Abstract: The advanced form of AMD, geographic atrophy, is associated with increased RPE oxidative stress and chronic inflammation. Here we evaluated the effects of delivering an anti-inflammatory viral gene by an AAV-vector in a mouse model of geographic atrophy. We measured changes in retinal function, structure, and morphology over nine months with electroretinography, optical coherence tomography, and fundoscopy, respectively. In addition, we used retinal tissue to quantify changes in markers of inflammation by multiplex ELISA, RT-qPCR, and immunofluorescence staining. Our AAV significantly delayed the loss of retinal function and structure and decreased retinal inflammation compared to the control AAV treatment. Our results suggest that modulating retinal inflammation could significantly slow the progression of geographic atrophy. Copy rights belong to original authors. In today's episode, we have Stu Massengil. He has accomplished more by the age of 20. He has an eight-figure business and beat cancer at 24. His whole mission is to change the statistics of 80% of people not loving what they do to the point where people fall in love with what they do. Today, Stu unpacks his story to help other people find their direction and mission in life [7:18] Why should we listen to you? I don't think you would end up listening to me. I think I'm an innately curious person and you would end up telling me about your life. I'm going to get you to talk about yourself, and people love talking about themselves. So you'll keep talking. I like to listen. I want to learn. I find people have amazing stories that I can unpack if I speak to them.   [8:36]  Lifetime story version I grew up with a beautiful family, two great parents, a great older brother, and just went throughout life, being more of the shy and introverted kid throughout my life. I got into action sports. I would skateboard, snowboard and always had that “little entrepreneurship” in me.  When I started a neighborhood business, I was young by selling candy in high school. I started diving into entrepreneurship in San Diego when a network marketing company was introduced to me. [9:49] I'm a big believer in following my heart and intuition and felt the calling to do something else. One of the biggest things I've learned in that process is I left not knowing what I was going to do. [10:48] Now I work with Tony Robbins, and I'm the founder of Finding Direction because there's always a story intertwined. I felt so much pain, feeling lost, and not knowing what I wanted to do with my life and figured out that I'm not the only one there.  [11:45]  How do you create that plan? Figuring out if you're in this place where maybe you're not super excited about what you're doing, or you've been pushing for this business, or you've been pushing in this career and feeling your internal guiding system going. I don't know if this is what you're supposed to do and if you're at that place, I would say the most significant part is starting with what do you genuinely feel that liked your soul up?  [12:32] I've noticed that it's easier to get overwhelmed in today's society than ever before. We got you to go on social media for five minutes, and five minutes later, you become envious of what your friends are doing—it's the shiny object syndrome. These are distractions. Sometimes, we forget who we are at our core and get overwhelmed with the possibilities.  [14:48] How do you approach an objection? The biggest thing you can do is build your network. One of the most prominent beliefs I have is through effective communication and networking; you can open any door of opportunity.  [18:19] When that fear pops up on those cold feet, you want to ask yourself innately why is this fear popping up? You're not trying to thrive; you're trying to survive. By pushing yourself to get into an opportunity and fear shows up, understand that it wants to protect you. Acknowledge it. [19:30] Anytime that fear pops back into your mind, start looking at those new connections you've made and push yourself over the edge—you now have more reasons to do it than not. [21:28] Remember you're tasting these different areas of life. It is all part of the process of figuring out your life rather than someone.  [26:07] What promise did God make to the world when he created you? He traveled to ‘find himself’ for eight months, but he got diagnosed with testicular cancer. If you want true, genuine, real, authentic, the purest connection and relationships you can have in your life, that you have to have vulnerability. [12:27] Find someone who has what you want and model them, and then reach out to them very, very persistently and see how you can add value to them. [14:00] Success without fulfillment is the ultimate failure. [15:30] By building the right connection and by being an effective communicator and networker, that you can literally open any door of opportunity. [21:44] Success breeds success and failure can't breed success. What are three things that I could take at the end of the day and go, man, I just dominated this today. And then you carry that into the next day, and that allows you to build success on success. [26:28] Just because someone else has something doesn't mean that they were born with that or everything can be learned [28:10] How to take action: 1. Visualizing process which will take 3 minutes. literally imagine your head as a file cabinet. And imagined thoughts of, I can't do this. I'm not good enough, whatever those negative thoughts, literally imagine them being pulled out of the cabinet file and being thrown away, and then imagine taking these positive, empowering beliefs, and then putting them now in your file cabinet, which is your head and just visualize that. Do over and over and you're literally rewiring your brain. 2. A shitty plan today is better than a perfect plan tomorrow because tomorrow doesn't exist. Gil meets Nancy, a Jew, and they elope, living with her parents until he graduates. He works for a print shop whose owner pays poorly, and the men try to unionize but lose their jobs. They plot to kill him; then Gil is diverted to Atlanta where he works and takes his wife and four children. He goes overboard with drinking and drug use. Some church ladies come to the door, inviting them to church. Gil slams the door on them three Thursdays in a row. Then he overdoses and spends a week in the hospital after which he takes his family to church and is saved. He goes to Bible college, Nancy is saved and they go to Brazil as missionaries. Now in ministry to Jews.

Gil's parents drink, fight and practice infidelity. His aunt provides each of the five siblings with a week of respite at her home. Gil's father, who spent five years in prison for bootlegging, punishes the children for their occasional church attendance. But Gil loves his dad more than he loves his mother; and when dad dies of liver failure, Gil chooses to go to a boys' home. His mother visits once to tell him his younger brother has accidentally shot and killed his youngest brother. He runs away but is caught and returned. At a church service at the home, he learns to pray. Gil's aunt brings him home after three years, but he rebels, drinking and doing drugs with school friends. The first part ends as he sees the girl he will marry.
