In this episode of the Epigenetics Podcast, we talked with Vijay Ramani from the Gladstone Institute to talk about his work on Single-Molecule Adenine Methylated Oligonucleosome Sequencing Assay (SAMOSA). Our discussion starts with Vijay Ramani's impactful contributions to the field during his time in Jay Shendure's lab, where he worked on several innovative methods, including RNA proximity ligation. This project was conceived during his graduate studies, aiming to adapt techniques from DNA research to investigate RNA structures—a largely unexplored area at the time. We delved into the nuances of his experiences in graduate school, emphasizing how critical it was to have mentors who provided room for creativity and autonomy in experimental design. Dr. Ramani then shares insights about his foray into developing more refined methodologies, such as in-situ DNA Hi-C, a revolutionary protocol tailored for three-dimensional genomic mapping. He explained the rationale behind his projects, comparing the outcomes with contemporaneous advancements in methods like Micro-C. The discussion highlighted the importance of understanding enzyme bias in chromatin studies and the need for meticulous experimental design to ensure the validity of biological interpretations. We further explored exciting advancements in single-cell genomics, specifically Ramani's work on developing sci-Hi-C. This innovative technique leverages combinatorial indexing to allow high-resolution mapping of chromatin architecture at the single-cell level, a significant leap forward in understanding the complexities of gene regulation. As we progress, Ramani detailed his transition from graduate student to independent investigator starting his own lab. He elaborated on the challenges and excitements associated with establishing his research focus in chromatin structure and function using advanced sequencing technologies. Employing various strategies, including the innovative SAMOSA assay, his research seeks to elucidate the mechanisms by which chromatin structure influences transcriptional regulation. We also discussed the heterogeneity of chromatin and its implications for gene expression. Ramani provided a fascinating perspective on how variations in chromatin structure could affect gene activity, highlighting potential avenues for future research that aims to untangle the complex dynamics at play in both healthy and diseased states.   References Ramani, V., Cusanovich, D., Hause, R. et al. Nach dem Ende von Led Zeppelin geht Robert Plant viele andere Wege. Legislation to improve federal agency oversight and management of software purchases passed the House on Wednesday, keeping top IT and software trade groups' hopes alive that the bill will get through the Senate and become law before this congressional term is up. The Strengthening Agency Management and Oversight of Software Assets Act (H.R.1695) – better known as the SAMOSA Act — was introduced by Rep. Matt Cartwright, D-Pa., last year and co-sponsored by a bipartisan group of 20 House lawmakers. Calling the rooting out of waste, fraud and abuse a “signal mission” of the House Oversight Committee, Cartwight said the bill would ensure that federal agencies are required to conduct a “comprehensive assessment of their current software assets and restructure their operations to reduce unnecessary costs.” A bipartisan group of senators introduced legislation to reauthorize and refocus the National Quantum Initiative after certain research and development activities in its initial five-year run lapsed in September. The National Quantum Initiative Reauthorization Act would authorize $2.7 billion for quantum R&D at federal agencies for the next five years and shift the initiative's focus “from basic research to practical applications.” It would also expand the initiative to include agencies like the National Institutes of Health, the State Department and the Small Business Administration, according to a news release. This week Justin replays one of his personal favorite episodes, number 26, when author and former special forces officer Greg Walker shared the little-known story of a David Baez, a green beret defector. Baez was a Nicaraguan national whose father was killed by U.S.-backed President Samosa after voicing his displeasure with inequalities that had risen under Samosa's rule. After immigrating the U.S. and joining the Army (and then Special Forces), Baez found himself in El Salvador watching the country he served try to re-institute the rule of a dynasty that had done personal harm to his own family. Baez defected to Nicaragua in support of Sandinista revolutionary forces--despite the danger it posed to his American brothers in arms.Connect with Greg:LinkedIn: Gregory WalkerCheck out Part 1 of Greg's series on Baez, A defector in place: The strange and terrible saga of a Green Beret Sandinista, here.https://www.linkedin.com/pulse/defector-place-strange-terrible-saga-green-beret-part-greg-walker/Connect with Spycraft 101:Get Justin's latest book, Murder, Intrigue, and Conspiracy: Stories from the Cold War and Beyond, here.spycraft101.comIG: @spycraft101Shop: shop.spycraft101.comPatreon: Spycraft 101Find Justin's first book, Spyshots: Volume One, here.Check out Justin's second book, Covert Arms, here.Download the free eBook, The Clandestine Operative's Sidearm of Choice, here.A podcast from SPYSCAPE.A History of the World in Spy Objects Incredible tools and devices and their real-world use.Support the show

Aperta o play e vamos juntos nesse debate tão urgente! _____ FALE CONOSCO .
