On this episode of The Business Mastermind Podcast, Gavin is joined by Robin Waite, the founder of Fearless Business Coaching, to talk about how to avoid under-pricing/under-selling yourself.  How to double business revenue in 3-6 months and why charging by the hour is unethical. KEY TAKEAWAYS Fearless isn't about being reckless. Fearless is about fearing the thing in business ever so slightly less that would ordinarily hold you back. Things like doing a 60 second pitch to a room full of strangers at a network meeting. Some people are terrified of doing that, but if you don't do it no one's ever going to know anything about you, and you won't be able to grow your business and sell stuff. Most business owners will only ever put their prices up once a year or two years, and even then just by 5-10%. They have a belief that if they put their prices up clients will leave. Actually that's not a bad thing. There are two different types of clients: your existing clients who will be anchored to a specific price point and are more price-sensitive, but you've also got prospective clients who haven't been exposed to your prices yet so aren't anchored, there's no tension, you can pitch whatever you want to them and if they feel that's a good value for money they'll go ahead. The money mindset that's been drilled into us by our parents between the age of 3-7 can be quite negative. The education system needs to talk about money and love more. These are two big questions in life: how do I find a partner to be with and how do I make more money? If you have a negative money mindset we seek evidence to back this up in our businesses. If you're always looking back at the past, you're always going to be looking at the blueprint that already exists. In order to change that you've got to look to the future; you can have abundance, you can charge more, you can have cash flowing through your business and you can create the opportunities that you want. This is something you can change in 30 days, maybe less. Too many business owners want to keep control over absolutely everything. But why would you hire someone just to look over their shoulder the whole time, then two people are doing the same job. It's pointless. Perhaps implement SOPs (standard operating procedures) and review them every month or two to make sure they work. I'll take family fit first, skills afterwards. They have to get on with the crew first, we can train them afterwards. In business there are only two things that can go wrong: Either you look a bit stupid, or you lose some money. That's it – unless you do something grossly negligent or nefarious. You can always come back from looking stupid and losing money.   BEST MOMENTS ‘We should try and bring in some of our core values into our business, it's much more authentic.' ‘The core belief you have to have is “I'm worth it”.' ‘Focus on your goals (how much you want to earn) and working backwards (what do you offer) is a good way of working out if you're charging your clients enough.' ‘Record all your sales calls and meetings (with the other person's permission) and listen back to them to see if you were communicating clearly and listening properly. Or do sales roleplay to increase your confidence.' ‘Delegate responsibly, don't delegate responsibility.' Teagasc specialist Catherine Keena joins Catherine Egan on this week's Beef Edge podcast with an update on the new Results Based Environment Agri Pilot Programme also known as REAP. REAP is an agri-environment pilot project that pays farmers to maintain and improve the environmental conditions of their land. Farmers will bring in between 2ha and 10ha of land which will be scored in year one to establish its environmental condition. Farmers will work with their advisers to undertake environmental improvement works on the land that can increase the environmental score in year two. REAP will focus on improving existing farm features rather than the creation of new features and habitats. Farms with higher environmental scores will receive larger payments. The pilot will test the ‘Results-Based' approach and prepare farmers, Advisors and DAFM for the Agri Environment Scheme in the new Rural Development Programme in 2023. There is no guarantee of entry. The deadline to apply is the 10th May and contact your advisor or local Teagasc office to apply. It also drives the direction of successful program design – for both your clients, and yourself. When it comes to program design, it's great to have as many tools in your toolbox as possible. Background: The Grading of Recommendations Assessment, Development and Evaluation (GRADE) approach has been adopted by many national and international organisations as a systematic and transparent framework for evidence-based guideline development. With reference to an ongoing debate in the literature and within public health organisations, this study reviews current experience with the GRADE approach in rating the quality of evidence in the field of public health and identifies challenges encountered. Methods: We conducted semi-structured interviews with individuals/groups that have applied the GRADE approach in the context of systematic reviews or guidelines in the field of public health, as well as with representatives of groups or organisations that actively decided against its use. We initially contacted potential participants by email. Responses were obtained by telephone interview or email, and written interview summaries were validated with participants. We analysed data across individual interviews to distil common themes and challenges. Results: Based on 25 responses, we undertook 18 interviews and obtained 15 in-depth responses relating to specific systematic reviews or guideline projects; a majority of the latter were contributed by groups within the World Health Organization. All respondents that have used the GRADE approach appreciated the systematic and transparent process of assessing the quality of the evidence. However, respondents reported a range of minor and major challenges relating to complexity of public health interventions, choice of outcomes and outcome measures, ability to discriminate between different types of observational studies, use of non-epidemiological evidence, GRADE terminology and the GRADE and guideline development process. Respondents' suggestions to make the approach more applicable to public health interventions included revisiting terminology, offering better guidance on how to apply GRADE to complex interventions and making modifications to the current grading scheme. Conclusions: Our findings suggest that GRADE principles are applicable to public health and well-received but also highlight common challenges. They provide a starting point for exploring options for improvements and, where applicable, testing these across different types of public health interventions. Several public health organisations are currently testing GRADE, and the GRADE Working Group is eager to engage with these groups to find ways to address concerns.

Background: Bacillus subtilis is a very important Gram-positive model organism of high biotechnological relevance, which is widely used as a host for the production of both secreted and cytoplasmic proteins. We developed a novel and efficient expression system, based on the liaI promoter (P-liaI) from B. subtilis, which is under control of the LiaRS antibiotic-inducible two-component system. In the absence of a stimulus, this promoter is kept tightly inactive. Upon induction by cell wall antibiotics, it shows an over 100-fold increase in activity within 10 min. Results: Based on these traits of P-liaI, we developed a novel LiaRS-controlled gene expression system for B. subtilis (the "LIKE" system). Two expression vectors, the integrative pLIKE-int and the replicative pLIKE-rep, were constructed. To enhance the performance of the P-liaI-derived system, site-directed mutagenesis was employed to optimize the ribosome binding site and alter its spacing to the initiation codon used for the translational fusion. The impact of these genetic modifications on protein production yield was measured using GFP as a model protein. Moreover, a number of tailored B. subtilis expression strains containing different markerless chromosomal deletions of the liaIH region were constructed to circumvent undesired protein production, enhance the positive autoregulation of the LiaRS system and thereby increase target gene expression strength from the P-liaI promoter. Conclusions: The LIKE protein expression system is a novel protein expression system, which offers a number of advantages over existing systems. Its major advantages are (i) a tightly switched-off promoter during exponential growth in the absence of a stimulus, (ii) a concentration-dependent activation of P-liaI in the presence of suitable inducers, (iii) a very fast but transient response with a very high dynamic range of over 100-fold (up to 1,000-fold) induction, (iv) a choice from a range of well-defined, commercially available, and affordable inducers and (v) the convenient conversion of LIKE-derived inducible expression strains into strong constitutive protein production factories.

Background: The Minho Integrative Neuroscience Database (MIND)-Ageing project aims to identify predictors of healthy cognitive ageing, including socio-demographic factors. In this exploratory analysis we sought to establish baseline cohorts for longitudinal assessment of age-related changes in cognition. Methods: The population sample (472 individuals) was strictly a convenient one, but similar to the Portuguese population in the age profile. Participants older than 55 years of age were included if they did not present defined disabling pathologies or dementia. A standardized clinical interview was conducted to assess medical history and a battery of neuropsychological tests was administered to characterize global cognition (Mini Mental State Examination), memory and executive functions (Selective Reminding Test; Stroop Color and Word Test; and Block Design subtest of the Wechsler Adult Intelligence Scale). Cross-sectional analysis of the neuropsychological performance with individual characteristics such as age, gender, educational level and setting (retirement home, senior university, day care center or community), allowed the establishment of baseline clusters for subsequent longitudinal studies. Results: Based on different socio-demographic characteristics, four main clusters that group distinctive patterns of cognitive performance were identified. The type of institution where the elders were sampled from, together with the level of formal education, were the major hierarchal factors for individual distribution in the four clusters. Of notice, education seems to delay the cognitive decline that is associated with age in all clusters. Conclusions: Social-inclusion/engagement and education seem to have a protective effect on mental ageing, although this effect may not be effective in the eldest elders.
