CMS121 Mitigates Aging-related Obesity and Metabolic Dysfunction

BUFFALO, NY- April 9, 2024 – A new #researchpaper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 6, entitled, “CMS121: a novel approach to mitigate aging-related obesity and metabolic dysfunction.” Modulated by differences in genetic and environmental factors, laboratory mice often show progressive weight gain, eventually leading to obesity and metabolic dyshomeostasis. The geroneuroprotector CMS121 has a positive effect on energy metabolism in a mouse model of type 2 diabetes. In this new study, researchers Alcir L. Dafre, Saadia Zahid, Jessica Jorge Probst, Antonio Currais, Jingting Yu, David Schubert, and Pamela Maher from Salk Institute for Biological Studies, National University of Sciences and Technology (NUST) and Federal University of Santa Catarina investigated the potential of CMS121 to counteract the metabolic changes observed during the ageing process of wild type mice. “This comprehensive analysis aimed to further understand how CMS121 influences the metabolic landscape, paving the way for potential therapeutic applications beyond its established geroneuroprotective benefits.” Control or CMS121-containing diets were supplied ad libitum for 6 months, and mice were sacrificed at the age of 7 months. Blood, adipose tissue, and liver were analyzed for glucose, lipids, and protein markers of energy metabolism. The CMS121 diet induced a 40% decrease in body weight gain and improved both glucose and lipid indexes. Lower levels of hepatic caspase 1, caspase 3, and NOX4 were observed with CMS121 indicating a lower liver inflammatory status. Adipose tissue from CMS121-treated mice showed increased levels of the transcription factors Nrf1 and TFAM, as well as markers of mitochondrial electron transport complexes, levels of GLUT4 and a higher resting metabolic rate. Metabolomic analysis revealed elevated plasma concentrations of short chain acylcarnitines and butyrate metabolites in mice treated with CMS121. “The diminished de novo lipogenesis, which is associated with increased acetyl-CoA, acylcarnitine, and butyrate metabolite levels, could contribute to safeguarding not only the peripheral system but also the aging brain. This is a new revelation for me.  Yes, we can help our own wellness with good habits, tending to our mental health, and rest, among others.  But when you are drowning in burnout, it is not the time nor the place to be taught how to help yourself.  We need the help of someone else at that point in time to jump in and give us a hand and pull us out so we can rest.  That is what I haven't seen in medicine.  We are missing the people that are supposed to be looking after the medical workers. M., Radhakrishnan, S. K. Abstract: Cells exposed to proteotoxic stress invoke adaptive responses aimed at restoring proteostasis. Our previous studies have established a firm role for the transcription factor Nuclear factor erythroid derived 2-related factor 1 (Nrf1, also called NFE2L1) in responding to proteotoxic stress elicited by inhibition of cellular proteasome. Following proteasome inhibition, Nrf1 mediates new proteasome synthesis, thus enabling the cells to mitigate the proteotoxic stress. Here we report that under similar circumstances, multiple components of the autophagy lysosomal pathway (ALP) are transcriptionally upregulated in an Nrf1-dependent fashion, thus providing the cells with an additional route to cope with proteasome insufficiency. In response to proteasome inhibitors, Nrf1-deficient cells displayed profound defects in invoking autophagy and clearance of aggresomes. This phenomenon was also recapitulated in NGLY1 knockout cells (a model for NGLY1 disease) where Nrf1 is known to be non-functional. Overall, our results significantly expand the role of Nrf1 in shaping the cellular response to proteotoxic stress. Copy rights belong to original authors. When Scott Widenmaier left high school, he wasn't sure what career path he wanted to pursue. He grew up in Alameda, Saskatchewan, and soon found work on oil rigs. But by the time he was in his early twenties, he knew it was time for a change. "I realized that winters are just too cold to continue doing that," said Widenmaier. "I wasn't sure what I was going to do with my life, but I was interested in biology and human physiology." In his third year as a science major at the University of Regina, he became fascinated by a lab experiment examining how neurotransmitters control heart rates. He then moved west, to graduate studies at the University of British Columbia, studying endocrinology and its role in diabetes. "I really like the elegance of the feedback circuits," said Widenmaier, who went on to land a post-doctoral position with Dr. Gökhan Hotamisligil at Harvard University's School of Public Health. As he discovered the emerging field of immunometabolism, Widenmaier began to see links between the way the gut and the human immune system talk to each other, especially around obesity. "That communication to the immune systems and the metabolic systems is dysfunctional and contributes to a lot of the diseases that we see linked to obesity," said Widenmaier, who studies the way the body manages cholesterol. That's the focus of his work at the University of Saskatchewan as an assistant professor in the College of Medicine's department of Anatomy, Physiology and Pharmacology. "There's this transcription factor that sits in a part of the cell and it recognizes when cholesterol's too high," he said. "It changes where it goes in the cell and it regulates a response by the cell to try to prevent the cholesterol from causing damage and stress." Widenmaier has now landed a number of awards, including the Heart and Stroke Foundation's National New Investigator Award in 2020/21. Widenmaier was also named that year's McDonald Scholar. "We've made a lot of progress with understanding the role of NRF1 and NRF2 in the liver," he said. "We've identified some molecules that may be important for protecting cells from too much cholesterol." In this episode, hear how this work has implications for patients with heart and stroke disease, obesity, liver disease, degenerative brain conditions, as well as various forms of cancer.  "The issue with cholesterol is that we absolutely need it, but we also need it to be exactly the right amount," said Widenmaier. "There's lots of times where that that capacity gets stretched, especially under conditions of obesity." "What are those natural adaptive systems? If we can find out what they are, can we make them work better?"

Abstract: The membrane-bound transcription factor Nrf1 (i.e., encoded by Nfe2l1) is activated by sensing glucose deprivation, cholesterol excess, proteasomal inhibition and oxidative stress, and then mediates distinct signaling responses in order to maintain cellular homeostasis. Here, we found that Nrf1 stability and transactivity are enhanced by USP19, a tail-anchored ubiquitin-specific protease in the endoplasmic reticulum (ER). Further experiments revealed that USP19 directly interacts with Nrf1 in proximity to the ER and acts as a deubiquitinating enzyme to remove ubiquitin moieties from this protein and hence circumvent potential proteasomal degradation. Such USP19-mediated effect takes place only after Nrf1 is retrotranslocated by p97 out of ER membranes. Conversely, knockout of USP19 causes significant decreases in Nrf1 abundance and its active isoform entering the nucleus, resulting in down-regulation of its target proteasomal subunits. This led to a modest reduction of USP19-/-derived tumor growth in xenograft mice, when compared with wild-type controls. Altogether, these demonstrate that USP19 serves as a novel mechanistic modulator of Nrf1, but not Nrf2. In turn, our additional evidence has also unraveled that transcriptional expression of endogenous USP19 and its promoter-driven reporter genes is regulated by Nrf2, as well by Nrf1, at distinct layers within a complex hierarchical regulatory network. In today’s episode, Ryanne Sullivan, shares her 20-year journey with IBD. Diagnosed with both Crohn’s Disease and Ulcerative Colitis, she’s learned to manage her symptoms and achieve remission through nutrition and lifestyle. At age 14, she started treatment with medications, like many others, using steroids and 6MP to try and control inflammation and symptoms. But, she often found the side effects of medications to be worse than the IBD itself.  Over the years, Ryanne has found stress to be a major trigger of her flare ups. Right out of high school she started working full time and living on her own, but the stress brought on yet another flare. Remicade helped to put her back into remission, however, it was short lived and soon after she was back to battling Crohn’s symptoms.  After the Remicade failed, her aunt introduced her to “The Maker’s Diet” by Jordan S. Rubin. It focused on bringing nutrition back to the basics of real food, grown in nutrient-dense soil and improving the microbiome. The book kicked off her journey to discovering how she could control inflammation from within. Since then, Ryanne has only had a few flare ups due to the stress from each of her three pregnancies and with the knowledge and experience she’s gained over the years (and continues to gain!), she was able to get right back into remission.  Ryanne focuses her diet around foods and supplements that help reduce inflammation, including plenty of fruits and vegetables and grass-fed meats. Her family eats the same and follows a healthy lifestyle with her, though she does try to let her kids be kids and enjoy the occasional bowl of ice cream and Goldfish crackers :) She’s learned to listen to her body and knows what the first signs of a flare up are for her, which gives her a chance to head off a full-blown flare up by sticking to the basics and giving her gut a chance to heal. Her go-to methods and supplements include intermittent fasting/gut rest, aloe vera capsules (or juice), Protandim® Nrf2 and Nrf1, prebiotics and probiotics, easy-to digest foods and soups, golden milk (made with almond milk, turmeric, black pepper, ginger, and cinnamon), and dandelion tea. As many of us with Crohn’s Disease and Ulcerative Colitis know, genetics play a role in IBD. Recently, Ryanne’s 8-year-old daughter was diagnosed with Crohn’s and they’re working together to help give her the same level of control and quality of health. Ryanne talks about what it was like to have open and honest conversations about taking a holistic route with her daughter’s pediatrician and gastroenterologist. While medications are not off the table, they are hoping to avoid them. Ryanne’s advice for other parents with children diagnosed with IBD: keep a food journal, learn everything you can about the disease, learn to talk to doctors openly, and  get involved in the community!
