Abstract: Tunneling nanotubes (TNTs) are open actin- and membrane-based channels, connecting remote cells and allowing direct transfer of cellular material (e.g. vesicles, mRNAs, protein aggregates) from cytoplasm to cytoplasm. Although they are important especially in pathological conditions (e.g., cancers, neurodegenerative diseases), their precise composition and their regulation were still poorly described. Here, using a biochemical approach allowing to separate TNTs from cell bodies and from extracellular vesicles and particles (EVPs), we obtained the full composition of TNTs compared to EVPs. We then focused to two major components of our proteomic data, the CD9 and CD81 tetraspanins, and further investigated their specific roles in TNT formation and function. We show that these two tetraspanins have distinct functions: CD9 participates in the initiation of TNTs, whereas CD81 expression is required to allow the functional transfer of vesicle in the newly formed TNTs, possibly by regulating fusion with the opposing cell. Abstract: The promise of extracellular vesicles (EVs)-based liquid biopsy resides in the identification of specific signatures of EVs of interest. Knowing the EV profile of a body fluid can facilitate the identification of EV-based biomarkers of diseases. To this end, we characterised purified EVs from paired human milk and serum by surface protein profiling of cellular markers in association with gold standard EV markers (tetraspanins CD9, CD63 and CD81). By using the MACSPlex bead-based flow-cytometry assay with pan-tetraspanin detection (i.e. simultaneous CD9, CD63 and CD81 detection), besides specific breast epithelial cell signatures in milk EVs and platelet signatures in serum EVs, we also identified body fluid-specific markers of immune cells and stem cells. Interestingly, comparison of pan-tetraspanin and single tetraspanin detection unveiled both body fluid-specific tetraspanin distributions and specific tetraspanin distributions associated with certain cellular markers, which were used to model the potential biogenesis route of different EV subsets and their cellular origin. Abstract: Extracellular vesicles (EVs) are released by all cells into biofluids and hold great promise as reservoirs of disease biomarkers. One of the main challenges in studying EVs and using them for diagnostics is a lack of methods to quantify EVs that are sensitive enough and can differentiate EVs from similarly sized lipoproteins and protein aggregates. We demonstrate the use of ultrasensitive assays to quantify EVs by immuno-isolating and detecting EV transmembrane proteins in microwell arrays. We developed single molecule array (Simoa) assays for the quantification of EVs using three widely expressed transmembrane proteins: the tetraspanins CD9, CD63, and CD81. Using Simoa to measure these three EV markers, as well as albumin to measure protein contamination, we were able to compare the relative efficiency and purity of several commonly used EV isolation methods in plasma and cerebrospinal fluid (CSF): ultracentrifugation, precipitation, and size exclusion chromatography (SEC). We further used these assays to rapidly optimize EV isolation using SEC from plasma and CSF. Our results highlight the utility of quantifying EVs using Simoa and provide a rapid framework for comparing and improving EV isolation methods from biofluids. Copy rights belong to original authors. Dr. Carolina Soekmadji studies different types of extracellular vesicles, specifically trying to understand their connection to prostate cancer. In this discussion, she describes  how different types of extracellular vesicles seem to react differently to the same substance, why the CD9 vesicle's proliferation under different androgen conditions is important, and what this means in how doctors can individualize prostate cancer treatment. Dr. Carolina Soekmadji works as a senior research officer at the University of California, Irvine. She studied for her Master's in Japan and completed her PhD work in Australia, where she studied the exocytosis and endocytosis of synoptic vesicles. She discusses the typical therapy for prostate cancer where doctors decrease the androgen presence, also called ADT. While this usually has the desired effect, there's always a group of patients that don't seem to show an effective response. While cancer cells initially die in this group, the cancer cells return and start growing again. Dr. Soekmadji has located a specific vesicle that appears to grow under both conditions: androgen presence or absence. She thinks that this vesicle may make the difference between these two populations and their response. Dr. Soekmadji covers general causes of prostate cancer as well. She continues to study the activity of extracellular vesicles, and how and why this particular vesicle responds as it does and why this happens in some patients but not others.  Dr. Carolina Soekmadji offers a general course on extracellular vesicles and health issues through Coursera and the University of California that's open to the general public. Teens have their own speak, using slang and lingo that can sometimes be difficult for adults to interpret. This week we are going to teach you a little bit about one of those languages, teen texting. Recently, I (Julie here) had my own encounter at a friend's house who has 3 teen daughters. As I was talking with the 17-year-old about her Snapchat communications, she shared that she hates to leave people "unread". She believes this means she saw their message but didn't reply with one. I tried to explain what the meaning of "unread" in general terms implies, that she never saw it...but she kept replying that "unread" means she saw it and didn't send a message back. After we each repeated our points multiple times, she finally shrugged her shoulders and said "I don't know, that's just what we call it". Teenagers, with the most impressive conviction, speak in a different language even when they don't really know what they are talking about. Recently, Darlene discovered a service called Bark that analyzes what teens are actually saying through their various channels: social media, texting, email, etc. There she uncovered some shocking short codes teens are using to say things you need to know about. Whether you're the parent of a teen or not, you likely have teens in your life; you're an Aunt, a teacher, a mentor or neighbor. In this episode we unpack what teens are saying and how adults can keep up to date and even practical insight on how to protect the teens in your life in this ever-changing social society. Die Apoptose ist ein genetisch codiertes Suizidprogramm, das es jeder eukaryontischen Zelle erlaubt, auf bestimmte endogene oder äußere Signale mit einem kontrollierten Degradations- und Absterbeprozess zu reagieren. Auf diese Weise werden dem Organismus überzählige oder geschädigte Zellen entzogen, ein Prozess, der z.B. bei Krankheiten wie Krebs zumindest partiell ausser Kraft gesetzt ist. Um neue Einblicke in das erst in Teilen aufgedeckte Netzwerk Apoptose-induzierender Gene zu erhalten, wurde im Rahmen dieser Arbeit ein Expressions-Screeningverfahren zur Identifikation neuer proapoptotischer Gene eingesetzt. Mit diesem Verfahren, das auf der Fähigkeit der Gene dominant Apoptose zu induzieren basiert, wurden mehr als 50 neue Apoptose-induzierende Gene isoliert, von denen einige Ursache bestimmter Krankheiten sein könnten. Aus diesen wurde das Metastasensuppressorgen C33/CD82 für weitere Untersuchungen ausgewählt, da seine Expression in zahlreichen stark metastasierenden Tumoren herabreguliert ist und es bislang nicht als proapoptotisch beschrieben wurde, obwohl es eines der bislang bestcharakterisiertesten Metastasensuppressorgene ist und die Apoptose ein kritischer Faktor bei der Metastasierung ist. C33/CD82-Apoptose erfolgt durch Aktivierung der Mitochondrien. Dadurch kommt es zum Zusammenbruch des mitochondrialen Membranpotentials und zur Freisetzung proapoptotischer Faktoren in das Cytoplasma, wo diese Caspasen aktivieren, sodass es zur Ausprägung charakteristischer Merkmale der Apoptose kommt: neben morphologischen Veränderungen, wie der Ausbildung apoptotischer Körperchen, wird Phosphatidylserin exponiert und die DNA und bestimmte Proteine werden degradiert. Ausgelöst wird die C33/CD82-Apoptose durch Superoxidanionen, die anders als bei Standard-Apoptosewegen nicht an der mitochondrialen Elektronentransportkette gebildet werden. Darüber hinaus wurde in dieser Arbeit demonstriert, dass auch einige andere Mitglieder der Tetraspanin-Superfamilie, der C33/CD82 angehört, Apoptose induzieren: hierzu zählen CD9, CD37, CD81 und CD151 und in geringerem Umfang auch CD53 und CD63. Mit Hilfe trunkierter Mutanten wurde der für die Apoptoseinduktion verantwortliche Bereich auf die ersten 119 Aminosäuren von C33/CD82 eingegrenzt. Nach den Ergebnissen dieser Arbeit ist die C33/CD82-Apoptose eine wesentliche neue Eigenschaft des Proteins: Sie könnte neben der Beeinflussung der Zellmotilität, die durch E-Cadherin und Integrine vermittelt wird, mit denen C33/CD82 interagiert, und der Internalisierung des EGF-Rezeptors, die durch C33/CD82 beschleunigt wird, zur Wirkung des Proteins als Metastasensuppressor beitragen. Die Überexpression von C33/CD82 induziert nicht nur Apoptose in allen getesteten Tumorzelllinien, sondern auch unter hypoxischen Bedingungen. In vivo sind derartige Bedingungen im Inneren nicht vaskularisierter Tumore und Metastasen zu finden. Dort könnten sie, bedingt durch die Hypoxie-aktivierbaren Transkriptionsfaktoren p53 und NF﷓IL﷓6, zu einer Verstärkung der C33/CD82-Expression führen. Die Vermeidung von Hypoxie-induzierter C33/CD82-Apoptose könnte ein Grund dafür sein, dass die Expression von C33/CD82 in vielen untersuchten stark metastasierenden Tumoren herabreguliert ist. Eine Besonderheit von C33/CD82 ist, dass nicht nur seine Überexpression dominant Apoptose induziert, sondern dass bereits das normale Expressionsniveau Zellen für wenigstens einen spezifischen proapoptotischen Stimulus sensitiviert. Im Fall dieser Arbeit war dies Cycloheximid, welches stellvertretend für weitere applizierbare und möglicherweise therapeutisch verwendbare Stimuli stehen könnte. Vielleicht existiert auch ein Cycloheximid entsprechender endogener Stimulus, der wie Cycloheximid ausschließlich in C33/CD82-positiven Zellen Apoptose auslöst. Die in dieser Arbeit entdeckten Fähigkeiten von C33/CD82, dominant Apoptose zu induzieren und Zellen für Apoptoseinduktion zu sensitivieren, sind neue Eigenschaften des Metastasensuppressors C33/CD82, die zu der in vivo beobachteten, in der Literatur vielfach beschriebenen Metastasensuppression beitragen können.
