UK politics are never boring - not even when it comes to research policy (which - let's face it - can appear quite sedate for non-research-geeks).In this episode, we explore the recent spat after UKRI announced that in the next REF - Research Culture would count for 25% of an institution's score which was closely followed by a tender to develop indicators to help them measure culture. That didn't go down well with a lot of universities, and on top of all that, UKRI dissolved their EDI committee based on governmental critique. That didn't go down well with the research community. But we liked the idea - so we decided to do an episode on why indicators in research culture are a good idea - and particularly why we should have indicators on EDI topics - and what they could look like. Unsurprisingly, we think universities should be held accountable in this field.We also go into some of the concerns about indicators - and address the questions and comments we had on LinkedIn for this episode.Finally, we argue that this is not just a UK thing. The term “research culture” is travelling to other countries, so we should all be aware of the possibilities and challenges - and have indicators on the topic. Abstract: Fragile X messenger ribonucleoprotein (FMRP) is an RNA-binding protein that can form granules within neurons. Although FMRP-containing Fragile X granules have been extensively characterized, visualizing changes in granule structure in relation to function has been difficult. We used high resolution imaging to detect discrete differences in puncta size, intensity and contrast in our mouse FMRP and serine 499 (S499) phospho-mutant reporters. Tracking analyses of FMRP particles showed that phospho-mutant granules traveled at rates consistent with active transport within dendrites, while GFP-FMRP exhibited a wide range of speeds owing to its ability to oligomerize when overexpressed. Taken together, our approach allows us to evaluate FMRP granules and elucidate the central role of FMRP phosphorylation and dephosphorylation in granule clustering and transport. It had been recently suggested that FMRP granules could be phase separated structures that may function as hubs for local protein synthesis. To test this idea, we performed fluorescence recovery after photobleaching (FRAP) on individual FMRP granules. Intriguingly, we observed only a partial recovery that occurred over minutes time-scale and was dependent on translation. Moreover, the phospho-mutants revealed differences in recovery kinetics reflecting the labile (S499A) or preferential (S499D) nature of their interaction with granules containing stalled ribosomes. Our high resolution approach can reveal how the phosphorylation-state of FMRP can modulate the clustering of FMRP granules and thereby regulate local protein synthesis at the level of individual granules. Your pup comes sprinting through your living room, running in large, wide circles with seemingly no desire to stop… It's the DOG ZOOMIES! And while this slightly ‘crazy' behavior can be confusing at first, we're here to break down everything you need to know about dog (or puppy) zoomies! While dog zoomies is the most common term you'll hear, there is actually a more technical term for this fascinating display of energy put on by your pup! FRAPs or FRAP. This stands for frenetic random activity periods. And while I think this term is spot on, “zoomies” is more fun! Just like the real name says, the zoomies are a spurt of energy (often at seemingly random times) from your dog. Dog (or puppy) zoomies often include (but is not limited to) things like this… ⬇️ -Running as fast as possible -Running in no clear direction or with no clear purpose -Barking, howling or making other vocal forms of communication -Jumping on and off of things like couches, beds, nearby toddlers, chairs, etc. -Spinning wildly in circles (yes, this happens) -Trying to get you (or other dogs) to chase them, often encouraged by a play bow -Just overall being a little bit hyper and uncontrollable for a short burst of time The question of why do dogs get the zoomies gets a lot of attention in our private Facebook community and on our social media pages! And while there may not be a hard and fast rule as to why it happens, there are some ideas as to why. When our pups encounter prolonged periods of boredom, confinement, or even stress, it can create a pent-up amount of energy. And when you get a pent-up amount of energy and a desire to relieve stress… THE ZOOMIES! Most of the time our dogs get the zoomies after they've been forced to be not active for quite a while. Additionally, it can happen after stressful situations. Think bath time, vet visits, and even new guests coming to your home! And while the zoomies can really happen to your pup at any time, you'll often see it after a period of boredom/confinement (like in a crate) or after stressful situations. Just like undesired car repairs, the zoomies can happen at any time! And, it's often when you aren't expecting it. While these random bursts of energy can happen spontaneously, there are instances when dogs are more likely to get the zoomies. Here are a few ⬇️ -After being crated -After visiting the vet -After visiting the groomer -After a bath/shower at home -After being on a leash for an extended period of time -When they hear you say something they love like walk, park, dinner, treats, etc. -When you come home after a long period of time -When your dog knows you are about to leave -When your dog becomes extremely bored -When your dog knows it is about bedtime -When your dog doesn't have enough outlets for physical and/or mental activity Those are just a few times when dogs get the zoomies! Before we answer how to stop the zoomies, we really should ask should we want to stop the zoomies? While it's understandable to sometimes need your dog to chill out, shouldn't we be excited when our pups get the zoomies?! I mean we all want a tired, relaxed, and chilled out pup and doesn't exercise help with that? When my dogs get the zoomies, I'm stoked! Why? Because they're making my life easier. And as a human being, I like when my life is easier. They get to run around, go crazy, and get some energy out all while I often sit and enjoy the spectacle from the comfort of my couch! Abstract: Nonmuscle myosin II (NM II) is an integral part of essential cellular processes, including adhesion and migration. Mammalian cells express up to three isoforms termed NM IIA, B, and C. We used U2OS cells to create CRISPR/Cas9-based knockouts of all three isoforms and analyzed the phenotypes on homogeneous and micropatterned substrates. We find that NM IIA is essential to build up cellular tension during initial stages of force generation, while NM IIB is necessary to elastically stabilize NM IIA-generated tension. The knockout of NM IIC has no detectable effects. A scale-bridging mathematical model explains our observations by relating actin fiber stability to the molecular rates of the myosin crossbridge cycle. We also find that NM IIA initiates and guides co-assembly of NM IIB into heterotypic minifilaments. We finally use mathematical modeling to explain the different exchange dynamics of NM IIA and B in minifilaments, as measured in FRAP experiments. Copy rights belong to original authors. Abstract: The brain-expressed ubiquilins, UBQLNs 1, 2 and 4, are highly homologous proteins that participate in multiple aspects of protein homeostasis and are implicated in neurodegenerative diseases. Studies have established that UBQLN2 forms liquid-like condensates and accumulates in pathogenic aggregates, much like other proteins linked to neurodegenerative diseases. However, the relative condensate and aggregate formation of the three brain-expressed ubiquilins is unknown. We report that the three ubiquilins differ in aggregation propensity, revealed by in-vitro experiments, cellular models, and analysis of human brain tissue. UBQLN4 displays heightened aggregation propensity over the other ubiquilins and, like amyloids, UBQLN4 forms ThioflavinT-positive fibrils in vitro. Measuring fluorescence recovery after photobleaching (FRAP) of puncta in cells, we report that all three ubiquilins undergo liquid-liquid phase transition. UBQLN2 and 4 exhibit slower recovery than UBQLN1, suggesting the condensates formed by the brain-expressed ubiquilins have different compositions and undergo distinct internal rearrangements. We conclude that while all brain-expressed ubiquilins exhibit self-association behavior manifesting as condensates, they follow distinct courses of phase-separation and levels of aggregation. This variability among ubiquilins along the continuum from liquid-like to solid likely informs both the normal ubiquitin-linked functions of ubiquilins and their accumulation and potential contribution to toxicity in various neurodegenerative diseases. Abstract: The transient voltage-gated K+ current (IA) mediated by Kv4.2 in CA1 hippocampal pyramidal neurons regulates dendritic excitability, synaptic plasticity, and learning. Here we report that Ca2+ entry mediated by the voltage-gated Ca2+ channel subunit Cav2.3 regulates Kv4.2 function in CA1 pyramidal neurons through Ca2+ binding auxiliary subunits known as K+ channel interacting proteins (KChIPs). We characterized an interaction between Cav2.3 and Kv4.2 using immunofluorescence colocalization, coimmunoprecipitation, electron microscopy, FRAP, and FRET. We found that Ca2+-entry via Cav2.3 increases Kv4.2-mediated whole-cell current due in part to an increase in Kv4.2 surface expression. In hippocampal neurons, pharmacological block of Cav2.3 reduced whole-cell IA. We also found reduced IA in Cav2.3 knockout mouse neurons with a loss of the dendritic IA gradient. Furthermore, the Cav2.3-Kv4.2 complex was found to regulate the size of synaptic currents and spine Ca2+ transients. Mientras la crisis del petróleo hacía de las suyas en todo el planeta, en España los titulares de los periódicos, vigilados estrechamente por el Ministerio de Información y Turismo, nos dejaban perlas como la boda de Julio Iglesias con la señorita, entonces, Isabel Preysler, o la famosa fuga del penal del Puerto de Santa María de Eleuterio Sánchez, alias El Lute que, aprovechando el relajo y el jolgorio de la celebración de final de año, se deslizó de su celda, saltó la tapia y, aunque fue tiroteado por la policía que custodiaba la prisión, logró fugarse y pasó varios años en libertad mientras abundaban las noticias contradictorias sobre su paradero. Un dato más para ubicarnos en este año 1971. El ínclito y omnipresente Ministerio de Información y Turismo, coto privado del Sr. Fraga, ordena el cierre del diario Madrid, un periódico que, al parecer, no estaba dispuesto a callar ante los abusos del poder ilegalmente establecido. Bueno, no se conformaron con cerrarlo, había, además, que volarlo. Y así lo hicieron. Pero, bueno, centrándonos en la música que es lo que nos ocupa, en España triunfaban los cantautores. Personajes como Mari Trini, Patxi Andión o Victor Manuel. No obstante, el cantautor por excelencia y que triunfó por encima de todos ellos fue Joan Manuel Serrat. La multinacional CBS decide instalarse en nuestro país y para iniciar su actividad edita un disco de Joan Manuel Serrat que, inmediatamente llega al número uno y se convierte, según encuestas de hoy, en el mejor disco de la música popular española. Evidentemente es Mediterráneo, un trabajo lleno de pequeñas joyas, todas compuestas por el músico barcelonés con la excepción de “Vencidos”, un poema de León Felipe pero con música de Serrat. El álbum, grabado en Milán, se editó definitivamente en 1971. A este disco pertenece la canción «Aquellas pequeñas cosas», que contiene una de sus letras más personales y evocativas. Estuvo casi un año, de forma continuada, en la lista de los 10 discos más vendidos de España, y varias semanas como el número 1 absoluto, y todo esto a pesar de la estricta censura que había en su contra después del asunto de Eurovisión. Y es que este hombre se empeñaba en dar “pol saco” al régimen todo lo que podía. A quien se le ocurre! En 1975, once militantes del FRAP y ETA fueron acusados del asesinato de varios policías y se les condenó a muerte en Consejo de Guerra, aunque posteriormente se les conmutó la pena por prisión a seis de ellos. Cuando se llevó a cabo la ejecución de los cinco restantes, Serrat estaba en México y en rueda de prensa condenó al régimen franquista y las medidas represivas. Además, se solidarizó con la postura del presidente de México, Luis Echeverría Álvarez, que había mantenido la postura mexicana de reconocer solo al gobierno de la Segunda República Española, en el exilio. A raíz de estas declaraciones tuvo que exiliarse durante un año en México, porque en España se había decretado orden de búsqueda y captura. Además, tal y como ya había ocurrido en 1968, sus trabajos fueron retirados de la difusión y censurados por el régimen. Hay que ver como nos hizo adelgazar ese puñetero régimen. No podemos pasar por alto, de este disco, una de las bellas canciones de amor que yo he oído. Lucia. María Trinidad Pérez de Miravete, más conocida como Mari Trini fue una cantante y compositora española que gozó de enorme popularidad y éxito comercial tanto en España como en países de habla hispana durante las décadas de 1970 y 1980. Vendió más de diez millones de discos. Mari Trini tenia una marcada personalidad y fue una ferviente defensora del feminismo en sus letras, y lo hacía en un momento en el que, hacerlo, no era fácil en absoluto. Su álbum, Escúchame, incluye canciones como "Yo no soy esa" que es posiblemente la que mejor define la personalidad de Mari Trini y el mensaje que quería transmitir: que las mujeres deben ser libres para ser y actuar al margen de las normas, y que no deben supeditarse a los deseos y expectativas de los hombres. El título "Yo no soy esa" es revelador porque cita (y rebate) otra canción: "Yo soy esa", tema clásico del género de la copla (cantado por, entre otras, Isabel Pantoja) cuya letra alude a una mujer utilizada por los hombres. En este grupo selecto de músicos cantautores, y en lugar privilegiado se encuentra Cecilia, bueno, Evangelina Sobredo Galanes, nacida en Madrid el 11 de octubre de 1948. Cecilia fue una carismática cantautora española de música popular. Desgraciadamente falleció en un accidente de automóvil a los 27 años cuando estaba en la cima de su éxito. Uno más para el maldito club de los 27. Su carrera artística fue corta, pero consiguió gran popularidad con canciones como "Un ramito de violetas", "Mi querida España", "Dama, dama" o "Amor de medianoche", que lograron ser un éxito en España en la década de los setenta. No se que lectura tendrá hoy una letra como esta Dama Dama, una crítica social sobre el falso puritanismo de algunas mujeres, pero en su momento y contexto, fue muy reveladora. Cecilia supo aunar la música británica y norteamericana que había escuchado en su juventud mientras residía en Reino Unido y Estados Unidos con la música y literatura españolas, y todo ello envuelto en un estilo absolutamente personal y único. El nacimiento en una familia de la carrera, su padre era diplomático, le facilitó una educación de un nivel superior a la media, y lo que fue más importante, un contacto con un ambiente internacional que le serviría para tomar una postura más exigente consigo misma de lo que en aquel momento había en la música nacional. Como muchos otros artistas, Cecilia tuvo algunos problemas con la censura existente en España durante la dictadura. Algunas de sus letras tuvieron que ser modificadas, ciertos temas fueron descartados de los discos y algunas de sus portadas fueron sustituidas. La canción "Dama, dama" fue suavizada en los versos que dicen "Puntual cumplidora del tercer mandamiento, algún desliz en el sexto" para dejar así el final del verso: "... algún desliz inconexo". El tema "Mi querida España" tuvo que ser modificado, y quedaron eliminadas las referencias a las Dos Españas (Esta España viva, esta España muerta, esta España blanca, esta España negra). Sin embargo, el texto fue publicado sin censura en la carpeta del LP. Cosas de la censura. Pero, bueno, en algunas de sus actuaciones en vivo, Cecilia colaba los textos originales. Entre los temas descartados para su publicación se encuentra la canción "Soldadito de plomo", que acabamos de escuchar, una crítica al ejército que nunca vio la luz en vida de Cecilia. El álbum más problemático fue Cecilia 2. Su título iba a ser "Me quedaré soltera" y una fotografía de Pablo Pérez-Mínguez ilustraría la portada, con una Cecilia que mostraba su embarazo. La compañía discográfica desechó la idea, cambiando el título al disco y utilizando una fotografía más discreta. El tema "Un millón de sueños" supuso a Evangelina pasar por el juzgado. Su título original era "Un millón de muertos" en referencia a la novela homónima de José Maria Gironella. Su alusión a la Guerra Civil española era directa. Cecilia declaró que se trataba de un tema acerca de la Guerra de los 6 días, que vivió en persona, y el tema no fue modificado salvo en el título, si bien recibió el calificativo de "no radiable" de los censores y quedó fuera de las emisoras de radio por un tiempo. Cantautor, doctor en Sociología, buen conversador, mejor comedor, vasco militante (de Madrid, «porque los vascos nacemos donde queremos»), cazador, motero... Patxi Andion. Como cantautor, que es lo que nos interesa, no tuvo la repercusión que a mi entender tuvieron otros con los mismos méritos. Una de sus muchas canciones con las que nos sorprendió fue esta semblanza de un maestro de escuela que, en la España de mediados del siglo pasado, tenía que pelearse con la ideología reinante en la época. Un retrato de la realidad cotidiana que nos tocó vivir. El apogeo de su carrera musical abarcó el periodo comprendido entre 1971 y 1978, durante el cual alcanzó notables cotas de calidad artística con temas como "Puedo inventar", "La casa se queda sola", "Tiempo, tiempo", entre otras. A partir de 1979, fue adquiriendo un estilo cada vez más hondo y personal, lo que le costó ser apartado paulatinamente de los circuitos comerciales. Por este motivo, o por otro, vete a saber, se centró en su faceta de actor, protagonizando alguna películas que, bueno, tampoco han marcado ningún hito en la historia del séptimo arte. Para mí, sigue siendo su música y la facilidad para realizar unos precisos y preciosos retratos sociales en los escasos versos de una canción, lo que da la medida de su calidad artística. Un retrato como el que hizo de una de las zonas más emblemáticas de Madrid, El rastro. La canción se titula Una, dos y tres. Nacido como Pedro Ample Candel, eligió el sobrenombre de Ruy-Blas debido a su admiración por la obra de Victor Hugo Ruy Blas, al grabar en 1970 su primer disco como cantante solista, trabajo con el que consiguió un importante éxito en España con la canción titulada A los que hirió el amor, la que con el paso de los años ha ido convirtiéndose en todo un clásico de la música popular española. Comenzó su prolífica carrera musical siendo apenas un adolescente. Primero formó parte de grupos como Los Príncipes y Los Grimm con quiénes grabó sus primeros discos para la compañía Philips. Poco después formó parte del legendario grupo Los Canarios (1968) durante el tiempo en que su director, Teddy Bautista, cumplía el servicio militar. Fue al término de su trabajo con este grupo cuando grabó A los que hirió el amor, producido por nuestro viejo conocido Alain Milhaud. El segundo disco de Pedro Ruy Blas, “Mi voz es amor”, estaba destinado a tomar el relevo de “A los Que Hirió el Amor”, y es que “Mi voz es amor”, la canción que da título al disco, era y es un gran tema que mezcla la clásica balada con un coro y una dicción solista góspel, con unos arreglos orquestales típicamente soul. También se recurrió a una canción foránea, que en 1969 había sido grabada por Derek Lawrence, productor de Deep Purple y por el propio grupo. En un principio fue bien acogido y vendió un número importante de ejemplares, pero a algún lumbreras del régimen le pareció que frases como “la paz que nadie consiguió yo no predico en el reino de los sordos, mi voz es amor” o “Yo soy el pueblo que cree en la libertad” escondían proclamas filocomunistas y el disco fue censurado en los medios de comunicación y borrado de las listas de éxitos, a pesar de no vender lo suficiente para siquiera haberse asomado a sus puestos bajos. Un varapalo que no tardaría en pasar recibo a la carrera del cantante. Fue a partir de ese momento cuando Pedro orientó su carrera hacia el jazz. Fundó el grupo de fusión y jazz-rock Dolores junto a jóvenes músicos como Jorge Pardo, el baterista José Antonio Galicia, Rubem Dantas y el pianista Tomás San Miguel, entre otros. En esta formación, descubrimos a Pedro como un excelente baterista y percusionista. Graban varios discos para Polydor, como Luna llena, Dolores, La Puerta Abierta, y Asa-Nisi-Masa. Al poco tiempo, Dolores se convirtió en uno de los más importantes grupos de la historia de la música progresiva en España, y así surgió la colaboración con el maestro Paco de Lucía y el mismísimo Camarón, impulsando de este modo una nueva época en el flamenco. «Cantemos como quien respira. Porque eso es la libertad, porque es decir que somos quienes somos, porque eso es el amor: respirar o cantar. Porque ambas cosas son la misma: Poesía» (Gabriel Celaya) Asi que… cantemos, cantemos y oigamos música porque eso nos hará libres y, si no nos hace libre, nos lo habremos pasado de miedo. Personajes como Mari Trini, Patxi Andión o Victor Manuel. No obstante, el cantautor por excelencia y que triunfó por encima de todos ellos fue Joan Manuel Serrat. La multinacional CBS decide instalarse en nuestro país y para iniciar su actividad edita un disco de Joan Manuel Serrat que, inmediatamente llega al número uno y se convierte, según encuestas de hoy, en el mejor disco de la música popular española. Evidentemente es Mediterráneo, un trabajo lleno de pequeñas joyas, todas compuestas por el músico barcelonés con la excepción de “Vencidos”, un poema de León Felipe pero con música de Serrat. El álbum, grabado en Milán, se editó definitivamente en 1971. A este disco pertenece la canción «Aquellas pequeñas cosas», que contiene una de sus letras más personales y evocativas. Estuvo casi un año, de forma continuada, en la lista de los 10 discos más vendidos de España, y varias semanas como el número 1 absoluto, y todo esto a pesar de la estricta censura que había en su contra después del asunto de Eurovisión. Y es que este hombre se empeñaba en dar “pol saco” al régimen todo lo que podía. A quien se le ocurre! En 1975, once militantes del FRAP y ETA fueron acusados del asesinato de varios policías y se les condenó a muerte en Consejo de Guerra, aunque posteriormente se les conmutó la pena por prisión a seis de ellos. Cuando se llevó a cabo la ejecución de los cinco restantes, Serrat estaba en México y en rueda de prensa condenó al régimen franquista y las medidas represivas. Además, se solidarizó con la postura del presidente de México, Luis Echeverría Álvarez, que había mantenido la postura mexicana de reconocer solo al gobierno de la Segunda República Española, en el exilio. A raíz de estas declaraciones tuvo que exiliarse durante un año en México, porque en España se había decretado orden de búsqueda y captura. Además, tal y como ya había ocurrido en 1968, sus trabajos fueron retirados de la difusión y censurados por el régimen. Hay que ver como nos hizo adelgazar ese puñetero régimen. No podemos pasar por alto, de este disco, una de las bellas canciones de amor que yo he oído. Lucia. María Trinidad Pérez de Miravete, más conocida como Mari Trini fue una cantante y compositora española que gozó de enorme popularidad y éxito comercial tanto en España como en países de habla hispana durante las décadas de 1970 y 1980. Vendió más de diez millones de discos. Mari Trini tenia una marcada personalidad y fue una ferviente defensora del feminismo en sus letras, y lo hacía en un momento en el que, hacerlo, no era fácil en absoluto. Su álbum, Escúchame, incluye canciones como "Yo no soy esa" que es posiblemente la que mejor define la personalidad de Mari Trini y el mensaje que quería transmitir: que las mujeres deben ser libres para ser y actuar al margen de las normas, y que no deben supeditarse a los deseos y expectativas de los hombres. El título "Yo no soy esa" es revelador porque cita (y rebate) otra canción: "Yo soy esa", tema clásico del género de la copla (cantado por, entre otras, Isabel Pantoja) cuya letra alude a una mujer utilizada por los hombres. En este grupo selecto de músicos cantautores, y en lugar privilegiado se encuentra Cecilia, bueno, Evangelina Sobredo Galanes, nacida en Madrid el 11 de octubre de 1948. Cecilia fue una carismática cantautora española de música popular. Desgraciadamente falleció en un accidente de automóvil a los 27 años cuando estaba en la cima de su éxito. Uno más para el maldito club de los 27. Su carrera artística fue corta, pero consiguió gran popularidad con canciones como "Un ramito de violetas", "Mi querida España", "Dama, dama" o "Amor de medianoche", que lograron ser un éxito en España en la década de los setenta. No se que lectura tendrá hoy una letra como esta Dama Dama, una crítica social sobre el falso puritanismo de algunas mujeres, pero en su momento y contexto, fue muy reveladora. Cecilia supo aunar la música británica y norteamericana que había escuchado en su juventud mientras residía en Reino Unido y Estados Unidos con la música y literatura españolas, y todo ello envuelto en un estilo absolutamente personal y único. El nacimiento en una familia de la carrera, su padre era diplomático, le facilitó una educación de un nivel superior a la media, y lo que fue más importante, un contacto con un ambiente internacional que le serviría para tomar una postura más exigente consigo misma de lo que en aquel momento había en la música nacional. Como muchos otros artistas, Cecilia tuvo algunos problemas con la censura existente en España durante la dictadura. Algunas de sus letras tuvieron que ser modificadas, ciertos temas fueron descartados de los discos y algunas de sus portadas fueron sustituidas. La canción "Dama, dama" fue suavizada en los versos que dicen "Puntual cumplidora del tercer mandamiento, algún desliz en el sexto" para dejar así el final del verso: "... algún desliz inconexo". El tema "Mi querida España" tuvo que ser modificado, y quedaron eliminadas las referencias a las Dos Españas (Esta España viva, esta España muerta, esta España blanca, esta España negra). Sin embargo, el texto fue publicado sin censura en la carpeta del LP. Cosas de la censura. Pero, bueno, en algunas de sus actuaciones en vivo, Cecilia colaba los textos originales. Entre los temas descartados para su publicación se encuentra la canción "Soldadito de plomo", que acabamos de escuchar, una crítica al ejército que nunca vio la luz en vida de Cecilia. El álbum más problemático fue Cecilia 2. Su título iba a ser "Me quedaré soltera" y una fotografía de Pablo Pérez-Mínguez ilustraría la portada, con una Cecilia que mostraba su embarazo. La compañía discográfica desechó la idea, cambiando el título al disco y utilizando una fotografía más discreta. El tema "Un millón de sueños" supuso a Evangelina pasar por el juzgado. Su título original era "Un millón de muertos" en referencia a la novela homónima de José Maria Gironella. Su alusión a la Guerra Civil española era directa. Cecilia declaró que se trataba de un tema acerca de la Guerra de los 6 días, que vivió en persona, y el tema no fue modificado salvo en el título, si bien recibió el calificativo de "no radiable" de los censores y quedó fuera de las emisoras de radio por un tiempo. Cantautor, doctor en Sociología, buen conversador, mejor comedor, vasco militante (de Madrid, «porque los vascos nacemos donde queremos»), cazador, motero... Patxi Andion. Como cantautor, que es lo que nos interesa, no tuvo la repercusión que a mi entender tuvieron otros con los mismos méritos. Una de sus muchas canciones con las que nos sorprendió fue esta semblanza de un maestro de escuela que, en la España de mediados del siglo pasado, tenía que pelearse con la ideología reinante en la época. Un retrato de la realidad cotidiana que nos tocó vivir. El apogeo de su carrera musical abarcó el periodo comprendido entre 1971 y 1978, durante el cual alcanzó notables cotas de calidad artística con temas como "Puedo inventar", "La casa se queda sola", "Tiempo, tiempo", entre otras. A partir de 1979, fue adquiriendo un estilo cada vez más hondo y personal, lo que le costó ser apartado paulatinamente de los circuitos comerciales. Por este motivo, o por otro, vete a saber, se centró en su faceta de actor, protagonizando alguna películas que, bueno, tampoco han marcado ningún hito en la historia del séptimo arte. Para mí, sigue siendo su música y la facilidad para realizar unos precisos y preciosos retratos sociales en los escasos versos de una canción, lo que da la medida de su calidad artística. Un retrato como el que hizo de una de las zonas más emblemáticas de Madrid, El rastro. La canción se titula Una, dos y tres. Nacido como Pedro Ample Candel, eligió el sobrenombre de Ruy-Blas debido a su admiración por la obra de Victor Hugo Ruy Blas, al grabar en 1970 su primer disco como cantante solista, trabajo con el que consiguió un importante éxito en España con la canción titulada A los que hirió el amor, la que con el paso de los años ha ido convirtiéndose en todo un clásico de la música popular española. Comenzó su prolífica carrera musical siendo apenas un adolescente. Primero formó parte de grupos como Los Príncipes y Los Grimm con quiénes grabó sus primeros discos para la compañía Philips. Poco después formó parte del legendario grupo Los Canarios (1968) durante el tiempo en que su director, Teddy Bautista, cumplía el servicio militar. Fue al término de su trabajo con este grupo cuando grabó A los que hirió el amor, producido por nuestro viejo conocido Alain Milhaud. El segundo disco de Pedro Ruy Blas, “Mi voz es amor”, estaba destinado a tomar el relevo de “A los Que Hirió el Amor”, y es que “Mi voz es amor”, la canción que da título al disco, era y es un gran tema que mezcla la clásica balada con un coro y una dicción solista góspel, con unos arreglos orquestales típicamente soul. También se recurrió a una canción foránea, que en 1969 había sido grabada por Derek Lawrence, productor de Deep Purple y por el propio grupo. En un principio fue bien acogido y vendió un número importante de ejemplares, pero a algún lumbreras del régimen le pareció que frases como “la paz que nadie consiguió yo no predico en el reino de los sordos, mi voz es amor” o “Yo soy el pueblo que cree en la libertad” escondían proclamas filocomunistas y el disco fue censurado en los medios de comunicación y borrado de las listas de éxitos, a pesar de no vender lo suficiente para siquiera haberse asomado a sus puestos bajos. Un varapalo que no tardaría en pasar recibo a la carrera del cantante. Fue a partir de ese momento cuando Pedro orientó su carrera hacia el jazz. Fundó el grupo de fusión y jazz-rock Dolores junto a jóvenes músicos como Jorge Pardo, el baterista José Antonio Galicia, Rubem Dantas y el pianista Tomás San Miguel, entre otros. En esta formación, descubrimos a Pedro como un excelente baterista y percusionista. Graban varios discos para Polydor, como Luna llena, Dolores, La Puerta Abierta, y Asa-Nisi-Masa. Al poco tiempo, Dolores se convirtió en uno de los más importantes grupos de la historia de la música progresiva en España, y así surgió la colaboración con el maestro Paco de Lucía y el mismísimo Camarón, impulsando de este modo una nueva época en el flamenco. «Cantemos como quien respira. Porque eso es la libertad, porque es decir que somos quienes somos, porque eso es el amor: respirar o cantar. Porque ambas cosas son la misma: Poesía» (Gabriel Celaya) Asi que… cantemos, cantemos y oigamos música porque eso nos hará libres y, si no nos hace libre, nos lo habremos pasado de miedo. Compartment models are a frequently used tool for imaging data gained with medical and biological imaging techniques. The solutions of the differential equations derived from a compartment model provide nonlinear parametric functions, based on which the behavior of a concentration of interest over time can be described. Often, the number of compartments in a compartment model is unknown. As the model complexity itself, which is, the number of compartments, is certainly an important information, it is desirable to estimate it from the observed data. Additionally, the unknown parameters have to be estimated. Therefore, methods dealing with both the parameter estimation and model selection in compartment models are needed. The methods proposed in this thesis are motivated by two applications from the field of medical and biological imaging. In the first application, the quantitative analysis of Fluorescence recovery after photobleaching (FRAP) data, compartment models are used in order to gain insight into the binding behavior of molecules in living cells. As a first approach, we developed a Bayesian nonlinear mixed-effects model for the analysis of a series of FRAP images. Mixed-effect priors are defined on the parameters of the nonlinear model, which is a novel approach. With the proposed model, we get parameter estimates and additionally gain information about the variability between nuclei, which has not been studied so far. The proposed method was evaluated on half-nucleus FRAP data, also in comparison with different kinds of fixed-effects models. As a second approach, a pixelwise analysis of FRAP data is proposed, where information from the neighboring pixels is included into the nonlinear model for each pixel. This is innovative as the existing models are suitable for the analysis of FRAP data for some regions of interest only. For the second application, the quantitative analysis of dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) of the breast, we use a compartment model which describes the exchange of blood between different, well-mixed compartments. In the analysis of such data, the number of compartments allows conclusions about the heterogeneity of cancerous tissue. Therefore, an estimation and model selection approach based on boosting, with which the number of compartments and the unknown parameters can be estimated at the voxel level, is proposed. In contrast to boosting for additive regression, where smoothing approaches are used, boosting in nonlinear parametric regression as described in this thesis is a novel approach. In an extension of this approach, the spatial structure of an image is taken into account by penalizing the differences in the parameter estimates of neighboring voxels. The evaluation of the method was done in simulation studies, as well as in the application to data from a breast cancer study. The majority of the program code used in the three approaches was newly developed in the programming languages R and C. Based on that code, two R packages were built.

Targeting the cytoskeleton (CSK) of cancer cells offers a valuable strategy in cancer therapy. Whereas drugs which address microtubule CSK such as vinca alkaloids or taxanes are well established in the clinic, compounds binding to the actin CSK are still far away from their therapeutical application. One reason might be the lacking knowledge on their mode of cytotoxicity and moreover their tumor specific mechanism of action. We used the myxobacterial compound Chondramide as a tool to first elucidate the mechanisms of cytotoxicity by actin targeting in different breast cancer cells, namely MCF7 and MDA-MB-231. Chondramide inhibits actin filament assembly and dynamics shown by a fluorescence-based analysis (FRAP) in whole cells and leads to apoptosis characterized by phosphatidylserine exposure, release of cytochrome C from mitochondria and finally activation of caspases (-9 and -3). Detailed analysis revealed, that Chondramide induces apoptosis by enhancing the occurrence of mitochondrial permeability transition (MPT). Known MPT-modulators were found to be affected by Chondramide: Hexokinase II (HkII) bound to the voltage dependent anion channel (VDAC) translocated from the outer mitochondrial membrane to the cytosol and the proapoptotic protein Bad was recruited to the mitochondria. Importantly, PKCε, a prosurvival serine/threonine kinase possessing an actin-binding site and known to regulate the HkII/VDAC interaction as well as Bad phosphoylation was identified as the link between actin CSK and apoptosis induction. PKCε which was found overexpressed in breast cancer cells accumulated in actin bundles induced by Chondramide and lost its activity. The second goal of our work was to inform on a potential tumor specific action of actin binding agents such as Chondramide. As the nontumor breast epithelial cell line MCF-10A in fact shows resistance to Chondramide induced apoptosis and notably express very low level of PKCε we claim that trapping PKCε via Chondramide induced actin hyperpolymerization displays tumor cell specificity. Our work provides a link between targeting the ubiquitously occurring actin CSK and selective inhibition of pro-tumorigenic PKCε, thus setting the stage for actin-stabilizing agents as innovative cancer drugs. This is moreover supported by the in vivo efficacy of Chondramide triggered by abrogation of PKCε signaling shown in a xenograft breast cancer model. For the actin targeting compound Doliculide we could show that Doliculide impairs the dynamics of the actin CSK similar to Chondramide. Moreover, it reduces the proliferation rate and migration of cancer cells and also leads to the induction of apoptosis, thus Doliculide is also an interesting lead structure for further preclinical investigations.

The peptidoglycan cell wall (CW) and the actin-like MreB cytoskeleton are the majordeterminants of cell morphology in non-spherical bacteria. Bacillus subtilis is a rod-shaped Grampositive bacterium that has three MreB isoforms: MreB, Mbl (MreB–like) and MreBH (MreBHomologue). Over the last decade, all three proteins were reported to localize in dynamic filamentous helical structures running the length of the cells underneath the membrane. This helical pattern led to a model where the extended MreB structures act as scaffolds to position CW-synthesizing machineries along sidewalls. However, the dynamic relationship between the MreB cytoskeleton and CW elongation complexes remained to be elucidated. Here we describe the characterization of the dynamics of the three MreB isoforms, CW synthesis and elongation complexes in live Bacillus subtilis cells at high spatial and temporal resolution. Using total internal reflection fluorescence microscopy (TIRFM) we found that MreB, Mbl and MreBH actually do not assemble into an extended helical structure but instead into discrete patches that move processively along peripheral tracks perpendicular to the long axis of the cell. We found similar patch localization and dynamics for several morphogenetic factors and CW-synthesizing enzymes including MreD, MreC, RodA, PbpH and PBP2a. Furthermore, using fluorescent recovery after photobleaching (FRAP), we showed that treadmilling of MreB filaments does not drive patch motility, as expected from the structural homology to actin. Blocking CW synthesis with antibiotics that target different steps of the peptidoglycan biosynthetic pathway stopped MreB patches motion, suggesting that CW synthesis is the driving force of patch motility. On the basis of these findings, we proposed a new model for MreB fuction in which MreB polymers restrict and orient patch motility to ensure controlled lateral CW expansion, thereby maintaining cell shape. To further investigate the molecular mechanism underlying MreB action, we next performed a site-directed mutagenesis analysis. Alanine substitutions of three charged amino 2 acids of MreB generated a B. subtilis strain with cell shape and growth defects. TIRFM analysis revealed that the mutated MreB protein displayed wild-type localization and dynamics, suggesting that it is still associated to the CW elongation machinery but might be defective in an interaction important for MreB morphogenetic function. Thus, this mutant appears as as a good candidate to start characterizing the interactions between the three MreB isoforms and components involved in CW elongation. It might also help to understand the function of components of theCWsynthetic complexes, and how they are coordinated to achieve efficient CW synthesis. Finally, to investigate how the integrity of the CW is maintained, we studied the localization and dynamics of the LiaIH-system, which i s t he t arget o f L iaRS, a t wo-component system involved in cell envelope stress response. We found that under stress conditions, when liaI and LiaH genes are expressed, the proteins form static complexes that coat the cell membrane. LiaI is required for the even distribution of the LiaH in the membrane. Taken together, these data suggest that LiaIH complexes may protect the cell from CW damage. Taken together, the findings described in this thesis provide valuable insights into the understanding of CW synthesis in B. subtilis, which may open new perspectives for the design of novel antimicrobial agents.

Dynamic imaging series acquired in medical and biological research are often analyzed with the help of compartment models. Compartment models provide a parametric, nonlinear function of interpretable, kinetic parameters describing how some concentration of interest evolves over time. Aiming to estimate the kinetic parameters, this leads to a nonlinear regression problem. In many applications, the number of compartments needed in the model is not known from biological considerations but should be inferred from the data along with the kinetic parameters. As data from medical and biological experiments are often available in the form of images, the spatial data structure of the images has to be taken into account. This thesis addresses the problem of parameter estimation and model selection in compartment models. Besides a penalized maximum likelihood based approach, several Bayesian approaches-including a hierarchical model with Gaussian Markov random field priors and a model state approach with flexible model dimension-are proposed and evaluated to accomplish this task. Existing methods are extended for parameter estimation and model selection in more complex compartment models. However, in nonlinear regression and, in particular, for more complex compartment models, redundancy issues may arise. This thesis analyzes difficulties arising due to redundancy issues and proposes several approaches to alleviate those redundancy issues by regularizing the parameter space. The potential of the proposed estimation and model selection approaches is evaluated in simulation studies as well as for two in vivo imaging applications: a dynamic contrast enhanced magnetic resonance imaging (DCE-MRI) study on breast cancer and a study on the binding behavior of molecules in living cell nuclei observed in a fluorescence recovery after photobleaching (FRAP) experiment.

Fluorescence microscopy provides an efficient and unique approach to study fixed and living cells because of its versatility, specificity, and high sensitivity. Fluorescence microscopes can both detect the fluorescence emitted from labeled molecules in biological samples as images or photometric data from which intensities and emission spectra can be deduced. By exploiting the characteristics of fluorescence, various techniques have been developed that enable the visualization and analysis of complex dynamic events in cells, organelles, and sub-organelle components within the biological specimen. The techniques described here are fluorescence recovery after photobleaching (FRAP), the related fluorescence loss in photobleaching (FLIP), fluorescence localization after photobleaching (FLAP), Forster or fluorescence resonance energy transfer (FRET) and the different ways how to measure FRET, such as acceptor bleaching, sensitized emission, polarization anisotropy, and fluorescence lifetime imaging microscopy (FLIM). First, a brief introduction into the mechanisms underlying fluorescence as a physical phenomenon and fluorescence, confocal, and multiphoton microscopy is given. Subsequently, these advanced microscopy techniques are introduced in more detail, with a description of how these techniques are performed, what needs to be considered, and what practical advantages they can bring to cell biological research.

Neural stem cells are defined by their unique ability to undergo self-renewal divisions. By dividing asymmetrically, a stem cell simultaneously produces a daughter cell that retains stem cell identity, whereas the other starts to differentiate and contributes to a continuous supply of neural cell types. Drosophila neuroblasts provide an excellent model system to study asymmetric stem cell divisions. The first part of this thesis will concentrate on the important adaptor protein Miranda which ensures the asymmetric segregation of cell fate determinants to the differentiating ganglion mother cell during neuroblast mitosis. The dynamic apical-then-basal localization pattern and the requirement for both Myosin II and Myosin VI suggested that Miranda is actively transported to the basal pole as a myosin cargo. However, immunofluorescence studies combined with time-lapse confocal microscopy and FRAP analyses revealed that Miranda reaches the basal cortex by passive diffusion throughout the cell rather than by long range myosin-directed transport. Instead, myosins play an indirect role in asymmetric Miranda localization. The formation of active Myosin II filaments in early prophase results in the exclusion of Miranda from the apical cortex. In the cytoplasm, Miranda diffuses three-dimensionally through the cell and becomes restricted to the basal half of the metaphase neuroblast by Myosin VI to facilitate its interaction with a putative basal cortical anchor. There is growing evidence that deregulation of the self-renewing process of stem cells may be an early event in tumorigenesis and that many cancers contain a small population of so called cancer stem cells which are responsible for maintenance and growth of tumors. The second part of the thesis will report on the isolation of cells with stem-like features from a murine mouse model of oligodendroglioma with activated EGFR signaling and loss of the tumor suppressor p53 in the postnatal stem cell lineage. Although oligodendroglioma-derived progenitor cells share many similarities with normal neural stem cells, they have increased self-renewing and proliferation capacities and in addition, undergo aberrant differentiation. They are multipotential, however, when induced to differentiate they preferentially generate cells of the oligodendrocytic lineage recapitulating the properties of the tumor they originate from. Brain cancer derived stem-like cells generate new tumors following intracranial injections that faithfully reproduce the phenotype of the parental tumor qualifying them as cancer stem cells. Interestingly, neural stem cells isolated from tumor prone mice long before oligodendroglioma occurrence show similar, but less severe alterations in their self-renewing and differentiation capacities. Importantly, they never form orthotopic tumors and thus were referred to as premalignant stem cells. The overproduction of oligodendrocytic cells is caused by a defect in asymmetric cell division that is very likely accompanied with genetic instabilities and epigenetic alterations. This results strengthen the hypothesis that early defects in neural stem cells, together with additional genetic alterations lead to the progression to a more malignant stem cell type which is responsible for tumor growth and maintenance.

Various stimuli like thrombin induce endothelial cell shape change and stress fiber formation via Rho/Rho-kinase-mediated reorganization of the actin cytoskeleton. LIM-kinases regulate actin cytoskeletal reorganization through phosphorylation of cofilin at Ser3. The LIMK family kinases possess characteristic structural features, consisting of two LIM domains, a PDZ domain and a C-terminal kinase domain. In cell transfection studies it has been shown that LIMK2 is phosphorylated at Thr505 by Rho-kinase thereby activating the enzyme. Recently it has been reported that nuclear LIMKs suppress cyclin D1 expression in a manner independent of cofilin phosphorylation and actin polymerization. In this study, we found that endothelial cells express both LIMK1 and LIMK2. By using live cell imaging, we confirm previous findings that thrombin induces stress fiber formation, ruffle formation and cell contraction. Furthermore, the cell-cell contacts were disrupted and F-actin fibers connecting two cells were broken. Thrombin induced a rapid and sustained Rho-kinase activation and subsequent phosphorylation of LIM-kinase and cofilin. Pretreatment of endothelial cells with the specific Rho-kinase inhibitor Y27632 inhibited MYPT1 phosphorylation, LIM-kinase and cofilin phosphorylation and blocked stress fiber formation in thrombin-stimulated cells. Notably, thrombin induced actin stress fiber formation was abolished in cells transfected with dominant negative LIMK2. LIMK2 was mainly localized in the cytoplasm. By using Leptomycin B (a specific inhibitor of CRM-1 dependent nuclear export) and FRAP and FLIP analysis, we demonstrate that LIMK2 in resting endothelial cells shuttles between the nucleus and cytoplasm. The LIM domains of LIMK2, but not of LIMK1 inhibited its nuclear import thereby keeping LIMK2 mainly in the cytoplasm. Mutational analysis of the unique basic amino acid-rich motif (amino acids 480-503) indicated that this motif regulates the nuclear and nucleolar localization of LIMK2. Activation of PKC in PMA-stimulated endothelial cells stimulated the phosphorylation of LIMK2 at Ser283 and the translocation of LIMK2 and the PDZ-kinase construct of LIMK2 from the nucleus to the cytoplasm. Of the various PKC isoforms, PKC- and PKC- were found to be mainly responsible for Ser283 phosphorylation and the regulation of translocation of LIMK2. Mutational analysis indicated that LIMK2 phosphorylation at Ser283 and Thr494 play a role in the regulation of nucleocytoplasmic shuttling of LIMK2 by PKC. These results show that LIM-kinase activation is mediated by Rho-kinase in stimulated endothelial cells, and that LIM-kinase-mediated cofilin phosphorylation plays an essential role in thrombin-induced stress fiber formation. LIMK2 shuttles between nucleus and cytoplasm in resting endothelial cells. Phosphorylation of LIMK2 at Ser283 and Thr494 by PKC regulates nucleocytoplasmic shuttling and suggests that LIMK2 might also have a function in the nucleus such as the suppression of cyclin D1 expression.
