To date, no effective drug is available to directly treat the disease and prevent virus spreading. In a search for a drug against COVID-19, we have performed a massive X-ray crystallographic screen of repurposing drug libraries containing 5953 individual compounds against the SARS-CoV-2 main protease (Mpro), which is a potent drug target as it is essential for the virus replication. In contrast to commonly applied X-ray fragment screening experiments with molecules of low complexity, our screen tested already approved drugs and drugs in clinical trials. From the three-dimensional protein structures, we identified 37 compounds binding to Mpro. In subsequent cell-based viral reduction assays, one peptidomimetic and five non-peptidic compounds showed antiviral activity at non-toxic concentrations. Interestingly, two compounds bind outside the active site to the native dimer interface in close proximity to the S1 binding pocket. Another compound binds in a cleft between the catalytic and dimerization domain of Mpro. Neither binding site is related to the enzymatic active site and both represent attractive targets for drug development against SARS-CoV-2. This X-ray screening approach thus has the potential to help deliver an approved drug on an accelerated time-scale for this and future pandemics. Copy rights belong to original authors. In der heutigen Folge geht es um das bzw. dein Digitales Erbe. Das ist ein sehr sensibles Thema, dass noch nicht viele auf dem Schirm haben. Wir alle leben inzwischen in einer digitalen Welt, in der wir einen Großteil online organisieren, bestreiten und arbeiten. Dazu richten wir eine Vielzahl von Kundenkonten und Accounts ein und speichern digitale Inhalte, wie z.B. Fotos, die wir täglich mit unserem Handy machen, online in der Cloud. Doch was passiert eigentlich damit nach meinem Tod? Wer hat dann darauf Zugriff und wer weiß überhaupt davon? Wie kommen meine Erben an das Vermögen, dass ich vielleicht über digitale Plattformen oder Börsen investiert habe? Ich spreche darüber mit dem Onlineexperten Nicolai Fleckenstein. Er hat dazu auch kürzlich erst durch einen Todesfall in der Familie schmerzliche Erfahrungen sammeln müssen und gibt praktische Empfehlungen zur Regelung des Digitalen Erbes zu Lebzeiten. Dabei sollte man es seinen Erben so einfach wie möglich machen und ihnen eine Übersicht über alle Nutzerkonten, die Login-Daten und die dazugehörigen Passwörter zur Verfügung stellen. Am einfachsten macht man das mit einer Excel-Tabelle und für die Passwörter nutzt man am besten einen Password Safe wie z.B. KeePass. Das ist dann mit einem Masterpasswort verschlüsselt und die Dateien können dann etwa per USB-Stick zusammen mit dem Testament bei einem Notar hinterlegt werden. Bill begins by discussing the magnitude of the attack on Saudi Arabia may have on markets depending on the actions taken in response to the incident and ponders about risk premiums and how agents have reacted to geopolitical tensions recently. Fleckenstein then talks about the possibility that we are experiencing a bubble in the financial markets and the role that central banks, trading algorithms and passive investment vehicles play in this context, all from a historical and economic perspective, elucidating situations of distortion in asset prices. On the possibility of a crash, Bill comments on Fed interventions, the impact of monetary policy on the market and what may be the catalyst for a strong correction, as well as aspects of the current scenario that are unprecedented in history and thus create more uncertainties. Now is a good time to be shorting stocks? Fleckenstein, who has been very successful in the past with a short only fund, explains his point of view about being short at the moment and justifies, based on his perceptions of current market conditions, the valuations and prices observed. Bill also talks about investing in gold and other precious metals, as well as gold mining, in contrast to other asset classes such as bonds and currencies, considering all the factors hitherto exposed. As for the US-China trade war, Fleckenstein discusses his views on the issue and his expectations of a resolution for the problem. He also talks about the way Trump and next year's US elections might impact those obstacles. Bill concludes with an analysis of risks that are not being properly priced by the market.

My guest is Jim Fleckenstein. He is a coach and educator on sexuality. He is also a researcher who focuses predominantly on non-exclusive relationships and how they affect the individuals involved. He is an expert in consensual non-monogamy and a wellspring of knowledge and insight, which he graciously shares in this episode. Jim shares stats and information on consensual non-monogamy, how those who practice it are reporting an overwhelming amount of satisfaction and happiness, how emotional needs have a lot to do with those who are drawn towards it, and how these relationship structures are actually much more common than you may have realized. This is just the tip of the iceberg of this discussion though. Listen along and learn a lot about this interesting topic! The Three Main Divisions of Consensual Non-Monogamy To start the interview, Jim breaks down consensual non-monogamy into polyamory, swinging, and open relationships. Polyamory is separate from the others because there is a chance for a deep emotional connection to be reinforced. While the others are more reserved for sexual acts, polyamory is can delve into the emotional as well as the sexual components of a relationship. Jim says that swinging is probably the oldest of the trio. And for those who don't know, it's a couple-centric act where couples work hard to establish an emotional wall to prevent emotional developments. They also don't have sex independently of one another. And in open relationships, partners are free to seek sexual liaisons outside of the primary relationship. Emotional connection is not established, and unlike swingers, they don't participate in sexual acts together but do their own thing instead. Jim says much more within the episode. Listen along! The Reasons Why Certain People Prefer Consensual Non-Monogamy For the reasons why people go the non-monogamous route, Jim says it has a lot to do with the emotional needs of the person. He says the question of “what is it that I am trying to attain here?” is a great question to ask to evaluate your emotional needs. Your needs will determine which version of consensual non-monogamy that you gravitate towards and want to eventually practice–if you do end up deciding that your emotional needs warrant the lifestyle, of course. But according to the stats, Jim says that somewhere between 2-7% of all relationships are practiced through one of the three divisions of consensual non-monogamy. The Importance of Education Jim shares a lot of important information on how people learn about polyamory or swinging by attending educational events where people discuss consensual non-monogamy in a non-sexual environment. Often the trepidations that occur can be alleviated through more understanding. And as is brought up during the episode, some of the concerns of being pressured by a partner into swinging or an open relationship can be addressed once more information is processed. He also makes it clear this needs to be a mutual decision. Who knows, maybe all it takes is a couple of events and you will comfortable enough with shifting your relationship dynamic! Maybe it will confirm your anxiety. Becoming educated is key! It destigmatizes and demystifies.
