Abstract: Precise control of pluripotency is a requirement for the safe and effective use of hPSCs in research and therapies. Here we report that pyruvate dehydrogenase upregulates histone H3 pan acetylation and levels of pluripotency marker NANOG in 5% O2. Pyruvate dehydrogenase (PDH) is an essential metabolic switch and a bottleneck for the glycolytic production of acetyl-CoA. Silencing of gene expression showed that PDH is regulated by the activity of its phosphatase PDP1. We show that PDP1 is sensitive to reactive oxygen species-mediated inactivation, leading to the downregulation of H3 pan acetylation and NANOG levels. Furthermore, we show that FGF2, a cytokine commonly used to maintain pluripotency activates pyruvate dehydrogenase through MEK1/2-ERK1/2 signaling pathway-mediated downregulation of ROS in 5% O2, thus promoting histone acetylation. Our results show the importance of pyruvate dehydrogenase in regulating energy metabolism and its connection to pluripotency. Furthermore, our data highlight the role of reactive oxygen species and redox homeostasis in pluripotency maintenance and differentiation. Abstract: Alzheimers disease (AD) is the most common cause of dementia, with no current cure. Consequently, alternative approaches focusing on early pathological events in specific neuronal populations, besides targeting the well-studied Amyloid beta (A{beta}) accumulations and Tau tangles, are needed. In this study, we have investigated disease phenotypes specific to glutamatergic forebrain neurons and mapped the timeline of their occurrence, by implementing familial and sporadic human induced pluripotent stem cell models as well as the 5xFAD mouse model. We recapitulated characteristic late AD disease phenotypes, such as increased A{beta} secretion and Tau hyperphosphorylation, as well as previously well documented mitochondrial and synaptic deficits. Intriguingly, we identified Golgi fragmentation as one of the earliest AD phenotypes, indicating potential impairments in protein processing and post-translational modifications. Computational analysis of RNA sequencing data revealed differentially expressed genes involved in glycosylation and glycan patterns, whilst total glycan profiling revealed minor glycosylation differences. This indicates general robustness of glycosylation besides the observed fragmented morphology. Importantly, we identified that genetic variants in Sortilin-related receptor 1 (SORL1) associated with AD could aggravate the Golgi fragmentation and subsequent glycosylation changes. In summary, we identified Golgi fragmentation as one of the earliest disease phenotypes in AD neurons in various in vivo and in vitro complementary disease models, which can be exacerbated via additional risk variants in SORL1. Why do we need so much money? Why not go do a job I love that pays peanuts? Some of the jobs in which you can make a difference or pursue a passion are not the best paying jobs. For example, acting. The average member of the screen actors guild makes $1000 per year. Musicians? I know a little about this from personal experience. I never made any money to speak of playing my own songs. Orchestra players, unless they are in a big city, do not make enough to live on, and maybe nothing at all. Playing in clubs, if you play cover music and not your own, you can make maybe a few hundred dollars for a weekend gig. All this is before expenses, taxes etc. More importantly, all artists have to hustle to get work. Even if you make $50 per hour, the average for film and tv actors, you're not guaranteed to get those hours. Teachers make about $45K per year, and police officers make $39K. Suppose you are passionate about your religion and want to make a difference there? Priests in my diocese get room and board plus $1,000 per month. Isn't this enough? Well, let's look at how much of this you get to keep while pursuing your dream job. Average living expenses for a single person add up to $13K not including rent, which is $17K on average. That is $30K for a single person. Average tax is 23% including Social Security and Medicare. So the average teacher, probably the most stable job I mentioned, is down to about $400 a month left over. No entertainment, no savings for retirement, no medical care, no student loan payments, no vehicles. We haven't included anything here except survival. When Tesla first announced its upcoming Model Y all-electric crossover in March of this year, it was forced to share the stage with Audi, who also unveiled plans for its Q4 e-Tron around the same time.Both vehicles have release dates in 2020 and both target the popular crossover segment with close to 300 miles per charge.And while the two brands, known for luxury, intuitive interiors, will find formidable competition in one another, they shouldn’t forget to swivel – as competition seems to be coming from all sides.The latest is from Fisker Inc., the second auto company started by designer Henrik Fisker… the first being Fisker Automotive, the electric vehicle maker that made more than one splash – its Fisker Karma extended range luxury electric was a first of its kind, garnering the company many high-profile investors, including celebs like Leonardo DiCaprio.Then, in 2012, they went bankrupt because, at that time, that’s what you did if you were an electric car company. But Fisker the man had gotten out by then and just moved right along with his plans for electric domination and now, it appears, he’s ready to strike.In the year 2021, Fisker will begin producing the unnamed crossover and has priced it “under $40,000” which is right in the Model Y sweet spot, as Elon Musk has promised they’d start around $39K.According to Digital Trends, the Fisker will feature “an 80-kilowatt-hour lithium-ion battery pack affording around 300 miles of range, but it’s keeping other details under wraps for now.” The company is reportedly hoping to build these crossovers here in America, and is scouting factory locations in several states. I get asked regularly about students loans. Usually, the question is "What can I do about these loans?" The percentage of college graduates who carry student loan debt is staggering, ranging from 66% to 88%, depending on the type of college or university they attended. The debt balances range from $23K to $160K, with the average in 2017 being $39K.   In This Episode, We Look At: The tough love answer to this question (which is Biblical truth) is to pay it.
