Parent-specific epigenetic marks (imprints) leading to parent-specific gene expression are crucial for normal growth and development, yet their mechanisms of establishment and maintenance are not fully understood. In humans, approximately 200 imprinted genes have been discovered, and improper imprinting can manifest in growth restriction, obesity, intellectual disabilities, behavioral abnormalities, and an increased risk of certain cancers. While the use of pluripotent stem cells, especially those in the naïve state, have advanced aspects of modeling early development, a persistent issue hampering bona fide naïve hPSCs is the erosion of imprints. Our guests on today's episode will discuss genomic imprinting, its function, impact, and a new reporter system of imprinted gene expression in hPSCs that enables real-time visualization of loss-of-imprinting at single-cell resolution. Children's genetic disorders and the non-profit Cool Cars for Kids were discussed by Sarah E. Raible, MS, LCGC, a genetic counselor and the clinical director of the Center for Cornelia de Lange Syndrome (CdLS), and rare disease core leader in the Roberts Individualized Medical Genetics Center (IMGC) at Children's Hospital of Philadelphia  and the Chief Operating Officer of Cool Cars for Kids, and Lainey Moseley, an Emmy® award winning journalist working for CBS and NBC News,  founder of CHOPS Syndrome Global; an organization raising awareness for her daughter Leta's rare disease CHOPS Syndrome, and Cool Cars for Kids Board Member.  We talked about how ‘rare' is rare?  One in 11 families are affected by some form of genetic disorder, ranging from clef lip, spina bifida, to heart malformations and other identifiable conditions.  Othe genetic mutations can be so rare that – as in the case of Leta, the exact cause can take decades to discover.  Her disorder was identified by a CHOP test when she was 18, her genetic condition named CHOPS Syndrome in honor of the hospital that discovered it.  Today there are some 40 patients found in Italy, Texas and around the globe, now connected by Lainey's organization.    We spoke about the extra roles the parents of these children must take on, caregiver, advocate and researcher.  Lainey is currently in post-production on her documentary RARE, an essay-style documentary in three acts, each delving into the lives of families grappling with rare diseases, the immense challenges they face, and the remarkable scientific progress they drive in the quest for cures. She has also organized their 2nd CHOPS Syndrome symposium in Philadelphia this summer, July 19th and 20th where Unravel Bioscience will be discussing their results for potential FDA repurposed drugs that may be treatment options for their kids.   Fundraising is vital to these families and to the research that helps identify their conditions.  Cool Cars for Kids was founded by former CHOP geneticist Ian Krantz , Sarah and others to provide help and hope to children with rare genetic diagnoses and their families, and to support research that will identify the best possible treatments and cures.  Their annual fundraiser, the Seventh Annual Philadelphia Concours d'Elegance kicks off with a Preview Gala on Saturday, June 22, followed by the main event on Sunday, June 23 at the Simeone Foundation Automotive Museum.  Tis year's featured model is the Jaguar, with special displays and a panel discussion about the vehicle.  There will again be the Cool Car Corral, an outdoor space where anyone can display their ‘cool' car, classic, rare or just beloved.  Kids activities, music, food trucks and admission to the Simeon racing car collection are part of the day's fun.   Tickets and corral registration available by visiting philadelphiaconcours.com or coolcarsforkids.org.  Walk-ins and drive-ins are welcome that day.

This discovery has conservatively saved 27,000 people from developing a severe form of stomach cancer. Parry is determined to continue to change the course of HDGC stomach cancer patients outcomes, he is invested in the CDH1 mutation families and feels their trials as if they were his own. We discuss genetic counselling, the age at which we test our children for hereditary cancers, his work on lobular breast cancer, bowel cancer and funding research.

Today's Episode Dr. Raj talks with Dr. Rudy Tanzi about his work in Alzheimer's Disease research, his acronym SHIELD for evading its progression, and how his background as a musician has intersected with this work.  Today's Guest Dr. Tanzi is the Vice-Chair of Neurology, Director of the Genetics and Aging Research Unit, Co-Director of the Henry and Allison McCance Center for Brain Health, and Co-Director of the MassGeneral Institute for Neurodegenerative Disease at Massachusetts General Hospital. He also serves as the Joseph P. and Rose F. Kennedy Professor of Neurology at Harvard Medical School. Dr. Tanzi received his BS (microbiology) and BA (history) at the University of Rochester in 1980 and his PhD (neurobiology) at Harvard Medical School in 1990. Dr. Tanzi co-discovered the first Alzheimer's disease (AD) gene, the amyloid precursor protein (APP) gene, and the two other early-onset familial AD genes, presenilin 1 and presenilin 2. As leader of the Cure Alzheimer's Fund Alzheimer's Genome Project, Dr. Tanzi identified several other AD genes, including CD33, the first AD gene shown to regulate neuroinflammation. He also discovered the Wilson's disease gene and other neurological disease genes. Dr. Tanzi's team was the first to use human stem cells to create three-dimensional neural-glial; cell culture organoids of AD, dubbed “Alzheimer's-in-a-Dish”. This model was the first to recapitulate all three key AD pathological hallmarks in vitro, and first to definitively show that amyloid plaques directly cause neurofibrillary tangles - and also made drug screening for AD faster and more cost-effective. Using this system, Dr. Tanzi has developed several novel therapies for AD including gamma secretase modulators targeting amyloid pathology and AMX0035, which was already successful in a clinical trial of ALS and now seeking FDA approval. Dr. Tanzi also discovered that beta-amyloid plays a functional role in the brain as an anti-microbial peptide, supporting a role for infection in AD pathology. Dr. Tanzi serves as Chair of the Cure Alzheimer's Fund Research Leadership Group and numerous advisory and editorial boards. He has been named to TIME magazine's list of TIME100 Most Influential People in the World. Dr. Tanzi is a New York Times bestselling author who has co-authored the books Decoding Darkness, and the bestsellers Super Brain, Super Genes, and The Healing Self with Dr. Deepak Chopra. Dr. Tanzi has hosted several shows on public television, regularly appears on television news programs, and has testified to Congress on both Alzheimer's disease and brain health. I have never wanted a “cure” for my albinism and corresponding visual impairment, but only recently found the words to explain why. Research regarding treatments for and the genetic origins of disabilities raises complex ethical questions. Does studying albinism in a laboratory genuinely improve people's lives, or does it merely satisfy the curiosity of medical professionals? In “I'm Not Broken,” I argue that albinism should be viewed as part of one's identity and a source of community rather than an undesirable mutation. I consider the emotional, social, and political impacts of seeking to reverse or cure blindness. I then argue how redirecting these funds towards developing assistive technology, advocating for legislative changes, and providing necessities (sunscreen, UV protective gear, etc.) to people with albinism would have a stronger impact. In today's episode, we discuss genetic research as it pertains to the African genome. Joining us today to talk about this topic is world-renowned geneticist, Ambroise Wonkam.  Ambroise Wonkam is a Professor of Genetic Medicine and director of the McKusick-Nathans Institute of Genetic Medicine at Johns Hopkins University. Professor Wonkam received medical training from the Faculty of Medicine and Biomedical Sciences at the University of Yaoundé I. He completed a thesis in Medical Sciences from the University of Geneva in Switzerland, and a Ph.D. in Human Genetics from the University of Cape Town in South Africa. He has also received training as a specialist medical geneticist from the University of Switzerland. Professor Wonkam is a well-seasoned researcher, with more than 190 peer-reviewed publications. His research interests hone in on 1) Genomics modifiers of sickle cell disease (SCD); 2) Genetics of hearing loss, and 3) Ethical and educational issues in human genetics in Africa. Over the past 10 years, Prof. Wonkam has successfully led numerous NIH- and Wellcome Trust–funded projects, accounting for about 20 million USD. His research projects span several countries across Africa including Tanzania, Cameroon, Nigeria, Zimbabwe, Zambia, Uganda, Mali, Sudan, Rwanda, and Ghana.Additionally, Professor Wonkam serves as president of the African Society of Human Genetics, the chair of the steering committee of H3 Africa, a board member of the International Federation of Human Genetics Societies, a steering committee member of the Global Genetic Medicine Collaborative (G2MC), and a faculty scholar of the Human Genome Organization (HUGO). In this third pandemic summer, it's difficult to say anything about Covid that hasn't been said before. More than two years into this transformative event, we've pretty much heard it all. But that doesn't mean we've reached a state of peaceful coexistence with the virus, or an acceptance of pandemic life. In spite of all that's unfolded since news of a novel respiratory virus emerged out of Wuhan, China in late 2019, the infection has continued to surprise us and catch us off guard. Even at this late stage, with many effective vaccines and therapies and a near global consensus from authorities that it's time to roll back measures and learn to “live with the virus”, we continue to struggle with new variants, massive numbers of infections and, in spite of the many effective means of prevention and treatment at our disposal, illness and death. While many of you have probably had Covid, or at the very least know someone who has, we are by no means done with this pandemic. I've been hesitant to dedicate an episode to Covid and uncertain of the point of expending yet more energy on what is certainly the most talked about subject of the last two years. But faced with the virus's persistent and ever-changing impact on our lives, and having so many unanswered and nagging questions about our response to it, I decided it needed to be done. I chose Donald Vinh, an infectious disease clinician and researcher, as my guest for this conversation. Don is an experienced medical communicator, and a measured, rigorous voice on Covid. I came to know of him because of his vocal and unflinching but always factual assessments of our local response to Covid here in Quebec. He's also actively engaged in research on Covid, and contributed to an international study on severe Covid that was published in the prestigious journal Science in 2020 and was named one of 2020's 10 remarkable scientific discoveries by the equally prestigious journal Nature. So, he seemed like the ideal person to have on for a deep dive on Covid. Donald Vinh is an Infectious Disease specialist and Medical Microbiologist at the McGill University Health Centre. He is Director of the Centre of Excellence for Genetic Research in Infection and Immunity, and Fonds de recherche du Québec Santé Senior Scholar with a translational research program on human immunodeficiencies and genetic susceptibility to infectious diseases at the Research Institute of the McGill University Health Centre. His frequent media appearances have earned him a reputation as a trusted voice on infectious diseases, such as Covid-19, monkeypox, and the rare diseases he studies. Rachel Chaikof was one week old when her mother, Melissa, suspected that something was wrong: she didn't startle at the sound of a slamming car door. Rachel was diagnosed with Usher Syndrome, a genetic condition which causes either inherent or progressive deafness.  But being deaf was not an absolute: as a child, Rachel became one of the first children to receive a Cochlear Implant by way of an FDA trial. Because of medical innovation, Rachel could now experience the world for how it was meant to be experienced.  But later, Rachel also began to lose her sight. At the age of 19, Rachel was diagnosed with Usher Type 1, more specifically categorized by progressive blindness in addition to loss of hearing. This time, there was no medical innovation currently available, and Melissa, an experienced advocate in the network of nonprofits dedicated to hearing loss, devoted herself to a new goal: finding a cure for Usher's Type 1.  The Chaikof's have seen all that medical innovation is capable of. That's why Melissa and her husband started the Usher 1F Collaborative, a nonprofit devoted to funding such research. And while their daughter remains optimistic about all that she is capable of, her loss of sight has compromised what she loves most about the world: seeing its beauty in color and having the freedom to navigate on her own.  While Rachel is thankful for all of the modern innovations that have allowed people with disabilities to navigate society, she also believes that sometimes, that can distract from the commitment to finding a cure for genetic conditions like Usher Syndrome. Dr. Di Forte is world renowned expert in cannabis induced psychosis research and risk factors for schizophrenia. Dr Marta Di Forti is a Clinical Reader in Psychosis Research at the Dept of Social, Developmental and Genetic Research, Institute of Psychiatry, and Honorary Consultant Adult Psychiatrist, Lambeth EI Community team, South London and Maudsley NHS foundation Trust. She leads the first Cannabis Clinic for patients with Psychotic disorders in UK. She was recently awarded the Royal College of Psychiatrist Researcher of the year prize.  In 2020 she was granted a MRC Senior Research Fellowship to expand her research in the role of cannabis use in psychosis and its underlying biology. With her team she showed for the first time that use of high potency types of cannabis e.g. "skunk" carries a higher risk of psychosis than use of traditional types and that it affects rates of Psychotic disorders across Europe. Though it still remains unclear who are those cannabis use most at risk. Her future work aims to investigate the interaction between cannabis use and genes predisposing to schizophrenia, and how cannabis changes the epigenome. ​Dr. Adriana Humanes is a coral reef scientist from Venezuela. She completed her Ph.D. in Australia, and now is pursuing a postdoc with the @Coralassist_Lab through Newcastle University in the UK….I met her on the ship to Antarctica in 2018.For the last four years Adriana, and her colleagues have conducted field work in Palau, which is an archipelago of more than 300 islands situated in the Pacific Ocean between Indonesia and the Philippines.As Adriana explains, coral reefs are facing and unprecedented decline due to marine heatwaves, and also because of mass coral bleaching events. In the last decade, there have been five large scale coral bleaching and mortality events which have affected hundreds of reefs around the world.During the last massive bleaching event, on the Great Barrier Reef of Australia, almost half of the coral cover lost. Over the last 30 years, worldwide, coral cover has decreased by an alarming 20 per cent.The research at the Coralassist lab (as well as Adriana's) focuses on testing the feasibility of assisting evolution by identifying coral colonies that have a high tolerance to heat stress, and then, sexually breeding them. The goal is to create next generation corals that have inherited heat tolerance from their parents.Adriana first went to Palau in 2018, which was not long after I met her, in Antarctica. Although the pandemic has restricted her ability to travel there to continue her onw field research, by coincidence, one lone Ph.D student working with the  Coralassist lab  flew to Palau on March 7, 2020 … which turned out to be a long stay for her, 18 months, to be exact, where she became the lone student there tasked with keeping the Lab research project going.Adriana and her colleagues hope to returned this year, for their final leg of field research...after being away for years. And good news! They found corals spawning! Hopefully this is one more step towards the goal to use these sexually produced corals to help increase their future resilience, and hopefully even restore some vreefs that have been damaged or degraded from heat waves.
