In this episode we feature the latest research by Richard Wong and colleagues at the Kanazawa University NanoLSI. The research described in this podcast was published in the Journal of Extracellular Vesicles, in November 2022 Kanazawa University NanoLSI website https://nanolsi.kanazawa-u.ac.jp/en/The offshoot of cells visualized in real timeIn a study recently published in the Journal of Extracellular Vesicles, researchers from Kanazawa University use high-speed microscopy to capture the dynamics of nanosized sacs released from cells.Small extracellular vesicles (sEVs) are tiny sacs released by cells to deliver chemical messengers to other cells. Since sEVs are compatible with biological tissue they are being investigated as carriers for nanodrugs. However, the impact of physiological stress—such as changes in temperature—on the structure of sEVs is obscure. A research team led by Richard Wong and Keesiang Lim at Kanazawa University has now used an advanced form of microscopy to elucidate these changes in real time.The temperature, acid, and salt levels in our bodies can fluctuate with factors such as disease. Thus, research on sEVs for drug development requires a deeper understanding of how stressful environments affect the vesicles' structure. For their study, the team first isolated sEVs from cells. Next, using a technique known as high-speed atomic force microscopy (HS-AFM) the structure of sEVs was revealed to be either spherical or ellipsoidal in shape. HS-AFM also enabled the researchers to accurately measure the sizes of sEVs without rupturing or damaging the vesicular membranes.The effect of varying temperatures on sEVs was the first parameter assessed. At temperatures higher than normal (37°C) body temperature the vesicles showed deformations in shape coupled with a loss of elasticity of their membranes. On the other hand, sEVs in cold conditions (4°C) had a reduced ability to release any internal material effectively.The researchers then studied the effects of pH (acid levels) on sEVs. The physiological pH of the bloodstream is 7.4. A pH less than 7 indicates acidic conditions and anything more than that is termed alkaline. The sEVs seemed to maintain their shape in acidic conditions (pH 4) but in alkaline conditions (pH 10) they were deformed. However, at a pH of 4 the sEVs were smaller in size suggesting their internal contents had been lost.Now, salt levels (known as osmotic pressure) at a concentration of 0.15 M are healthy. However, changes in osmotic pressure can have detrimental effects on cells. As conditions were gradually changed it was found that the spherical nature of sEVs decreased at high salt concentrations (1.8 M) but seemed to remain intact at low concentrations (0 M). After a while, vesicles in high osmotic conditions showed ruptured membranes.An understanding of these dynamics is imperative to formulating sEVs as pharmaceutical aids in different disease conditions. Easy.I've been thinking these last few weeks, about penning some thoughts on rap lyrics; specifically, around content, when working in a workshop-type-educational setting with young people; and the type of conflict I often come up against when doing this. So here it is. A bit of-preamble before, thoughI've been facilitating workshops now for a number of years; mainly around poetry, rap, beatbox and theatre. Sometimes all together. I work on the premise that it's about people expressing themselves and being able to tell own stories, in their own way. Most of which, I'd come to learn from working with my good pal Conrad Murray (you can find out in more detail about the processes we use in THIS here BOOK making Hip Hop theatre) Regardless of the medium, that's what I'm trying to do; in a nutshell.In the world of youth-arts, you'll often hear the much talked about category of ‘hard-to-reach-young-people.' Ie- kids that don't engage with the arts so much, if at all. A lot of the time, these are the young people I'm working with. Sometimes I'm able to engage with them, using a rap context as lots of them enjoy rap music and I've been fortunate enough to work on some really meaningful projects.  Rap music has been popular my entire life but gradually over the last twenty years, for a lot of British kids it's become something that isn't just a popular form of entertainment in the UK, it's a thing you can actually do, in the UK.I can remember being young and loving hip-hop but thinking that it's something that Americans do. UK Hip Hop was a small niche but UK Garage and Jungle MC's really helped to break down the idea that a kid from these shores could spit too and in their own accent.These days most young people I encounter are more likely to listen to British rappers than American rappers, often rappers from their own areas even; so at the very least, it can be a starting point just to get them talking, when I'm initially trying to engage them. Rap is a broad church, I'm 38, I like what I like and though I try and keep an ear out for what's currently popping, most of the time I don't know; so I get them to tell me about who they're listening too. The whole process is anchored on earning their trust and for what are probably good reasons, they don't tend to give that out easily; hence the ‘hard-to-reach' tag. It takes timeIn this sort of work, there's normally some kind of loose objective involved (which is often due to funding requiring an outcome and isn't always helpful) – produce some music, work towards a performance, that kind of thing. Sometimes, it's enough just to engage them in the street, in conversation for ten minutes, let alone getting them into a building and into some kind of workshop environment, which is what the well-meaning but often naive funders want.Building up the type of trust required to even get them into a building and into a some of kind of structure can take a lot of time; weeks, months, years even.  So after all of that, if you're able to get even one young person into a room to start writing lyrics, you've done well but you're now into the tricky-waters of content.I'm no psychologist but particularly with boys, they're going to naturally want to emulate what their peers, or perceived elders are doing. In the last few years, the most popular form of rap, has been Drill. Which often talks about violence and street-crime. This is their main reference point, it's what they know (in terms of the music), so it's natural this is the sort of stuff they're going to try and create; these rappers, are who they look up to.A lot of it, to be fair, is just bravado and metaphor, which as far as I'm aware, has been around for time immemorial. Whether a rapper is rapping about how violent they are, or how many drugs they've sold, to how many books they've read and how much wisdom they have, it's still just bravado. And it's part of rap. This is hip-hop after all. (Where the line gets very blurred and serious, is when actual incidences are being referenced but that's a whole another discussion.)The problem for me comes when they start writing this stuff and it includes violence, crime and sexist lyrics etc. I operate on the basis that it's a win just getting them into the room and giving it go. For a lot of them, it might be their first time, so the content, to me, doesn't matter so much in those early stages. Also, they're often doing it around their peers, so there's all those group-dynamics to take into account. But as long as they're doing something, I think that's a good thing, regardless of what they've said, they're using their voices and learning the craft of constructing lyrics and rhymes.The trick is to build their trust over time and work with them, to broaden their horizons a bit and challenge them, but like I said, this takes a long time and often these projects will only run over a short period.If I started wagging a moral finger at them from the off they'd be gone, never to be seen again; which defeats the whole object of dong this work. Lots of these projects, organisations and funders are well-meaning but naive in their aims and objectives, and increasingly idealistic. Talking to these young people about distant academic concepts, like toxic-masculinity, or heteronormativity, is rarely going to end well, yet sometimes it feels like that's what some of these leaders in charge want me to do. They can walk in a room, whilst I'm running a session and look at or hear some of the lyrics that are being created and tick me off about it, it's happened before; which makes me think if that's all they care about? As opposed to the process of slowly developing a relationship with these young people over time and encouraging them to explore their thoughts and expand their thinking, whilst taking creative risks in a safe environment; without castigating them, which more often than not, is what they expect from adults with any kind of authority. I'd be interested to hear alternative thoughts on this, as I'm sure there's people out there who disagree with me. I can only do what I know and what I think works and where I've been allowed the space to do this, I've seen positive results.There are times when I've had to draw a line. With most young people who are used to engaging in these sorts of projects, setting boundaries is one of the first things you do but with these so called hard-to-reach kids, who already feel like they've been written off; it's never that simple. A little patience and less judgement, please.So there you go, some thoughts. I mentioned last week that I started writing this article and trashed it, because I thought it was no good. But then I reminded myself about why I started this blog, to put my work out but also to learn and try and take a few risks. I want to get better at writing this sort of stuff but it doesn't come easily to me, it's a craft that I‘m learning and hopefully improving.It'll most likely be back to the regularly scheduled program next week, ie me just putting up bits of writing that I'm working on from this show, or maybe I'll start that Peep Show article I was going on about.Either way, large up for reading / listening. Not all genetic variations are associated with threats or harms to human health. Some even  protect us, such as genetic variations that have been shown to make bones harder or the heart more impervious to disease. But while some genetic variations are positive, others can cause or contribute to disease. In this episode, we answer the question of how does gene therapy work, and learn how gene therapy replaces and repairs certain gene variants, and is changing the trajectory of genetic diseases.For several years, Dr. Jean Bennett at the University of Pennsylvania's Department of  Ophthalmology Center for Advanced Retinal and Ocular Therapeutics investigated the possibility of replacing gene variants in the retina – which cause blindness – with copies of healthy ones.After successfully treating blind puppies, Dr. Bennett and her colleagues turned their attention to treating human eyesight.The retina turned out to be a good target site for gene therapy. In most parts of the human  body, our cells keep dividing after we're born. But not the rods and cones in our retina – these photoreceptor cells don't regenerate.A major obstacle in the development of gene therapy is the tendency of cloned genes to get lost in the process of cell division before they have a chance to integrate into the host DNA. Because retinal cells don't divide after birth, the cloned gene may be expressed for a prolonged timeIn 2017, after decades of painstaking research, building on the efforts of countless scientists  throughout history, Dr. Bennett and her colleagues had the evidence that gene therapy may be used to treat genetic conditions in humans.Since then, gene therapy has undergone giant leaps in the treatment of specific diseases. And record numbers of gene therapy trials are ongoing, including potential treatments for conditions such as sickle cell disease and Parkinson's disease. It's just one of many ways that the field of gene therapy is poised to change the world in the years to come.For more education on gene therapy, visit www.genetherapynetwork.com.

It's having a plan that enables you to do the things that matter most. In the inaugural episode of this podcast, I went over where happiness is ultimately derived. In this episode, I dive deeper into two of those areas: meaning and purpose.  You may be baffled by the idea that retirement can feel a bit depressing for some people. For early retirees, it can feel more so. We forget that we get a lot of meaning and purpose from our careers. Finding that same level of meaning and purpose in retirement doesn't happen accidentally. It requires significant planning. When you look up the word retirement, you'll see images of people relaxing on a tropical beach or traveling the world. These images convey feelings of liberation, relaxation, happiness, and joy. But reality can be very different from what you see online. However, your retirement can be even more meaningful with the right type of planning. Finding deep meaning As I shared in the first episode, much of what makes us happy has nothing to do with money. True meaning doesn't have to be something grand like solving world hunger. It can be as simple as spending more meaningful time with your family or volunteering for your community. Ideally, we can find an activity that provides meaning, purpose, and true fulfillment.  Another way retirees plan for success is by staying relevant and connected. As an early retiree, many of your friends and family will still work. Losing connections at the workplace can lead to increased feelings of loneliness. Because of that, it's crucial to become involved with organizations before you retire so you can look forward to spending time with people you already know. The exponent of timeIn the episodes leading up to the 2022 personal tax filing deadline, April 18th, I'm going to be going over strategies to not only prepare you for early retirement but to do so in ways that may reduce the taxes you pay in the process. While our focus is on early retirement planning, you may also want to consider helping your children get started on their early retirement planning as well. We always want to pay taxes when it's to our advantage. Obviously, that would mean paying when income tax rates are the lowest. As a child, that rate could be as low as zero. Therefore, contributing to a Roth IRA as a child can mean never having to pay taxes on the amount contributed during this period. As an investor, the single most powerful thing you can do is increase your time horizon. So there was a lot of optimism in the air. Roshan decided to go all in. He believed they could build the app successfully just as they had done with their previous offerings, the digital assets. It was not as easy as it seemsRoshan and his team grossly underestimated the time, effort, resources, money, patience, and skills required to build a consumer app. Roshan soon found out that App Store discovery is quite hard, and an app has to either go viral or spend a ton of money on acquisition. And once you’ve acquired these consumers, you still have to retain them and then make money out of them, which is not a trivial thing. Having to wind upRoshan had to wind up after a couple of years of trying to make the app a success. This was quite humiliating for him as he had to let go of people he had hired and nurtured. The failure of the app drained all of Roshan’s self-confidence. He hit a real low point after this venture. You have 24 hours in a day. And on average, you’ll probably sleep anywhere from 7-9 hours, 10 probably on weekends. 60 minutes in an hour, so if you remove the amount of time you spend sleeping, on avg im gonna say that probably is going to equate to about 1,000 minutes left (easy to remember) to make something happen every day. You have 1,000 minutes, and you get to choose how you spend those minutes each day.Audit yourself every daySome things that I find value in are making appointments on my calendar for work that I need to get done. If I have a contract to write or a meeting I need to prep for i put that on the calendar so it gets done and I dont get double booked. Fill your calendar with things you need to do and make people work around your schedule. If you value your time so will others. By doing this if you get something done early you can either start the next task ahead of time or take a littl3 break, scroll through IG and mot feel bad about it.Multi-task when you canI do these strategically, but during the day I can get away with multi-tasking and still be effective. IE I am constantly learning… When I run in the morning, instead of just listening to music, which occasionally I do because it helps me relax mentally or motivate me temporarily, but most of the time I am listening to Audiobooks, learning new concepts, new perspectives, and continuously challenging my knowledge. Meal Prep your lunchesSaves you moneyAdds discipline to your diet, creating minimal room for cheating or becoming lazy with your diet. Don’t waste time driving to grab food or choose a poor meal optionMeal prep on sundays for an hour saves you timeIn addition to what cooper said other things thay you can do are clean while you cook, pick things up as you go.If you are in a meeting where you don't have to be 100% involved answer emails.I've told contractors thay im very busy and don't have time to attend all of their meetings. We comprised and when they need me they address my name to get my attention, I answer the question or provide feedback and them go back to doing whatever else I needed to do.You can wake up earlierI don’t wanna hear it, matter of fact, if you are OK with living a mediocre life, then go ahead and enjoy your beauty sleep. That’s OK with me, and that’s OK with all the rest of us working our ASS OFF waking up at 530-6am everyday working towards someday making our dreams our reality because look… I don’t know a single successful person that started with nothing and created something that didn’t wake up early PISSED OFF or FIRED UP to make/create something epic. Not a single one. They sacrifice sleep, maybe not every single day, but they go through the week with 5-7 hours of sleep and are accomplishing WAY MORE than someone that sleeps 8-9 hours a day. I can promise you that. When I need to get something done the first thing that goes is sleep. Your goals are worth your time. I don't want to miss out on life to sleep. It is important but making memories and hitting goals is more important to me.If you take anything from this, remember this:YOU are the CEO of your own bank of time and YOU get to decide where that time gets spent. Time is a commodity that you don’t get back. It is not like money you can’t make more of it. Make each minute count,We’re out. MAKE TODAY COUNT.

Becoming a mother is an evolutionary process. It is one of the most beautiful and rewarding experiences of womanhood. This transition also comes with the challenges of hormonal changes, figuring out how to manage your new life, accepting your new body, and learning a new human. We aren’t given an instruction manual after delivery, but we can rely on the resources that are provided to us in this day in age and ask for help when needed. Our guest mom in this episode is Jabina Coleman. She is a mother of two, a board-certified lactation consultant, and therapist. In her line of work, she provides counseling and lactation support to mothers in hopes to help ease their transition into motherhood. Jabina shares how her motherhood journey created her passion for maternal health and wellness support. She has a wealth of knowledge that ranges from psychotherapy to resources for moms who are in food deserts. In this episode, Jabina mentions Kimberly Seals Allers, a breastfeeding advocate and author of a research report on the access to breastfeeding. Jabina also shared other resources such as the Maternal Wellness Village, located in Philadelphia, PA (also offers virtual support) and The Perinatal Mental Health Alliance for People of Color, www.PMHAPOC.org. You can find a silicon breast pump online (the one mentioned in this episode is called a Haakaa) at stores like buybuy Baby, Target, or Amazon). In this episode, Kelly Brown joins the podcast to talk about God’s enduring goodness- regardless of what we can see, what we feel, or the depths of our suffering. We all have something sacred that Satan will touch in our lives, but during the storms we have to look for the faithful hand of God and hold tightly to truth. Satan can give us his best shot, but God’s goodness still reigns. This episode helps us evaluate and redirect our hearts back to truth as Kelly asks the question: “Where do you go when life hurts?” About the Speaker: Kelly Brown has been a committed disciple for over 45 years. She and her husband, Tom, were baptized while attending the University of Florida where Kelly received her BS in Mathematics. She married Tom in 1975 and then moved to Boulder, CO to begin a campus ministry at the University of Colorado that grew from 1 to over 250 students while the Browns served in Boulder. They have continued to help build dynamic campus ministries and churches in San Francisco, Boston, Los Angeles and now in Atlanta. One of Kelly’s greatest convictions is that the young women in our churches are the future and that mature sisters in Christ must continue to teach the younger so they can benefit from all that we have learned through the years. She is known for her youthful spirit and creativity and has two children and two grandchildren. Kelly and Tom currently serve the North River Church of Christ in Atlanta. There are times our view of God changes when we suffer and don’t see God’s goodness in the pain.There are blessings in disguise in the midst of these storms. But we have to look for them. Mel B'S former hubby trashed the Spice Girl's “Brutally Honest” memoir as full of lies and omissions — including no mention of the young nanny who she conned into getting an intimate tattoo that read 'Property of Melanie and Stephen!' Speaking exclusively to "All Rise," Stephen Belafonte — the singer's 43-year-old ex-husband — pegged manipulative Mel as the driving force behind their kinky sex games during their 10-year marriage that has exploded into one of Hollywood's nastiest divorces ever.The pervy pop star penned a memoir blasting Stephen as a wife-beater.But instead, Stephen, who requested the no-holds barred interview with Dylan Howard, claimed:- He never had a physical altercation with Mel B- Mel B was once hospitalized after a booze and drugs bender and suffered organ failure- After being hospitalized again, claiming he'd thrown her head into a door and beat her — the truth was she was suffering from kidney failure because of a drinking problem- They were forced to get a divorce because Mel B's addictions spiraled out of control- She fleeced their bank accounts prior to the split- She orchestrated a "sick plot" to have their house searched by law enforcement- She cheated on him with a British actor- When fellow Spice Girl, Geri Halliwell, suggested they do a charity show to announce the band was reforming, Mel responded: "F*ck charity, charity doesn't pay the bills."- Mel B's daughter Phoenix lied when she said he'd beat her mother- The pop star seduced Lorraine Gilles — the couple's live-in nanny of eight years — in 2009 when she was just 18- Mel B told the blond teen she would make her a “superstar” — but instead used her as a sexual plaything- He never got Lorraine pregnant- Mel B had an affair with a Beverly Hills Cop whose wife was pregnant at the timeIn one of the most candid tell-all interviews ever, Stephen alleged his ex-later bought Lorraine's silence, adding: “She knew about Mel's drug use, how bad she was at parenting ... so Melanie paid to keep her quiet.” Contacted for comment, Lorraine, now 27, said she had no comment on the blockbuster charges. The movie producer also fired off on his bitter divorce, scoffing at Mel's story that he used their bedroom sex tapes as leverage to settle their split quickly.Instead, Stephen told Howard he is in possessions of evidence to prove Mel B — whose slogan is “Girl Power” — candidly acknowledging he never touched her.“Where is that girl power to come in and break up a family?”  “She's a bully,” he added. “Melanie is not a person who gets beat.” Guests:Stephen Bellafonte

Happy September, lightworker!I know fall is coming up because my ever-eager, inner Virgo has already lured me to Target a few times for ritualistic hunts for fresh journals, highlighters and pens.(Speaking of rituals, check out this post for tips on creating your own fall ritual to get you in the fall mood.)This month is when dreams and plans become form, when iced coffee and flip flops are replaced by pumpkin spice lattes and cozy blankets. It's time to get down to business.I'm ready.Harvest the Universe's Supply of Love, Money & TimeIn keeping up with the theme of fall, this month's first Intuitive Tuesday is dedicated to helping you receive the harvest of love, time and money from the Universe that you could be unconsciously blocking.Not an audio learner? That's OK. Read the article transcription below.Today, I want to talk about receiving, and give you two simple practices that will help you easily start the flow of receiving from the Universe.I was inspired by a recent call with one of my clients where she opened up about being an over-giver. She said she has always been a giver, in her friendships, as well as in romantic relationships.Giving Is Depleting If You're Not ReceivingGiving is amazing, and benefits both the giver and the receiver. But it can quickly become draining and depleting if you don’t allow yourself to receive.Think of the cycle of giving and receiving as a natural cycle of the Universe. In order to be in balance, you have to do both.We Receive in Many Ways - Energy, Time, Money, & LoveThe thing is, so many of us are caught in this “give, give, give” cycle and feel like a deer in headlights when it’s time for us to receive. I can definitely relate to this, and have a feeling many of you can, too. Receiving is crucial, and if you’re blocking yourself from receiving, you’re blocking yourself from abundance. Think about it – where do you feel a lack right now- is it money, love, time? Consider the ways you might be blocking yourself from receiving in these areas.Deflection is a Form of Self SabotageHere is an example of how you might deflect receiving love. Say that you’ve been working out, toning up, and have seen noticeable improvement in your energy and body. You’ve been giving a lot of yourself, and feel proud of your accomplishments.Now, imagine that your signficiant other tells you that you look amazing, better than you’ve ever looked before. That you’re beautiful and shining, inside and out!You can respond 2 ways – you can either receive the compliment, say thank you and genuinely feel and receive the love of that statement, or you can deflect, be embarrassed, and either change the topic or mumble a “thank you, I’ve JUST been working out more” (notice how the word “just” downplays the tremendous effort you’ve been giving).The first way allows you to receive the love from the Universe through your partner, and the second way deflects it.2 Practices to Start Receiving More TodaySo, the first practice I want to share with you is to practice saying “thank you” and pausing after to genuinely feel and soften into your “thank you”. It’s a way to start receiving in a small way, that will open doors to higher levels of receiving.The second practice I’d like to share with you is a mantra with my favorite receiving crystal – rose quartz!
