This Day in Legal History: Civil Rights Act of 1957On August 29, 1957, the U.S. Congress passed the Civil Rights Act of 1957, marking the first federal civil rights legislation enacted since the Reconstruction era. This landmark act aimed to address racial discrimination and was a significant step in the ongoing struggle for civil rights in America. The law established the U.S. Commission on Civil Rights, a bipartisan body tasked with investigating voter discrimination and other civil rights violations. Additionally, it created the Civil Rights Division within the U.S. Department of Justice, empowering federal prosecutors to seek injunctions against those who violated voting rights. Although the act faced significant opposition and was weakened by compromises, it paved the way for future, more comprehensive civil rights legislation. It also symbolized the federal government's renewed commitment to addressing racial inequality, setting the stage for the civil rights movements of the 1960s. By way of background, the Biden administration's FY 2025 budget proposes a 25% wealth tax on unrealized gains for individuals owning over $100 million in assets. Unrealized gains are the increase in an asset's value that hasn't been sold or converted into cash, meaning the gains exist only on paper. This proposal largely affects the ultra-wealthy, with the average taxpayer not impacted.To illustrate, consider someone who invests $250,000 in a stock, and its value increases 40 times within a year. Even with this substantial gain, the investor wouldn't face the unrealized gains tax unless their shares appreciated another ten times, reaching a value of $100 million. At that point, they would owe $24.9 million in taxes, requiring them to sell a portion of their shares. Despite the hefty tax bill, they would still retain significant wealth.Critics worry that such a tax might lead to a stock market downturn if large shareholders sell off assets to cover their tax obligations. However, selling shares to fund other projects is common, and similar concerns about market instability aren't typically raised in those cases. In sum, you almost certainly needn't worry about an unrealized gains tax if you are listening to this–and should be more concerned about what kinds of services you can expect to receive from the revenue raised from such a tax. If I may briefly editorialize: think about the earlier story, where student debt relief has been shelved–or possibly killed entirely. The only time the national discourse revolves around what a policy will cost–the cost would be borne by the ultra-wealthy and benefits would flow to low and middle-income households. Abstract: Ribosomes catalyze protein synthesis by cycling through various functional states. These states have been extensively characterized in vitro, yet their distribution in actively translating human cells remains elusive. Here, we optimized a cryo-electron tomography-based approach and resolved ribosome structures inside human cells with a local resolution of up to 2.5 angstroms. These structures revealed the distribution of functional states of the elongation cycle, a Z tRNA binding site and the dynamics of ribosome expansion segments. In addition, we visualized structures of Homoharringtonine, a drug for chronic myeloid leukemia treatment, within the active site of the ribosome and found that its binding reshaped the landscape of translation. Overall, our work demonstrates that structural dynamics and drug effects can be assessed at near-atomic detail within human cells. Copy rights belong to original authors. In this episode we feature the latest research by Richard Wong and colleagues at the Kanazawa University NanoLSI. The research described in this podcast was published in the Journal of Extracellular Vesicles, in November 2022 Kanazawa University NanoLSI website https://nanolsi.kanazawa-u.ac.jp/en/The offshoot of cells visualized in real timeIn a study recently published in the Journal of Extracellular Vesicles, researchers from Kanazawa University use high-speed microscopy to capture the dynamics of nanosized sacs released from cells.Small extracellular vesicles (sEVs) are tiny sacs released by cells to deliver chemical messengers to other cells. Since sEVs are compatible with biological tissue they are being investigated as carriers for nanodrugs. However, the impact of physiological stress—such as changes in temperature—on the structure of sEVs is obscure. A research team led by Richard Wong and Keesiang Lim at Kanazawa University has now used an advanced form of microscopy to elucidate these changes in real time.The temperature, acid, and salt levels in our bodies can fluctuate with factors such as disease. Thus, research on sEVs for drug development requires a deeper understanding of how stressful environments affect the vesicles' structure. For their study, the team first isolated sEVs from cells. Next, using a technique known as high-speed atomic force microscopy (HS-AFM) the structure of sEVs was revealed to be either spherical or ellipsoidal in shape. HS-AFM also enabled the researchers to accurately measure the sizes of sEVs without rupturing or damaging the vesicular membranes.The effect of varying temperatures on sEVs was the first parameter assessed. At temperatures higher than normal (37°C) body temperature the vesicles showed deformations in shape coupled with a loss of elasticity of their membranes. On the other hand, sEVs in cold conditions (4°C) had a reduced ability to release any internal material effectively.The researchers then studied the effects of pH (acid levels) on sEVs. The physiological pH of the bloodstream is 7.4. A pH less than 7 indicates acidic conditions and anything more than that is termed alkaline. The sEVs seemed to maintain their shape in acidic conditions (pH 4) but in alkaline conditions (pH 10) they were deformed. However, at a pH of 4 the sEVs were smaller in size suggesting their internal contents had been lost.Now, salt levels (known as osmotic pressure) at a concentration of 0.15 M are healthy. However, changes in osmotic pressure can have detrimental effects on cells. As conditions were gradually changed it was found that the spherical nature of sEVs decreased at high salt concentrations (1.8 M) but seemed to remain intact at low concentrations (0 M). After a while, vesicles in high osmotic conditions showed ruptured membranes.An understanding of these dynamics is imperative to formulating sEVs as pharmaceutical aids in different disease conditions. Abstract: Retinal inflammation accelerates photoreceptor cell death (PCD) caused by retinal degeneration. Minocycline, a semisynthetic broad-spectrum tetracycline antibiotic, has previously been reported to show PCD rescue effect in retinal degeneration. The purpose of this study was to assess the effect of minocycline on Cx3cr1 and Ccr2 expression in retinal degeneration. Mertk -/- Cx3cr1 GFP/+ Ccr2 RFP/+ mice, which enabled observation of Cx3cr1 - and Ccr2 - expression pattern in inherited retinal degeneration, were used to test the effect of minocycline. Minocycline was systemically administered to Mertk -/- Cx3cr1 GFP/+ Ccr2 RFP/+ mice. For observing the effect of minocycline on Cx3cr1 and Ccr2 expression, administration was started on 4-week-old mice and continued for 2 weeks. To assess the PCD rescue effect, minocycline was administered to 6-week-old mice for 2 weeks. The expression pattern of Cx3cr1-GFP and Ccr2-RFP were observed on retinal and retinal pigment epithelium (RPE) flat-mounts. The severity of retinal degeneration was assessed on retinal sections. Minocycline administration suppressed Ccr2 expression in Mertk -/- Cx3cr1 GFP/+ Ccr2 RFP/+ mice as observed in retinal and RPE flat-mounts. On the contrary, Cx3cr1 expression was not affected by minocycline administration. Retinal degeneration is ameliorated in minocycline administered Mertk -/- Cx3cr1 GFP/+ Ccr2 RFP/+ mice. There's a lot of talk in the data visualization, We thought it might be helpful to speak with someone who is leading voice on information visualization, and so we were thrilled to get a chance to talk with Manuel Lima, Who is Fellow of the Royal Society of Arts, named "one of the 50 most creative and influential minds" by Creativity magazine, Manuel Lima is the founder of VisualComplexity.com, Senior UX Manager at Google, and a regular teacher of data visualization at Parsons School of Design. He has over fifteen years of experience designing digital experiences and leading product teams at companies like Google, Codecademy, Microsoft, Nokia, R/GA, Kontrapunkt, and the Museum of the Moving Image. Over the past 20 years, my guest on today's show, , has been trying to recommend pillows for his patients that would put their head in the correct position so that they could get a great night's sleep and wake up rested. He could not find one, and he would have patients purchase soft down pillows and stuff them under their necks. He found that a conventional pillow would loose its shape and end up on the floor. The problem is that a pillow is for your head and you can not support your head to get a great night sleep. But one day he was watching the movie “Crouching Tiger Hidden Dragon” in 2000 and the two actors came out of the room after “cuddling” all night. The camera gave a quick glimpse of the room they came out of. It showed what the two actors slept on, and to his amazement it was just a thin pad for their body and a block of wood in lieu of a pillow for their head. In that lightbulb moment, Dr. Martone invented the exact sleeping tool we discuss on today's podcast. He's one of the smartest guys I've ever talked to when it comes to sleep, so prepare to optimize your rest and recovery biomechanics like never before! During our discussion, you'll discover: -How Crouching Tiger, Hidden Dragon inspired Dr. Martone's invention... One of the props was a piece of wood, not as a pillow, but for their neck Peter had shoulder issues, couldn't identify the cause While camping, he used a shoe under his neck for sleeping; woke up well rested Used a shoe, and other odd objects for sleeping; shoulder and other health issues were resolved -Surprising problems caused by sleeping on one's side... 90% of Peter's patients have some type of damage to their cervical curve in the neck We lose the curve doing day to day activities, and there's nothing to repair it at night 98% of patients who lose the curve in their neck develop an unnatural curve in their lower back This eventually leads to scoliosis in the lower back The portion of the brain: main function is to control the body's 80% of proprioception into the brain comes from the spinal cord The vermis atrophies due to loss of proprioception It affects the pre-frontal cortex (executive functioning) Decreasing the curve in the spine severely limits glymphatic drainage Why we toss and turn at night: Pain centers and nerve centers are very close together When you fall asleep on your side, you can only stay in that position for a short period of time Body should be in a "neutral sleeping position" Alcohol inhibits pain sensors. You stay in the same position longer, and wake up in pain OURA ring If you didn't get all that, get this: Loss of the cervical curve = degeneration of the spine. It also causes the vermis to atrophy, which negatively affects the pre-frontal cortex. Side sleeping causes pain and discomfort, which in turn affects the quality of our sleep. The advantages of sleeping on the back versus the side or stomach... Use a pillow for the neck rather than the head Restore the cervical curve and decrease lumbar tension -How to train yourself to sleep on your back... Just start on your back 1-2 hours per night will be enough to help restore the cervical curve in your neck : 5-10 degree angle, should go to the base of your spine The body will eventually be able to go through the night on your back Total 2-3 month transition Sleeping on your back, you distribute your body weight over the greatest amount of space -The little-known hack called "Remembering" that may help with sleep problems... Peter struggled to sleep due to numerous things he was thinking about Needed to be "in control" of something Visualized people sitting in a specific seat after giving a talk; he went right to sleep Focus on something, but don't think about it You can't think yourself to sleep; you have to remember yourself to sleep Dreams are how the mind categorizes the day's events The further back in time you can remember, the more efficacious this technique will be -Ways to control your core temperature while sleeping... Allostatic load: Hierarchy of needs in bodily functions (heart pumping vs. growing fingernails) Your body cares more about your core temperature than even the immune system We get sick because there's a finite amount of energy our body can distribute Maintaining the body's core temperature maintains the allostatic load in our entire body Keep your core as warm as possible; keep hands, feet and head exposed People like to sleep on their side so they can curl up, feel protected Use a for a similar effect "Head garage" Place a pillow over your head, just above the nose -More little-known sleep hacks... Keith grew up in a Chicago suburb and attended college at Drake University when he landed his fist internship in radio at KISS FM in Chicago. Since then, Keith finished school and moved to Cincinnati where he worked on a radio show that was heard in 15 different cities, daily. He got tired working for a large radio company and found an amazing job at Hubbard Broadcasting in Minneapolis, where he worked for 5 years. While in radio, Keith got to meet & hang out with big names like, 50 Cent, Nick Leshay, Britney Spears, Christina Aguilera, Rob Thomas, Jewel and his date to the company Christmas party one year was the cast of the Real World Las Vegas!. He fell in love, got a Bernese Mountain dog named Roxy, got married, and then Keith and his wife decided to open a doggy daycare in 2008… That was an instant success, and in 2011 they took another leap and opened Bubbly Paws which is a self-service dog washing/grooming salon which now has 3 (and growing) locations in the Minneapolis area. The focus of both businesses was to make sure they provided and amazing service, but also gave back to the pet community. Keith and his wife are actively involved in the day to day operations of each business, and they now have a 13 month old son! Not feeling completely fulfilled in his radio career, Keith decided to take a leap of faith into something new. From radio DJ to business owner, him and his wife have made it successfully in the entrepreneur world. “In radio, we would pre-record each episode. Basically all that did for me, was teach me how to be a great liar and BS my way through each day.” Like many others who have dreams to start their own business, Keith’s biggest fear was failure. But with a year full of strategic planning, they finally decided to quit their mundane jobs and open a doggy daycare! Keith and his wife are the epitome of entrepreneurs. They first THOUGHT and VISUALIZED their dream, took MASSIVE ACTION even before they were ready, and finally made a dream their reality. “Writing my first rent check for $10,000 especially during the recession and only having one client was scary, but you just need to do it!” Having no idea where the business would take them today, they both still took the leap of faith, knowing there had to be more to live then their previous situation. Becoming insane with their marketing and advertising ideas, the doggy daycare has been mentioned even through Fed Ex’s National campaign and QuickBooks! “We didn't just want to open a business, we wanted to give back and have fun with it!” As we all know, having a successful business means having A1 customer service. Keith’s doggy day care has blown competitors out of the water, with their love for the customer and their pets (of course). In this podcast, the Editor-in-Chief of JNIS, Robert Tarr, is joined by David Fiorella, the corresponding author of the study, "Final results of the US humanitarian device exemption study of the low-profile visualized intraluminal support (LVIS) device". Dr Fiorella, from the Department of Neurological Surgery, Stony Brook University Medical Center, New York, concludes that the LVIS device facilitates coil embolization of wide-necked intracranial aneurysms with high rates of technical success. The process also granted an excellent safety profile, and very high rates of complete and near-complete occlusion at follow-up. Jessica Hagy is a cartoonist and writer best known for her insightful and hilarious webcomic Indexed, in which she draws charts and Venn diagrams connecting completely different ideas and getting us to think about them in different ways. Her latest book is The Art of War Visualized. It's the Sun Tzu classic reworked and with illustrations.
