The Government has announced the reversal of speed limit reductions on a large number of New Zealand roads, including many in the northern regions of the country. The move will result in speed limits being increased on 38 sections of key routes, including parts of State Highway 1 in Northland, SH16 near Auckland and SH3 in Hamilton. Also on the list are changes to SH2 between Featherston and Masterton, and SH3 Whanganui. Making the announcement in Wairarapa, where the speed limit increase will save drivers around three minutes on their journey, Transport Minister Chris Bishop said: “It all adds up - that is productivity and growth.” Viastrada Certified Safe System Assessor and Level Crossing Safety Impact Assessor Glenn Koorey says the move goes against evidence that lower speeds make for safer roads, but there are some areas where putting the limit up can be justified. Kenyan doctors have vowed to continue their 23-day-old strike to demand better pay and improved working conditions. This, after a labor court in Nairobi Wednesday ordered the doctors to suspend the strike and complete negotiations with the government within 14 days. The doctors say the government has failed to implement a collective bargaining agreement signed in 2017. The government has accused the doctors of refusing to compromise. On Wednesday, the government offered to make available Sh2.4 billion (a little over $184 million) to cover the deployment of interns or junior doctors. Meanwhile, the strike has paralyzed health services at some hospitals. Dr Simon Kigondu, President of the Kenya Medical Association, tells VOA's James Butty, the strike will continue until the government completely addresses of all their concerns.

BUFFALO, NY- November 8, 2023 – A new editorial #paper was #published in Oncotarget's Volume 14 on March 31, 2023, entitled, “Impact of SHP2 tyrosine phosphorylation on the development of acquired resistance to allosteric SHP2 inhibitors.” The SH2 domain-containing tyrosine phosphatase 2 (SHP2) is a ubiquitously expressed non-receptor protein tyrosine phosphatase, encoded by the PTPN11 gene. It is positively regulated by upstream receptor tyrosine kinases (RTKs) to activate downstream the RAS-ERK pathway. In this new editorial, researchers Giulia Franciosa and Jesper V. Olsen from the University of Copenhagen discuss potential leukemia therapies that effectively prevent adaptive resistance. Acute myeloid leukemia (AML) is a bone marrow malignancy characterized by a blockage of differentiation and an uncontrolled proliferation of myeloid hematopoietic progenitor cells. The internal tandem duplication (ITD) in the juxtamembrane domain of the RTK FLT3 is an oncogenic driver mutation that leads to constitutive activation of its tyrosine kinase activity. Consequently, FLT3 inhibitors that block its tyrosine kinase activity represent the targeted treatment option for patients with FLT3-ITD AML, often administrated in combination with induction chemotherapy. “Nevertheless, the short duration of remission urges the development of novel combinatorial therapies for FLT3-ITD AML.” Since 2016, several potent and selective allosteric, noncovalent SHP2 inhibitors have been developed and tested in clinical trials for solid tumors. A recent study reported the effectiveness of short-term treatment with the allosteric SHP2 inhibitor SHP099 as a single agent in clinically relevant mouse models of Flt3-ITD AML. This observation was in contrast with published data showing that allosteric SHP2 inhibition is only effective as combination treatment with inhibitors of other nodes of the RAS-ERK pathway. Abstract: RNA sequencing and genetic data support spleen tyrosine kinase (SYK) and high affinity immunoglobulin epsilon receptor subunit gamma (FCER1G) as putative targets to be modulated for Alzheimer's disease (AD) therapy. FCER1G is a component of Fc receptor complexes that contain an immunoreceptor tyrosine-based activation motif (ITAM). SYK interacts with the Fc receptor by binding to doubly phosphorylated ITAM (p-ITAM) via its two tandem SH2 domains (SYK-tSH2). Interaction of the FCER1G p-ITAM with SYK-tSH2 enables SYK activation via phosphorylation. Since SYK activation is reported to exacerbate AD pathology, we hypothesized that disruption of this interaction would be beneficial for AD patients. Herein, we developed biochemical and biophysical assays to enable the discovery of small molecules that perturb the interaction between the FCER1G p-ITAM and SYK-tSH2. We identified two distinct chemotypes using a high-throughput screen (HTS) and orthogonally assessed their binding. Damaged highways are stopping produce getting to market, and forcing at least one freight company to add a surcharge to account for the major detours they have to take. State Highway Two between Gisborne and Opotiki will be closed until at least the end of the week, because of a drop out from more wild weather. Countdown has already told customers to be prepared for shortages of some vegetables. Further north in the Coromandel, another lifeline highway is out of action because of persistent slips. With the school holidays around the corner, they're already counting their losses. Krystal Gibbens has more.

Commuters are frustrated with heavy congestion on State Highway 2 between Ōmokoroa and Tauranga. The stretch of highway is used by more than 25-thousand vehicles a day and in recent weeks commuters have complained about huge delays. The Western Bay of Plenty District Council has been counting vehicles using side roads to skip the congestion. Councillor Don Thwaites told Heather du Plessis Allan the congestion is caused by the building of 800 metres of cycle track near Tauranga City. "The most significant weather event this century." That's the way Prime Minister Chris Hipkins has described the impact of ex-tropical cyclone Gabrielle. Communication and physical access to many communities, particularly in Hawke's Bay and Tairawhiti, remain difficult, with power outages adding to the strain. Hipkins said he is particularly concerned about Wairoa, where the Wairoa River has burst its banks, inundating 10-15 percent of the town which is home to around half of the town's 8000 population.  Communication was only partly restored late last night through the use of a Star Link terminal. Hawke's Bay Civil Defence said Wairoa has only one day's worth of food, and enough drinking water for two days. They have made a request to NEMA for enough food and water to supply the district for seven days. Travel between Napier and Hastings is not possible. SH2 is out due to flooding, with the Ngaruroro River Bridge closed due to flooding. SH51 is also closed due to severe weather conditions".  Further north, Wairoa is cut off as State Highway 2 Gisborne to Ōpōtiki and Napier to Gisborne is closed as is State Highway 35 Gisborne to Hicks Bay A Tokomaru Bay business owner said the highway through Tairāwhiti has sustained incredible damage.  Peter Cunningham of Highway 35 Cafe spent last night sheltering at an uphill marae along with most of the town's population.  He said Tokomaru Bay has been flooded - but Uawa/Tolaga Bay appears to be the worst-affected.  Gisborne residents are being asked to urgently conserve water after a break in the city's main supply, somewhere between the dams and the treatment plant.   The Government estimates roughly 2500 people have been displaced by Cyclone Gabrielle so far. This includes 1000 people in the Far North, 400 households in Auckland, 93 in Bay of Plenty, 75 in Waikato and roughly 1000 people in Hawke's Bay.  But with communication sparse, officials have struggled to get an idea of the damage caused throughout the country. Northland Civil Defence said hundreds of people were evacuated in Dargaville yesterday after the Awakino River burst its banks.   Further south, hundreds of people were rescued from raging flood waters in Hawke's Bay after rivers burst their banks. Hawkes Bay civil defence reported late last night that a woman in Putorino had died after a bank collapsed onto her home. Police visited the property following a missing person's report and undertook rescue efforts but were unsuccessful. And a body was found on the shore in Bay View, Napier shortly before eight-PM last night. It is not yet known if the death is connected to the flooding. Kevin McCormack spent most of the day on his roof with his wife and two neighbours after the Tutaekuri River in Napier burst its banks. In Muriwai, near Auckland, Fire and Emergency will resume the search today for the volunteer firefighter missing in a landslide. Dave van Zwananberg is a vet at Vets North in Kumeu. He is trapped in the wreckage of a house that collapsed. A person is in a critical condition in hospital after being hit by a a falling tree fell in a Kohimarama playground yesterday afternoon. AT's Executive General Manager of Safety, Stacey Van Der Putten said: "Aucklanders should be prepared for delays and disruption to travel today as flood damage, sea conditions, power outages and fallen trees continue to affect our roads and public transport network." Work is well underway to assess and repair damaged roads, with over 500 crew members working across Auckland. While dozens of roads have been reopened, there are currently more than 100 roads closed or partially closed. Many thousands of people still don't have power this morning and face extended outages. Power outages have been reported for 60,000 customers across Hastings, Havelock North, Napier, north along east coast to Tūtira and south to Waimārama. It is expected to take some time before power is fully restored across the region. At its peak earlier yesterday, across the country close to 225,000 people were without power, authorities said.

The sun might be out but the job isn't done in the Coromandel, according to Coromandel-Thames Civil Defence controller Gary Towler. The region has experienced massive slips and land instability, compromising its entire roading network. ”Every time our land guys clear a slip, another one comes down right behind them,” Towler told Kerre Woodham Mornings on Newstalk ZB today. A number of houses in the Coromandel have already been red-stickered, Towler confirmed, as staff worked their way through affected areas to check the condition of housing. The other focus is power, which most of the area is still without. Powerco said it had 7462 customers without power in the Coromandel as of 8am today. ”We have the luxury of the New Zealand Defence Force, which we're deploying on to the east coast today to do a lot of review work and drone work to get a good picture of what carnage was created,” Towler said. Meanwhile, critical assets such as generators and wastewater plants in the Coromandel are running out of diesel, which helicopters are bringing in for stock-ups as they can't be accessed by road. If people are wanting to visit their homes and baches, Towler has told residents to “take a breath” and check the NZTA website and affiliated social media pages for information on road conditions. ”People need to have a go-bag and Plan B in case a slip blocks their path. That will be the case for the next five or six days,” he said. ”But people should check their properties as quickly as possible so we know if we're dealing with any other stretches. If you noticed a drop in pressure around Waitakaruru, the Water Treatment Plant is now back up and running.” The council says it is continuing to support the clean-up effort around the district and evaluating damage. It is compiling a list of affected properties and completing a welfare needs assessment. “Please call our team on 07 862 8609 or 0800 734 834. Our team will then pass the information onto our Civil Defence Welfare team.” Building inspectors will be out checking damaged properties today. Abstract: Class IA phosphoinositide 3-kinase (PI3K) galvanizes fundamental cellular processes such as migration, proliferation, and differentiation. To enable multifaceted roles, the catalytic subunit p110 utilizes a multi-domain, regulatory subunit p85 through its inter SH2 domain (iSH2). In cell migration, their product PI(3,4,5)P3 generates locomotive activity. While non-catalytic roles are also implicated, underlying mechanisms and its relationship to PI(3,4,5)P3 signaling remain elusive. Here, we report that a disordered region of iSH2 contains previously uncharacterized AP-2 binding motifs which can trigger clathrin and dynamin-mediated endocytosis independent of PI3K catalytic activity. The AP-2 binding motif mutants of p85 aberrantly accumulate at focal adhesions and upregulate both velocity and persistency in fibroblast migration. We thus propose the dual functionality of PI3K in the control of cell motility, catalytic and non-catalytic, arising distinctly from juxtaposed regions within iSH2. Abstract: Suppressor Of Cytokine Signaling (SOCS) 1 is a critical negative regulator of cytokine signaling and required to protect against an excessive inflammatory response. Genetic deletion of Socs1 results in unrestrained cytokine signaling and neonatal lethality, characterised by an inflammatory immune infiltrate in multiple organs. Overexpression and structural studies have suggested that the SOCS1 kinase inhibitory region (KIR) and Src homology 2 (SH2) domain are important for interaction with and inhibition of the receptor-associated JAK1, JAK2 and Tyk2 tyrosine kinases, which initiate downstream signaling. To investigate the role of the KIR and SH2 domain in SOCS1 function, we independently mutated key conserved residues in each domain and analysed the impact on cytokine signaling, and the in vivo impact on SOCS1 function. Mutation of the SOCS1-KIR or SH2 domain had no impact on the integrity of the SOCS box complex, however, mutation within the phosphotyrosine binding pocket of the SOCS1-SH2 domain specifically disrupted SOCS1 interaction with phosphorylated JAK1. In contrast, mutation of the KIR did not affect the interaction with JAK1, but did prevent SOCS1 inhibition of JAK1 autophosphorylation. In human and mouse cell lines, both mutants impacted the ability of SOCS1 to restrain cytokine signaling, and crucially, Socs1-R105A and Socs1-F59A mice displayed a neonatal lethality and excessive inflammatory phenotype similar to SOCS1 null mice. This study defines a critical and non-redundant role for both the KIR and SH2 domain in endogenous SOCS1 function. Copy rights belong to original authors. Studio Spider zaoferowało całkiem przyjemny rpg-soulslike w XVI-to wiecznym Paryżu a debitutancki projekt Reply Games zaserwował nam slashera w stylu DMC. Poza tym newsy, Resident Evil Showcase i to na co bardzo czekaliśmy czyli Silent Hill Showcase. Gdzie zaprezentowano remake SH2 od polskiego Bloober Team oraz trzecią część filmu i inne gry z tego uniwersum które zadebiutują w przyszłości.

Star and Cluster Formation in the Sh2-112 Filamentary Cloud Complex by Alik Panja et al. on Tuesday 20 September We present the star formation activity around the emission nebula Sh2-112. At a distance of $sim2.1$~kpc, this ion{H}{2} complex, itself 3~pc in radius, is illuminated by the massive star (O8,V) BD$+$45,3216. The associated molecular cloud extends in angular scales of $2fdg0times0fdg83$, corresponding to linear sizes of 73~pc by 30~pc, along the Galactic longitude. The high-resolution ($30arcsec$) extinction map reveals a chain of dust clumps aligned with the filament-like structure with an average extinction of $A_{V} sim 2.78$~mag, varying up to a maximum of $sim17$~mag. Our analysis led to identification of a rich population ($sim 500$) of young (average age of $sim 1$~Myr) stars, plus a numerous number ($sim 350$) of H$alpha$ emitters, spatially correlated with the filamentary clouds. Located near the edge of the cloud, the luminous star BD$+$45,3216 has created an arc-like pattern as the ionizing radiation encounters the dense gas, forming a blister-shaped morphology. We found three distinct young stellar groups, all coincident with relatively dense parts of the cloud complex, signifying ongoing star formation. Moreover, the cloud filament (excitation temperature $sim 10$~K) traced by the CO isotopologues and extending nearly $sim 80$~pc is devoid of ionized gas except at the dense cores (excitation temperature $sim$ 28--32~K) wherein significant ionized emission excited by OB stars (dynamical age $sim$ 0.18--1.0~Myr) pertains. The radial velocity is dynamic (median $sim -3.65$~km~s$^{-1}$) along the main filament, increasing from Galactic east to west, features mass flow to form the massive stars/clusters at the central hubs. arXiv: http://arxiv.org/abs/http://arxiv.org/abs/2209.09609v1

Silent Hill: Secuelas: “Silent Hill: Revelation” (2012). Presupuesto: $50 M USD. Recaudación: $100 M USD. Año: 2006. Director: Christophe Gans. Actores: Radha Mitchell (Rose DaSilva), Sean Bean (Christopher DaSilva), Laurie Holden (Cybill Bennet) y Jodelle Ferland (Alessa Guillespie). Cronología:No hay que olvidar que para el 2006, cuando salió la película, ya existía el videojuego original del 99, también SH2 de 2001 y SH3 del 2003. Es decir, había más contexto en cuanto a la historia, personajes y sobre el pueblo. Esto tendría un impacto en la primera adaptación cinematográfica.Reparto: Radha Mitchell (Rose DaSilva): esta actriz no es muy popular, la recuerdo en el remake de George A. Romero “The Crazies”. Sean Bean (Christopher DaSilva): Actor de gran trayectoria y hasta donde se dice, todos sus personajes mueren. Tal vez lo recordemos más como Boromir en “TLOTR” y Ned Stark en “GOT”. Laurie Holden (Cybil Bennet): recordada por su trabajo en la serie “X-Files” y después en “The Walking Dead”. ¿De qué trata la película?El matrimonio DaSilva empieza a preocuparse debido a que su hija adoptada, Sharon (Alessa), sufre de pesadillas y sonambulismo, donde a menudo pronuncia las palabras “Silent Hill”, sin recordar nada al despertar.Rose investiga un poco sobre ese pueblo y decide llevar a su hija con el propósito de ayudarla a recordar sobre su vida antes de ser adoptada. Al llegar a Silent Hill, descubre un sitio cubierto de cenizas y neblina, sufre un aparatoso accidente y Sharon desaparece. Rose se adentrará hasta las entrañas del lugar para localizar a su hija, sin embargo, encontrará un terrible secreto.Antecedentes:¿Qué tanto se acerca la adaptación al juego? De entrada, en el juego es el esposo, Harry, quien se dirige a Silent Hill con su hija Cheryl para tomar unas vacaciones. Al entrar al pueblo, una niña se atraviesa haciendo que, de un volantazo, el hombre pierda el control del vehículo y queda inconsciente por el impacto. Al despertar, Cheryl no está y va a buscarla.Toda esta introducción es casi igual al juego en la película. Con la diferencia de que, en lugar de Harry, tenemos a Rose y el nombre de la hija adoptiva es Cheryl en lugar de Sharon.Además, contamos con el pueblo lleno de niebla y ceniza, en la película se explica que esto se debe al carbón ardiente bajo la ciudad. Sabemos que Rose escapaba de la policía, para llegar al pueblo. En ambas propuestas, logran el encuentro entre Cybil y Rose.¿Qué es lo que ocasiona que el pueblo esté lleno de neblina?Igual se preserva bastante bien este hecho, el horror detrás de la historia, muy crudo. En ambos casos, Alessa sufrió quemaduras fatales debido a un ritual que traería al dios del culto al que Dahlia lideraba. Como muestran en la película, Alessa es inmolada a sus siete años mediante un sacrificio horrible y lento.En su desesperación, la joven utiliza una proyección astral para separarse en dos versiones: la que está en sufrimiento y la feliz. Esto interfiere con el ritual porque, sí, logran poner la semilla del embrión maléfico en Alessa, pero no puede nacer porque su alma está dividida.Las heridas fatales de Alessa son sanadas por medio de un hechizo que la mantiene con vida. Dahlia quiere que su contraparte, Cheryl (en la película Sharon) regrese a Silent Hill para que se unan y pueda nacer la deidad prometida. Sin embargo, mantiene escondida a su hija en el sótano del hospital, a cargo de una enfermera. El médico –cultista– le administra alucinógenos y Dahlia la hace sufrir para que atraiga a Cheryl. ¿Cómo? Quiere intensificar el deseo de cometer un suicidio, pero en su estado, no puede hacerlo.En la película vemos a Dahlia, la madre vagabunda, aislada y viviendo en remordimiento por entregar a su hija. Sin embargo, en el juego, Dahlia era la líder del culto. De igual forma, la hija de Dahlia lleva el nombre de Alessa, la niña demonio que ha puesto la maldición sobre Silent Hill.La parte de Alessa que está enojada quiere venganza y se encuentra en continuo sufrimiento y alucinaciones.La enfermera que aparece en la película participa en el juego. Lisa, quien estaba a cargo de Alessa mientras era torturada y drogada, empieza a sangrar por los ojos, ya que no puede soportar verla.En el juego, Harry salva a Cybill (opcional) de morir y después de derrotar a la deidad reencarnada, huyen juntos.Es bien sabido que, según las decisiones del jugador, hay diferentes finales.En el caso de la película, Cybill termina inmolada también, quemada como bruja hasta morir.Derrotada la deidad, con la última energía de Alessa, reencarna un bebé, el cual Harry se lleva de regreso a su casa.En la película, Rose salva a Sharon de ser ejecutada. No existe la deidad como tal, sino la líder del culto que vive en el limbo sin aceptar que ellos son los pecadores. Cuando Alessa quiere vengarse de ellos, no puede porque se resguardan en la iglesia. Por eso guía a Rose con el propósito de que entre y por fin pueda hacerlos pagar.Al final, se entiende que Rose regresa a su casa con Sharon, sin embargo, se quedó en el Otro Mundo, ese limbo extraño.Mitología:Previo a la colonización de USA, el sitio se llamaba “El lugar del Silencio” o “Espíritus silenciados”. La leyenda dice que un espíritu no es solo de almas vivas, sino también rocas, agua, etc. El lugar no era malo por naturaleza, pero debido a las cosas ocurridas, cambió.El pueblo manifiesta los pensamientos más oscuros de las personas que están ahí.La conquista expulsa a los nativos, los nuevos habitantes le cambian el nombre. Una epidemia arrasa con ellos y los pocos sobrevivientes huyen. Más adelante construyen una prisión y un hospital debido a otra epidemia. La prisión cierra, se descubre la mina de carbón, entra en operación y la zona crece económicamente.Durante la guerra civil americana se construye otra prisión. Gente comienza a desaparecer.La prisión cierra y se vuelve un lugar turístico. Más gente muere, incluyendo gobernadores.Los cultistas son los culpables de las desapariciones.Debido a tanta corrupción en ese espacio sagrado y espiritual, es que el poder de esa tierra comenzó a reaccionar con más fuerza hacia sus ocupantes. Cada vez que alguien la toca, se expresaría lo que tuviera reprimido en lo más profundo de su ser.La neblina y el otro mundo:Es una delgada línea entre la realidad y los sueños.Los desarrolladores del juego explican estos cambios entre “el lado correcto” y “el lado inverso”. Ponen de ejemplo que, cuando uno duerme, puede estar en la fase de “sueño ligero” y “sueño REM (rapid eye movement)” que es cuando estamos soñando. De la misma forma actúa Silent Hill, hay momentos en la zona ligera, con la neblina y algo de luz... Pero al momento de alcanzar la zona REM, ahí nos adentramos a la peor pesadilla, junto a la oscuridad. Así, el ciclo se repite.Según los desarrolladores, ninguno de estos espacios es la realidad. La neblina representa los pensamientos que tiene y cómo los espíritus se aparecen en ellos. Conforme empiezas la espiral descendiente a las angustias y represiones, caes en el lado inverso y las manifestaciones se vuelven horribles.El mundo inverso:Según qué tan corrupto es su espíritu, será el lado inverso. ¿Por qué se llama así? En teoría es porque toma algo que conoces, algo que existe y lo transforma en su polo opuesto, en su lado más terrible.Dios (teoría de Max Derret):El dios al que La Orden (el culto) hace referencia, es muy probable que sea Baphomet –dios de lo absoluto–, se le conoce así porque es todo: tiene cara de cabra, cuerpo humano, tiene partes femeninas y masculinas, representa la luz y la oscuridad, tiene escamas y alas.Por otro lado, Carl Jung en su libro “Aion” dice que todos los humanos quieren convertirse en cosas mejores. Le llamó a la cumbre de este proceso de mejora como “Sí-mismo” o “The Self”. Este es el ideal máximo, equivalente a Dios.En varias religiones hay representaciones de Dios que alcanzan ese estado al llegar a un balance último entre lo mejor y peor de ellos. Por ejemplo, los cristianos quieren llegar a ser como Jesús, quien tuvo que vivir un infierno, ser crucificado y asesinado para resucitar como Dios, en su nivel más poderoso. Al lograr esto, se vuelve un todo: fue material y espiritual, fue malo y bueno, fue único (Jesús), pero universal porque es todo lo que existe, fue temporal como humano, pero también es eterno.Así, en “Silent Hill” vemos ese concepto en la deidad que quieren revivir y lo que se manifiesta en el pueblo. Está basada en el absoluto. El absoluto puede tomar varias formas según quién lo perciba. Por otro lado, La Orden, como culto está dispuesta a recorrer el camino del sufrimiento, sacrificios y demás, a cambio de llegar a un paraíso libre de sufrimiento.¿Cuál fue la contribución de la película al cine de horror?Las adaptaciones de videojuegos a películas por lo general no tienen buenos resultados, pero con “Silent Hill” se hizo un excelente trabajo.Basada en el primer título del videojuego del mismo nombre, nos presenta un gran tributo a la historia y personajes. Para el año del estreno ya contaban con cuatro títulos publicados.Tuvo buena recepción, pero tal vez se esperaba más por el potencial del universo de los videojuegos y por lo popular que estos son.Aunque mantiene la base de la historia, poco a poco la película se va alejando de Harry Mason. Inclusive toma elementos de otros títulos de la franquicia como las enfermeras y Pyramid Head del “Silent Hill 2” o la música de “Silent Hill 3”. Tal vez una estrategia para atraer a los fans.¿Por qué incluir a Pyramid Head?Pyramid head es un personaje icónico de “Silent Hill” y presenta una simbología importante: es la encarnación de la culpa y la ira.Es evidente que el personaje sale en “Silent Hill 2” por primera vez y que, muchos fans se enojaron porque no tenía nada que ver en la primera parte. Pero, si lo vemos desde un punto de vista simbólico, más allá de ser fan service e inspirar un gran miedo, representa todos los pensamientos oscuros que tenemos y que reprimimos.Como referencia, en “SH2”, el protagonista vive con una culpa terrible por haber asesinado a su esposa. No solo eso, previo al acto, lo acosan sentimientos de desesperación, enojo o represión sexual.Cuando vemos a Pyramid head, en varias ocasiones está realizando actos sexualizados y físicamente es un carnicero.En la película, la aparición de Pyramid head representaría aquella culpa de los actos horrendos realizados a Alessa y es el verdugo que va a castigar al pueblo. Actúa como protector.Se dice que también representa la figura paterna de Alessa. De cierta forma, en la película, debido a que Alessa nació del pecado, pues Dahlia era madre soltera, el culto decidió castigarla. Esto no hubiera sucedido si Alessa hubiera nacido de un matrimonio típico.Producción:Christopher Gans (“Brotherhood Of The Wolfes”) intentó por casi cinco años obtener los derechos de Konami para filmar una película de “Silent Hill”. Tanto Konami Japan y Team Silent, en ese entonces equipo de desarrollo de los videojuegos, se involucraron en la producción del filme.El escritor Roger Avary explicó que cuando su padre era niño, un ingeniero de minas solía contarle historias sobre el pueblo de Centralia, en Pensilvania, EE. UU., donde los depósitos de carbón de una mina local se incendiaron, lanzando gases tóxicos que obligaron a los habitantes a evacuarlo indefinidamente. Esta historia sirvió como base para “Silent Hill” en la película.La producción también contrató al compositor de la música de los videojuegos, Akira Yamaoka, y eso le da una mayor atracción, sin duda te transmite la sensación de estar jugando algún título de la saga.Gans pensó en hacer la película un universo de mujeres. La inclusión del esposo y el detective fueron elementos que la productora les hizo introducir.Al hacer el personaje principal mujer, Rose, considera que la película tiene una dirección clara: es un matriarcado, las mujeres están a cargo de toda decisión relevante y existe hermandad entre ellas, pueden sentir simpatía a través del hecho de ser madres, explorar desde la concepción divina. Lo vemos en la religión cristiana, pero en la historia se aborda como una deidad maligna, además de la fuerte simbología que existe respecto a la caza de brujas. Son concepciones con las cuales la audiencia está familiarizada.Akira Yamaoka menciona que el juego de “Silent Hill” se basa en todos los miedos y ansiedades reprimidas que tenemos. Cada persona sufre de manera diferente. Esas figuras deformadas representan dichos aspectos.Los efectos especiales son muy buenos, pero, además, el director buscó en la medida de lo posible que las criaturas humanoides fueran como esculturas, evitando el uso de CGI. Todas son actores y realzan la película, inspiran mayor miedo debido al impacto visual y por lo viscerales que son.Curiosidades:El director Gans, durante las grabaciones, tenía una TV con un PS2 y jugaba “Silent Hill” para que los actores y cinematógrafos pudieran ver lo que quería emular en algunos ángulos y movimientos.La película no cuenta con el elemento “JumpScare”. Federated Farmers President David Clark said floodwaters in Canterbury have exceeded warnings "by a huge magnitude" and multiple rescue operations are taking place.One person is awaiting rescue by helicopter to get out of a flooded area as torrential rain hits Canterbury."It's a really serious situation here in Canterbury, the rivers are breaking in multiple places," he said.Clark has been helping with rescues, including a car that has been swept out in the floods at 4.30am where fire and emergency crews couldn't get to, and a farmer being trapped in a tractor by the floodwaters."Key message from Federated Farmers today is that safety is paramount, don't put your own life in danger," he said."We've seen multiple rescues, with machinery having to go out in deep floodwaters to rescue people."Police were called to assist a fire crew in a rescue emergency on Walkhams Rd at 8am this morning, a police spokeswoman said."It appears one person needs assistance to get out of a flooded area," she said."No injuries, they're just awaiting rescue at the moment. A helicopter has been called."A storm chaser at Canterbury Snow and Weather Watch is expecting multiple rivers to burst their banks today after the worst weather he has seen in a decade.The region is being lashed by torrential rain, causing widespread flooding and road closures, including the stretch from Springfield to Castle Hill on SH73, the main route to the west coast."We're hearing that a lot of the rivers on the Canterbury Plains, which people live very close to and normally run dry, are running at the highest levels on record," Josh Oliver said."People are pretty concerned. Abstract: Protein engineering through directed evolution is an effective way to obtain proteins with novel functions with the potential applications as tools for diagnosis or therapeutics. Many natural proteins, largely antibodies as well as some non-antibody proteins, have undergone directed evolution in vitro in the test tubes in the laboratories around the world, resulted in the numerous protein variants with novel or enhanced functions. In this study, we constructed a Fyn SH2 variant library by randomizing the 8 variable residues in its phosphotyrosine (pTyr) binding pocket. Selection of this library by a pTyr peptide from MidT antigen led to the identification of SH2 variants with enhanced affinities to the peptide, compared to the wild type SH2, by EC50 assay. Fluorescent polarization (FP) was then applied to quantify the binding affinity of the newly identified SH2 variants. As a result, three SH2 variants, named V3, V13 and V24, have comparable binding affinities with the previously identified SH2 triple-mutant superbinder (refer to Trm). Biolayer Interferometry (BLI) assay was employed to disclose the kinetics of the binding of these SH2 superbinders, in addition to the wild type SH2, to the phosphotyrosine peptide. The results indicated that all the SH2 superbinders have two-orders increase of the dissociation rate when binding the pTyr peptide while there was no significant change in their associate rates. The previously identified SH2 superbinder Trm as well as the V13 and V24 discovered in this study have cross-reactivity with the sulfotyrosine (sTyr) containing peptide while the wild type SH2 does not. Intriguingly, though binding the pTyr peptide with comparable affinity with other SH2 superbinders, the V3 does not bind to the sTyr peptide, implying it binds to the pTyr peptide with a different pattern from the other superbinders. The newly identified superbinders could be utilized as tools for the identification of pTyr-containing proteins from tissues under different physiological or pathophysiological conditions and may have the potential in the therapeutics. Abstract: Mutations of PTPN11, the gene coding for the Src homology 2 domain-containing phosphatase 2 (SHP2), cause childhood leukemias and developmental disorders. SHP2 inhibitors targeting the catalytic site or an allosteric pocket lack specificity or are ineffective on pathogenic variants. In addition, several data indicate that increased association with cognate proteins, through its SH2 domains, rather than enhanced catalytic activity, is the main effect of mutations causing hyperactivation of SHP2-mediated signaling. We developed peptide-based molecules with low nM affinity to the N-SH2 domain and high specificity. These molecules bind to pathogenic variants of SHP2 with an affinity up to 20 times higher than to the wild-type protein, in contrast to allosteric inhibitors, and were able to revert the effects of a pathogenic SHP2 mutation in zebrafish embryos. Our results provide a novel route for SHP2-targeted therapies and a tool to investigate the role of protein-protein interactions in the function of SHP2. Copy rights belong to original authors. Now we are going to cover Space Rabbits! This conversation took place between Houston and the Apollo 11 crew just before the first Moon landing in 1969:         Houston: Among the large headlines concerning Apollo this morning, is one asking that you watch for a lovely girl with a big rabbit.         An ancient legend says a beautiful Chinese girl called Chang-O has been living there for 4,000 years. It seems she was banished to the Moon because she stole the pill of immortality from her husband.         You might also look for her companion, a large Chinese rabbit, who is easy to spot since he is always standing on his hind feet in the shade of a cinnamon tree. The name of the rabbit is not reported.         Michael Collins: Okay. We’ll keep a close eye out for the bunny girl. Having said that, lets look a little deeper into Rabbit's in space. Now from Wikipedia, Animals had been used in aeronautic exploration since 1783 when the Montgolfier brothers sent a sheep, a duck, and a rooster aloft in a hot air balloon (the duck serving as the experimental control). The limited supply of captured German V-2 rockets led to the U.S. use of high-altitude balloon launches carrying fruit flies, mice, hamsters, guinea pigs, cats, dogs, frogs, goldfish and monkeys to heights of up to 144,000 feet (44,000 m). These high-altitude balloon flights from 1947 to 1960 tested radiation exposure, physiological response, life support and recovery systems.  The U.S. high-altitude manned balloon flights occurred in the same time frame, one of which also carried fruit flies. Animals in space originally served to test the survivability of spaceflight, before human spaceflights were attempted.  Later, animals were also flown to investigate various biological processes and the effects microgravity and space flight might have on them.  Bioastronautics is an area of bioengineering research which spans the study and support of life in space. To date, seven national space programs have flown animals into space: the Soviet Union, the United States, France, Argentina, China, Japan and Iran. A wide variety of animals have been launched into space, including monkeys, dogs, and insects.  The United States launched flights containing monkeys and primates primarily between 1948-1961 with one flight in 1969 and one in 1985.  France launched two monkey-carrying flights in 1967.  The Soviet Union and Russia launched monkeys between 1983 and 1996.  During the 1950s and 1960s, the Soviet space program used a number of dogs for sub-orbital and orbital space flights. Back in the late 1950’s space programs in the Soviet Union (Russia) and United States used animals to make high altitude and orbital flights.  Tragically, many times they were one-way trips. It was inhumane on a lot of levels. Marfusha, the first rabbit astronaut, made a high altitude test flight on July 2, 1959 aboard an R2-A rocket.  Following this on August 19, 1960, another rabbit was sent into space on the Soviet Sputnik 5 and returned alive along with its fellow animal astronauts, which included dogs and mice. There are a few photo's of unlikely space travelers, such as a white dog and a grey rabbit, ad two dogs and a rabbit available on line. Otvazhnaya (the dog) and Marnushka (the rabbit) braved a high altitude test flight on July 2, 1959 aboard an R2-A rocket.  Another dog, Snezhinka, also went on the flight.  Fortunately, all three animals were recovered successfully.  This rabbit went on to make five more successful high altitude test flights in the following year. Then in August 1960, two dogs, Belka and Strelka, an unnamed gray rabbit, 40 mice and 2 rats, were launched in Sputnik 5. Again in September 1962 a rabbit was aboard the Artemis that made 12 complete earth orbits. However, Artemis collided with space debris and sustained critical damage. Fortunately, the rabbit was recovered unharmed 933 miles off the east coast of Brazil. NASA’s History of Animals in Space web page states, after the manned lunar landing of Apollo 11, the role of animals was limited to the status of "biological payload." The range of species broadened to include rabbits, turtles, insects, spiders, fish, jellyfish, amoebae, and algae. Although they were still used in tests dealing with long-range health effects in space, tissue development, and mating in a zero-g environment, etc., animals no longer made the front pages. One exception to this was one of the last Apollo flights, Skylab 3, which launched on July 28, 1973. On board were Anita and Arabella, two common Cross spiders. Tests were set up to record the spiders' successful attempts to spin webs in space. Now this is also from NASA. Most recently, Animals go into space to help conduct scientific research only when absolutely necessary. Researchers prefer to research with computer models, or by involving the astronauts directly. For some experiments, however, only animals will work. Sometimes the situations need to be closely controlled-such as a monitored diet. Human astronauts generally aren't willing to agree to eat the same amount and type of food, so this experiment would be a burden to them.  Animals, however, always have monitored feedings. Taking animals into space requires special considerations. If a group of laboratory mice were to fly aboard the next Space Shuttle mission, what would be needed? Traditional aquarium-style cages don't provide enough traction for mice to walk around; instead, space mice have wire mesh cages so their toes can grip a rougher surface. Wood chips couldn't be used for bedding; they wouldn't stay in place. Gravity-feed water bottles wouldn't work; pressurized water containers are needed instead. Bowls of dry food aren't practical, so compressed food bars are provided instead. As for how to clean the cages, a special waste containment system has been created to keep everything in its place. Do the animals like living in microgravity? Does floating instead of walking confuse them? "Amazingly, they adapt very quickly," says Laura Lewis, a member of NASA Ames Institutional Animal Care and Use Committee. "Within 5 minutes, mice are floating in their living spaces, grooming themselves, and eating, just as they would on Earth." "Good science sets up hypotheses for an experiment, but sometimes the result is not what you expect," says Lewis. "While we test our projects on the ground and in simulators, once we get into space, we are sometimes surprised by what we learn." Baby mammals have a hard time in space because they normally huddle for warmth-and in space, it's hard to huddle when bodies drift and float. It's also difficult for babies to nurse when they can't locate their mother's nipple. Animals that travel in space are cared for ethically and humanely, Lewis says. "The Institutional Animal Care and Use Community looks at the most humane alternatives for taking animals into space," she says. "Regulations for animal research are more intense than for using people in research because people can give consent. Animals can't object, so people need to work on their behalf. Animal housing rules are more extensive than the requirements for human children day care centers. NASA facilities that house animals for research are accredited by an organization that requires proof that animals are cared for in a facility that meets those standards." The United States Department of Agriculture Animal Welfare Act and the Public Health Services Policy Act protect research animals and set minimum standards. "Animals don't go into space very often," Lewis says. "There are so few flight opportunities for a mission to include animals, so the project has to be pretty important to earn a spot on any trip into space. When animals do make the trip, their welfare is a key concern." There are a couple of websites touting the use of rabbits as a food source for human space colonies on Mars. Over the past 50 years, American and Soviet scientists have utilized the animal world for testing. Despite losses, these animals have taught the scientists a tremendous amount more than could have been learned without them. Without animal testing in the early days of the human space program, the Soviet and American programs could have suffered great losses of human life. These animals performed a service to their respective countries that no human could or would have performed. The Micmacs are original natives of the Nova Scotia/New Brunswick region. They also settled in locations in Quebec, Newfoundland, and Maine. Today, most Mi'kmaq people live on the Canadian side of the border, but the Aroostook Micmacs live in northeastern Maine. Long ago, Rabbit was a great hunter. He lived with his grandmother in a lodge which stood deep in the Micmac forest. It was winter and Rabbit set traps and laid snares to catch game for food. He caught many small animals and birds, until one day he discovered that some mysterious being was robbing his traps. Rabbit and his grandmother became hungry. Though he visited his traps very early each morning, he always found them empty. At first Rabbit thought that the robber might be a cunning wolverine, until one morning he found long, narrow footprints alongside his trap line. It was, he thought, the tracks of the robber, but they looked like moonbeams. Each morning Rabbit rose earlier and earlier, but the being of the long foot was always ahead of him and always his traps were empty. Rabbit made a trap from a bowstring with the loop so cleverly fastened that he felt certain that he would catch the robber when it came. He took one end of the thong with him and hid himself behind a clump of bushes from which he could watch his snare. It was bright moonlight while he waited, but suddenly it became very dark as the moon disappeared. A few stars were still shining and there were no clouds in the sky, so Rabbit wondered what had happened to the moon. Someone or something came stealthily through the trees and then Rabbit was almost blinded by a flash of bright, white light which went straight to his trap line and shone through the snare which he had set. Quick as a lightning flash, Rabbit jerked the bowstring and tightened the noose. There was a sound of struggling and the light lurched from side to side. Rabbit knew b the tugging on his string that he had caught the robber. He fastened the bowstring to a nearby sapling to hold the loop tight. Rabbit raced back to tell his grandmother, who was a wise old woman, what had happened. She told him that he must return at once and see who or what he had caught. Rabbit, who was very frightened, wanted to wait for daylight but his grandmother said that might be too late, so he returned to his trap line. When he came near his traps, Rabbit saw that the bright light was still there. It was so bright that it hurt his eyes. He bathed them in the icy water of a nearby brook, but still they smarted. He made big snowballs and threw them at the light, in the hope of putting it out. As they went close to the light, he heard them sizzle and saw them melt. Next, Rabbit scooped up great pawfuls of soft clay from the stream and made many big clay balls. He was a good shot and threw the balls with all of his force at the dancing white light. He heard them strike hard and then his prisoner shouted. Then a strange, quivering voice asked why he had been snared and demanded that he be set free at once, because he was the man in the moon and he must be home before dawn came. His face had been spotted with clay and, when Rabbit went closer, the moon man saw him and threatened to kill him and all of his tribe if he were not released at once. Rabbit was so terrified that he raced back to tell his grandmother about his strange captive. She too was much afraid and told Rabbit to return and release the thief immediately. Rabbit went back, and his voice shook with fear as he told the man in the moon that he would be released if he promised never to rob the snares again. To make doubly sure, Rabbit asked him to promise that he would never return to earth, and the moon man swore that he would never do so. Rabbit could hardly see in the dazzling light, but at last he managed to gnaw through the bowstring with his teeth and the man in the moon soon disappeared in the sky, leaving a bright trail of light behind him. Rabbit had been nearly blinded by the great light and his shoulders were badly scorched. Even today, rabbits blink as though light is too strong for their eyes; their eyelids are pink, and their eyes water if they look at a bright light. Their lips quiver, telling of Rabbit's terror. The man in the moon has never returned to earth. When he lights the world, one can still see the marks of the clay which Rabbit threw on his face. Sometimes he disappears for a few nights, when he is trying to rub the marks of the clay balls from his face. Then the world is dark; but when the man in the moon appears again, one can see that he has never been able to clean the clay marks from his shining face. News stories of the Week:  2-4-17 Forty-foot Peter Rabbit statue built to mark Beatrix Potter’s 150th anniversary burnt down in suspected arson.  A 40ft straw statue of Peter Rabbit built for Beatrix Potter’s 150th anniversary has been burnt down in a suspected arson attack.  The tribute to the children's book character was unveiled last year to mark the anniversary of the Lake District writer's birth.  Police and fire chiefs are investigating the cause of the blaze that destroyed the artwork in a Cheshire field, But now there is nothing left after it caught fire at around 5pm on Thursday, in its field at Snugburys farm in Hurleston, Cheshire.  "It's wonderful to see that Peter Rabbit was loved by so many.  We certainly won't let this stop us!"family that run Snugburys farm.  For more than 10 years, staff at Snugburys have been building sculptures in their field. The giant Peter Rabbit was made last summer.  The farm wrote on its website: "The whole Snugburys family grew up listening to the stories of Peter and his friends.  "They therefore felt it was only fitting to dedicate this year's straw sculpture to Beatrix Potter. In total around 1,000 man hours have been spent creating the feature.  "The inspiration for the blue jacket came from recycling the blue bags our packaging arrives in.  We hand wove a thin layer of blue bags over the straw and we are really pleased with the outcome.  "The whole structure weighs in at an impressive 8 tonnes and Peter stands at 38 ft tall, with his 10 ft carrot and 10 ft ears!".  In a statement, the farm said: "We are very sad to confirm Peter was set alight last night by an arsonist.  "Thank you all - we are truly overwhelmed by all the support from everyone far and wide in the last few hours.  "It's wonderful to see that Peter Rabbit was loved by so many. We certainly won't let this stop us!"  Cheshire Fire and Rescue Service confirmed a crew from Nantwich was sent to the scene. Flt3 is the most recently discovered member of the platelet-derived growth factor receptor (PDGFR) subfamily of receptor tyrosine kinases (RTKs). It is expressed on early hematopoietic progenitor cells and is involved in their growth. Activating mutations of Flt3, such as internal tandem duplications (ITD) in the juxtamembrane (JM) region of Flt3, are among the most common genetic alterations found in patients with acute myeloid leukemia (AML). For this reason Flt3 is an attractive target for specific tyrosine kinase inhibitors, and a number of such compounds are currently being tested in clinical trials. However, the mechanisms of Flt3-mediated signal transduction are poorly understood. Src family tyrosine kinases (SFKs) are involved in the signaling processes of various RTKs, including those of the PDGFR subfamily . Most SFKs are expressed predominantly or exclusively in hematopoietic cells, and at least Hck and Lyn were found to be activated in AML patients. These observations indicate that SFKs could also be involved in Flt3 signaling. In this work the role of SFKs in signal transduction of the Flt3 receptor was investigated. A panel of Hck and Flt3 mutants was used in biochemical and biological assays to deduce the contribution of kinase activity, SH2 and SH3 binding domains and tyrosine phosphorylation status to Flt3-SFK interaction. The work presented shows that SFKs interfere with the maturation of wild type and Flt3 ITD receptors in human embryonic kidney 293 (HEK- 293) cells in a kinase-dependent manner. The SFK-mediated effect on receptor maturation is reflected by accumulation of the immature, intracellular form of Flt3 at the expense of the mature, plasma membrane-inserted form of the receptor. This effect of SFK kinase activity on receptor maturation is not limited to Flt3, because analogous results were also obtained for the Kit receptor, another member of the PDGFR subfamily. The demonstration of the role of SFKs in regulation of receptor maturation is novel and the exact mechanism underlying this effect requires further investigation. Using the HEK-293 cells it was also shown that Hck is able to phosphorylate Flt3 on tyrosine residues and to associate with autophosphorylated Flt3 in an SH2 domain-dependent manner. Hck-mediated tyrosine phosphorylation occurs in the JM region of Flt3. Tyrosine residues 589 and 591 in the JM region, when phosphorylated, were also identified as the docking sites for Hck. Although Hck phosphorylation and binding sites on Flt3 overlap, Hck-mediated phosphorylation of Flt3 is not sufficient for Flt3-Hck association. In contrast to the situation in HEK-293 cells, Hck neither interferes with Flt3 maturation nor phosphorylates Flt3 on tyrosine residues, and does not detectably associate with the Flt3 receptor in the hematopoietic murine cell line 32D clone 3 (32D cl.3). However, the level of the ectopic expression of Hck in 32D cl.3 cells was lower than in HEK-293 cells. This difference in Hck expression level probably accounts for both the lack of the Hck-mediated phosphorylation of Flt3 and the interference with its maturation in 32D cl.3 cells. The lower Hck expression level, however, does not account for the lack of detectable association of Hck with Flt3 in 32D cl.3 cells. Using biological assays in which activation of Flt3 can partially overcome the requirement for cytokine stimulation in 32D cl.3 survival and proliferation, it was shown that Hck is not involved in Flt3 signal transduction leading to Flt3-mediated cell survival and Flt3 ITD-dependent cell growth. Therefore, biological relevance of SFKs in Flt3 signaling remains unclear. In summary, although the binding and phosphorylation of Flt3 by Hck can be shown, the biological relevance of Hck in Flt3 signaling remains to be formally demonstrated. This research led to the novel finding that, at least when over-expressed or hyperactivated, SFKs interfere with the maturation process of RTKs. Various studies have shown that the ligandindependent activation of RTKs as well as their premature phosphorylation can interfere with maturation of RTKs. This study reveals that, by virtue of their phosphorylating of Flt3, Kit and possibly other RTKs, SFKs can regulate maturation of these RTKs and consequently alter their transport to the plasma membrane. Further studies are required to investigate the biological relevance of this function of SFKs.
