He received his PhD from the University of Sussex in 1978. In 1991, Dr. Rose published seminal work per his text 'Evolutionary Biology of Aging' exploring a view of the subject based on antagonistic pleiotropy, the hypothesis that aging is caused by genes that have two effects, one acting early in life and the other much later. The genes are favored by natural selection as a result of their early-life benefits, and the costs that accrue much later appear as incidental side-effects that we identify as aging. Dr. Rose's laboratory has conducted the longest-running artificial selection experiment designed to test the theory of antagonistic pleiotropy using Fruit flies (Drosophila melanogaster) bred for longevity by collecting eggs from the longest-lived flies in each generation. The experiment has run since 1981, and has produced flies with quadruple the original life span. In 1997, Dr. Rose was awarded the Busse Research Prize by the World Congress of Gerontology. He received his PhD in 2009 in Biotechnology from University of Pune for his work demonstrating microRNA regulation of cytokines invovled in allergic inflammation in mice models. Dr. Sharma's postdoctoral research at the Buck Institute, Novato, California involved investigating novel molecular regulatory pathways in regards to genotoxic stress and cellular senescence in invertebrate and mammalian models. Aubrey David Nicholas Jasper de Grey is an English author and biomedical gerontologist. He is the author of The Mitochondrial Free Radical Theory of Aging (1999) and co-author of Ending Aging (2007). He is known for his view that medical technology may enable human beings alive today not to die from age-related causes. As an amateur mathematician, he has contributed to the study of the Hadwiger–Nelson problem in geometric graph theory, making the first progress on the problem in over 60 years. De Grey is an international adjunct professor of the Moscow Institute of Physics and Technology, a fellow of the Gerontological Society of America, the American Aging Association, and the Institute for Ethics and Emerging Technologies. In the fifth episode of the Longevity & Aging Series, Dr. Amit Sharma, Group Lead from SENS Research Foundation, Mountain View, CA, discusses a research paper he co-authored that was published in Volume 14, Issue 5, of Aging (Aging-US), entitled, “Enhanced co-culture and enrichment of human natural killer cells for the selective clearance of senescent cells.” DOI - https://doi.org/10.18632/aging.203931 Corresponding Author - Amit Sharma - amit.sharma@sens.org Video - https://www.youtube.com/watch?v=WQR8_gm2gUI Abstract In the context of aging and age-associated diseases, Natural Killer (NK) cells have been revealed as a key cell type responsible for the immune clearance of senescent cells. Subsequently, NK cell-based therapies have emerged as promising alternatives to drug-based therapeutic interventions for the prevention and treatment of age-related disease and debility. Given the promise of NK cell-mediated immunotherapies as a safe and effective treatment strategy, we outline an improved method by which primary NK cells can be efficiently enriched from human peripheral blood across multiple donors (ages 20-42 years old), with a practical protocol that reliably enhances both CD56dim and CD56bright NK cells by 15-fold and 3-fold, respectively. Importantly, we show that our co-culture protocol can be used as an easily adaptable tool to assess highly efficient and selective killing of senescent cells by primary NK cells enriched via our method using longer co-culture durations and a low target to effector ratio, which may be more physiological than has been achieved in previous literature. Dr. Kevin Perrott, Ph.D. is Founder and CEO, OpenCures (https://opencures.org/), Adjunct Professor, University of Alberta, Co-Founder and Advisor, Oisin Biotechnologies, President, of Global Healthspan Policy Institute, and Co-Founder and Treasurer, SENS Research Foundation. Kevin is a successful entrepreneur and owner of the largest motorcycle and snowmobile dealership in Canada, Riverside Honda and Skidoo Sales in Edmonton, Alberta. He became a cancer survivor, an experience which clearly highlighted the deficiencies of the current health technology development paradigm where the customer has almost no input in the development of their own health solutions. Armed with the realization that nothing is more valuable than health and the time to enjoy it with those you love, Kevin resolved to put his energies towards addressing these deficiencies. In 2003, Kevin joined Aubrey de Grey and David Gobel to form the Methuselah Foundation which offered competitive prizes for advances in longevity science. A few years later he became a co-founder with Aubrey de Grey and others of SENS Research Foundation to develop interventions able to repair or make harmless the damage that accumulates and underlies multiple age-related disorders. Mostly recently, Kevin formed OpenCures as a for-benefit corporation helping individuals perform self-directed research, generate data, and focus the value of data on the health solutions they are interested in. Kevin has authored and co-authored several peer-reviewed articles all focused on accelerating the science underlying the development of therapies for degenerative diseases. He is a co-founder of multiple non-profit and for-profit entities whose missions are aligned with that purpose. PD results from specific kinds of aging damage accumulating in key parts of the brain and nervous system, but similar damage accumulates in other aging tissues, causing dysfunction in those local tissues; these degenerative processes get labeled "diseases" when they reach a tipping point and the tissue can no longer carry out its normal function. The good news is that we now have a strategy for preventing, delaying, and one day even reversing Parkinson's and other diseases of aging through the revolution in rejuvenation biotechnology: therapies that directly remove, repair, replace, or render harmless the damage of aging. Aubrey de Grey, Ph.D., Chief Science Officer and Co-founder of SENS Research Foundation, delivers an overview of aging and the many health problems that develop in our advanced years. Dr. de Grey is a respected member of the science community; he is the noted biomedical gerontologist who devised the innovative SENS platform and co-founded the SENS Research Foundation to further it. Dr. de Grey has written about his work and as an established researcher, he has been appointed to the editorial and scientific advisory boards of many journals, organizations, etc. Dr. de Grey is a Fellow of the Gerontological Society of America as well as the American Aging Association. He holds a BA in Computer Science and a Ph.D. in Biology from the prestigious University of Cambridge.  Dr. de Grey discusses his research in aging and the motivations for tackling the career. As he states, aging is the number one medical problem as it causes more suffering. He was motivated to research in this area because he found that not enough was being done to focus on aging and the myriad of problems that come with it. He talks about the many excuses that are given as reasons to simply accept aging as it is, or not focus on it at all, such as “it's inevitable…everything ages,” or the philosophical—“death gives meaning to life,” or social—“maybe we could do this, but it would create new problems worse than the problem we are solving.” And as the Ph.D. states, none of these excuses stand up to even the faintest scrutiny, however, they still remain quite popular.  Dr. de Grey explains that ‘why' and ‘how' we age are fairly well understood, but it is the understanding of what we already know that is important, to take the information we know to be true and utilize that to design and implement therapies that will prevent the health problems of old age. And this is the core of what Dr. de Grey's foundation, SENS Research, seeks to accomplish. SENS Research Foundation works to create, promote, and provide wide-ranging access to all kinds of therapies that can cure and prevent the diseases and detrimental disabilities of aging by essentially repairing the damage that builds up within our bodies over long periods of time. SENS Research seeks to change the way the world researches and thus cares for age-related ill health. The Ph.D. discusses stem cell therapy and its ability to repair the damage, loss of cells, etc. As he explains, stem cells can be injected that target areas within the body and repair damage, replace lost cells, and thus keep the body area, organ, etc. functioning in a healthier state. He explains in detail how cells function and the processes used to make genetic modifications. Additionally, Dr. de Grey expounds upon the kinds of complex research they engage in at his foundation, and the types of conditions that they seek to bring more understanding to in order to combat them.

Dr. Aubrey de Grey is a biological gerontologist on a crusade to defeat aging. He thinks we are within a couple of decades of rejuvenation medicine capable of adding another 30 years to human lifespan, proposing that the first person to live to 1,000 years old may already be alive today. In the field of longevity science, there are a few roadblocks, but the biggest one is funding. They discuss why it isn’t a higher priority for average people to further this research.   Mark and Dr. de Grey cover a number of concepts at the forefront of anti-aging, such as whether increasing telomere lengths may be beneficial against aging or may cause side effects. Dr. de Grey suggests we learn more about the subject of defeating aging. Then become an advocate for the research, to bring about these medicines faster. Mark also asks Dr. de Grey about the morality of pursuing greater longevity for humans. Also on the podcast, Mark and Dr. Gladden discuss how the typical medical approach isn’t designed to promote longevity but treat the immediate problem. Medicine is still, after all, a business. Dr. De Grey is a biomedical gerontologist who researched the idea for and founded SENS Research Foundation. SENS Research Foundation is a public charity that is transforming the way the world researches and treats age-related disease. SENS asserts that two thirds of all deaths worldwide, and about 90% of all deaths in the developed world, are from causes that only rarely kill young adults. If we look at the entire world, then the number of deaths that occur each day is roughly 150,000 and about two-thirds of them are because of aging. These causes include Alzheimer's, cardiovascular disease, Type II diabetes and most cancers. They are age-related because they are expressions of the later stages of aging, occurring when the molecular and cellular damage that has accumulated in the body throughout life exceeds the level that metabolism can tolerate. Before it kills us, aging, imposes on most elderly people, a long period of decline, debilitation and disease. For these reasons, aging is unarguably the most prevalent medically-relevant phenomenon in the modern world and the primary ultimate target of biomedical research. In this episode, we have the distinct honor and privilege to talk to one of the world’s foremost experts on aging and learn about his assertion that the first human beings who will live to be 1,000 years old have already been born.

Clearly throughout recorded history we’ve been fascinated by things like the Fountain of Youth and anything that would reverse the aging process. Let’s face it, if you’re over 40, you feel it physically. Your systems just start to break down. In modern medicine, this process has been seen as disease but this week’s guest, Aubrey de Grey, sees it more like an engineering challenge. Aubrey is a very controversial biomedical gerontologist and crusader against aging. He has a very specific plan that identifies the various components that cause human tissue to age, and he has very specific remedies for each of them. That’s what his non-profit, The SENS Research Foundation (a 501C-3) focuses on and it’s got some interesting people behind it, like Peter Thiel (one of the founders of PayPal) and Aubrey himself who has invested a large part of his inheritance to the cause.  Grow Big Always often has conversations with rebels that help us reshape the points of view that we take for granted, so Aubrey de Gray is a perfect guest.
