The fire that led to 39 deaths and nine injuries at a complex building in Jialeyuan community in Xinyu, Jiangxi province, on Jan 24 was caused by illegal construction of a cold storage facility in its basement, according to an investigation report released on Saturday.根据9月21日公布的一份调查报告,江西省新余市佳乐苑小区的一栋综合楼于1月24日发生火灾,造成39人死亡、9人受伤,起火原因是该楼地下一层违规建设冷库。Polyurethane foam sealant was used during construction at the first floor underground, releasing flammable gases that accumulated to reach combustible conditions. While laying plastic film on extruded polystyrene boards, static electricity was generated, igniting the gases, the foam and other flammable materials. On the second floor, students and teachers were having classes organized by a training institute. The process of rebuilding communities that have taken a hit during the weather events of this summer is going to be a long and arduous process. Tough decisions will have to be made. Personal decisions will have to be made. Do you want to go back and live in the place where your worst nightmare occurred? I'm thinking of the families whose homes came down in landslides, whose homes were flooded and had to flee in the middle of the night. And there are many Auckland families, and indeed Hawkes Bay families, who have been flooded not once, not twice, not three times, but more than that. I'm thinking of the lovely lady who rang us who had been washed away in the floods, her home had been destroyed that they'd worked so hard on. Her mum was living with them - she had to go into a rest home because while they were in the process of rebuilding because it was cold and it was damp there was Polyurethane covering some of the walls so she couldn't live there. So all of a sudden life changed. Well, I just wonder what happened to her in these latest floods, if she's been hit again, will she want to go back there? A group of West Auckland homeowners, and this will not be the first group, has set up agroup asking authorities to step in and buy their flood affected properties so they can leave. Basically, execute the managed retreat that's being talked about, a process where people are relocated and their properties are turned into parks or reserves. The campaign is backed by Te Atatu MP Phil Twyford who's been working with families in about eight neighbourhoods since the floods of 2021. A report found the flooding was partly due to blockages in the Waimoko stream, partly due to geography and the overpowering force of the one-in-100-year flood, which then came along in 2022. These one in 100 year events seem to be occurring far more often! Should the same thing happen in Esk Valley? Should clifftop homes be part of a managed retreat, something like the King's chain, whereby the shoreline belongs to the public and you have access to the beach, the cliff tops are the same. You get access to the spectacular views and walk along there and houses are moved back from cliff edges. When we talk about buying back houses? That's the taxpayer. That's us trying to pay our mortgages as well as other people's homes. It might be the way to do it. I mean we are going to have to take a long hard look, not just stick a Band-Aid on communities, spend billions of dollars and then do it all again in another couple of years. When it comes to the managed retreats and the rebuilding of communities, do we rebuild them in the same places? Do we restore homes and put people back in there after they've been flooded once, twice, three times? That doesn't seem like sane advice, when we're being told that the weather's only going to get worse. We're spending billions anyway, about every couple of years on natural disasters, or do people just have to cut their losses and clear out? And it's on the individual and not the community. Stephen Mayfield, a professor of Biology at UC San Diego and director of the California Center for Algae Biotechnology, invented Soleic, an algae-based rubbery foam material that can be used in footwear, surfboards and other products in the place of petroleum-based polyurethane foam. He launched Algenesis, a biotechnology-based materials science company to commercialize the Soleic.But shoe companies did not come running to use Soleic, which biodegrades completely in sea water and compost piles. Along with Algenisis president Tom Cooke, a footwear and apparel industry veteran who had worked for Reef and Vans, Steve launched Blueview Footwear, maker of the world's first compostable shoe. Steve and Tom join me today to talk about the evolution of Algenesis and Blueview, as well as the many materials Soleic could replace across a variety of product categories. C is for Condom Welcome to the A to Z of Sex. I'm Dr Lori Beth and I am your host. We are working our way through the erotic alphabet one letter at a time. Just a reminder this podcast deals with adult content, so if you don't have total privacy, you might want to put on your headphones. Today the letter is C and C is for Condom. This week – the history of the condom. There are two cave paintings in France from 11,000 BC that scientists believe illustrate the first evidence of condoms. Having looked at the illustrations it is unclear to me as to why they believe that. Around 1,000 BC, the Egyptians used a linen sheath around the penis to prevent insect bites and tropical diseases. The Chinese used oiled silk paper and the Japanese used leather and tortoiseshell sheaths. The Romans developed condoms made of goat's bladders. None of this is terribly appealing… In the 1400's in China and Japan, condoms first began to be used for birth control and to protect against infection. Safe sex was understood even back then. It is amazing that we still have trouble engaging people in safe sex given how long we have known how to engage in it! The Chinese favoured silk and lamb's intestines. The Japanese favoured horn and tortoiseshell which seems tremendously uncomfortable to me! In the 1500's Gabrielle Fallopius invented a chemical soaked linen sheath to prevent the spread of syphilis. It was tied with a ribbon around the shaft of the penis. He conducted the first research that proved that condoms prevent the spread of sexually transmitted infections. The study had 1100 participants and used the linen condoms he invented. None of the people who used these contracted syphilis. By the time we reached the 1700's, the use of condoms for the prevention of pregnancy and sexually transmitted infections is more widely known but now there is controversy as to whether or not they work. Some people believe that the availability of condoms encourages people to have more sex with more partners and so encourages immorality. During this time period, condoms became available in pubs and markets and were either made of intestines or bladder and soaked in lye or sulphur or made of linen and soaked in chemicals. In the 1800's members of parliament in the UK tried to get condoms banned because they didn't do enough to prevent syphilis and encouraged lots of sex. The first rubber condom arrived in 1855. Rubber condoms were reusable so saved money in the long run but they were individually fitted. Despite this, they often fell off because they only covered the glans of the penis. Skin condoms were still cheaper. In the 1900's, lots of development occurred primarily because the military began to issue soldiers with condoms to prevent STI's. In the late 1920's latex was invented, and the first latex condoms appeared. But early latex condoms were not very effective at all – many leaked. As a result, the FDA in the US decided that a condom was a drug so that every condom had to be tested. In 1957, Durex created the first condom with lubrication. When condoms were found to prevent the spread of HIV, sales increased dramatically. Condoms remain the mainstay of safe sex. Modern condoms are made of a variety of materials including natural latex, polyurethane, A-10 resin, and lambs intestines. Latex are the most flexible however some people have allergies and they cannot be used with oil based lubricants. Polyurethane has the disadvantage of being less elastic than latex and are more likely to slip or break than latex, but they can be used with oil based lubricants. Polyisoprene is a synthetic version of natural rubber latex. It has the advantage of being softer and more elastic like latex but without the protein that causes latex allergies. It transmits body heat well. But it cannot be used with oil based lubricants. Lambskin are said to feel more natural, but they...

The aim of the study was to compare the behavior of seeded cells on synthetic and natural aortic valve scaffolds during a low-flow conditioning period. Polyurethane (group A) and aortic homograft valves (group B) were consecutively seeded with human fibroblasts (FB), and endothelial cells (EC) using a rotating seeding device. Each seeding procedure was followed by an exposure to low pulsatile flow in a dynamic bioreactor for 5 days. For further analysis, samples were taken before and after conditioning. Scanning electron microscopy showed confluent cell layers in both groups. Immunohistochemical analysis showed the presence of EC and FB before and after conditioning as well as the establishment of an extracellular matrix (ECM) during conditioning. A higher expression of ECM was observed on the scaffolds' inner surface. Real-time polymerase chain reaction showed higher inflammatory response during the conditioning of homografts. Endothelialization caused a decrease in inflammatory gene expression. The efficient colonization, the establishment of an ECM, and the comparable inflammatory cell reaction to the scaffolds in both groups proved the biocompatibility of the synthetic scaffold. The newly developed bioreactor permits conditioning and cell adaption to shear stress. Therefore, polyurethane valve scaffolds may offer a new option for aortic valve replacement.
Background: Tissue engineering represents a promising new method for treating heart valve diseases. The aim of this study was evaluate the importance of conditioning procedures of tissue engineered polyurethane heart valve prostheses by the comparison of static and dynamic cultivation methods. Methods: Human vascular endothelial cells (ECs) and fibroblasts (FBs) were obtained from saphenous vein segments. Polyurethane scaffolds (n = 10) were primarily seeded with FBs and subsequently with ECs, followed by different cultivation methods of cell layers (A: static, B: dynamic). Group A was statically cultivated for 6 days. Group B was exposed to low flow conditions (t(1)=3 days at 750 ml/min, t(2)=2 days at 1100 ml/min) in a newly developed conditioning bioreactor. Samples were taken after static and dynamic cultivation and were analyzed by scanning electron microscopy (SEM), immunohistochemistry (IHC), and real time polymerase chain reaction (RT-PCR). Results: SEM results showed a high density of adherent cells on the surface valves from both groups. However, better cell distribution and cell behavior was detected in Group B. IHC staining against CD31 and TE-7 revealed a positive reaction in both groups. Higher expression of extracellular matrix (ICAM, Collagen IV) was observed in Group B. RT-PCR demonstrated a higher expression of inflammatory Cytokines in Group B. Conclusion: While conventional cultivation method can be used for the development of tissue engineered heart valves. Better results can be obtained by performing a conditioning step that may improve the tolerance of cells to shear stress. The novel pulsatile bioreactor offers an adequate tool for in vitro improvement of mechanical properties of tissue engineered cardiovascular prostheses.

Objective: Hepatocytes are increasingly used as functional units in bioartificial liver devices. The objective of the present study was to investigate the feasibility of culturing porcine hepatocytes in high density on a novel polyurethane-based nonwoven three-dimensional matrix. We investigated (1) the optimal cell density within this culture configuration, (2) the maintenance of liver-specific morphology and cell functions over long-term periods and (3) the necessity to apply an additional extracellular matrix component (collagen gel). Methods: Nonwoven polyurethane matrices were manufactured by a specially developed fiber extrusion technology. Pig hepatocytes were cultured at various cell densities of 0.1, 0.25, 0.5, 0.75, 1 and 2 x 10(6) cells/cm(2) on three-dimensional networks of nonwoven polyurethane matrices and cell adhesion as well as functional parameters (DNA of nonattached/attached cells, lactate dehydrogenase release and cytochrome P450 activity) were determined. To assess the performance of cells within this configuration albumin and urea excretion was measured over 8 days. The potentially beneficial effect of an additional extracellular matrix configuration was evaluated by comparing the average albumin synthesis in groups of identical cell numbers. Results: The optimal cell density in this three-dimensional culture configuration was 1 x 10(6) cells/cm(2). The functional capacity of hepatocytes was stable for 8 days at an average level of 53.7 +/- 5.6 ng/h/mug DNA and of 1.8 +/- 0.14 mug/h/mug DNA for albumin and urea excretion, respectively. The supplementation of an extracellular matrix configuration did not improve functional activity of cells. Average albumin synthesis was 35.6 ng/h/mug DNA (28.7, 42.8) and 32.7 ng/h/mug DNA (23.4, 49.2) for collagen-immobilized and control cultures, respectively, Conclusion: The results of the study indicate that nonwoven polyurethane sheets supply a biocompatible support structure for functionally active high density cultures. Thus, nonwoven polyurethane matrices should be further investigated on with respect to their role in the development, optimization and design of bioartificial liver systems.
