SCP-047 is a heavily rusted, breached gas cylinder made of an iron-[REDACTED] alloy. When exposed to open air, the material of the cylinder evaporates slowly, producing a previously undocumented mutagenic gas. This gas has no effect on eukaryotic organisms (e.g. humans), but profoundly alters prokaryotes, showing preference for common human microbiota - the natural microorganisms that live on the skin and throughout the body. On rare occasions these mutations produce a "superbug" (collectively known as SCP-047-1), a natural commensal with enhanced survivability and therefore opportunistic pathogenicity. There’s an extra green mole on his face not accounted for in our planetary system, and that’s with all the moons! Look at the elliptical; its orbit takes it in around the back of our moon! Ever since that day, Lilo McGregor disappeared from my head and this new sound built slowly up in my head over twelve fuckim' years... more and more... That’s why I got into conspiracy theories, and reporting the Soupalon invasion. Which was REAL! And I was RIGHT. And FUCK You!! Busher: Peah... I always had moles and I was nowhere near the factory twelve year ago, but but... PEAH! Somehow with Baldy next door and Laura... I must have picked it up from Laura... Eh… Well, I suppose… I-I-I used to steal her used toothbrushes an’ n’ n’ other things right, from their rubbish bin... I had me a horde under the bed and I may have put one or two in me mouth... once or twice like... Tucker: WOT? They may have shared a toothbrush Busher! ... Did you ever think of that? Busher: AH Gross! WHA? PEAH! AHRGH! SPIT IT OUT! Baldy in me mouth! No! NO! Jagger: What did he just say? Arlee: He may have gone other places too with the toothbrush I reckon hehheheh. Busher: MAMAAAY! Tucker: That’s how Baldy became pregnant! Of course! Makes logical sense now! He got jellied like the rest of us. Fack! Dickus: The Vat of Mutagen. The factory foreman Gerry Freely set it up for us... that was for the... ah... actually I don’t really know... but I’m sure it was... eh... for the good of Earth Society! Busher: PEAH! MMMNNLLLMMMLLL. LIAR! Tucker: What was that green jelly for Dickus? Dickus: That’s not important now, what is, is you lads have been infected with a MUTAGEN! You are all MUTANTS! Skilled up and all powerful! Jagger: I slow down time! Heeheheheeeee!!! Good for workin’ out the riffs in songs! Don’t need to go to Waltons anymore... Arlee: Why did you get a good one? I got schizophrenia! Tucker: Oh no... Gabbler: What Tucks? What is it now? Are ya alright? Tucker: NO! I’ve realised that I don’t know how to control my superior intelligence! Oh Look! Rigel... It’s a bright one... on the scale of magnitudes it’s in the negative numbers which means it’s very bright and numbers on the opposite end up are very faint. So Sirius has an MV of -1.42, our Moon -12.5 and the Sun is -26.5! Oiiright Rigel! -1.36! Gabbler: Yeah yeah and Brussel Sprouts are like pubic hair Arlee: Aha good one Gabbler- Jagger: What was the joke lads? Dickus: So we have the United Mutations heh? And Spikus, we have a map! Busher, get your face into the cockpit till we get to the Savage Garden! This is going to tear the skin off your eyeballs... 12 years on from 'first contact' gone wrong, Baldy Kendall has been snatched by the Soupalon race. Baldy’s neighbour Busher sees his own cat Ginger climb aboard the departing alien orb. Busher hates Baldy but loves Ginger. He will find her! This is a quest for Ginger the Cat. Die Transplantation gesunder Organe, insbesondere der Nieren, zur Therapie unheilbarer Erkrankungen stellt eine der bedeutendsten Errungenschaften der modernen Medizin dar. Die zur Vermeidung der Organabstoßung erforderliche langzeitige Immunsuppression bein-haltet jedoch auch ein gegenüber der Normalbevölkerung erhöhtes Risiko für die Entstehung maligner Erkrankungen, so vor allem von Plattenepithelkarzinomen der Haut und malignen Lymphomen. Auffällig ist weiterhin auch die vermehrte Aggressivität dieser Malignomerkran-kungen, die sich in rascher Progredienz und Metastasierung und damit verbundener erhöhter Letalität äußert. Als mögliche Ursache dieser Phänomene kommen eine erhöhte Mutagen-sensitivität der DNS, wie auch eine reduzierte Reparaturkapazität der DNS-Reparaturmechanismen durch die chronische Behandlung mit immunsuppressiven Medika-menten in Betracht. Ziel dieser Arbeit sollte es daher sein, anhand der etablierten Verfahren Comet Assay und Mikrokerntest, Aussagen über die Reparaturkapazität und die Genomstabilität bei nieren-transplantierten Patienten zu ermöglichen. Untersucht wurde ein Kollektiv von 40 nierentransplantierten Patienten, darunter 21 Männer und 19 Frauen, denen ein Kontrollkollektiv von ebenfalls 21 Männern und 19 Frauen zum Vergleich gegenübergestellt wurde. Als zu untersuchendes Zellmaterial dienten Lymphozy-ten des peripheren Blutes. Der Comet Assay wurde als quantitatives Verfahren zur Bestimmung fremdstoffinduzierter DNS-Schäden an einzelnen, nicht proliferierenden Zellen eingesetzt. Die Untersuchung von DNS-Reparaturereignissen über einen definierten Zeitraum wurde hierdurch ermöglicht. Nach Induktion eines DNS-Schadens durch Zugabe eines mutagenen Agens wurden über einen Zeitraum von 60 Minuten Reparaturereignisse an der DNS von Lymphozyten quantitativ erfasst. Der Mikrokerntest wurde als quantitatives Verfahren zur Untersuchung erhöhter Genomin-stabilität sowohl aufgrund individueller Disposition als auch als Folge der Einwirkung muta-gener Substanzen verwendet. Eine erhöhte Genominstabilität, wie sie in der Bildung von Mikrokernen zum Ausdruck kommt, kann ihre Ursache sowohl in einer erhöhten Mutagen-sensitivität als auch in einem Defizit der DNS-Reparaturmechanismen haben. In der vorliegenden Arbeit wurde der Mikrokerntest an unbehandelten Lymphozyten der Pa-tienten- und der Kontrollgruppe durchgeführt, um Aussagen über die spontane Brüchigkeit des Genoms in beiden Gruppen zu erlauben. Die Ergebnisse der mit der Methode des Comet Assays durchgeführten Lymphozytenrepara-turversuche zeigten bei Nierentransplantierten weder eine erhöhte Sensitivität gegenüber Mutagenen noch eine dem Normalkollektiv gegenüber reduzierte DNS-Reparaturkapazität. Vielmehr wurden beim Vergleich der Mediane der DNS-Migrationen mitunter signifikant ver-minderte Reparaturaktivitäten der Lymphozyten-DNS des Kontrollkollektivs beobachtet. Beim Vergleich der Mittelwerte dagegen ergab sich zu den meisten Versuchszeitpunkten kein sig-nifikanter Unterschied zwischen Nierentransplantierten und Kontrollpersonen. Bei der Durchführung des Mikrokerntests wurden bei Nierentransplantierten wie auch bei Kontrollpersonen keine signifikanten Unterschiede der Mikrokernfrequenzen verzeichnet. Bei den weiblichen Studienteilnehmern fanden sich insgesamt signifikant höhere Mikrokernraten als bei den männlichen, unabhängig davon, ob diese zum Patienten- oder Kontrollkollektiv gehörten. Somit konnte die Hypothese, dass bei Nierentransplantierten mit einer erhöhten Sensitivität der DNS gegenüber schädigenden Substanzen oder mit einer Reduktion der DNS-Reparaturfähigkeit zu rechnen sei, durch die Ergebnisse dieser Studie nicht gestützt werden. Da es sich bei der Karzinogenese jedoch um einen komplexen Prozess handelt, bei dem zahlreiche Faktoren beteiligt sind, kommt auch in Zukunft der Forschung nach möglichen Ursachen große Bedeutung zu. Nur durch Kenntnis der beteiligten Mechanismen und durch die Einschränkung der damit verbundenen Risiken ist ein verbessertes Langzeitüberleben organtransplantierter Patienten möglich.

In der vorliegenden Arbeit wurde ein neuer Vektor (pGrec) zur Messung der homologen Rekombination (HR) in mitotischen Säugerzellen kloniert und etabliert. pGrec enthält eine HR-Kassette mit zwei Allelen des GFP (green fluorescent protein)-Gens, die durch differentielle Mutagenese inaktiviert wurden. Diese Allele können nur durch HR zu einem Wildtyp-Allel rekonstituiert werden, dessen zelluläre Expression mittels Fluoreszenz-Mikroskopie und Durchflusszytometrie nachweisbar ist. Die Detektion mittels Durchflusszytometer erlaubt es, seltene GFP+-Zellen zu isolieren und als rekombinante Subklone zur weiteren molekularbiologischen, biochemischen und zellulären Analyse zu etablieren. Das pGrec-HR-Meßsystem wurde an zwei isogenen Zellpärchen angewendet. Nach chromosomaler Integration in die Nager-Zelllinien V79B und deren mutierte Variante CL-V4B (Rad51C(-/-) (Godthelp et al., 2002)) konnte die vermutete Störung der HR in den Rad51C-defizienten CL-V4B-Zellen (Masson et al., 2001; Takata et al., 2001; French et al., 2002; Godthelp et al., 2002) eindeutig nachgewiesen werden: Im Vergleich zur Wildtyp-Zelllinie V79B ist die intrachromosomale Rekombinationsfrequenz der HR-Kassette in der Rad51C-Mutante statistisch signifikant erniedrigt (6-fach). Ebenso scheint die Rate der homologen Rekombination in Fluktuations-Analysen reduziert zu sein (2,7-fach). Somit konnte erstmalig direkt eine Beteiligung von Rad51C an HR gezeigt werden, deren Störung mit der Mutagenüberempfindlichkeit (MMS) und chromosomalen Instabilität von CL-V4B-Zellen korreliert. Kultivierte Zellen von A-T-Patienten sind charakterisiert durch chromosomale Hyper-Rekombination (Meyn, 1993; Luo et al., 1996). Das pGrec-System wurde in AT-Zellen auf seine Anwendbarkeit zur Messung extrachromosomaler Rekombination getestet, um ungleiche Einflüsse der Chromatinstruktur an einem spezifischen Integrationsort zu umgehen. Das Zellpärchen AT-pEBS (Atm(-/-), transfiziert mit leerem Kontroll-Vektor) und AT-YZ (Atm(-/-), ATM ektopisch komplementiert (Ziv et al., 1997)) wurde mit der für humane Zellen episomalen Version des pGrec-Vektors transfiziert. Die Rekombinationsrate in zwei untersuchten Subklonen der AT-pEBS-Zelllinie war im Vergleich zu zwei AT-YZ-Subklonen statistisch signifikant erhöht (34 - 43-fach). Somit wäre erstmals eine Hyperrekombination von AT-Zellen auch in episomalen Loci gezeigt. Die Bestimmung der Rekombinationsfrequenz ergab jedoch, dass diese in der Gesamtpopulation ATM-komplementierter AT-Zellen erhöht zu sein scheint (6-fach). Mögliche Gründe für die unterschiedlichen Ergebnisse, die mit verschiedenen experimentellen Ansätzen in den AT-Zelllinien erhalten wurden, werden diskutiert. Das pGrec-System ist als leicht modifizierbarer Vektor konzipiert und somit prinzipiell in allen Säugerzellen anwendbar. Insgesamt stellt pGrec also ein sehr sensitives und nützliches Werkzeug zur Charakterisierung von HR auf molekular-biologischer, biochemischer und zellulärer Ebene dar.
