In this episode of the Epigenetics Podcast, we talked with Christa Bruecker from the Max Perutz Laboratories in Vienna about her work on transcriptional regulation during early embryonic development. Christa Buecker unravels the differences between naive and primed pluripotency states, showcasing how OCT4 relocalization and enhancer chromatin landscapes play pivotal roles during this transition. The conversation delves into the intricate interplay of transcription factors like OCT4 and OTX2, shedding light on their collaborative efforts in regulating gene expression during differentiation. Dr. Bücker then shares insights from her study on enhancer elements controlling FGF5 expression and discusses the surprising revelation that individual enhancers show no intrinsic activity but work together in a super additive fashion. She also touches upon her research on IRF1's connection to the gene regulatory network and its role in protecting cells against viral infections. The conversation shifts to Dr. Bücker's current research endeavors, focusing on exploring the strength of enhancers and their impact on gene expression at different distances from promoters. She shares her vision for future experiments and the integration of enhancers to decipher their impact on transcription regulation.   References Buecker, C., Srinivasan, R., Wu, Z., Calo, E., Acampora, D., Faial, T., Simeone, A., Tan, M., Swigut, T., & Wysocka, J. On August 9th, 2021 they announced Pooja Hegde as heroine along with technical crew AS Prakash art, Navin Nooli editing, and Madhie as DOP.  Originally supposed to start shooting in November 2021 but due to SVP shooting delays they never started. Then Madhie was replaced with PS Vinod and on Feb 3rd, 2022 muhurtam pooja happened at Ramanaidu Studios.  But filming only started on  September 12th, 2022 with what they called a high octane action entertainer that was choreographed by Anbariv master. What I heard was the original story was revenge drama action thriller in which Tabu plays Mahesh's mom, he is fatherless, and Anil Kapoor is a huge Mumbai based gangster who Mahesh wants to take down. They filmed a 6 minute heavy fight with the footage Babu in full form. 2nd schedule was supposed to start on Dussera 2022 with Mahesh and Pooja Hegde but it never happened. Naga Vamsi removed Anbariv masters from the project  And then they scrapped the original action entertainer script and converted it into a commercial family entertainer with action. Full confusion started on the project and everybody wondered why they scrapped it to begin with. Then in Butta Bomma promotions in early 2023, Naga Vamsi said Pooja Hegde and Sreeleela are the heroine August 11th release or so we thought  New script and movie finally started filming on Jan 18th, 2023 in Saradhi Studios glimpse fight was filmed by PS Vinod and now Ram lakshman as fight masters.  Then as with any Trivikram movie, the huge house set you see in the movie was built by AS Prakash and they built it in Shankarapalli Telangana for a cost of 10 crores so they claimed. 2nd schedule started in Feb and ended in April. March 26th, 2023 first look poster released with Sankranthi release date on it Babu smoking beedi.  And then all the problems started. Filming stalled, people said the output wasnt good and they were gonna do reshoots. The only content they had were first look poster of Babu smoking beedi with chillies and then first fight so they released that in the glimpse on May 31st. Then filming never continued due to delays after delays.  Nobody knew what was happening. Pooja Hegde left and then Sreeleela took her role and then Meenakshi Chaudhury was cast in Sreeleela 2nd heroine role. Then due to more delays PS Vinod left the project and no DOP. Became like a big boss house set with elimination every week. Talk was also that Thaman was going to get replaced with GV Prakash Kumar but Naga Vamsi said no Thaman is our music director and they kept him. Filming continued on June 24th, 2023 but then a month later July 22nd, PS Vinod walked out of the movie.  Then Manoj Paramahamsa replaced PS Vinod and then filming continued. Somehow they were able to finish the entire film and shooting finished on December 28th, 2023. Movie did not release any actual content about the movie besides the glimpse in may and finally trailer came on Jan 7th, 2023.  First song Dum Masala on Nov 7th and then Oh my Baby on Dec 13th and then Kurchi Madhapetti on Dec 30th, 2023. Only Dum Masala got a good response but nowhere near AVPL range songs. So very disappointing stuff from Thaman. Negativity mainly from disappointing songs.

For Hi Nanna thats apt for the most part.  The story seems like a rom com between 3 characters but much like Jersey it actually does genre shifts and its purely a drama movie. The writing is actually very good for all the emotional scenes and director has written some good threads.  Good part first is that the movie is all about relationships and how it affects others. Nani daughter one, Nani Mrunal one, Nani mother in law, Mother and Jayaram and how it affects Mrunal, all of these bring conflict into he story. Also everybody wants something  Mahi wants to know who her mom is, Yashna wants just wants to be happy, Viraj wants to be free from his guilt of what happened to his wife, Jayaram doesnt want to fail as a parent.  The writing is very strong especially in emotional scenes. The hospital scene, pre climax, and climax are so strong that you will get tears from your eyes. If you got cry then you eyes definitely will get wet for sure. Its that strong and relatable. Through Yashna's character, director has created a lot of relatability and actually her character is the best.  I felt this is the movie Kushi wanted to be in terms of husband wife conflict but failed miserably at. The brilliance of Hi Nanna actually is that the whole flashback sequence its not convincing their love story its just there and I'll come back to that but their conflict after marriage is very convincing so you relate to their marriage and love story during that part,. Due to strong emotions, things work.  I felt the most emotional part is that Viraj is such a dedicated father that he attended every ceremony based on cystic fibrosis and lives his daughter so much hes willing to any extent and fight withe everybody to make sure that she lives. That part hit hard.  Also loved the thread between Pluto and Yashna. She doesn't like dogs but then the dog gave her confirm, she likes dogs and then that day Pluto still remembered her after all these years despite her not remembering him.  Interval twist was expected but accident scene was heartbreaking man. So crazy. Also liked the two mass warning scenes basically when Yashna's mom tries to insult Viraj in the hotel and then at wedding party in 2nd half he also gives her warning. There was no fight.  Also the symbolism of doors closing. Idk if you guys noticed. Every time Mahi asks Nani to tell her about her mom, Viraj always shuts the door implying that chapter is closed and he doesnt want to talk about it. But Mahi does. And then in operation room, the door starts to close and last thing Mahi sees is her mom and dad outside the ER ward showing that this chapter has finally been opened and now its closed. She knows the truth.Very cool.  2nd half has a trope of bakra track and initially I thought why is Angad Bedi there. Greatest bakra ever asalu the level of class and respect was too good. We got whistles in our theater when he came in climax to do the operations.  Interestingly, the movie keeps going just based on emotion. Theres no traditional commercial elements or entertainment. I wish there was more because movie has inconsistent pacing. Towards the interval and then post interval movie really felt flat and slow but then last 30 mins is outstanding with emotion that you leave the theater with a feel good feeling. The two scenes are when they hide in the cabinet and Nai runs away and then their fight after they drunk in Odiyamma song, Wish more scenes were there like that.

Therefore he's supposed to have animalistic qualities about human nature. It's hard to explain this stuff without using English words so please forgive me this time.  Writing is actually the biggest villain of the movie not Bobby Deol I will explain why. Given the core point and movie was promoted as such, you would think its a family drama with strained relationship between father and son. Movie is not about that. It's about a man's descent into madness because of father's neglect and he never gave him the love and affection that deserved so he is like this. BUt it's not convincing at all. Thats the biggest problem. There's no reason/nothing shown for us to connect as a father son relationship or for us to care. I will take example of a movie that Im not a big fan of but in Ala Vaikuntapuram Lo he clearly shows Murali Sharma has made his life a living hell since he was young with a grudge on Jayaram so we feel for Allu Arjun's character and relate to him and buy that father son relationship.  Here Vanga doesnt show anything. He tells you but doesn't show it much. Yet this guy still loves his dad. The opportunity to make a commercial film is there but it didnt work for me.  Family drama part is nonexistent, romance part is comical she falls for one dialogue about alpha males and then next scene cancels her engagement and then Hua Main song, and then action part about grandfather wealth wars. But he villains have no proper motives etc. nothing is properly established or explained about the family heir wars to begin with. Another example a lot pf people didn't like KGF2 but its same like animal yenti ante, whatever Rocky Bhai does since you connect with him, it seems believable. Because I didnt connect here, you dont connect with what he's doing so Vanga's paithyam is in full affect.  To positives, Hua Main to interval is next level movie. Masterpiece level if you ask. Best sequence in the movie is revealing the suspense of Varun and that flashback. Too good. That entire stretch was so entertaining and amazing. Also the body double part was so ridiculous but so entertaining. But then rest of the movie never reached that level again. And then the post credit scene didnt really work. Remembered oh Zoya said that right but it felt forced and then im not sure theres even scope to make a sequel to be honest. But thats just me.   Direction by Sandeep Reddy Vanga is decent. Hero characterization throughout the movie is superb and 1st half taking is very good for that stretch but rest of the movie doesnt work or feel convincing. Due to the lack of conviction you feel a disconnect and as said before 2nd half was like his piathyam whatever he wanted to show was more for triggering audiences and critics rather than telling a emotional and relatable story with an interesting hero characterization. That being said whether you disagree or agree with a film or whatever the content is, Sandeep Reddy has created a brand for himself making unique movies which star controversy and open for a lot of discourse. This is for sure. So while I might agree or disagree with what he showed, its the audience responsibility to take it however they do. That is for sure.

This problem has been re-current and exacerbated for the duration of the Covid-19 pandemic. Across the pond, much attention focused on the trial of child murdered White Woman, Lucy Letby. One important component of this scandal is that her non-white colleague, Dr. Ravi Jayaram, allegedly reported concerns about Letby well in advance. Mani Ratnam's Ponniyin Selvan Part 1 released a little over a week ago and has taken the box office by storm. A critical and commercial success, the film has attracted a wide demographic to the theatres. Largely due to its source - Kalki R. Krishnamurthy's serialized novel Ponniyin Selvan published in the 1950s, a historical fiction based on the Cholas and the conflicts within and outside the kingdom for the throne. The film, like most Mani Ratnam films, is also an example of technical mastery which is one of the attractions for widespread theatre viewing with almost no one choosing to wait for the streaming release like it is the norm of late. The film is star studded - Karthi, Aishwarya Rai, Vikram, Trisha, Jayam Ravi, Jayaram, Sobhita Dhulipala, Aishwarya Lekshmi, Sarath Kumar, Parthiban, Prabhu, Lal, Prakash Raj and more. There are stars behind the camera as well apart from the director and writing team (Mani Ratnam, Jeyamohan and Elango Kumaravel). It is edited by Sreekar Prasad, art direction by Thotta Tharani with cinematography by Ravi Varman. Music is by A.R Rahman and sound design by Anand Krishnamoorthi. Foley Artist is Prathap and re-recording mix by Craig Mann. In this episode, we talk about one aspect of the technical mastery - sound design. Sound Designer/Engineer/Editor Anand Krishnamoorthi joins Aditya and Ashoka to talk about the sound of Ponniyin Selvan Part 1 and the nuances in the art of sound design. We talk about sound design in general in cinema as well as the work that has gone into Ponniyin Selvan. Anand gives both eli5 explanations for some aspects of his work and also some nerdy and deeply technical things that go into a film's sound. Listen on. Welcome to Khandaan: A Bollywood Podcast where we discuss two of this week's mega releases - PONNIYIN SELVAN - 1 and VIKRAM VEDHA. But first, our musical correspondent Asim went to the Pasoori concert (04m20s) in London and has many thoughts about the production. Next, we saw the brand new teaser for ADIPURUSH (11m40s)starring Prabhas, Kriti Sanon, and Saif Ali Khan as Ram, Sita, and Raavan respectively and… it's quite something. Sujoy and Amrita both watched Mani Ratnam's Ponniyin Selvan (18m25s) - I starring Aishwarya Rai, Vikram, Karthi, Trisha, Jeyam Ravi, Prakash Raj, Jayaram and many others, and had slightly divergent thoughts while agreeing that it is a cinematic achievement. For those who're unfamiliar with the source material, we've worked out a strategy that you should first read the plot wiki before watching the film, just to keep up with all the characters. And lastly, all three of us watched Vikram Vedha(45m30s) starring Hrithik Roshan, Saif Ali Khan, and Radhika Apte. This (very loud) remake of the Tamil movie by its original directors is bound to work better for those who have not seen the version with Vijay Sethupathi and Madhavan. As a bonus, you can hear Asim interview the director duo Gayatri and Pushkar (1h16m20s) and Radhika Apte (1h28m10s)at the end of this episode. Welcome to another episode of OTTplay Lost and Found, one podcast that talks about critically acclaimed films and shows that are lesser-known. I'm your host NikhilSeveral hit Malayalam films from the 90s and 2000s have come under scrutiny in recent years for their regressive themes on numerous socio-political themes. However, one film stood out in terms of being far ahead of its time, the 1991 satire Sandesham. The movie stars Jayaram as Prakashan and Sreenivasan as Prabhakaran in lead roles as brothers, and the plot revolves around the two who belong to opposite ends of the political spectrum. Sreenivasan's character, an ardent follower of a fictionalized version of the Communist Party, is at odds with his brother, who belongs to the fictionalized version of the Congress Party. Written by Sreenivasan himself, and directed by Sathyan Anthikad, the film uses humour as a plot device and as a narrative tool to deliver one of the most thought-provoking films in the Malayalam film industry. It critiques the partisan following of political parties and the lack of objective discourse regarding politics in society - something which is still relevant to contemporary politics.Thilakan, Siddique, Kaviyoor Ponnama, Oduvil Unnikrishnan, Innocent, Mathu, Mammukoya, Sankaradi, and others play excellent supporting roles. The film has aged like fine wine and is widely regarded as one of the best films in Malayalam. If you haven't watched this hidden gem yet then do so it's streaming on HotstarWell, that's the OTTplay Lost and Found film for today's podcast, until the next episode it's your host Nikhil signing out.Aaj kya dekhoge OTTplay se poocho

Listen to the first part here - A Comprehensive Overview of the History of Malayalam Cinema - Part 1 The 1970s witnessed a new wave of Malayalam cinema where the bridging of parallel and commercial cinema was at its best. It was also the time when political and action movies gained a following. Adoor Gopalakrishnan's directorial debut, Swayamvaram in 1972 is known for pioneering the new wave of cinema movement in the industry. Chattakari (1974) and Avalude Ravukal (1978) were some of the boldest Malayalam movies of that time with the latter being the first A certified movie. The movies were based on the themes of teenage pregnancy and prostitution, respectively, which were topics that were profoundly frowned upon during those times. The latter half of the 1970s witnessed the emergence of the legendary actor Jayan. He reformed the industry with his macho image and action sequences never seen before. Like who can forget the “We are not beggars…” dialogue from the movie Angadi (1980). It still gives me goosebumps. Notable movies from the 1970s Swayamvaram (1972) Arakkallan Mukkalkkallan (1974) Chattakari (1974) Kodiyettam (1977) Avalude Raavukam (1978) Thacholi Ambu (1978) The 1980s was the decade of combining common man's problems with comedy. The rise of geniuses like Sreenivasan spearheaded this genre of movies to the forefront with movies such as Odaruthammava Aalariyam (1984), Sanmanassullavarkku Samadhanam (1986), Nadodikkattu (1987), and Varavelpu (1989). 1981 was a milestone year for Malayalam cinema with the release of Elippathayam and Manjil Virinja Pookkal. The former being screened at the London Film Festival and winning the Sunderland Trophy and the latter propelling Mohanlal Viswanathan, our very own Lalettan, to stardom. Though it is almost impossible to imagine that he started his career predominantly playing antagonistic characters. M T Vasudevan Nair made his presence known with his screenplay for movies like Nakhakshathangal (1986), that helped Monisha win the National Award for Best Actress at the age of 15, and Oru Vadakkan Veeragatha (1988), an epic historical drama which showed Chandu Chekavar as a misunderstood hero. Director P Padmarajan also made his mark with the 1987 romantic drama, Thoovanathumbikal, and the 1988 mystery thriller, Aparan. The decade also delivered Malayalam's own investigative thriller detective series with Oru CBI Diary Kurippu (1988), the first movie from the four-part series, with Mammootty playing the role of Sethurama Iyer. The 80s observed many firsts with Padayottam (1982), a film based on the novel The Count of Monte Cristo, becoming the first Malayalam film to be shot in 70 mm, My Dear Kuttichathan (1984), the first Indian 3D film, and Adipapam (1988) gaining the crown of being the first Malayalam softcore film. Notable movies from the 1980s Angadi (1980) Manjil Virinja Pookkal (1981) Elippathayam (1981) Padayottam (1982) Odaruthammava Aalariyam (1984) My Dear Kuttichathan (1984) Nakhakshathangal (1986) Sanmanassullavarkku Samadhanam (1986) Nadodikkattu (1987) Thaniyavartanam (1987) Aparan (1988) Oru CBI Diary Kurippu (1988) Varavelpu (1989) Oru Vadakkan Veeragatha (1989) The 1990s had a great start with movies such as the comedy-thriller In Harihar Nagar (1990) and comedy-drama Godfather (1991) running for over a year in theatres gaining cult status. It also saw a few larger than life stars take on a majority of screentime. With Mohanlal and Mammooty taking lead roles in almost all genres, Suresh Gopi shining in the action movie landscape, and Jayaram starring in your next-door neighbour, family-friendly characters. Siddique-Lal's Vietnam Colony was released in 1992 and it is noteworthy for having plot similarities with the 2009 movie Avatar — the second highest-grossing movie in the world. What does James Cameron have to say about that, I wonder? The time also witnessed movies like Manichitrathazhu (1993) where the female
