Abstract: Nociceptive habituation is a conserved process through which pain-sensitivity threshold is adjusted based on past sensory experience and which may be dysregulated in human chronic pain conditions. Noxious heat habituation in C. elegans involves the nuclear translocation of CaM kinase-1 (CMK-1) in the FLP thermo-nociceptors neurons, causing reduced animal heat sensitivity and avoidance responses. The phosphorylation of CMK-1 on T179 by CaM kinase kinase-1 (CKK-1) is required for nuclear entry. Recently, we identified a specific Nuclear Export Sequence (NES) required to maintain CMK-1 in the cytoplasm at rest (20 {degrees}C) and showed that Ca2+/CaM binding is sufficient to enhance CMK-1 affinity for IMA-3 via a specific Nuclear Localization Signal (NLS) in order to promote nuclear entry after persistent heat stimulation (90 min at 28{degrees}C) (Ippolito et al., 2021). Here, we identified additional functional NES and NLS on CMK-1, whose activity can counteract previously identified elements. Furthermore, we clarify the relationship between the CaM-binding-dependent and T179-dependent effects. T179 phosphorylation can promotes nuclear entry both downstream of CaM-binding and as part of an independent/parallel pathway. Moreover, T179 phosphorylation can also produce the opposite effect by promoting nuclear export. Taken together, our studies show that multiple calcium-dependent regulatory mechanisms converge to bias the activity pattern across a network of NES/NLS elements, in order to activate CMK-1 nucleo-cytoplasmic shuttling, change its localization equilibrium and actuate stimulation-dependent nociceptive plasticity. Heute ist sie allerdings die Inhaberin dieser Werbeagentur «Contexta», wo sie dreissig Angestellte führt. Seit über drei Jahren sind ihre Crewmitglieder als «Nomaden» unterwegs. Es gibt keinen festen Arbeitsort mehr. Mit mobilen Büros arbeitet die Agentur mal in einer Badi, mal in einer Jugendherberge oder bei einem Pizza-Kurier Die ehemalige Schweizermeisterin im Rückenschwimmen ist leidenschaftliche Taucherin, Mutter von zwei Töchtern und lebt mit ihrem Partner in Zürich. Am Computer und in Meetings sitzend: So hat er viele Jahrzehnte von seinem Berufsleben verbracht. Dann kam der Wunsch nach einer Richtungsänderung auf. Seit bald 5 Jahren ist er nun als Fährmann tätig und transportiert mit der Bodenackerfähre (Muri bei Bern) Leute über die Aare. Das sei für ihn mehr als Arbeit. Es sei ein Lebensgefühl, das ihm erlaubt, ganz in der Gegenwart zu leben. Der Vater einer erwachsenen Tochter lebt abwechselnd in Bern und Basel. In der Stadt am Rhein ist seine Partnerin daheim. Heute spreche ich mit Raoul Biltgen, dem gebürtigen Luxemburger, der inzwischen in Wien als Schriftsteller, Schauspieler und Psychotherapeut arbeitet. Nicht unerwähnt bleiben darf der Umstand, dass Raoul am vergangenen Samstag, dem 24.04. einen der größten Preise im deutschsprachigen Krimi, den Glauser des Syndikats, für den besten Kurzkrimi des Jahres 2020 erhalten hat. Er schreibt Theaterstücke, Romane, Kurzgeschichten und hat auch einen wirklich interessanten und lustigen ... tja man könnte sagen ... Ratgeber geschrieben, der den Titel trägt „Adam spricht" In diesem Buch gibt er, begleitet von erotischen Zeichnungen, in 69 Kapiteln Einsichten in das Sex- und Liebesleben der Männer ... man achte auf die Anzahl der Kapitel, also ... nein, ich will das hier nicht vertiefen. Er wurde für seine Krimikurzgeschichten bereits 5 Mal und einmal für einen seiner Romane als Preisträger für den Glauser, den deutschsprachigen Krimipreis nominiert ... und dieses Jahr hat es dann auch geklappt. Ein neuer Friedrich-Glauser-Band von Christa Baumberger, ein Klassiker von George Orwell in neuer Übersetzung und eine ergreifende Familiengeschichte aus der Basler Arbeiterbewegung von Béatrice Schmid – dies das Angebot im heutigen Buchzeichen. «Matto regiert» - den Krimi von Friedrich Glauser kennen wir alle, und sei es nur vom Hörensagen. Wie kam es, dass Glauser die Wachtmeister-Studer-Krimis, mit denen er Furore machte, mehrheitlich in psychiatrischen Kliniken schrieb? Wie kam es, dass er von seinem 22. Altersjahr bis zu seinem Tod mit 42 entmündigt war? Ein gewichtiger Materialienband zu Glausers 125. Geburtstag wirft ein teils neues Licht auf diese Fragen. «Big brother ist watching you» - diesen Spruch kennen wir alle. Aber was hat uns der Klassiker «1984» von George Orwell heute immer noch zu sagen? «1984» ist DER Roman zum Überwachungsstaat schlechthin, geschrieben 1948. Before this he was President and Managing Partner of the Glauser Group and President at Glauser Mercedes-Benz. In today’s episode we discuss his approach and how he uses volatility.
Beide haben in ihrem Alltag intensiv mit Menschen zu tun: Die Psychologin Sibylle Glauser berät Angehörige von psychisch kranken Menschen. Der Coiffeur-Unternehmer Marc Riedo führt mehrere Coiffeurbetriebe und arbeitet selbst als Coiffeur. Sibylle Glauser (59), Psychologin Für Sibylle Glauser war in der Jugend klar: Sie wollte Tierärztin werden. Dann hat sie «Amors Pfeil» getroffen. Nach der Matura ist sie einer Jugendliebe wegen nach Spanien gezogen. Zurück in der Schweiz studierte sie dann Psychologie, was aus heutiger Sicht genau richtig war. Selber geprägt von einer schweren psychischen Erkrankung ihres Bruders, berät sie heute Angehörige von psychisch kranken Menschen. Die zweifache Mutter ist auch Mitglied des Care-Teams des Kantons Bern, wo sie bei Notsituationen psychologische Hilfe leistet. Sibylle Glauser ist ein neugieriger Mensch, dem soziale Kontakte sehr wichtig sind. Für ihre beste Freundin hat sie vor einem Jahr eine Niere gespendet. Marc Riedo (49), Coiffeur-Unternehmer Seine Grosseltern und auch seine Eltern waren es – Marc Riedo ist sozusagen im Coiffeursalon aufgewachsen. Längstens führt er in der dritten Generation die Coiffeur-Tradition der Familie fort. Und auch einer seiner Söhne ist mittlerweile in die Fussstapfen seiner Vorfahren getreten. Marc Riedo ist nicht nur zu 100 Prozent als Coiffeur tätig, sondern trägt eine grosse Portion Unternehmergeist in sich. Er leitet heute drei Unternehmen, verteilt auf acht Standorte, mit über 80 Mitarbeitenden. Auffallend in seiner Biografie: Alles hat er in seinem Leben sehr früh angepackt. Auch mit seiner heutigen Ehefrau ist er bereits seit dem 13. Lebensjahr zusammen. In dieser Woche beginnt für den Coiffeur-Unternehmer die Zeit nach den Corona-Einschränkungen: eine grosse Herausforderung! Techtonica offers six month, full time engineering apprenticeships to low income learners with a living stipend and laptop provided. We'll hear about what they've accomplished, what's next and how you can help.
Welcome back to Tech Forward, listeners! On this week’s episode, I spoke with Michelle Glauser, founder and CEO of Techtonica. Based in San Francisco, Techtonica is a nonprofit that offers free tech training, plus living and childcare stipends, to local women and non-binary adults with low incomes. Michelle is also a software engineer and the lead organizer for PyLadiesSF — the world's largest chapter of women who code in Python. In 2015, she spearheaded the #ILookLikeAnEngineer ad campaign. We discussed her motivation behind founding Techtonica, what makes it different from other boot camps, as well as her journey into tech and entrepreneurship. Despite a lifelong interest in computers, Michelle found herself working full time at a startup doing everything except software engineering. Noticing the high demand for engineers, she signed up for a training program in 2012. While her experience was overwhelmingly positive — she had a new job within 2 weeks of graduation — she also witnessed, firsthand, the lack of diversity in the tech world. She attributes part of this to the high cost of tuition (which, on average, has since tripled). But that isn’t the only barrier: many programs require participants to already have their own computer or tech network, or impose age limits. Through her crowdfunding work with the #ILookLikeAnEngineer ad campaign, Michelle had the revelation that would steer her towards entrepreneurship: “It was such a big challenge, but at the end of it I thought, I can do hard things! What other hard things can I do?” She turned her attention to socioeconomic diversity. With the bootcamp model growing in popularity, Michelle didn’t want to reinvent the wheel. By providing living and childcare stipends to participants, Techtonica supports locals in danger of displacement. “We want to train the people who are already here, alleviate the high and rising cost of living, and have the tech companies who are interested in building diversity support that training.” Companies partner with Techtonica and sign up as sponsors for participants in the 6-month apprenticeship program. After 5 months, sponsors and apprentices interview each other in order to find the best match, and the apprentices have their final month to prepare for their new role within that company. The companies who partner with Techtonica also have diversity and inclusion training to ensure the apprentices enter a welcoming environment. The curriculum at Techtonica is open sourced and created with input from the sponsor companies and industry professional volunteers. From their rigorous application process, to workshop activities designed to evaluate the characteristics of a successful engineer, Techtonica strives to ensure that every participant ends up in the best possible role. In fact, Techtonica has now graduated its first class of apprentices, and placed 100% of them with partner companies! Part of building to sell is knowing who you are going to sell to. If you don’t start thinking about your potential buyers list early, you may end up growing an entire appendage of your business that an acquirer will neither want nor value. Take Northern Lights as an example: Michael Glauser started Northern Lights to offer low-fat frozen yogurt through a growing wholesale distribution network of stores selling his desserts. At the same time, he built up a network of 60 company-owned stores under the Golden Spoon brand. The company-owned stores were expensive to start and complicated to manage. Glauser went on to sell Northern Lights to Cool Brands International for five times net income. Cool Brands turned around and immediately sold or shut down the 60 company-owned stores because they wanted Northern Lights’ wholesale distribution channel – not a bunch of expensive retail stores. During our interview, I couldn’t help but wonder how much more Glauser and his shareholders would have gotten for their company had Glauser figured out what a buyer would value and then invested all of his limited resources into building his brand and its wholesale distribution channel from the start.
