Dr. Amit Sharma was awarded a Master's degree in Biomedical Sciences from Delhi University, India. He received his PhD in 2009 in Biotechnology from University of Pune for his work demonstrating microRNA regulation of cytokines invovled in allergic inflammation in mice models. Dr. Sharma's postdoctoral research at the Buck Institute, Novato, California involved investigating novel molecular regulatory pathways in regards to genotoxic stress and cellular senescence in invertebrate and mammalian models. Dr. Sharma has recently joined SENS Research Foundation as Group Lead in the Senescence Immunology Research Group. His research focus involves studying how aging and senescence affects the immune system and his research group will also investigate strategies to harness the immune system in mitigating deleterious effects of senescent cells with translational focus. Combining the toxic and corrosive composition of its atmosphere with the crushing pressure exerted on anything that attempts to venture too far into it, Venus is the last place most of us would choose for a date. These conditions proved to be disastsrous for missions that sent traditional craft to the planet. To date, the only "craft" to have survived for more than a few hours are the 1985 Soviet Vega 1 (https://solarsystem.nasa.gov/missions...) and Vega 2 (https://solarsystem.nasa.gov/missions...) balloon missions where each survived just over 46 hours before their batteries ran out. It would seem that baloons are the way to go!   This week we are joined by Dr. Jacob Izraelevitz, Principal Investigator of the JPL project that is developing robotic balloons  — currently called aerobots — that will eventually (hopefully) lead to successful exploration of Venus. In July, 2022, a one-third scale prototype aerobot successfully completed two test flights and achieved controlled flight at more than 4000 feet. In the fifth episode of the Longevity & Aging Series, Dr. Amit Sharma, Group Lead from SENS Research Foundation, Mountain View, CA, discusses a research paper he co-authored that was published in Volume 14, Issue 5, of Aging (Aging-US), entitled, “Enhanced co-culture and enrichment of human natural killer cells for the selective clearance of senescent cells.” DOI - https://doi.org/10.18632/aging.203931 Corresponding Author - Amit Sharma - amit.sharma@sens.org Video - https://www.youtube.com/watch?v=WQR8_gm2gUI Abstract In the context of aging and age-associated diseases, Natural Killer (NK) cells have been revealed as a key cell type responsible for the immune clearance of senescent cells. Subsequently, NK cell-based therapies have emerged as promising alternatives to drug-based therapeutic interventions for the prevention and treatment of age-related disease and debility. Given the promise of NK cell-mediated immunotherapies as a safe and effective treatment strategy, we outline an improved method by which primary NK cells can be efficiently enriched from human peripheral blood across multiple donors (ages 20-42 years old), with a practical protocol that reliably enhances both CD56dim and CD56bright NK cells by 15-fold and 3-fold, respectively. Importantly, we show that our co-culture protocol can be used as an easily adaptable tool to assess highly efficient and selective killing of senescent cells by primary NK cells enriched via our method using longer co-culture durations and a low target to effector ratio, which may be more physiological than has been achieved in previous literature. This piece is a continued deep dive into the subject, of 'fitness, function, or frailty' and, perhaps, "fixing". It's essential listening for anyone interested in the latest thinking on this crucial topic and was a highlight of the London conference. How can we integrate frailty and cognitive scores in risk assessment tools to facilitate inclusion in shared decision making? How can we improve a surgical colleague's ability to identify frailty? Most patients will have an operation sometime in their life; should surgical school be renamed and moved to being a "well being" clinic and offered to all patients? What screening tools would you recommend if only minimal time or money is available? Is pre-op screening actually 'public health' and should it really be being done for everyone? What are the differences and where is the overlap between fitness functional capacity, frailty, and sarcopenia? In mild cognitive impairment, should we tell patient that it will work and postop and not be expected to fully recover? What strategies have you found most helpful in engaging surgeons with identifying their frail patients? How are we tackling the workforce shortage among geriatricians? What has your experience been in advising achieving physical fitness in patients who present to surgery with persistent pain which is a limitation? This is part two of a two part piece. How can we integrate frailty and cognitive scores in risk assessment tools to facilitate inclusion in shared decision making? How can we improve a surgical colleague's ability to identify frailty? Most patients will have an operation sometime in their life; should surgical school be renamed and moved to being a "well being" clinic and offered to all patients? What screening tools would you recommend if only minimal time or money is available? Is pre-op screening actually 'public health' and should it really be being done for everyone?
