconnectivity, and document that this connectivity fingerprint is enriched for autism-dysregulated genes interacting with mTOR or TSC2. Finally, we show that the identified transcriptomic signature is predominantly expressed in a subset of children with autism, thereby defining a segregable autism subtype. Our findings causally link mTOR-related synaptic pathology to large-scale network aberrations, revealing a unifying multi-scale framework that mechanistically reconciles developmental synaptopathy and functional hyperconnectivity in autism. Abstract: fMRI-based measurements of functional connectivity are commonly interpreted as an index of anatomical coupling and direct interareal communication. However, causal testing of this hypothesis has been lacking. Here we combine neural silencing, resting-state fMRI and in vivo electrophysiology to causally probe how inactivation of a cortical region affects brain-wide functional coupling. We find that chronic silencing of the prefrontal cortex (PFC) via overexpression of a potassium channel paradoxically increases rsfMRI connectivity between the silenced area and its thalamo-cortical terminals. Acute chemogenetic silencing of the PFC reproduces analogous patterns of overconnectivity, an effect associated with over-synchronous fMRI coupling between polymodal thalamic regions and widespread cortical districts. Notably, multielectrode recordings revealed that chemogenetic inactivation of the PFC attenuates gamma activity and increases delta power in the silenced area, resulting in robustly increased delta band coherence between functionally overconnected regions. The observation of enhanced rsfMRI coupling between chemogenetically silenced areas challenges prevailing interpretations of functional connectivity as a monotonic index of direct interareal communication, and points at a critical contribution of global rhythm generators to the establishment of brain-wide functional coupling. Abstract: The evolutionarily conserved default mode network (DMN) is characterized by temporally correlated activity between brain regions during resting states. The DMN has emerged as a selectively vulnerable network in multiple disorders, so understanding its anatomical composition will provide fundamental insight into how its function is impacted by disease. Reproducible rodent analogs of the human DMN offer an opportunity to investigate the underlying brain regions and structural connectivity (SC) with high spatial and cell type resolution. Here, we performed systematic analyses using mouse resting state functional magnetic resonance imaging to identify the DMN and whole brain axonal tracing data, co-registered to the 3D Allen Mouse Common Coordinate Framework reference atlas. We identified the specific, predominantly cortical, brain regions comprising the mouse DMN and report preferential SC between these regions. Next, at the cell class level, we report that cortical layer (L) 2/3 neurons in DMN regions project almost exclusively to other DMN regions, whereas L5 neurons project to targets both in and out of the DMN. We then test the hypothesis that in- and out-DMN projection patterns originate from distinct L5 neuron sub-classes using an intersectional viral tracing strategy to label all the axons from neurons defined by a single target. In the ventral retrosplenial cortex, a core DMN region, we found two L5 projection types related to the DMN and mapped them to unique transcriptomically-defined cell types. Abstract: Both macroscale connectome miswiring and microcircuit anomalies have been suggested to play a role in the pathophysiology of autism. However, an overarching framework that consolidates these macro and microscale perspectives of the condition is lacking. Here, we combined connectome-wide manifold learning and biophysical simulation models to understand associations between global network perturbations and microcircuit dysfunctions in autism. Our analysis established that autism showed significant differences in structural connectome organization relative to neurotypical controls, with strong effects in low-level somatosensory regions and moderate effects in high-level association cortices. Computational models revealed that the degree of macroscale anomalies was related to atypical increases of subcortical inputs into cortical microcircuits, especially in sensory and motor areas. Transcriptomic decoding and developmental gene enrichment analyses provided biological context and pointed to genes expressed in cortical and thalamic areas during childhood and adolescence. Supervised machine learning showed the macroscale perturbations predicted socio-cognitive symptoms and repetitive behaviors. Our analyses provide convergent support that atypical subcortico-cortical interactions may contribute to both microcircuit and macroscale connectome anomalies in autism. Abstract: Cholinergic drugs acting at M1/M4 muscarinic receptors hold promise for the treatment of symptoms associated with brain disorders characterized by cognitive impairment, mood disturbances or psychosis, such as Alzheimer's disease or schizophrenia. However, the brain-wide functional substrates engaged by muscarinic agonists remain poorly understood. Here we used a combination of pharmacological fMRI (phMRI), resting-state fMRI (rsfMRI) and resting-state quantitative EEG (qEEG) to investigate the effects of a behaviorally-active dose of M1/M4 agonist xanomeline on brain functional activity in the rodent brain. We investigated both the effects of xanomeline per se and its modulatory effects on signals elicited by the NMDA-receptor antagonists phencyclidine (PCP) and ketamine. We found that xanomeline induces robust and widespread BOLD signal phMRI amplitude increases and decreased high frequency qEEG spectral activity. rsfMRI mapping in the mouse revealed that xanomeline robustly decreased neocortical and striatal connectivity but induces focal increases in functional connectivity within the nucleus accumbens and basal forebrain. Notably, xanomeline pre-administration robustly attenuated both the cortico-limbic phMRI response and the fronto-hippocampal hyper-connectivity induced by PCP, enhanced PCP-modulated functional connectivity locally within the nucleus accumbens and basal forebrain, and reversed the gamma and high frequency qEEG power increases induced by ketamine. Collectively, these results show that xanomeline robustly induces both cholinergic-like neocortical activation and desynchronization of functional networks in the mammalian brain. These effects could serve as a translatable biomarker for future clinical investigations of muscarinic agents, and bear mechanistic relevance for the putative therapeutic effect of these class of compounds in brain disorders. Så det här blev ett snack om längtan att åter få jobba med Roger Melin och Stefan Persson – vad som är lika respektive annorlunda nu. Vi pratade pengar och spelarbudget, organisation och möjligheter som finns nu som inte fanns förr. PLAYWRIGHT | FICTION WRITER -James Magruder is a playwright, translator, and fiction writer. His stories have appeared or will appear in The Gettysburg Review, The Harrington Quarterly, Bloom, Subtropics, The Normal School, Mary, and the anthologies Boy Crazy and New Stories from the Midwest. He teaches translation and adaptation at the Yale School of Drama, where he received his doctorate. His produced translations and adaptations of works by Marivaux, Moliere, Lesage, Labiche, Gozzi, and Dickens have been produced on and off Broadway, across the country, and in Japan and Germany. He also teaches dramaturgy at Swarthmore College. His plays have been staged in Baltimore, Atlanta, and New York and published in The Art of the One-Act, Arts & Letters, and Third Coast. His writing has been supported by the Maryland State Arts Council, the New Harmony Project, the MacDowell Colony, where he was named a Thornton Wilder Fellow, the Ucross Foundation, the Blue Mountain Center, and the Jerome Foundation. The University of Wisconsin Press published his debut novel, Sugarless, last October. It was named one of “Twenty Indies to Watch” by Publishers Weekly. It is one of five nominees for a Lambda Literary Award and a semi-finalist for the Cabell First Novelist Award.
