B., Campos, J. C., Ferreira, J. C., Haileselassie, B., Mochly-Rosen, D. Abstract: Cardiac troponin I (cTnI) is a sarcomeric protein critical to myocyte contraction. Unexpectedly, we found that some cTnI localized to the mitochondrial matrix in the heart, inhibited mitochondrial functions when stably expressed in non-cardiac cells and increased opening of the mitochondrial permeability transition pore under oxidative stress. Direct, specific, and saturable binding of cTnI to ATP synthase was demonstrated in vitro, using immune-captured ATP synthase, and in cells using proximity ligation assay. cTnI binding doubled F1F0 ATPase activity, whereas skeletal troponin I and several human mutant cTnI variants associated with familial hypertrophic cardiomyopathy did not. A rationally-designed ten amino acid peptide, P888, inhibited cTnI binding to ATP synthase and cTnI-induced increase in ATPase activity in vitro and reduced cardiac injury following transient ischemia in vivo. We therefore suggest that mitochondria-associated cTnI may inhibit cardiac ATP synthase under basal conditions; pharmacological agents that release this inactivating effect of cTnI and thus preventing ATP hydrolysis during cardiac ischemia may increase the reservoir of functional mitochondria to reduce cardiac injury. Johan has a M.Sc. in Landscape Planning from the Swedish University of Agricultural Sciences (2008), a PhD in Landscape Planning also from the Swedish University of Agricultural Sciences (2013) and has been working as a researcher and teacher since 2014. Apart from his academic career he is also and ISA Certified Arborist and active as a consultant, where he is running a consultant networking comprising of consultants from a different profession, working together to create management plans for larger tree populations in Sweden.

Alex Ostberg is currently entering his 5th year at Stanford University, where he has been an All-American in both cross country and track. This podcast is rooted in a presentation Alex did at the 2019 Boulder Running Camps titled, "The Ten Things I Wish I Knew As A High School Runner." Alex is passionate about both the sport and helping high school athletes improve, which makes this podcast is a must-listen for high school coaches and athletes. Alex has been running competitively for 11 years and has developed a passion for understanding the physiological, emotional, and psychological demands of running. He competed for 2 years in middle school with the Wilton Running Club. After that, Alex ran for Darien High school where he set 6 state records. He was a two-time qualifier at Footlocker Nationals, and the Northeast Footlocker Regional Champion in 2014. These performances earned him the Connecticut Gatorade Player of the Year twice. In track, Alex was the Penn Relays 3000m champion in 2015 and the 2014 New Balance Outdoor Nationals champion. He finished his high school career with PRs of 4:06 (1600), 8:43 (3200), and 14:16 (5K). Alex was recruited to Stanford University where he has competed for 4 years and is entering his fifth year. He has steadily improved on his performances with four NCAA All-American certificates. He has excelled on the cross country circuit, placing 16th and 13th in the past two NCAA XC Championships. On the track his PRs currently stand at 3:59 (mile), 7:51 (3K), and 13:42 5K. In 2018, Alex was elected by his peers to serve as captain of the Stanford Track Team. In addition to his training, Alex has developed a passion for learning about the science behind training and peak performance. He is pursuing a bachelors degree in Human Biology with an emphasis on Exercise Physiology and will also be getting a minor in Psychology. He has had the opportunity to be a TA for 4 different Stanford classes, including Exercise Physiology, Exercise and Aging, Clinical Anatomy, and EMT class. One day, when his competitive aspirations are over, Alex aspires to be a coach himself.

MINI John Cooper Works WRC La tappa italiana del Mondiale Rally si è svolta sugli impegnativi e tormentati tracciati sterrati della Sardegna. Piloti, vetture e meccanici sono stati messi a dura prova dai salti, i guadi e le asperità che hanno fatto di questa gara una delle più spettacolari ed impegnative del Mondiale. MINI John Cooper Works WRC Team ha scelto proprio questo scenario da brividi, per iniziare la sua avventura nel WRC. Ecco come è andata: MINI John Cooper Works WRC Team porta a termine la sua prima prova nel Mondiale Rally con l'ottimo sesto posto assoluto ottenuto da Dani Sordo in coppia con il navigatore Carlos Del Barrio, che hanno concluso i 18 stage a 3’27”6 dal leader Sebastian Loeb. Solo la sfortuna che ha costretto Kris Meeke al ritiro per un guasto meccanico ha impedito di piazzare anche la seconda vettura a ridosso dei migliori. Il rientro di MINI nel Campionato Mondiale Rally è stato salutato da grandi manifestazioni di entusiasmo dal pubblico che ha fin dalla prima speciale dimostrato grande interesse per la vettura anglo tedesca. La prova si è aperta il 6 maggio con la speciale di Lago Omodeo al termine della quale sia Kris Meeke che Dani Sordo hanno espresso la propria soddisfazione per le prestazioni della vettura che si è dimostrata subito all'altezza della situazione sugli insidiosi sterrati sardi. La seconda prova speciale Monte Grighini Nord che ha registrato una grande affluenza di spettatori, ha messo a dura prova le vetture a causa dei molti salti e del terreno scivoloso. In queste condizioni estreme Dani Sordo ha mantenuto il suo ritmo costante interpretando al meglio la prova. Kris Meeke, subito velocissimo ha chiuso al quarto posto pur accusando una forte toccata. Nello stage 3 Alta Marmilla Dani sordo è apparso molto soddisfatto della vettura, mentre Kris Meeke è stato costretto al ritiro per un'uscita di strada che ha danneggiato la ruota anteriore sinistra. Nella quarta speciale Monte Grighini Sud Dani Sordo e la sua MINI numero 37 hanno proseguito con grande regolarità. Al termine della prova il pilota iberico si è dimostrato sempre più entusiasta della MINI WRC che ha dimostrato grande affidabilità e buone doti di velocità e trazione. Per la quinta speciale le vetture si ripresentano a Monte Grighini Nord. Tutto procede per il meglio e Dani Sordo sente la sua MINI progredire continuamente raccogliendo importanti informazioni per lo sviluppo. Le speciali 6 e 7 confermano le buone prestazioni delle precedenti tanto da indurre Dani Sordo ad osare un po' di più senza tuttavia dimenticare la giusta cautela per cercare di preservare meccanica e pneumatici per portare a termine la prova. Al termine della settima prova Dani conferma di sentirsi al volante di una vettura in ottime condizioni. La giornata inaugurale si chiude con la speciale Monte Grighini Sud. L'ottimo feeling con la vettura inducono Dani Sordo a forzare ulteriormente l'andatura ricevendo ottimi riscontri in termini di performance ed affidabilità della sua MINI WRC. La seconda giornata del Rally d’Italia Sardegna si apre sul tracciato asciutto e compatto di Coiluna. Le due MINI appaiono subito pimpanti. Dani si dichiara molto soddisfatto della vettura e della messa a punto effettuate il giorno precedente. Kris Meeke accusa invece da subito problemi all'acceleratore e non riesce a spingere come vorrebbe. Il secondo stage della giornata si svolge sullo spettacolare tracciato di Monte Lerno, celebre per il suo “jump” tra i più alti di tutto il Mondiale. Sordo accusa un leggera perdita di grip rispetto alla prova precedente, ma dimostra ancora una volta un passo veloce e molto regolare. Kris Meeke purtroppo vede svanire le sue aspirazioni dovendosi ritirandosi per problemi meccanici. La gara di Dani Sordo continua senza inconvenienti dimostrando l'ottimo lavoro dei tecnici del MINI John Cooper Works WRC Team nella messa a punto della vettura. La speciale 12 vede la MINI di Dani Sordo accusare qualche problema ai freni, senza impedire al pilota spagnolo di mantenere il suo passo inarrestabile. Anche nello stage 13 la vettura di Dani Sordo registra un piccolo problema all'acceleratore che però non pregiudica la sua prestazione. Nella prova successiva i problemi vengono risolti, ma Dani Sordo, dimostrando una grande intelligenza strategica, evita di spingere al massimo. Risolti i piccoli inconvenienti all'acceleratore, Dani riprende a tirare nella prova 15. La possibilità di chiudere la prova con un ottimo piazzamento per una vettura esordiente si fa sempre più concreta. L’ultima giornata si apre con la speciale Monte Olia. I guadi presenti sul tracciato mettono in evidenza qualche problema di regolazione sul differenziale, e quindi di trazione sul bagnato. Un'ulteriore esperienza che servirà per rendere sempre più competitiva MINI WRC ormai entrata nei cuori del pubblico. L'ultimo stage è anch'esso caratterizzato da un fondo molto scivoloso, ma Dani Sordo riesce comunque a conservare il suo piazzamento presentandosi 6° alla partenza del Power Stage finale. L'obiettivo di portare a termine la prova macinando il maggior numero di chilometri è stato raggiunto ed il 6° posto finale è un risultato che regala grande soddisfazione a tutto il MINI John Cooper Works WRC Team. L'esperienza sui durissimi sterrati sardi servirà per continuare la messa a punto di una vettura che ha dimostrato fin dal suo esordio un grande potenziale. Evidentemente buon sangue non mente e a distanza di 42 anni dai trionfi al Rally di Monte Carlo, MINI è pronta a continuare la tradizione.
