Open Philanthropy's Global Aid Policy (GAP) team, which is devoted to increasing government aid and guiding it toward more cost-effective approaches, believes there may be opportunities to increase the impact of this aid through targeted advocacy efforts. They estimate that in Western nations like the UK, for every $1,000 spent on ODA, aid advocacy funders spend around $2.60 attempting to support and inform its allocation. Meanwhile, in Japan, advocacy spending is a mere $0.25 for the same amount - more than 10 times less. Accordingly, the GAP program has prioritized work in Japan. The following case study highlights one grantee helping to drive this work forward. ***** One day in March 2023, in the district of Wayanad near India's southern tip, hundreds of villagers lined up for an uncommon service from an unexpected source: a check-up on their lung health, courtesy of Fujifilm. The Japanese company, best known for its cameras, was taking a different kind of picture. Its portable, 3.5 kg battery-powered X-ray machine, designed to deliver hospital-grade diagnostics, enables tuberculosis screenings in regions where medical facilities usually lack the necessary technology. This scene was just one stop on an illuminating trip to India for a group of Japanese journalists and youth activists. From Toyota Tsusho's Sakra World Hospital to Eisai's efforts to combat neglected tropical diseases (NTDs) in Yarada village, each site visit highlighted Japanese businesses and researchers contributing to global health initiatives. Recognizing this opportunity, Open Philanthropy supported Platinum, a Tokyo-based PR firm, in organizing a trip across India aimed at boosting the Japanese public's awareness of urgent global health issues, particularly tuberculosis and neglected tropical diseases (NTDs). Sixteen people attended: six journalists, representing outlets ranging from a long-running daily newspaper to a popular economics broadcast, and 10 youth activists sourced from PoliPoli's Reach Out Project, an Open Philanthropy-funded initiative that incubates charities focused on global health advocacy. Our Senior Program Officer for Global Aid Policy, Norma Altshuler, thought the initiative was timely given recent trends in Japan's ODA spending. Between 2019 and 2022, the share of Japanese ODA allocated to global health doubled (or tripled, including COVID-19 relief). To sustain this momentum, Open Philanthropy is supporting Japanese groups that aim to preserve or grow Japan's commitment to prioritizing global health initiatives. In a post-trip interview with Open Philanthropy, Soichi Murayama, who helped organize the trip, says one challenge of Japan's media landscape "is that Japanese media doesn't cover global health very often." Murayama attributes the dearth of dedicated coverage to limited reader interest, creating a feedback loop where minimal reporting leads to low awareness, which in turn reduces appetite for such stories. Ryota Todoroki, a medical student who participated in the trip, echoes this sentiment: "NTDs are often seen as a foreign issue with no relevance to Japan, so changing this perception is a major challenge." The Fujifilm initiative in Wayanad provides an example of how connecting Japanese companies to global health efforts can help illustrate the impact of foreign aid. En entrevista para MVS Noticias con Juan Manuel Jiménez en ausencia de Manuel López San Martín, El Dr. Ciro Murayama, ex consejero del INE, habló sobre el riesgo de la sobrerrepresentación en el Congreso. Murayama comenzó señalando una "distorsión profunda de la voluntad popular" en el actual contexto político. “La ciudadanía le dio el 54.7 por ciento de los votos a Morena en la Cámara de Diputados y quieren casi 20 por ciento más de diferencia de lo que les dieron", afirmó Murayama, criticando la intención del partido oficial de aumentar su representación más allá de los votos obtenidos. ¿Qué implica la sobrerrepresentación en el Congreso de Morena? El ex consejero del INE alertó sobre las implicaciones de esta sobrerrepresentación, indicando que podría llevar a una consolidación autoritaria. “Quieren comprimir a la oposición, es inflar el peso del voto a favor de Morena y desinflar otras opciones, todo tiene un límite y hay que proteger el voto”. En particular, expresó su preocupación por cómo esta mayoría calificada podría utilizarse para influir en el proceso de selección de jueces, un poder que, según él, debería estar más balanceado. “Lo grave es que esta mayoría calificada se pretendería usar para lo que se nos dice, será para que jueces sean electos, sean votados por la corriente mayoritaria”. El ex consejero del INE también subrayó que cualquier intento de imponer una sobrerrepresentación iría en contra de los principios constitucionales y la decisión del electorado. “Lo que amerita es lo que la gente decidió lo que la Constitución establece, coalición y partido se entiende que son sinónimos, pero no se puede tener un 8 por ciento de sobrerrepresentación, es leer lo que quieren de un artículo constitucional y dañar la voluntad popular”, comentó Murayama. Además, Murayama advirtió que la situación podría llevar a resultados absurdos, comparando la situación con un ejemplo de injusticia. “Si se concreta lo que dicen, el Partido Verde tendría en la segunda bancada con mayor número de diputados, es como si al corredor que llegó en quinto lugar se le da la medalla de plata por capricho de un juez”, afirmó. A widely used model system to study this process is to seed amyloid filament formation in cultured cells using human brain extracts. Here, we report the electron cryo-microscopy (cryo-EM) structures of tau filaments from undifferentiated seeded SH-SY5Y cells, that transiently expressed N-terminally HA-tagged 1N3R or 1N4R human tau, using brain extracts from individuals with AD or CBD. Although the resulting filament structures differed from those of the brain seeds, some degree of structural templating was observed. Studying templated seeding in cultured cells, and determining the structures of the resulting filaments, can thus provide insights into the cellular aspects underlying neurodegenerative diseases. El consejero Ciro Murayama pide a la Suprema Corte –a través del ministro Alberto Pérez Dayan– que suspenda el ‘plan B' de la reforma electoral. Murayama explica que la Corte tiene hasta el 2 de junio para que la Corte la suspenda y así el Instituto Nacional Electoral mantenga su funcionamiento tal y como se encuentra actualmente. Si la reforma entra en vigor, por ley se despedirá a 2 mil personas, muchas de ellas del servicio profesional de carrera y eso provocaría que las elecciones de 2024 se realicen con el mínimo de personal y presupuesto, lo que complicaría los comicios presidenciales. El consejero asegura que el INE no cuenta con el dinero para indemnizar al personal que sería despedido y mucho menos para su recontratación, por lo que esperan se logre suspender mientras la Corte resuelve la inconstitucionalidad del ‘plan B' promovido por el presidente López Obrador.

Abstract: cAMP is a pivotal second messenger regulated by various upstream pathways including Ca2+ and G protein-coupled receptors (GPCRs). To decipher in vivo cAMP dynamics, we rationally designed cAMPinG1, an ultrasensitive genetically encoded green cAMP indicator that outperformed its predecessors in both dynamic range and cAMP affinity. Two-photon cAMPinG1 imaging detected cAMP transients in the somata and dendritic spines of neurons in the mouse visual cortex on the order of tens of seconds. In addition, multicolor imaging with a highly sensitive new red Ca2+ indicator RCaMP3 allowed simultaneous measurement of population patterns in Ca2+ and cAMP in hundreds of neurons. We identified Ca2+-induced cAMP responses that represented specific information, such as direction selectivity in vision and locomotion, as well as GPCR-induced cAMP responses. Overall, our multicolor suite revealed that information encoded in Ca2+ and GPCRs signaling is integrated and stored as cAMP transients for longer periods in vivo. Both monetary rewards/incentives and curiosity have been found to motivate and facilitate learning by dopaminergic midbrain projections to the hippocampus during encoding. In this study, we examined potential brain mechanisms during early consolidation period that jointly or independently contribute to these facilitating effects. Participants (N = 50) watched 36 videos of magic tricks and rated their "subjective feelings of curiosity" while the availability of extrinsic incentives was manipulated between groups. Functional magnetic resonance imaging (fMRI) data were acquired before, during, and after learning, and memory for magic tricks was assessed one week after. Our analysis focused on the change in resting-state functional connectivity (RSFC) between the dopaminergic midbrain and the anterior hippocampus, a dopaminergic consolidation mechanism previously reported in the context of extrinsically motivated learning. Changes in RSFC were correlated with behavioural measures of learning, i.e., the total number of items encoded and the curiosity-driven memory benefit. We found that brain-behaviour correlations differed depending on the availability of extrinsic incentives. More specifically, the correlation between the total number of items encoded and RSFC change was significantly different in the incentivised compared to the control group. The curiosity-driven memory benefit, however, did not correlate with changes in RSFC in either of the groups. In sum, this suggests that curiosity-motivated learning might be supported by different consolidation mechanisms compared to extrinsically motivated learning and that extrinsic incentives influence consolidation mechanisms supporting learning. El productor Epigmenio Ibarra afirma que Ciro Murayama miente al decir que las leyes se votan primero y se explican después en este sexenio. El consejero electoral Ciro Murayama afirma que la reforma a las leyes secundarias se hizo sin discusión, sin escuchar las voces de la oposición, sin diálogo y sin consenso. Murayama explica que en las 330 elecciones organizadas por el INE no han tenido problemas para determinar a los ganadores entre ellos los del partido Morena. Una de los argumentos dados por Morena es el ahorro de 3 mil millones de pesos, sin embargo, Ciro Murayama destaca que no hubo ningún análisis financiero para determinar cómo se lograrán esas metas. Murayama señaló que el poder legislativo está actuando de manera sumisa como en el presidencialismo priista y que es falso que los consejeros electorales ganen más que el presidente López Obrador.

Pedro Ferriz Híjar en su Editorial señala que el presidente Andrés Manuel López Obrador pone como pretexto el costo del Instituto Nacional Electoral para acabar con la democracia del país. Además, mencionó que Ciro Murayama y Lorenzo Córdova no son el INE, el INE somos todos y es el único organismo que nos queda de nuestra precaria democracia, de igual forma, señala que si es necesario que los quiten a Córdova y Murayama está bien, pero su lugar lo tiene que tomar alguien que tenga la capacidad y no alguien que le haga los favores a AMLO.

El debate por la Reforma Electoral propuesta por AMLO llegó hasta las encuestas, entre ellas una que levantó el propio INE donde la ciudadanía aprueba las modificaciones que propone el ejecutivo. Pese a esto, la legislación es vista por grupos opositores como un ataque a la democracia y un retroceso para México.

Experimental studies on information-seeking have found that curiosity facilitates memory encoding and exhibits similar reward,ng properties as extrinsic rewards/incentives by eliciting a dopaminergic response. However, it is not clear whether these findings hold with more naturalistic dynamic stimuli and how the joint effect of curiosity and extrinsic incentive manifests in learning and neural activation patterns. Herein, we presented participants with videos of magic tricks across two behavioural (N1 = 77, N2 = 78) and one fMRI study (N = 50) and asked them to rate subjective feelings of curiosity, while also performing a judgement task that was incentivised for the half of participants. Incidental memory for the magic trick was tested a week later. The integrated results showed that both curiosity and availability of extrinsic incentives enhanced encoding but did not interact with each other. However, exploratory analyses showed that curiosity and monetary incentives were associated with recollection and familiarity differently, suggesting the involvement of different encoding mechanisms. Analysis of the fMRI data using the intersubject synchronisation framework showed that, while the effects of curiosity on memory were located in the hippocampus and dopaminergic brain areas, neither the effects of curiosity nor incentives themselves were found in the often-implicated reward network, but instead were associated with cortical areas involved in processing uncertainly and attention. These results suggest that curiosity recruits broader brain networks than what was implicated in the previous literature when investigated with dynamic stimuli. Así como existe el Olimpo del cine mexicano, donde figuran en lo más alto ídolos sufrientes como Dolores del Río, Marga López o Pedro Infante, también está el averno en el que, al menos en el imaginario colectivo, purgan sus canalladas los villanos del celuloide, aquellos que provocan las lágrimas de los honrados, torturan al débil, abusan del menesteroso, explotan al necesitado, mancillan a la inocente, pisotean al valiente y otras infamias indescriptibles. El cine mexicano, tan afecto al melodrama y a sus transiciones que pasan en un santiamén de la bondad más inaudita a la crueldad más ufana, ha dado vida a villanos de antología, encarnados por actores que, en algunos casos, hicieron del cinismo su sello y de la saña su divisa. Después de todo ¿qué sería de las películas protagonizadas por un gran héroe si no existiera un gran villano? Dentro de este grupo, existen nombres de actores imprescindibles, especializados en roles negativos, algunos de los cuales hemos incluido en esta selección. A Carlos López Moctezuma (1909-1980) le salían muy bien los papeles de cacique atroz. Su fuerte presencia, sus ojos claros y sus cejas arqueadas contribuían a resaltar sus rasgos felinos, que se pronunciaban con su fuerte voz y sus modos groseros Por su parte, Miguel Inclán (1900-1956) entregaba notas diferentes. Su presencia torva y perversa, más propia de un sujeto de extracción humilde. En el género del cine de cabaret, de los fascinantes ambientes mundanos llenos de humo, música, alcohol y sensualidad, sobresale la figura de Rodolfo Acosta (1921-1974). Y si de vividores se trata, mezquinos y arrogantes, un peculiar hamponzuelo que encarnó el actor Víctor Parra (1920-1994). Arturo Martínez (1918-1992) hizo una larguísima carrera como villano en el que abarcó en cinco décadas prácticamente todos los géneros del cine nacional. Abstract: Cellular heat-sensing is a universal strategy for avoiding thermal damage and adapting to environments by regulating thermogenic activities. If heat-sensing results in the acceleration of processes governing cellular thermogenesis, hyperthermia can occur. However, how this positive feedback loop contributes to hyperthermia development, especially the gap between heat-sensing and thermogenesis, remains largely unknown. Here, we show that an optically controlled local heat pulse induces an intracellular Ca2+ burst in cultured HEK 293 cells overexpressing ryanodine-receptor-type-1 (RyR1) mutants related to the life-threatening illness malignant hyperthermia (MH), and that the Ca2+ burst originates from heat-induced Ca2+-release (HICR) because of the mutant channels' heat hypersensitivity. Furthermore, the heat hypersensitivity of the four RyR1 mutants was ranked, highlighting the complexity of MH. Our findings reveal the novel cellular heat-sensing mechanism, HICR, is essential for the functional positive feedback loop causing MH, suggesting a well-tuned HICR is fundamental for heat-mediated intracellular signaling. In a virtual call, what is the maximum number of people who should be joining on the buyer side, to make the meeting effective? There is a correlation between the number in the group and the number who will contribute during the meeting. The more people on the call, the greater tendency for people to become less engaged. Research by the Rain Group says that with up to four people on the call, the level of engagement can be quite good. Once we get over five buyers, it becomes much more difficult. The virtual world revolves around both camera and audio based communication. If there are multiple attendees on the buyer side, there is also a strong chance that not all will turn their cameras on. This is a downer because you now have no chance of reading their body language, fraught as that might have been anyway. This is not so different to the face to face world though, when there are many on the Japanese buying side in the room. The most senior people are either actually sleeping or are sitting there with their eyes closed, doing a spectacular job of pretending to be sleeping. Usually, there will only be around three people who will even speak up. Everyone is there because they have to protect their turf and want to influence the outcome of the meeting. They are expecting you to make your pitch. Their shotgun breeches are duly snapped shut and then they drill your pitch full of holes, exposing what a travesty it is. The online world will be the same and with the exposure to all of the multi-tasking going on in the background, because their cameras are turned off. Normally, it is extremely hard to get Japanese buyers to do anything for you in a sales meeting. They think they are there to listen, then critique everything you just said. Online it is even worse. We have to go beyond using the communication platform as a tool to speak to each other and must use it to facilitate an experience. If we were having a face to face meeting and the buyer was highly engaged, asking questions, seeking more detail, then there would be a pretty good chance they would buy. We need that same level of engagement in the online world. How do we get them to participate online? Remember, the buyers all see themselves as God and we are just pesky, unwashed salespeople. We need to direct the buyers and guide them to take certain actions, which will allow us to engage with them. We need to tell them what we want them to do and then tell them how to do it. We next ask for a response, using a follow up question. This is where the preparation is critical. For example, if with a flourish, we magically pull up a whiteboard on screen and expect a bunch of Japanese buyers to grab the text tool or the arrow tool and start interacting with us, their complete lack of response will be debilitating. If it was just a one to one online meeting, they might feel some pressure to take some action, but as a collective, they will just sit there and completely and shamelessly ignore us. They are totally happy to let hell freeze over, before they make a move for any tool. They will blank you and force you back to giving your pitch. If you have already given it, they will rip into your pitch, telling you why it is crap. We need to be super well organised and anticipate their resistance. For example, we have slides that have options numbered on screen. We need to set up the participation. “There are many of us on the call today, so we really appreciate everyone joining us. To make this meeting as effective as possible would you please give me some feedback on these four options. The best way to do that is go to the chat box, under the participant panel and write in the number of the option you think would generate the most value for your part of the business?”. Their names will appear in the chat showing the number chosen. We just refer to them by name and thank them for selecting their option. This is a way of encouraging the remainder to follow suit. For those who don't participate, the “I am not here to do anything” brigade, we need to draw them out by name. “Tanaka san, would you mind sharing your selection with everyone please, because we really value your insights?. I would really appreciate it”. We can then ask individuals to share their thinking behind their selection. “Murayama san, I see you chose option three. Thank you for doing that. Would you please come off mute and help me to understand why this is important to you. That will really help me to better understand how we can serve you best, so please share your thoughts with us. Thank you Maruyama san”. We try and do this with a number of the participants on the call, but it must be done in a deferential, “we are really trying to help you” way or resistance will be major and unmerciful. Following this we might ask a further expansion question to the group. “Could I ask everyone else to give me a green check please if they think we have sufficiently understood your situation. If you think we have missed something important, please choose the red cross. The green check and the red cross are located in the little panel, just below the participant list in the right hand panel on your screen. Please choose green check or red cross”. Don't expect to get everyone involved. You wouldn't get that in a face to meeting either, so it is an even more outlandish idea for the virtual world. But you will get engagement if you are properly prepared. Anticipate you will draw God's wrath and prepare accordingly. Make it super easy to get engagement with you in the sales call. If you can do that, then your chances of making a sale go up dramatically. Your rivals won't have a clue about any of this and will be drowning in their own blood as the buyers cut their pitch to ribbons, as usual.

The result is a practically aberration-free, fast scanning high optical invariant two-photon microscopy (FASHIO-2PM) that enables calcium imaging from a large network composed of ~16k neurons at 7.5 Hz in a 9 mm2 contiguous image plane including more than 10 sensory-motor and higher-order regions of the cerebral cortex in awake mice. Through a network analysis based on single-cell activities, we discover that the brain exhibits small-world-ness rather than scale-freeness. FASHIO-2PM will enable revealing biological dynamics by simultaneous monitoring of macroscopic activity and its composing elements. Copy rights belong to original authors. Many brain magnetic resonance imaging (MRI) studies have been conducted; however, imaging-based diagnostic and therapeutic biomarkers are not yet well established. The Brain/MINDS Beyond human brain MRI project (FY2018 ~ FY2023) is a multi-site harmonization study aiming to establish clinically-relevant imaging biomarkers using multiple high-performance scanners, standardized multi-modal imaging, and a study design that includes traveling subjects. This project began with 13 clinical research sites that collect MRI data on psychiatric and neurological disorders across the lifespan and three research sites that design and develop measurement procedures, neuroimaging protocols, data storage and sharing, and analysis tools. Brain images obtained with the Harmonization protocol (HARP) are preprocessed and analyzed using approaches developed by the Human Connectome Project, generating preliminary cortical structure, function, and connectivity measures that are comparable across scanners. The use of 'travelling subjects', in which study participants travel to multiple sites to undergo scanning with standardized neuroimaging techniques, enable us to minimize the measurement bias between scanners and protocols and to increase the sensitivity and specificity of case-control studies. All the imaging and demographic and clinical data are shared between the participating sites and will be made publicly available in 2024. To the best of our knowledge, this is the first multi-site human brain MRI project to explore multiple psychiatric and neurological disorders across the lifespan. The Brain/MINDS Beyond human brain MRI project will help to identify the common and disease-specific pathophysiology features of brain diseases and develop imaging biomarkers for clinical practice.
